Mass spectrometric analyses and characterization of Kepone in environmental and human samples.

A specific portion of our environment has been contaminated with Kepone, or chlordecone. Additionally, some specific human exposures to high concentrations of Kepone have been confirmed. Gas chromatography mass spectrometry involving chemical ionization and high resolution mass spectrometry were used to detect, identify and confirm the presence of Kepone, Kepone photoproducts and a reduction product of Kepone in environmental and human samples. Field desorption, field ionization and electron impact mass spectrometric methods, as well as infrared and nuclear magnetic resonance techniques were used to characterize and identify Kepone hydrate and hemiketal in benzene and methanol solutions, respectively.

Continuous-flow system for determination of diffusion coefficients: use of a natural membrane.

A natural membrane was employed in an automated diffusion system. A mature male Mongolian gerbil sebaceous gland pad was excised and mounted into a suitable retainer so that the external surface was oriented toward the concentrated aqueous drug solution. Aqueous solutions of benzoic acid and the three commonly used parabens were studied. The gerbil sebaceous pad effectively prevented any diffusion of these drug solutions within 15 hr. Water by itself, however, was transported through the skin even against a pressure gradient. Although no apparent diffusion of these compounds occurred, a significant amount of drug was retained by the sebaceous pad. An expression for membrane-water partition coefficients could be calculated. Based upon thicknesses of natural and synthetic membranes, theoretical approximations of diffusion rates were found using lag time calculations.

The lipometre: an easy tool for rapid quantitation of skin surface lipids (SSL) in man.

The purpose of this study is to present a rapid, simple, and accurate method for quantitation of SSL in man. The technique which is derived from the works by Schaefer and Kuhn-Bussius, employs a series of clearn ground glass plates, and an apparatus, the Lipometre, for the determination of the variation of light transmission across the plates as a function of the fat deposited on them. Results on 32 adults, males and females, show that the first four iterative samplings, at constant pressure, on the same forehead area follow an identical physical law independently of whether the skin is fatty or dry. This law affords a precise definition of the Casual-Level, its quantification and its correlation with skin patterns. It is consequently possible, either as an investigative or a routine technique, to predict the total amount of SSL with good accuracy within 2 min. The SSL, for non-pathological skins, fall within the range 150 microgram/cm2 to 900 microgram/cm2 which is in good agreement with values quoted in the literature.

Effect of topical application of 13-cis retinoic acid on skin of hairless rats and hairless mice.

The potential effectiveness of topical 13-cis retinoic acid (13-cis RA) as a sebosuppressive agent was evaluated in hairless ("fuzzy") rats and hairless mice. At nontoxic dosages (i.e., concentrations which induced no weight loss), topical 13-cis RA had no detectable sebosuppressive effects in either of these species. In hairless rats, the topical application of 0.2% 13-cis RA induced more severe symptoms of toxicity than was induced by the administration of equivalent amounts of the drug by either oral or subcutaneous routes. Due to variability in species sensitivity to 13-cis RA, the potential effectiveness of the topical use of this retinoid can probably only be evaluated in human volunteers.

17 alpha-Propylmesterolone (SH 434): an antiandrogenic sebosuppressive substance not influencing circulating testosterone concentrations. Experimental studies in Syrian hamsters.

17 alpha-Propylmesterolone is a new synthetic 5 alpha-reduced steroid with a propyl group in C-17 position and a methyl group in the A ring. The antiandrogenic action of 17 alpha-propylmesterolone on the sebaceous glands, testes weights and plasma testosterone concentrations were examined in the animal model of the Syrian hamster. The substance was given systemically (3, 5 or 10 mg/kg) and topically (3 or 5 mg/kg). 17 alpha-propylmesterolone reduced both sebaceous gland size and sebogenesis significantly in a dose-dependent manner. The topical administration was more effective than the systemic treatment. There was no influence of 17 alpha-propylmesterolone on testosterone concentration in plasma or testes weights although a diminished capacity or absence of free cytoplasmatic androgen receptor sites was detected in the sebaceous glands of the systematically or topically treated Syrian hamster. 17 alpha-Propylmesterolone exerts a potent topical sebosuppressive effect in the animal model. These findings should give rise to human studies and clinical trials.

[Sebaceous filaments (author's transl)].

Sebaceous filaments are cylindrical tubes of whitish-yellowish color, which can be expressed from areas of the face rich in sebaceous follicles by pinching the skin or by the cyanoacrylat-technique. Sebaceous filaments are most commonly found in the centrofacial areas and the alae nasae in postpuberal individuals with large facial pores and seborrhea. Sebaceous filaments are composed of a skeleton of 10-30 horny cell layers which enclose a mixture of bacteria, sebaceous lipid, corneocyte fragments and one hair. Following the expression of the filament the follicle refills within 30 days. The various portions of sebaceous follicles (acro- and infrainfundibulum) and the follicular contents are presented at the light microscopial and ultrastructural level. Follicles containing sebaceous filaments have a conspicuous granular layer and no acanthosis. Sebaceous filaments should be differentiated from a microcomedo and trichostasis spinolosa. They are a common morphological variant of sebaceous follicles.
