Chiasma formation in Arabidopsis thaliana accession Wassileskija and in two meiotic mutants.

Meiotic chiasmata were analysed in metaphase I pollen mother cells (PMCs) of wild-type Arabidopsis thaliana and in two meiotic mutants. Fluorescence in situ hybridisation (FISH) with 45S rDNA and 5S rDNA as probes was used to identify the five chromosome pairs. A wild-type chiasma frequency of 9.24 per cell was found, consistent with estimated genetic recombination values. Individual bivalent chiasma frequencies varied according to chromosome size; chromosome 1 had the highest mean chiasma frequency (2.14) while the short acrocentric chromosomes had the lowest frequencies (1.54 and 1.56). FISH analysis was extended to two meiotic mutants (asy1 and dsy1) having low residual bivalent and chiasma frequencies. Mutant dsy1 gave no indication of chromosome preference for residual bivalent formation; instead it showed a general reduction in bivalent and chiasma frequencies. In asy1, the longest chromosome (1) had the lowest bivalent frequency and chiasma frequency while the short acrocentric chromosome 2 had the highest frequencies. This chromosome pair may be preferentially involved in synapsis and chiasma formation because of their association with the nucleolus. However, other factors may be operating since the other acrocentric chromosome (4), with similar size and structure to chromosome 2, did not share these chiasma properties.

Experimental studies on the bridging hypothesis of anaphylaxis: haptenic determinants required to elicit immediate-type reactions in calf skin by separate or concurrent sensitization with reagins of different specificity.

The present study has established, that cows suitably immunized with either DNP-edestin (DNP-Ed), di-DNP-gramicidin-J [(DNP)(2)-Gram], respectively, or p-azobenzenearsonate-Ed (ABA-Ed) synthesized and secreted reaginic antibodies (IgE) into colostrum. Whereas ABA-Ed failed to elicit more than a low response, there was however a persistent and increased antibody synthesis between 10 and 56 days after priming with DNP-Ed. Bivalent and multivalent DNP haptens differing in molecular size, degree of substitution, and rigidity were compared for their effectiveness in eliciting Prausnitz-Küstner (P-K) reactions in either newborn colostrum-deprived calves or in those 4 wk of age. The sensitization with reaginic anti-DNP antibody has been accomplished either by feeding colostrum of the immunized dam or by intradermal injection of reaginic serum or colostral whey. It could be demonstrated that equimolar doses of the bivalent alpha,N-(epsilon,N-DNP-aminocaproyl-)-epsilon,N-DNP-L-lysine and the multivalent dinitrophenylated bovine serum albumin were equally effective in eliciting reactions in skin sites provided that a high affinity antibody was used for sensitization. By contrast, the comparatively rigid, bivalent hapten, (DNP)(2)-Gram consistently failed to induce comparable reactions. Furthermore, it was clearly shown that optimal distances of determinant groups on the haptenic molecule are a prerequisite for positive P-K reactions, since alpha,epsilon,N-bis-DNP-lysine failed to induce comparable reactions. Concurrent sensitization of skin sites with reaginic anti-DNP and anti-ABA antibodies provides the final proof that cross-linking of two adjacent reaginic molecules on the mast cell surface by a bivalent hapten is required for effective elicitation of immediate-type reactions. This has been accomplished by utilizing the bivalent epsilon,N-DNP-alpha,N-[(4-hydroxy-3-azobenzenearsonic acid)-phenacetyl]-L-lysine (DNP-ABA) carrying noncross-reactive haptenic groups, which was consistently effective in eliciting P-K reactions in doubly but never in singly sensitized skin sites. It is apparent from the results that equimolar doses of monovalent haptens could completely inhibit the response to DNP-ABA. The present studies finally establish that mast cells of newborn colostrum-deprived calves lack IgE molecules on their surface. Thus, mast cells of newborn calves may be unique, to investigate the molecular mechanisms involved in immediate-type reactions more precisely.

A mixed polyploid model for linkage analysis in outcrossing tetraploids using a pseudo-test backcross design.

Based on how chromosomes pair at meiosis, the nature of polyploids can be described by bivalent polyploids, multivalent polyploids, and mixed polyploids. In bivalent polyploids, only two chromosomes pair, during which two more similar chromosomes have a higher pairing probability (preferential pairing) than two less similar chromosomes, whereas in multivalent polyploids more than two chromosomes pair at a time, which results in double reduction. Preferential chromosome pairings and double reduction affect the frequencies of gamete formation and, therefore, linkage analysis of polymorphic markers in bivalent and multivalent polyploids, respectively. For mixed polyploids, in which both bivalent and multivalent formations occur simultaneously, linkage analysis is affected by both preferential pairings and double reduction. In this study, we develop a hierarchical maximum likelihood model for discerning gamete genotypes derived from different pairing mechanisms and different formation modes. The first-stage model in the hierarchy is formulated to characterize the relative frequencies of bivalent and multivalent pairing configurations in terms of the preferential pairing factor. The second-stage model is derived to rule out identical gamete genotypes into their different formation modes with relative probabilities determined by the recombination fraction. The first-stage pairing mechanism and second-stage formation mode are integrated to provide the simultaneous maximum likelihood estimates of the preferential pairing factor, the frequency of double reduction, and the recombination fraction, by implementing the EM algorithm. We performed extensive simulation studies to demonstrate the statistical properties of our hierarchical model for linkage analysis in tetraploids. The implications of our model for polyploid linkage mapping are discussed.

Chromosome morphology during meiosis of normal and Robertsonian translocation-carrying rams (Ovis aries).

A detailed description is given of the meiotic chromosomes of the normal ram (54,XY) and of the chromosomes at diakinesis-metaphase I in rams heterozygous for the t1, t2 and t3 and homozygous for the t1 and t3 Robertsonian translocation chromosomes. Analyses were made on diakinesis-metaphase I chromosomes in 309 cells from normal rams and 2402 cells from the Robertsonian translocation carriers. Hypomodal cell counts were due to missing chromosomal elements and hypermodal counts were due either to separation of the chromosomes of the sex bivalent or to separation of the components of an autosomal bivalent. Trivalent formation was characteristic for the heterozygous Robertsonian translocation carriers while the homozygous carriers had an additional large bivalent. An association between the sex bivalent and a small autosomal bivalent at diakinesis-metaphase I is reported.

JAK2 activation and cell proliferation induced by antibody-mediated prolactin receptor dimerization.

Cytokines that interact with receptors of the hematopoietin super-family have recently been reported to stimulate receptor-associated JAK tyrosine kinases, including PRL activation of JAK2. Unlike other tyrosine kinases, none of the JAK kinases has thus far been implicated in oncogenesis, and their involvement in growth signaling has not been established. Using the PRL-dependent pre-T-cell line Nb2, the present study provided a link between bivalent dimerization of a hematopoietin receptor and activation of its associated JAK kinase, and demonstrated a strong positive correlation between the mitogenic potency of a series of bivalent anti-PRL receptor antibodies and the degree of induced tyrosine phosphorylation of JAK2. Antibody bivalency was required for JAK2 phosphorylation. Monovalent anti-PRL receptor Fab fragments alone were inactive, but their activity could be partially restored by cross-linking with bivalent anti-Fab antibodies. Additional evidence for antibody-induced receptor dimerization was provided by a bell-shaped dose-response curve for the most potent receptor agonist, monoclonal antibody T6. This phenomenon is typically seen at pharmacological concentrations of bivalent ligands, when bound ligand molecules fail to adjoin a second receptor due to occupancy. The present study provided functional support for a model of PRL receptor triggering by ligand-induced receptor homodimerization and subsequent activation of the associated tyrosine kinase JAK2.

Phosphorylated proteins are involved in sister-chromatid arm cohesion during meiosis I.

Sister-chromatid arm cohesion is lost during the metaphase I/anaphase I transition to allow homologue separation. To obtain needed information on this process we have analysed in grasshopper bivalents the sequential release of arm cohesion in relation to the behaviour of chromatid axes. Results show that sister axes are associated during early metaphase I but separate during late metaphase I leading to a concomitant change of chromosome structure that implies the loss of sister-kinetochore cohesion. Afterwards, homologues initiate their separation asynchronously depending on their size, and number and position of chiasmata. In all bivalents thin chromatin strands at the telomeres appeared as the last point of contact between sister chromatids. Additionally, we have analysed the participation of phosphoproteins recognised by the MPM-2 monoclonal antibody against mitotic phosphoproteins in arm cohesion in bivalents and two different kinds of univalents. Results show the absence of MPM-2 phosphoproteins at the interchromatid domain in mitotic chromosomes and meiotic univalents, but their presence in metaphase I bivalents. These phosphoproteins are lost at the onset of anaphase I. Taken together, these data have prompted us to propose a 'working' model for the release of arm cohesion during meiosis I. The model suggests that MPM-2 phosphoproteins may act as cohesive proteins associating sister axes. Their modification, once all bivalents are correctly aligned at the metaphase plate, would trigger a change of chromosome structure and the sequential release of sister-kinetochore, arm, and telomere cohesions.

Non-random chromosome segregation in Neocurtilla hexadactyla is controlled by chromosomal spindle fibres: an ultraviolet microbeam analysis.

Single spindle fibres of Neocurtilla spermatocytes were irradiated by means of an ultraviolet microbeam. Irradiations were with monochromatic ultraviolet light. The single sex chromosome (the X1 univalent) reoriented after irradiation of its spindle fibre or of any of the spindle fibres associated with the heteromorphic bivalent (the X2Y bivalent): the X1 moved toward the Y half-spindle, and sometimes rotated as it moved. Irradiations of autosomal spindle fibres did not induce X1 movements, and hence the induction of reorientation is specific to irradiation of the spindle fibres associated with X1 or X2Y. In no case did the X2Y bivalent reorient; hence, the X1 is the active chromosome in ensuring that there is non-random segregation in Neocurtilla spermatocytes. The irradiations sometimes caused the X2Y bivalent to contrast, but the reorientation movements of the X1 were independent of the contraction of the X2Y bivalent. We suggest that the X1 and X2Y chromosomal spindle fibres form a network that is able to send signals to the X1 univalent to cause it to reorient.

Cutting edge: monovalency of CD28 maintains the antigen dependence of T cell costimulatory responses.

CD28 and CTLA-4 are the major costimulatory receptors on naive T cells. But it is not clear why CD28 is monovalent whereas CTLA-4 is bivalent for their shared ligands CD80/86. We generated bivalent CD28 constructs by fusing the extracellular domains of CTLA-4 or CD80 with the intracellular domains of CD28. Bivalent or monovalent CD28 constructs were ligated with recombinant ligands with or without TCR coligation. Monovalent CD28 ligation did not induce responses unless the TCR was coligated. By contrast, bivalent CD28 ligation induced responses in the absence of TCR engagement. To extend these findings to primary cells, we used novel superagonistic and conventional CD28 Abs. Superagonistic Ab D665, but not conventional Ab E18, predominantly ligates CD28 bivalently at low CD28/Ab ratios and induces Ag-independent T cell proliferation. Monovalency of CD28 for its natural ligands is thus essential to provide costimulation without inducing responses in the absence of TCR engagement.

Identification of human male meiotic chromosomes.

The study is based on examinations of surgical testicular biopsies performed in eleven men aged from 19-79 years (300 cells at leptotent stage, 19 at zygotene, 300 cells at pachytene, 490 spermatocytes at diakinesis/first metaphase and 23 cells at the second metaphase of meiotic division), as well as post-mortem necropsies taken from six men aged from 19-51 years (6,000 cells at the first meiotic prophage). Identification of chromosomes at leptotene and zygotene stages is limited to the determination of X and Y chromosomes enclosed in the sex vesicle. At the pachytene stage, identification of chromosomes can make use of the differences in their length and number of chromomeres, but is feasible only in figures with good chromosome spreading. Identification of chromosomes at diakinesis/first metaphase in preparations stained by classical methods rests on the size and shape of the bivalents. Application of centromeric heterochromatin staining technique enables us to differentiate among bivalents Nos. 1, 2, and 3, to recognize bivalents belonging to the B group, to identify bivalents Nos. 9, 16, 17-18, and to distinguish between bivalent No. 21 and 22. It further permits the modality of pairing of the X and Y chromosomes to be determined by their short arms. Chromosomes of secondary spermatocytes at metaphase show typical morphological characteristics essential for karyotyping, so that it is possible to arrange them into the haploid karyotype, analogous to the karyotype of somatic cells. Male germinal cells undergo very rapid autolytic changes. No spermatocytes at diakinesis/first metaphase stage could be detected in specimens taken as early as 2 hours after death. The morphology of chromosomes of cells at earlier stages of the first meiotic prophase was markedly altered. Post-mortem testicular material was found unsuitable for an analysis of meiotic chromosomes.

A 3-step pretargeting strategy to image infection.

UNLABELLED: A new 3-step approach to imaging infectious and inflammatory foci was developed and optimized in a rat model. The approach relies on the nonspecific localization of an anti-diethylenetriaminepentaacetic acid (DTPA) antibody in inflamed tissue. In this study, the 3-step strategy was optimized by selecting the most suitable radiolabeled hapten and tuning the dosing schedule. METHODS: Wistar rats with Staphylococcus aureus infection in the left calf muscle were primed with the anti-DTPA antibody DTIn-1 (0.67, 2, or 6 nmol per rat). In the second step (1-24 h later), the anti-DTPA activity in the circulation was blocked with unlabeled bovine serum albumin DTPA-In (0.3, 1, or 3 nmol per rat). In the third step (5-30 min later), the radiolabeled hapten (monovalent or bivalent 111In-DTPA) was administered. The in vivo distribution of the radiolabel was monitored by scintigraphic imaging and by ex vivo counting of dissected tissues. RESULTS: Scatchard analysis revealed that the affinity of DTIn-1 for bivalent DTPA-111In (111In-diDTPA) was 6 times higher than the affinity for monovalent 111In-DTPA (K(a) = 0.87 x 10(-9) mol/L vs. 5.3 x 10(-9) mol/L). The uptake of the bivalent chelate in the abscess was 2.5-fold higher than that of monovalent 111In-DTPA. Most important, the bivalent chelate was completely retained in the abscess over time. Using the bivalent chelate, the optimal dosing scheme was determined with respect to the DTIn-1 dose (2 nmol per rat), the blocking agent dose (1 nmol per rat), and radiolabeled chelate dose (40 pmol per rat). The procedure was rapid; the infectious focus was clearly visualized 1 h after injection of the 111In-labeled diDTPA, which was 5 h after administration of the anti-DTPA antibody. The nontargeted radiolabel rapidly cleared to the urine, only being retained in the abscess and the kidneys (4-6 percentage injected dose). Finally, an N2S2 core was attached to the diDTPA compound, allowing the use of 99mTc. CONCLUSION: This 3-step approach enables rapid imaging of infectious foci with minimal uptake in noninflamed tissues.

[Meiotic chromosomes of green frogs (Rana esculenta complex) of the Ukraine territory].

In this work meiotic chromosomes of R. ridibunda, R. lessonae and their hybrid form R. esculenta of the Ukraine territory are described for the first time. These chromosomes are connected in diakinesis more often by two chiasma, rarely by one or three chiasma in 13 bivalents. Bivalents with one chiasma in R. ridibunda are typical only for large bivalents, in R. lessonae only for small ones. Three chiasma in parental species are typical only for small bivalents. In the hybrid form R. esculenta chromosomes with two, one and three chiasma are typical both for large bivalents, and for small ones. In the hybrid form (R. esculenta) chiasma frequency noticeably decreases, comparatively with parental species that, possibly, specifies the aberration of normal meiosis realization. The chiasma frequencies of the hybrid form of the eastern and the southern parts of Ukraine are almost the same, but the types of chromosomes are different in such regions. In the eastern region chromosomes of the hybrid form are represented by two or one chiasma, in the southern region by two, one or three chiasma.

Residualizing iodine markedly improved tumor targeting using bispecific antibody-based pretargeting.

UNLABELLED: Previous studies have shown that pretargeting allows rapid visualization of renal cell carcinomas (RCC) with an (111)In-labeled bivalent peptide. For radioimmunotherapy, a beta-emitting radionuclide labeled to a bivalent peptide is required. Therapeutic efficacy of these radionuclides depends on the E(max), physical half-life, and residence time of the radiolabel in the tumor. The (131)I radiolabel generally clears rapidly from the tumor after internalization and subsequent degradation of the bivalent l-amino acid peptide (l-a.a. peptide) in the tumor cells. To improve the residence time of the iodine label in the tumor, a new bivalent peptide was synthesized that is peptidase resistant and consists of 4 d-amino acids (d-a.a. peptide). Here we investigated the characteristics of the residualizing iodine label in SK-RC-52 RCC tumors. METHODS: The d-a.a. peptide was manually synthesized according to standard solid-phase Fmoc/HBTU (2-[1H-benzotriazole-1-yl]-1,1,3,3-tetramethyluronium hexafluorophosphate) chemistry. The uptake and retention in the tumor of (111)In-/(125)I-labeled bivalent peptides (l-a.a. peptide and d-a.a. peptide) were studied in female BALB/c athymic mice with subcutaneous SK-RC-52 RCC tumors. Tumors were pretargeted with the bispecific monoclonal antibody (bs-mAb) G250xDTIn-1 and, 72 h later, mice were injected intravenously with one of both radiolabeled peptides. The effect of bs-mAb-diDTPA-bs-mAb (DTPA is diethylenetriaminepentaacetic acid) bridging at the tumor cell surface on the internalization of the bs-mAb-diDTPA complex was investigated in SK-RC-52 tumor-bearing mice. RESULTS: The maximum uptake and retention of (125)I-labeled l-a.a. peptide in the tumor were significantly lower compared with that of the (111)In-labeled l-a.a. peptide. In contrast, the tumor uptake and retention of the (125)I-labeled d-a.a. peptide) were similar to that of the (111)In-labeled l-a.a. peptide but were superior at later time points. The biodistribution of the radioiodinated d-a.a. peptide was highly similar to that of the (111)In-labeled d-a.a. peptide, and both radiolabeled peptides were retained significantly better in the tumor than the (111)In-labeled l-a.a. peptide. bs-mAb-diDTPA-bs-mAb bridge formation did not affect internalization of the bs-mAb-diDTPA complex. CONCLUSION: Uptake and retention in the tumor of the iodinated peptide after pretargeting with a bs-mAb can be significantly improved using d-a.a. peptides. Accordingly, the radiation dose to the tumor, correlating with the therapeutic efficacy of pretargeted RCC, can be enhanced substantially.

Desensitization of the neutrophil aggregation response to chemotactic factors.

In the presence of Ca2+ and Mg2+, the chemotactic fragment of C5, the synthetic chemotactic oligopeptide formyl-methionyl-leucyl-phenyl-alanine, and the ionophore A23187 aggregated human neutrophils. Aggregation induced by the two chemotactic factors was transient and reversed within 2 to 4 minutes after exposure; aggregation induced by A23187 was sustained and continued to increase over 15 minutes. In the absence of the bivalent cations, none of these three agents aggregated the cells. If bivalent cations were added after cell contact with a chemotactic factor, aggregation was detected after, but not before, addition of the cations. Under these conditions, the magnitude of the aggregation response was sharply reduced: cells preincubated with a chemotactic factor for longer than 2 to 4 minutes aggregated minimally after addition of bivalent cations. Moreover, cells preincubated with a chemotactic factor for 4 minutes, exposed to bivalent cations, and then rechallenged with the same chemotactic factor also showed a minimal aggregation response, ie, the cells were "desensitized" to the original stimulus. However, cells desensitized to one of the chemotactic factors still aggregated prominently when exposed to the other chemotactic factor or to A23187. Cells could not be desensitized to the ionophore A23187. Desensitization of the neutrophil aggregation response closely resembles desensitization of mast cell and leukocyte degranulation. Degranulation and aggregation appear to be closely related cellular responses to immunologic stimuli. Both responses may reflect alterations in surface membrane permeability to bivalent cations and/or changes in surface membrane adhesiveness to other biologic membranes.

Use of an inhibitor-resistant live attenuated influenza vaccine in normal and asthmatic adults.

The efficacy of a monovalent live attenuated influenza A (H3N2) vaccine (an inhibitor-resistant recombinant strain named "Alice") and of a bivalent vaccine composed of "Alice" and influenza B strain R75 (also inhibitor-resistant), was tested in healthy and asthmatic adults. Two intranasal doses of the monovalent "Alice" vaccine were given to 95 healthy adults in the winter of 1973-74. Ninety-three % of 68 subjects with initial serum hemagglutination (HI) titers of less than or equal to 1:40 had a significant (4-fold or greater) antibody increase in post-vaccination sera. Overall, 77% of the vaccinees had significant antibody rises. Two doses of the bivalent (A/B) vaccine were given to 53 healthy adults in the winter of 1974-75. Eighty-two % of 34 subjects with initial HI titers of less than or equal to 1:40 had 4-fold or greater antibody rises to influenza A, and overall 57% of the vaccinees responded. The B component of the bivalent vaccine was less effective; only 56% of persons with initial HI titers of less than or equal to 1:40, and 28% of all vaccinees had significant antibody rises. Both tf asthmatics who received "Alice" and one of 10 (10%) who received the bivalent vaccine had serologic responses to influenza A. None of the asthmatics responded to the B component of the bivalent vaccine. Analysis of the incidence of febrile respiratory illness during an influenza outbreak in the winter of 1974-75 revealed no differences in the attack rates of placebos and vaccines. In conclusion, both the "Alice" and bivalnet (A/B) vaccines were effective in eliciting serologic responses to influenza A in healthy persons. They were less effective in asthmatics. The lack of protection observed may have been due to the onset of influenza before the vaccine could take effect or to the failure of the vaccine itself.

Antibody-induced epidermal growth factor receptor dimerization mediates inhibition of autocrine proliferation of A431 squamous carcinoma cells.

We previously reported that anti-epidermal growth factor (EGF) receptor monoclonal antibody (mAb) 225 can block receptor activation and inhibit proliferation of tumor cells bearing EGF receptors. To further explore the mechanism of mAb-mediated growth inhibition, we compared the capacities of bivalent 225 mAb and 225 F(ab')2, and monovalent 225 Fab' fragment to block ligand binding to EGF receptors, inhibit activation of receptor tyrosine kinase by exogenous and endogenous ligand, produce receptor dimerization, down-regulate receptors, and inhibit proliferation of cultured A431 squamous carcinoma cells. Unlike 225 mAb and 225 F(ab')2, 225 Fab' fragment was a poor inhibitor of A431 cell proliferation. The weak antiproliferative capacity of 225 Fab' was not due to depletion of active fragment from cultures. When cells were exposed to exogenous EGF, monovalent 225 Fab' remaining in conditioned culture medium could act as well as the bivalent forms of mAb to block binding and tyrosine kinase activation by exogenous EGF. However, unlike the bivalent forms, 225 Fab' fragment was unable to induce receptor dimerization and down-regulation, and it lacked the capacity to block autocrine activation of EGF receptors by endogenous ligand. These deficiencies were corrected by addition of rabbit anti-mouse IgG antibody, which also enabled 225 Fab' fragment to inhibit cell proliferation. We conclude that in A431 cells, inhibition of autocrine-stimulated proliferation by anti-EGF receptor mAbs requires antibody bivalency, which provides the capacity to produce EGF receptor dimerization accompanied by receptor down-regulation. These properties may explain the greater efficacy of bivalent mAb and F(ab')2, compared with monovalent Fab' fragment, in inhibiting proliferation of a variety of malignant and nonmalignant cultured cell lines.

[Synaptonemal complexes of chromosomes of male mice after prolonged administration of neoaquasept].

A long-term (during 4 months) daily peroral injections of neoaquasept (NA) (a drinking water disinfections agent) at a dose corresponding to the normal level of consumption increases the number of spermatocytes in which the sexual bivalent associates with normal autosomal bivalents (up to 18.2%). In single cells a premature desynapsis of sex chromosomes at late pachytene was observed. There was the appearance of single cells with the X0 karyotype. A 3-fold excess of the normal consumption of NA causes an increase in the number of cells with the above disturbance in the SC structure and behaviour and causes the appearance of heteromorphic bivalents. On the 7th day after a single MTD injection of NA male mice displayed a sharp (up to 27.9%) increase in the number of cells in which the sexual bivalent associated with autosomal bivalents, formation of univalents of autosomal and sex chromosomes, SC degeneration.

Malorientation and abnormal segregation of chromosomes during recovery from colcemid and nocodazole.

Reversal of meiotic arrest in crane-fly spermatocytes by U. V. irradiation of Colcemid-arrested cells or by rinsing Nocodazole-arrested cells in fresh buffer results in the induction of chromosome malorientation. Malorientations observed among Colcemid-recovering and Nocodazole-recovering spermatocytes at frequencies higher than normally observed in untreated cells included associations of sister kinetochores of half-bivalents with both spindle poles (amphitely), in contrast with associations of sisters with only one pole (syntely) as is usually found during the first meiotic division. In several cases, prior to anaphase onset, maloriented bivalents appeared unusually tilted with respect to the spindle axis, and during anaphase they gave rise to laggard half-bivalents that did not segregate during anaphase along with half-bivalents having proper syntelic orientation. The results parallel previous findings obtained during cold recovery, and the properties of the drugs used here suggest that their action on microtubules, although reversible, induces malorientation during recovery from meiotic arrest.

Meiotic nondisjunction in oocytes from aged Djungarian hamsters correlates with an alteration in meiosis rate but not in univalent formation.

The effect of maternal ageing on the meiotic rate, on chiasma and univalent frequency as well as on heteroploidy in secondary oocytes from Djungarian hamsters was examined. The frequency of hyperhaploid oocytes increased from 0.6% in young (8-14 weeks) to 2.8% in middle-aged (26-46 weeks) and reached 3.6% in the oldest females (49-75 weeks). On the basis of malsegregated bivalents per oocyte, nondisjunction occurred most often in the middle-aged group (5.42 X 10(-2) bivalents per oocyte). Hereby, the large meta- and submetacentric A-D chromosomes were preferentially involved. Furthermore, the pattern of nondisjunction was not different from that expected on the basis of chromosome length or induced by colchicine. The large A-D chromosomes did not show any alteration in chiasma or univalent frequency. Terminalized chiasmata were only detected in the E group and univalents increased slightly, but not significantly in the small chromosomes (G group). At higher ages, both chromosome groups were not preferentially involved in nondisjunction. Presegregation slightly increased with age and affected more or less all bivalents, whereas the incidence of diploidy significantly decreased. With respect to the rate of meiosis in oocytes from aged females, the resumption was delayed at metaphase I. Our data suggest that failures in the control of oocyte proliferation are involved in nondisjunction rather than the "production-line." Furthermore, a model is proposed to explain nondisjunction of specific bivalents at certain maternal ages.
