Morphology, histochemistry, and the biological significance of the lymphatic system of the trematode Orthocoelium scoliocoelium.

The gross morphology and histological and cytological architecture of the lymph system of Orthocoelium scoliocoelium were studied. Each main vessel, near the intestinal bifurcation forms a spindle-shaped sinus. Lymphocytes, similar to the primitive blood cells, have been seen to form aggregates in certain regions of the lymph system. In addition, detailed histochemical observations were made on the localization and distribution pattern of various phosphatases, esterases and dehydrogenases. It is suggested that the lymph system, in addition to the functions already known, may also selectively reabsorb certain important outgoing higher fatty acids from the excretory system. Furthermore, the lymphocytes may also be involved in the defense mechanism of the worm. The nature and function of fine granules in the lymph remain speculative.

[Involvement of the lymphatic system in post-thrombotic syndrome].

The involvement of the lymphatic drainage in patients with postthrombotic syndrome can be verified by (quantitative) isotopic lymphography (99m Tc labelled microcolloids) and by indirect lymphography (water-soluble contrast medium Iotrolan). Both methods point to an augmented praefascial lymph-transport. In the region of lipodermatosclerotic skin changes dermal backflow can be explained by a local damage of peripheral skin-lymphatics ("localized lymphoedema"). No working lymphatics can be found in the border of ulcers. As could be demonstrated by intramuscular injection of the radioactive tracer subfascial lymphtransport is decreased not only in the postthrombotic stage but already in the acute phase of a deep vein thrombosis. The alterations of the lymph-drainage in a postthrombotic syndrome may have some pathophysiological importance for the development of the tissue changes and may also explain some clinical signs.

Current topics of physiology and pharmacology in the lymphatic system.

We have reviewed physiological significance of rhythmical spontaneous contractions of collecting lymph vessels, which play a pivotal role in lymph transport and seem to control lymph formation through changing the pacemaker sites of the rhythmic contractions and contractile patterns of the lymphangions. A characteristic feature that the rhythmic pump activity works in vivo physiologically under the specific environment of lower oxygen tension in lymph (25-40 mm Hg) has been evaluated. With the characteristic feature, generation of endogenous nitric oxide (NO) from lymphatic endothelial cells and/or activation of ATP-sensitive potassium channels (K(ATP)) are reviewed to play crucial roles in the regulation of lymph transport at physiological or pathophysiological conditions. Chemical substances released from malignant tumor cells and tumor-derived parathyroid hormone-related peptide (PTHr-P) are also shown to cause a significant reduction of lymphatic pump activity through generation of endogenous NO and activation of K(ATP) channels. Finally, we have discussed physiological significance and roles of the lower oxygen tension in lymph, generation of endogenous NO, and activation of K(ATP) in lymph formation, lymph transport, and the functions of lymph nodes.

Ultrastructure of the cerebrospinal fluid outflow along the optic nerve into the lymphatic system.

OBJECT: To explain the spontaneous CSF outflow into the orbit, the ultrastructure of the perineural meningeal layers at the distal and the proximal portions of the optic nerve were compared. METHODS: Ten cats were perfusion fixated and the orbital content removed for transmission and scanning electron microscopy. In five animals a 60-min cisternal infusion of contrast medium at low intracranial pressure was performed before perfusion fixation. RESULTS: In the contrast-infused animals it was possible to demonstrate the leakage of contrast medium in the distal portion of the optic nerve sheath (ONS) from the subarachnoid space (SAS) into the orbit and find it in the conjunctival lymphatics. Electron microscopy revealed that in the distal portion of the ONS the neurothelial layers are significantly thinner, some consisting of only one layer. Pore-like openings in the neurothelial covering are seen in the distal portion. Excavations of the SAS are far more numerous in the distal portion of the ONS. The excavations reach the neurothelial layer. Intracellular and extracellular filaments are more numerous in the distal portion of the ONS. There is no significant difference in the dura mater between the distal and proximal ONS. The results show the existence of an arachnoid window area in the distal portion of the ONS. It is characterised by a continuous, but thinned neurothelial barrier layer, with few pore-like openings. CONCLUSIONS: The main differences between distal and proximal ONS are a thinned neurothelial barrier layer and an increased number of intercellular filaments and pore-like openings. The findings explain the lymphatic CSF outflow pathway along the optic nerve.

[Lymphatic system of the human testis].

The distribution of lymph vessels in the human testis was investigated using ink injection methods, light and electron microscopy. Lymph capillaries occur in the septula testis but are absent in the intertubular tissue. They consist of endothelial cells provided with an incomplete basal lamina and bound by anchoring filaments to the adjacent connective tissue. Frequently, the endothelial cells are separated by gaps (measuring up to 2 microns). The lymph capillaries of the septula testis are connected to lymph vessels in the rete testis and tunica albuginea. These vessels have occasional smooth muscle cells and valves. At the posterior margin of the testis, the network of lymphatics merges into collecting ducts that together with vessels derived from the rete testis are drained by lymphatics in the spermatic cord.

[Effects of endolymphatic lymph stimulation on draining and transporting function of the lymphatic system in experimental peritonitis].

Bacterial aggression and tissue destruction during experimental peritonitis cause progressive disorders of blood and lymph microcirculation, paralleled by increase of the coagulation potential of the blood and lymph, activation (during the first 12 hr) of the drainage function of the lymph system, followed in 24 h by a drop of lymph flow and its complete blocking in 48-72 h. This is caused by disseminated microhemo- and lymphothrombosis with decompensation of the drainage and transporting functions of the lymph system during this period of the disease. Lymphogenic effect of endolymphatic stimulation by drugs in experimental peritonitis is notably (by 1.8-4.5 times) higher than the effect of routine lymph stimulation with the same drugs and their combinations.

Role of macrophages as possible transporters of Plasmodium yoelii nigeriensis merozoites through the lymphatic system. Preliminary note.

Vesicles containing apparently healthy merozoites from mature schizonts were observed in the spleen and lymph nodes of mice parasitized by Plasmodium yoelii nigeriensis. They differed from all parasitic stages undergoing digestion by the macrophage and from mature schizonts of the blood. Up to 40 merozoites from three mature schizonts may be seen in the same compartment. They are thought to be accumulations of latent merozoites.

Who discovered the lymphatic system.

The 17th century saw several emerging and almost simultaneous discoveries in the field of lymphology by Asselli, Pecquet, Bartholin and possibly Joliffe. However, Olof Rudbeck (1630-1708) of Sweden, a true scientific genius, who mastered botany, chemistry, physics, mathematics, astronomy, music, drawing, architecture and engineering, and became the Rector of the Faculty of Upsala, was probably the first anatomist to consider correctly the lymphatic circulation as an integrated system of the whole body.

Cannulation of the cardiac lymphatic system in swine.

OBJECTIVE: Cardiac lymph is the most direct medium for analyzing metabological changes in the myocardial cell. Currently, dogs are the animals used for investigation of myocardial lymphatic function. However, questions arise when comparing and interpreting the human system to the experimental model, since the dog coronary anatomy is different from human anatomy and pulmonary lymph contamination is found in up to 81% of the cases. Swine, having similar coronary anatomy to humans, are a proven model for cardiovascular research. The purpose of this study was to investigate the cardiac lymphatic anatomy of the swine and to develop a reliable cannulation technique to collect the lymph. METHODS AND RESULTS: The lymphatic anatomy of 60 pigs was studied and classified and a new technique for lymphatic cannulation was developed. The cannulation success rate was 55%. In addition, no pulmonary lymph contamination was found at the cannulation site. CONCLUSION: We conclude that porcine myocardial lymphatics can be successfully cannulated for the investigation of myocardial lymphatic function.

[Pathomorphology of the intestine and regional lymphatic system in pseudotuberculosis].

Available are data obtained at light and electron microscopy of operative specimens from patients with abdominal pseudotuberculosis and animals challenged orally with Yersinia pseudotuberculosis. The authors are the first to outline detailed characteristics of the intestinal and regional lymph node lesion arising in response to the infection and reflecting growing resistance to it. These features of pathological process involve marked tissue eosinophilia, necrosis due to phagocytes rexis, and granulomatosis suggesting a pronounced role in the pathogenesis of the body allergization in the course of infection. It is proposed to consider pseudotuberculosis-related changes in lymph nodes as lymphoblastic (early affection) and granulomatous-necrotic (advanced infection) lymphadenitis. The evidence obtained can promote differential diagnosis of pseudotuberculosis.

[Dynamics of indices of the rat endocrine and lymphatic systems during cold adaptation].

A study was made of the content of corticosterone, thyroxine and triiodothyronine in the blood, of the thymic weight and cytomorphological characteristics of rat lymph nodes during prolonged cold (4-5 degrees C) adaptation. Interrelated alterations in the characteristics under study were revealed at different stages of adaptation. The morphofunctional recovery of the lymphoid tissue occurs in the phase of the increased specific stability in spite of the fact that the action of cold is continued and the blood corticosterone concentration is elevated. A new type of the endocrine-lymphoid relationships is supposed to be formed during lengthy adaptation, with this type being different from that both in the starting state and acute stress.

Zinc transport by the heart lymphatic system after acute myocardial infarction.

Open-chest dog preparations were used to determine divalent cation transport following acute myocardial infarction. Cardiac lymph flow, lymph and plasma protein, zinc, calcium, and magnesium content and hemodynamic measurements were recorded every 20 min before and after coronary artery occlusion in sham operated (N = 4), infarcted (N = 6), and lymph-ablated animals (N = 4). During the 4-hr occlusion period, with constant blood pressure, lymph flow increased from 1.53 +/- 0.25 to 2.15 +/- 0.44 mg/hr (SEM), P less than 0.01. Zinc decreased in plasma from 0.69 +/- 0.10 to 0.41 +/- 0.08 micrograms/ml, P less than 0.01, and in lymph from 0.69 +/- 0.08 to 0.40 +/- 0.02 micrograms/ml, P less than 0.01. Zinc to protein ratio decreased similarly to total zinc in plasma and lymph. Changes in calcium and magnesium were insignificant. Lymph to plasma concentration ratios increased for protein from 0.57 +/- 0.05 to 0.62 +/- 0.02, P less than 0.05, and for zinc from 1.10 +/- 0.26 to 1.21 +/- 0.14, P less than 0.05. Heart lymph clearance (lymph:plasma ratio X lymph flow) steadily rose for protein from 0.31 to 0.06 to 0.50 +/- 0.08, P less than 0.05, and for zinc from 0.59 +/- 0.18 to 0.92 +/- 0.15, P less than 0.05. Lymph and plasma measurements did not change significantly in sham-operated animals. Plasma zinc remained unchanged from baseline after coronary occlusion in all lymph-ablated animals. The increased clearance of protein and zinc suggests that plasma proteins are zinc carriers after acute myocardial infarction and that the reduction of plasma zinc is dependent upon an intact cardiac lymphatic circulation.

The response of the lymphatic system to the human skin resident bacteria.

We designed a study to identify skin bacterial strains producing most intensive changes in lymphoid tissue of the draining nodes and analyse the node cell phenotypes reacting to bacterial antigens. The obtained results were expected to help in establishing rational therapy in recipients of hand allografts. The highest level of activation of lymphoid tissue was found after inoculation with S. epidermidis, Acinetobacter, Enterobacter and Aerococcus. There was an increase in weight and cell concentration of nodes, intense expression of W3/25+ (CD4), OX6+ (class II), EDI+ (CD14), CD54+ and OX62+ cells in the paracortex and medulla 7 days after bacterial inoculation. The most advanced changes were seen after recall bacterial stimulation. Taken together, evident differences in the intensity of response to the investigated bacterial strains were found. The obtained information can be useful for establishing rational antibiotic therapy after composite graft transplantation.

[The diffuse lymphatic system of the nasal mucosa in allergic rhinitis].

Morphologic and immunologic study were performed on the mucosa associated lymphoid tissue (MALT) of patients with allergic rhinitis. Scraping from 14 healthy subjects and 36 allergic ones were used. Besides ordinary hystological methods immunohistochemical ones wilk polyclonal antibodies were employed to study IgG, IgA, IgM, IgE and monoclonal antibodies used for B and T lymphocyte and subset T-suppressor cell identification. In a comparison between normal and allergic mucosa, the morphopathology show an accentuated edema and a slight fibrosis. Among the IgG, IgA and IgM do not show any substantial differences in the samples in normal or in allergic subjects; white traces of IgE are found in allergic patients, but are totally absent in normal ones. In the lymphocyte populations does not show any substantial changes between the two groups. Was also analyzed the mechanism and the majority of the factors by which were obtained the results.

[Morphological changes in the lymphatic system of children with hereditary immunologic deficiency syndromes].

The morphology of lymphatic tissues in 43 autopsy cases of children with inherited immunodeficiency states were analysed. Among the more common diseases, such as Di-George-syndrome, CID-patients, congenital agammaglobulinemia Bruton, CVID, selective IG-A deficiency, Wiskott-Aldrich-syndrome, tissue sections of very rare conditions associated with immunodeficiency, e.g. fetopathia diabetica and leprechaunismus, were investigated by routine and immunohistochemical stainings. Clinical history and laboratory data, augmented by the characteristic pathomorphology of lymphatic tissue sections, will establish or at least suggest a definite diagnosis. Since true thymic dysplasia is very rare (or even non-existent) in the human, this term should be abandoned. Severe thymic tissue alterations in SCID-patients, occur secondary to enzyme defects in lymphatic cells. If patients are successfully treated by bone marrow transplantation, the thymus will subsequently develop into a functionally normal organ.
