This report describes the assay of collagenolytic activity, particularly activity of tissue collagenases, using the reconstituted fibrils of fluorescein-labeled collagen type I as substrate. Labeling of soluble rat skin collagen was carried out by the modified method of Baichi et al 1980. This method is very sensitive, reproducible and useful for screening large number of samples.

The subcellular distribution of the 3H--2,6-dimethyl-3,5-dicarbomethoxy-4(2-isothiocyano) phenyl-1,4-dihydropyridine (DPSCN) binding to guinea-pig ileal smooth muscle was studied by subcellular fractionation. Initial experiments on subcellular fractionation of 3H-DPSCN-labelled tissues by differential centrifugation showed that there was an excellent correlation between the levels of the label present in a fraction and the plasma membrane marker phosphodiesterase I (r = 0.98) but not between the label and the putative endoplasmic reticulum marker NADPH: cytochrome-c-reductase (r = 0.56) or the inner mitochondrial marker cytochrome-c-oxidase (r = 0.36). Centrifugation of the microsomes on a continuous sucrose density gradient showed an excellent correlation of the migration of the label with phosphodiesterase I activity (r = 0.93) but not with the activities of NADPH: cytochrome-c-reductase (r = 0.66) or cytochrome-c-oxidase (r = 0.44). Treatment of microsomes with digitonin (1 mg/ml) followed by centrifugation on continuous sucrose density gradients increased the weighted mean densities of the phosphodiesterase activity (plasma membrane marker) and the labelling by similar magnitudes (0.04 to 0.06 g/ml). The weighted mean densities of NADPH: cytochrome-c-reductase and the cytochrome-c-oxidase were not altered significantly. It is concluded that in the guinea-pig ileal smooth muscle, DPSCN labels the plasma membrane specifically.
As previous studies have suggested a hormone dependence of binding sites for peanut agglutinin in mammary neoplasm, this feature has been thought to be correlated to steroid receptor status. The present investigation was undertaken on a well-established ovarian-dependent cancer model in order to check this hypothesis. Sections of primitive tumor transplants as well as of tumors induced in vivo by injection of cell clones were analyzed with the use of three fluorescent lectins. The lectin binding sites were evaluated semi-quantitatively and compared with estrogen and progesterone receptor levels. Using non-parametric statistical tests, the results revealed a strong correlation between the expression of peanut agglutinin (PNA) binding sites and steroid receptor status, but only in primitive tumor transplants. No such correlation was observed in tumors induced in vivo, by injection of cell clones. No correlation between the steroid receptor status and the two other lectins (Concanavalin A and Dolichos biflorus) was observed. These data suggest that PNA can be used as a valuable histochemical tool in steroid hormone dependence study.
Intact porcine ova and mechanically isolated zonae pellucidae from the ova were treated with a limiting amount of FITC in isotonic solutions at different pHs. The modified zona proteins were fractionated into three families (FTC-PZP1-3) by HPLC on a gel filtration column. It was found by this HPLC that the amino groups of PZP3 hardly reacted with FITC, whereas those of PZP1 and 2 fairly reacted, reflecting the organization of these families in the zona structure. The difference in reactivity of the three families with FITC suggested the presence of a structural unit in the zona, thus supporting the filamentous model of Wassarman.

The qualitative differences in the affinity of concanavalin A (Con A), wheat-germ agglutinin (WGA) and Phaseolus vulgaris lectin to the surface of 10 microbial strains inducing various diseases in humans and agricultural animals have been demonstrated by means of the indirect immunofluorescence tests. Enterobacteria, Coxiella burnetii and Bacillus anthracis have been found to possess pronounced affinity to Con A and WGA, while Rickettsia prowazekii, Francisella tularensis and Brucella abortus, as well as Treponema pallidum, have proved to be resistant to lectins. WGA has been found to bind specifically to Brucella abortus and Treponema pallidum. Con A and WGA are seemingly suitable for use in the preliminary test for the total capacity of lectin receptors to come in contact with biological macromolecules.

Murine cytotoxic responses to TNP-modified syngeneic cells (TNP-self) have been shown to exhibit preferential recognition of K or D end self products encoded by the H-2 complex. In the present study, a number of B10 congenic and recombinant mouse strains were investigated to determine the H-2K and H-2D-restricted FTC-self CTL response patterns, and these were compared with the CTL response patterns obtained for TNP-self. The results indicate that for strains possessing the H-2k,d,h2,h4 haplotypes, respectively, preferential CTL responses were observed against FTC recognized in association with Kk over Dk, Dd over Kd, and Kk over Db. These patterns of preferential CTL responses were the same as those reported for TNP-self as well as several anti-viral CTL responses. In contrast to the results obtained in the B10.A strain, in which Kk preference was observed over Dd for TNP-self CTL, no preferential CTL response was observed when FTC was recognized in association with Kk and with Dd. In this context, it was observed that the CTL response to FTC recognized in association with Dd was particularly strong. This strong D end-associated response was shown to involve D locus products, and no evidence was obtained indicating that L locus self products were involved. These studies are discussed with respect to the possibility that different haptens can be recognized by CTL in association with different self determinants encoded by the same H-2 gene products.
