TUNEL-positive staining in white and grey matter after fatal head injury in man.

Paraffin sections from the hippocampus, the cingulate gyrus and the insula of 18 head-injured patients who survived between 5 hours and 10 days, and 18 age-matched controls, were stained by the terminal deoxynucleotidyl transferase mediated biotinylated deoxyuridine triphosphate nick end labelling (TUNEL) technique for evidence of in situ DNA fragmentation. Additional staining techniques (HE, combined LFB/CV and immunohistochemistry for GFAP and CD68) were used to characterize any lesions and their time course. Only the occasional TUNEL+ cell per area was seen in the control brains. TUNEL+ cells were identified in both grey and white matter of the head-injured material and their numbers peaked between 24 and 48 hours and were still present at 10 days. Within the hippocampus, fewer TUNEL+ cells were seen in grey (between 3-5 per area) than in the white matter, (up to 51+ per area) whereas in the cingulate gyrus and in the insula, the number of TUNEL+ cells was always greater in the cortex (between 11-20 per area) than in white matter (6-10 per area). In the grey matter, most TUNEL+ cells had the morphology of necrosis. However, the histological appearances of some of the neurons (2-3%), and of oligodendroglia and macrophages in white matter (about 5%) were those of apoptosis.

[White matter changes and cognitive performance in aging].

OBJECTIVE: In this paper we review the main magnetic resonance studies to show a possible relationship between changes in the white matter of the brain or leukoaraiosis, and the neuropsychological profile of elderly persons without dementia. DEVELOPMENT: The articles published to date show contradictory data, and in nearly half the cases reviewed no clear relationship could be established between leukoaraiosis and conduct. However, by using sensitive cognitive tests it is possible to detect and association between the presence and degree of change in the white matter and decline in frontal function such as speed of processing information, visuomotor function, verbal fluency, classification and mental sequences. Other cognitive areas such as language, memory or visuospatial, visuoconstructive and visuoperceptive functions appear less frequently related to the presence or intensity of lesions of the white matter of the brain. From a neuropsychological point of view, periventricular localization of the leukoaraiosis seems to be more important than subcortical localization. CONCLUSIONS: The neuropsychological functions most frequently associated with the presence of leukoaraiosis are those dependent on the frontal lobes, and are a disconnection favoured by the presence of the white matter of the brain, the most probable underlying physiopathological mechanism. Although there is evidence showing a genetic effect in the appearance of the white matter of the brain, study of the genes associated with cognitive deterioration in normal ageing has not given conclusive findings.

High-resolution line scan diffusion tensor MR imaging of white matter fiber tract anatomy.

BACKGROUND AND PURPOSE: MR diffusion tensor imaging permits detailed visualization of white matter fiber tracts. This technique, unlike T2-weighted imaging, also provides information about fiber direction. We present findings of normal white matter fiber tract anatomy at high resolution obtained by using line scan diffusion tensor imaging. METHODS: Diffusion tensor images in axial, coronal, and sagittal sections covering the entire brain volume were obtained with line scan diffusion imaging in six healthy volunteers. Images were acquired for b factors 5 and 1000 s/mm(2) at an imaging resolution of 1.7 x 1.7 x 4 mm. For selected regions, images were obtained at a reduced field of view with a spatial resolution of 0.9 x 0.9 x 3 mm. For each pixel, the direction of maximum diffusivity was computed and used to display the course of white matter fibers. RESULTS: Fiber directions derived from diffusion tensor imaging were consistent with known white matter fiber anatomy. The principal fiber tracts were well observed in all cases. The tracts that were visualized included the following: the arcuate fasciculus; superior and inferior longitudinal fasciculus; uncinate fasciculus; cingulum; external and extreme capsule; internal capsule; corona radiata; auditory and optic radiation; anterior commissure; corpus callosum; pyramidal tract; gracile and cuneatus fasciculus; medial longitudinal fasciculus; rubrospinal, tectospinal, central tegmental, and dorsal trigeminothalamic tract; superior, inferior, and middle cerebellar peduncle; pallidonigral and strionigral fibers; and root fibers of the oculomotor and trigeminal nerve. CONCLUSION: We obtained a complete set of detailed white matter fiber anatomy maps of the normal brain by means of line scan diffusion tensor imaging at high resolution. Near large bone structures, line scan produces images with minimal susceptibility artifacts.

Compartmental model for organic matter digestion in facultative ponds.

A model has been developed for the digestion of organic matter in facultative ponds in tropical regions. Complete mixing has been assumed for the aerobic and anaerobic compartments. Settling, aerobic layer oxidation, and anaerobic layer methanogenesis are the main processes for organic matter removal in the water column. Exchange processes between layers are dispersive or soluble exchange, solubilization and transport of organic matter from sediments to water column are also taken into account. Degradation of organic matter in the sediments produces gaseous emissions to the water column. The exchange between bubbles ascending and the water column was measured. The model was calibrated with data obtained from a pilot facultative pond built in Muña Reservoir in Bogotá. The pond was sampled during 4 months to compare data between its water hyacinth covered section and uncovered section. The results clearly show the relative importance of different BOD removal processes in facultative ponds and suggest modifications to further improve performance. The results from the model suggest that internal loadings to facultative ponds due to solubilization and return of organic matter from the sediments to the aerobic layer greatly influence the soluble BOD effluent concentration. Aerobic degradation activity in the facultative pond does not affect significantly the effluent concentration. Anaerobic degradation activity in the facultative pond can more easily achieve increases in the removal efficiencies of BOD.

Corpus callosum, pons, and cortical white matter in alcoholic women.

BACKGROUND: To measure the effect of alcohol abuse on white matter brain macrostructure in women with alcoholism and to determine whether observed abnormalities interact with age. METHODS: Quantitative measures of corpus callosum area, cortical white matter volume, and pons volume were derived from magnetic resonance imaging scans obtained from 34 women with DSM-III-R alcoholism (aged 28-64, mean 41 years) and 35 healthy women (aged 22-65, mean 42 years). Transverse relaxation time of the pons was also obtained. RESULTS: No significant group differences in any brain measures were observed. However, in alcoholics greater length of sobriety was associated with more cortical white matter, and higher lifetime levels of alcohol consumption were associated with smaller volumes and prolonged transverse relaxation time in the pons. CONCLUSIONS: Despite a lack of overall deficits in white matter macrostructural size in alcoholic women, certain white matter structures showed alcohol exposure vulnerability whereas others showed evidence of recovery with abstinence.

Suspended particulate matter in an office and laser smoke particles in an operating room.

Suspended particulate matter in an office and laser smoke particles in a laser operative room of the Otolaryngology Department, Ramathibodi Hospital were compared. Suspended particulate matter sizes of PM15, PM10 and PM2.5 were selected due to their impact on health. The amount and sizes of the particles were measured by a laser diode portable dust monitor. The mean and standard deviation were measured every hour for 6 periods and calculated by specific computer software. The amount of suspended particulate matters in the office were within the accepted safety level. The amount of suspended particulate matter including laser smoke particles in the operative room before, during and after each laser evaporative procedure was much higher than that of the office. The amount of suspended particulate matter was dangerous for all personnel in the operative room. Risk management for patients in the operative room should be stressed. The ventilation system of the operative room will be assessed further.

[The effects of organic matter and hydrous metal oxides on the anaerobic degradation of gamma-666, p,p'-DDT in Liaohe River sediments].

Effects of organic matter and active Fe, Al, Mn oxides on the anaerobic degradation of gamma-666, p,p'-DDT were investigated by means of removing organic matter and hydrous metal oxides in Liaohe River sediments sequentially. The results showed that the anaerobic degradation of gamma-666, p,p'-DDT followed pseudo-first-order kinetics in different sediments; but, the extents and rates of degradation were different, even the other conditions remained same. Anaerobic degradation rates of gamma-666, p,p'-DDT were 0.020 d-1, 0.009 d-1 respectively for the sediments without additional carbon resources. However, with additional carbon resources, the anaerobic degradation rates of gamma-666, p,p'-DDT were 0.071 d-1 and 0.054 d-1 in the original sediments respectively. After removing organic matter, the rates were decreased to 0.047 d-1, 0.037 d-1. In the sediments removed organic matter and hydrous metal oxides, the rates were increased to 0.067 d-1, 0.059 d-1. This results indicated that organic matter in the sediments accelerated the anaerobic degradation of gamma-666, p,p'-DDT; the hydrous metal oxides inhibited the anaerobic degradation of gamma-666 and p,p'-DDT.

Impact of long-term exposure to air particulate matter on life expectancy and survival rate of Shanghai residents.

OBJECTIVE: To evaluate the impact of long-term air particulate matter exposure on the life expectancy and survival rate of Shanghai residents. METHODS: Epidemiology--based exposure-response function was used for the calculation of attributable deaths to air particulate matter in Shanghai, and the effect of long-term exposure to particulate matter on life expectancy and survival rate was estimated using the life table of Shanghai residents in 1999. RESULTS: It was shown that in 1999, the long-term air particulate matter exposure caused 1.34-1.69 years reduction of life expectancy and a decrease of survival rate for each age group of Shanghai residents. CONCLUSION: The effect of long-term exposure to air particulate matter on life expectancy is substantial in Shanghai.

MRI brain T1 relaxation time changes in MS patients increase over time in both the white matter and the cortex.

OBJECTIVE: To test the sensitivity of whole-brain T1 relaxometry to the evolution of pathological changes in multiple sclerosis (MS). BACKGROUND: T1-weighted hypointense lesion load in the brains of patients with MS is associated with axonal loss. Other work has shown that T1 measurements may provide information complementary to existing imaging techniques, such as magnetization transfer imaging. METHODS: The authors studied 14 MS patients twice over a median time interval of 19.5 months (range, 14-22 months). Structural images and whole-brain T1 maps using a novel rapid-scanning technique (3 min/study) were performed at 3 T. Analysis focused on defining changes separately in the lesional and normal-appearing white matter (NAWM) and in the cortical gray matter. RESULTS: At baseline, there was an inverse relationship between disease duration and the NAWM T1 histogram peak height (r = -0.75, P = .03). The total white matter T1 histogram peak height decreased over time (P < .001). This could be accounted for by changes in the NAWM (P < .03). There also was a decrease (6%) in the mean (11 of 14 patients, P = .004) and in the median (7%) (13 of 14 patients, P < .001) neocortical gray matter T1 over the follow-up period. CONCLUSIONS: Brain T1 maps can be generated quickly and are sensitive to pathological changes over time. T1 values in both the gray and the white matter at the baseline visit were related to disease duration, suggesting that the T1 changes are clinically relevant. Although the absolute values will be different, it is likely that similar changes will be able to be detected at 1.5 T. The role of T1 measurement as a magnetic resonance imaging outcome measure in clinical trials now should be explored.

Dementia associated with periventricular and deep white matter alterations: a subtype of subcortical dementia.

This research examined the neuropsychological functioning of demented patients with periventricular and deep white matter alterations. Thirty-three outpatients with NINCDS-ADRDA probable Alzheimer's disease (AD) and 27 outpatients with probable/ possible ischaemic vascular dementia (IVD, Chui et al., 1992) associated with periventricular and deep white matter alterations matched for age, education, level of dementia, and functional disability were studied. White matter alterations were measured using a 40-point scale previously described by Junque et al. (1990). Subjects with cortical CVAs were excluded. On executive control tests, IVD subjects made more preservations on tests of mental control and response set, and produced fewer responses on phonemic controlled oral word association tests (letters: F,A,S). IVD subjects also made more preservations and graphomotor errors on clock drawings. On the California Verbal Learning Test the IVD group performed better than AD subjects on the short delay free recall test condition, the recognition discriminability index, and made fewer intrusion errors on both free and cued recall conditions. We conclude that neuropsychological assessment can differentiate AD from IVD associated with white matter alterations, and that the neuropsychological profile of demented subjects with significant periventricular and deep white matter alterations is similar to other subcortical dementing illnesses.

[Concepts and relative analytical techniques of soil organic matter].

The research of soil organic matter (SOM) has been highlighted in soil science. In the past 50 years, new perspectives in the relationship between SOM and sustainability of atmosphere and biomosphere, and strong motivation to find a vivid index for soil quality variation induced the transformation in concepts and analytical techniques of SOM: the curiousness to humic substances faded off since they were dull to anthropogenic activities, and interests were focused on the light fraction of organic matter (LFOM), organic carbon associated with different mineral particles in size, particulate and intra-particulate organic matter (POM and iPOM), water soluble organic matter (WSOM), and microbial biomass carbon (MB-C). The relative fractionation procedures have been developed, and the main research activities on SOM are transformed from the products of microorganisms (humus) to the organic matter comprised in plant residues at their various decomposition stages and the organic carbon in microorganisms, since they are biologically active and immediately respond to soil cultivation and tillage, crop rotation, and fertilizer application, etc.

Enhancing gray-to-white matter contrast in 3T T1 spin-echo brain scans by optimizing flip angle.

BACKGROUND AND PURPOSE: Compared with MR imaging at 1.5T T1-weighted spin-echo imaging at 3T shows up with reduced gray-to-white matter contrast. The purpose of the present study was to show the effects of alterations of different flip angles as an easily accessible parameter to increase gray-to-white matter contrast. METHODS: Spin-echo T1 sequences of 6 healthy volunteers were acquired in a 3T head scanner with 5 different flip angles. Observer-independent contrast-to-noise ratios for gray versus white matter from different flip angles, as well as subjective ratings of image quality from 2 blinded neuroradiologists, were compared statistically. RESULTS: Gray-to-white matter contrast increased significantly with decreasing flip angle. No artifacts were introduced by decreasing flip angles, and T1 contrast characteristics were robust and stable at lowered flip angles. Also, specific absorption ratios significantly decreased with decreasing flip angles. CONCLUSION: Using a flip angle of 50 degrees significantly increases gray-to-white matter contrast in T1 spin-echo brain scans at 3T B0 field strength.

Some opinions about matter and material substances: from inanimate system -- to living according to A. Einstein general theory of relativity.

During transformation from inanimate to living, change of the space position of the matter causes the change of the field, as the space does not exist without the field, therefore the time-space as the properties of material substances, should undergo certain changes. The outside inanimate system, in this case a matrix, has its own time. The living system, in this case a cell, where the matter undergoes space conformation with the change of field and space-time, has its own time and it has begun to flow more rapidly than in matrix. From the surface of the body, from different energetic reservoirs oppositely charged matter substances following from special transport systems from the life system transmitted into lifeless one and change their matter space conformation, create transmission gradient that is the gradient border of time from lifeless system into live. In the case of a human, hypothetically, the gradient system of time must be of a complex scheme counting the inter-transformation and interaction gradients of outer and inner abdominal systems. Subconscious and consciousness by means of special links and messages, information selection interact and form unique connection between the systems. Subconscious serves for accelerated time system. Conscious by means of permanent contact with the environment collects and reacts in matrix time system By interconnection of these two systems ideal adaptation with the environment takes place. Time difference gradient system is an additional energy factor, by means of which respective ordered geometrical structures special for the given types are formed. The living organism is an inter-regulated interconnection global system resulting from the changes of matter and material substances space configuration.

Leukoencephalopathy with vanishing white matter due to homozygous EIF2B2 gene mutation. First Polish cases.

Leukoencephalopathy with vanishing white matter (VWM), also called childhood ataxia with central nervous system hypomyelination (CACH), is an autosomal recessive disease caused by mutations in any of the five genes encoding subunits of the eukaryotic translation initiation factor eIF2B. Neuropathological findings comprise a severe, cavitating orthochromatic leukodystrophy with only small amounts of myelin breakdown products, and predominantly involving the cerebral hemispheric white matter. Within the white matter abnormal oligodendroglial cells are present with abundant "foamy" cytoplasm. In some regions oligodendroglial cells are increased in numbers. We present three sisters, 18, 11 and 8 years old, with the early to late childhood phenotype. The first signs of the disease were gait disturbances at 4, 2 and 6 years of age, respectively. Neurological examination showed mild tremor of hands and head, truncal ataxia, dysarthria, and hypotonia, after several years followed by spasticity. The course of the disease was slowly progressive. Intellectual abilities are relatively spared. The MRI showed diffusely abnormal white matter of the cerebral hemispheres. The FLAIR images revealed rarefaction of the affected white matter with some stripe-like structures, suggesting the presence of remaining tissue strands. The abnormalities were most pronounced with the middle sister, who had the earliest onset of the disease. A homozygous point mutation in the EIF2B2 gene was found, 638A>G. Both the parents were found to be carriers of this mutation. This is the first description of a Polish family with VWM.

[Change of humic-like fluorescence characteristics of dissolved organic matter from Dagu River to Jiaozhou Bay].

Fluorescence excitation-emission matrix spectroscopy was employed to characterize the change of humic-like fluorescence from Dagu River to Jiaozhou Bay. The relationships of humic-like fluorescence characteristics with salinity, pH, chlorophyll a, dissolved organic carbon, chemical oxygen demand and carbohydrate were discussed. The results show that humic-like fluorescence intensities are high in Dagu River but low in Jiaozhou Bay. The input of Dagu River is important origin of humic-like fluorescent dissolved organic matter of Jiaozhou Bay. High humic-like fluorescence intensity and high chlorophyll a are not coexist always, which suggests that humic-like fluorescent dissolved organic matter doesn't mainly come from phytoplankton. Good relationships of humic-like fluorescence intensity with dissolved organic carbon, chemical organic demand and carbohydrate suggest that humic-like fluorescent matter is the main component of dissolved organic matter. The distribution of humic-like fluorescence reflects the distribution of dissolved organic matter to some extents.

Assessment of white matter damage in subacute sclerosing panencephalitis using quantitative diffusion tensor MR imaging.

BACKGROUND AND PURPOSE: Subacute sclerosing panencephalitis (SSPE), a rare progressive degenerative disease, is caused by persistent infection with a defective measles virus. The correlation between the clinical staging and MR imaging is usually poor. The aim of the study was to investigate the role of diffusion tensor imaging (DTI) in the early detection of white matter damage in SSPE in the presence of normal findings on conventional imaging. METHODS: DTI was performed in 21 patients in stage II SSPE and 10 age/sex-matched healthy controls. Fractional anisotropy (FA) and mean diffusivity (MD) values were calculated in the periventricular white matter, corpus callosum, and posterior limb of the internal capsule in patients with normal and abnormal findings on conventional imaging as well as healthy controls. RESULTS: The patients were grouped into those with normal (n = 11) and abnormal (n = 10) findings on conventional imaging for the purpose of quantitative DTI analysis. Abnormal- and normal-appearing white matter on T2-weighted images showed significantly decreased FA values in all the regions compared with those in healthy controls. MD values were significantly increased in the periventricular white matter region of the frontal and parietooccipital lobe in patients with normal as well as abnormal findings on conventional imaging compared with those in healthy controls. CONCLUSION: DTI detects early white matter abnormalities that may have significant therapeutic implication, even in the presence of normal findings on conventional imaging, in patients with SSPE.

Association of deep white matter infarction with chronic communicating hydrocephalus: implications regarding the possible origin of normal-pressure hydrocephalus.

The coexistence of cerebrovascular disease leading to deep white matter infarction and normal-pressure hydrocephalus has been noted previously in clinical studies, as both diseases can present with the triad of gait disturbance, dementia, and incontinence. The purpose of this MR study was to determine if the two diseases demonstrated a statistical association. Evidence of patchy periventricular hyperintensity representing presumed deep white matter infarction was sought in 20 patients shunted for normal-pressure hydrocephalus and in 35 additional consecutive patients with clinical symptoms and MR findings consistent with normal-pressure hydrocephalus. Deep white matter infarction was also sought in 62 consecutive age-matched control subjects. There was a statistically significant (p less than .001) higher association (58%) of marked infarction in the 55 patients with normal-pressure hydrocephalus than in the age-matched controls (24%). MR findings of communicating hydrocephalus (ventriculomegaly and increased aqueductal CSF flow void) were sought in 78 consecutive patients with presumed deep white matter infarction, and the degree of severity of the two diseases was also found to be statistically significant (p less than .05). In view of this association, the possibility that the two diseases are related was considered. A potential mechanism is discussed whereby deep white matter infarction leading to decreased periventricular tensile strength could result in communicating hydrocephalus. It is plausible that normal-pressure hydrocephalus may result from a number of different insults to the brain.

Incidental white matter hyperintensities on magnetic resonance imaging in HIV-1 infection. Multicenter AIDS Cohort Study.

Magnetic resonance (MR) scans were performed as part of a prospective neuropsychological study within the Multicenter AIDS Cohort Study. Fifty HIV-1-seronegative men, 85 HIV-1-seropositive men without constitutional symptoms, and 14 with symptomatic HIV disease underwent MR imaging using a uniform protocol. Scans were rated by neuroradiologists blinded to all clinical details except age. The majority of MR scans were normal in all of the clinical groups and no covert mass lesions or diffuse white matter abnormalities were identified. Focal hyperintensities in the white matter were observed in 24% of the HIV-1 seronegatives, 26% of HIV-1 asymptomatic seropositives (CDC II/III), and 17% of those with ARC/AIDS. No significant associations were noted between the white matter hyperintensities and HIV-1 serostatus, neurological abnormalities, CD4 count, alcohol or drug use, hypertension, or smoking. In one individual classified with early HIV-1 dementia, MR demonstrated several hyperintensities in the deep parietal white matter, but at autopsy no microscopic abnormalities corresponding to the MR findings were identified. Our studies imply that focal white matter hyperintensities identified on MR are not specific for HIV-1 infection and are probably incidental and of no clinical significance.
