Chemical composition, seasonal variability, and antifungal activity of Lavandula stoechas L. ssp. stoechas essential oils from stem/leaves and flowers.

Essential oils from the stems/leaves (L) and flowers (F) of Lavandula stoechas L. ssp. stoechas growing wild in southern Sardinia (Italy) were extracted by hydrodistillation and analyzed by gas chromatography coupled with flame ionization detector and ion trap mass spectrometry. The major compound was fenchone, accounting for, on average, 52.60% in L and 66.20% in F, followed by camphor (13.13% versus 27.08%, in L and F, respectively). F essential oil yields (volume per dry weight) decreased from the beginning to the end of the flowering stage, whereas L yields remained constant during the year. The nine main compounds derived from two different subpathways, A and B. The compounds that belong to the same subpathway showed a similar behavior during the year. The essential oils were tested for their antifungal activity using the paper disk diffusion method. The essential oils tested were effective on the inactivation of Rhizoctonia solani and Fusarium oxysporum and less effective against Aspergillus flavus. Among the single compounds tested, fenchone, limonene, and myrtenal appeared to be the more effective on the inhibition of R. solani growth.

Composition, enantiomeric distribution, and antibacterial activity of the essential oil of Achillea ligustica All. from Corsica.

The essential oil of Achillea ligustica from Corsica was investigated by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS). A total of 82 compounds representing 94.0% of the oil were tentatively identified. The main constituents were the camphane derivatives, representing >30% (camphor, 21.3%; borneol, 6.2%; bornyl acetate, 3.5%) of the whole oil, and santolina alcohol (19.3%). The enantiomeric distribution of 8 chiral constituents was determined by GC-MS using two enantioselective stationary phases (DIME-beta-CD and Lipodex-E). Racemic santolina alcohol, required for optimization of the enantioselective GC conditions, was prepared by an original two-step synthesis from 2,5-dimethylhexa-2,4-diene. The whole essential oil was tested for its antibacterial activity against a wide range of bacteria using a paper disk method. The results show a promising activity against Streptomyces species.

Dynamics of yield components and essential oil production in a commercial hybrid sage (Salvia officinalis x Salvia fruticosa cv. Newe Ya'ar no. 4).

The fresh yields, the essential oil content, and the quality of a sage hybrid (Salvia officinalis x Salvia fruticosa, cv. Newe Ya'ar No. 4, Lamiaceae) as affected by development and harvest time were determined. Marked increases in plant height and in the number of nodes developed per plant together with a modest increase in leaf size were accompanied by dramatic increases (more than 20-fold) in the fresh yields throughout a 50-day growth period. No major changes in the essential oil content per fresh weight and its composition were detected throughout the growth period. In contrast, the compositions of the essential oils obtained from stems, as compared to leaves and leaf-primordia, had marked differences. Developmentally controlled changes in the extractives from individual leaf pairs from the same plant were also noted. In upper young leaves, the oxygenated diterpene manool and the sesquiterpene hydrocarbons alpha-humulene and beta-caryophyllene constituted up to 20%, 8%, and 4% of the total extractives, respectively. In older leaves, the abundance of these components steadily dropped to roughly half their levels in young leaves. Conversely, the proportions of the monoterpenes, particularly the ketones camphor and alpha-thujone, steadily increased with leaf position. Minor changes in the levels of other extractives were also recorded. These studies imply independent regulatory patterns for di-, sesqui-, and monoterpenes in this sage hybrid, and suggest possible agrotechnical means to obtain preferred chemical compositions of its essential oil.

Synthesis of substituted 1-norbornylamines with antiviral activity.

The reaction of (+/-)-camphor (7) with triflic anhydride (Tf2O) yields the bridgehead triflate 8. The Nametkin rearrangement of 8 to 3 was realized by treatment with triflic acid (TfOH). The solvolysis of the bridgehead triflates 3 and 8 in acetonitrile affords the N-acetyl-1-norbornylamines 4 and 9. The Pd(0)-catalyzed hydrogenation of 4 and 9 gives the amides 5 and 10. The corresponding 1-norbornylamines 2 and 13 and the N-ethyl derivatives 1, 6, 11, and 12 were obtained by basic hydrolysis or reduction with LiAlH4, respectively, of the amides 4, 5, 9, and 10. The antiviral activity of the hydrochlorides of some of the obtained 1-norbornylamines was evaluated against influenza A, herpes simplex 2, and African swine fever virus. Particularly noticeable is the activity of the hydrochlorides of 1 and 11 against influenza A virus (SI (selectivity index) = 1000).

Synthesis and resolution of (+-)-7-chloro-8-hydroxy-1-(3'-iodophenyl)-3-methyl-2,3,4,5-tetrahydro- 1H-3- benzazepine (TISCH): a high affinity and selective iodinated ligand for CNS D1 dopamine receptor.

The synthesis and resolution of (+-)-7-chloro-8-hydroxy-1-(3'-iodophenyl)-3-methyl-2,3,4,5-tetrahydro-1 H-3- benzazepine, (+/-)-TISCH (8) has been achieved by resolution of intermediate 4, the O-methoxyl, 3'-bromo derivative, as the diastereomeric camphor sulfonate salt. The final products, R-(+)-8 and S-(-)-8, were prepared by treatment of R-(+)- or S-(-)-7, the 3'-tributyltin intermediates, with iodine in chloroform, followed by O-demethylation. By using HPLC with a chiral column, the optical purity (greater than 99%) of the intermediates and the final compounds was determined. Radioiodination was achieved by an iodo-destannylation reaction with sodium [125I]iodide and hydrogen peroxide. As expected, the R-(+)-[125I]-8 (the active isomer) displayed high affinity and selectivity to the CNS D-1 receptor in rat striatum tissue preparation (Kd = 0.205 nM). The rank order of potency was as follows: SCH-23390 (1a) greater than (+/-)-8 greater than (+)-butaclamol greater than spiperone, WB4101 greater than dopamine, 5-HT. After an iv injection, the R-(+)-[125I]-8 penetrated the blood-brain barrier with ease and displayed specific regional distribution corresponding to the D-1 receptor density, while the S-(-)-[125I]-8 showed no specific uptake. The data suggest that the ligand may be useful as a pharmacological tool for characterizing the D-1 dopamine receptor. When labeled with I-123, this ligand is a potential agent for in vivo imaging of CNS D-1 dopamine receptor.

Resolution, absolute configuration, and antiarrhythmic properties of the enantiomers of disopyramide, 4-(diisopropylamino)-2-(2-pyridyl)-2-phenylbutyramide.

Disopyramide was resolved by fractional crystallization of its diastereomeric bitartrate salts from methanol-acetone. Diastereomeric sulfonamides prepared from (+)-camphor-10-sulfonyl chloride and the primary amines obtained by LiAlH4 reduction of the resolved bases were separable by high-performance LC and were used to show that within experimental error resolution of disopyramide was complete. The absolute configuration was determined by X-ray crystallography. (+)-[(2R)-(-)-4-(Diisopropylamino)-2-(2-pyridyl)-2-phenylbutyramide (+)-(2R,3R)-bitartrate] crystallizes in space group P212121: a = 14.789 (4), b = 18.151 (4), c = 9.878 (2) A; Z = 4: Dx = 1.225, Dm (flotation C6H6/CCl4) = 1.226 g cm-3. The structure was solved by direct methods. The enantiomeric bitartrates were tested for antiarrhythmic properties. The enantiomeric bitartrate salts were equally effective in prolonging the effective refractory period (ERP) of rabbit ventricle. At 3 x 10(-6) M, the (-)-bitartrate [from (2S)-(+)-disopyramide] increased the ERP 21.8 +/- 6.3 ms and the (+)-bitartrate [from (2R)-(-)-disopyramide] increased the ERP 25.8 +/- 3.6 ms. At 1.5 x 10(-5) M no significant difference was noted, as the increases in the ERP were 41.2 +/- 8.9 and 50.5 +/- 6.3 ms for the (-)- and %+)-bitartrate, respectively.

Stereoelectronic, torsional, and steric effects on rates of enolization of ketones.

The stereoselectivities of base-catalyzed enolizations of ketones have been studied by quantum mechanical methods. Transition structures of exo and endo deprotonation of camphor, norcamphor, and dehydronorcamphor have been located with two model bases. Stereoelectronic, torsional, and steric effects on activation energies were assessed. These calculations demonstrate the importance of torsional strain between partial bonds and vicinal bonds on the rates of deprotonation and on related reactions involving the formation of enolates or reactions of enols and enolates.

Friedländer synthesis of chiral alkyl-substituted 1,10-phenanthrolines.

The synthetic scope of the Friedländer condensation in the preparation of chiral alkyl-substituted 1,10-phenanthrolines has been investigated. A range of chiral [x,y-b]-cycloalkeno-condensed phenanthrolines has been prepared in one step from steroidal or other cyclic ketones from the chiral pool and 8-amino-7-quinolinecarbaldehyde (1) via base-catalyzed condensation. Phenanthroline derivatives are formed in good yields with unhindered ketones, but the reaction proceeds even with sterically congested substrates such as camphor, albeit in low yield. The utility of the Friedländer condensation has been extended to the synthesis of chiral 3-alkyl-substituted phenanthrolines from monoalkyl-substituted acetaldehydes.

Asymmetric methoxyselenenylations and cyclizations with 3-camphorseleno electrophiles containing oxime substituents at C-2. Formation of an unusual oxaselenazole from an oxime-substituted selenenyl bromide.

Di[(1R)-2-Oximo-endo-3-bornyl] diselenide (4) and its benzoate derivative 5 were prepared from the corresponding known 2-keto diselenide 1. Treatment of 4 and 5 with bromine, followed by silver triflate in methanol-dichloromethane, generated the corresponding selenenyl triflates 6b and 7b. The latter reagents reacted with a variety of mono-, di-, and trisubstituted alkenes to afford the corresponding 1,2-addition products (beta-methoxy selenides) in a highly diastereoselective manner. The free oxime 6b was particularly effective in such methoxyselenenylations, giving diastereomeric ratios (d.r.s) ranging from 86:14 to > 98:2. Even cis-disubstituted alkenes, which typically give poor d.r.s in similar additions with other chiral selenium electrophiles, underwent highly stereoselective additions with this reagent. Reductive deselenizations of the adducts obtained from styrene and cis- and trans-stilbene provided the corresponding methyl ethers, whose absolute configurations were determined by comparison with authentic samples. As expected, the dominant enantiomers thus obtained from cis- and trans-stilbene, using either 6b or 7b, had opposite configurations. Moreover, each geometrical isomer of stilbene produced methyl ethers with the same configuration when treated with either the oxime 6b or the benzoate 7b. Coordination effects between the substituents at the 2-position of the camphor molecule and the positive selenium atoms in the intermediate seleniranium ions are believed to play an important role in determining the stereochemical outcome of methoxyselenenylations. Selenenyl triflate 6b and selenenyl chloride 7c were also investigated in the electrophilic cyclizations of several unsaturated alcohols and carboxylic acids. However, diastereoselectivities were typically much lower than in the methoxyselenenylations. When the selenenyl bromide 6a, derived from the addition of bromine to the corresponding diselenide 4, was allowed to stand in the absence of an alkene, it underwent intramolecular cyclization with the oxime hydroxyl group, followed by further bromination, to afford the unusual oxaselenazole 11, whose structure was determined by spectroscopic means as well as by X-ray crystallography.

A short synthesis of (+)-narciclasine via a strategy derived from stereocontrolled epoxide formation and SnCl(4)-catalyzed arene-epoxide coupling.

A facile construction of the typical framework of narcissus alkaloids has been realized by virtue of the development of a practical route involving stereocontrolled epoxide formation and SnCl(4)-catalyzed arene-epoxide coupling. To achieve this goal, it proved to be necessary to devise a strategy that would enable chemical transformations to install an epoxy moiety in a congested environment. The successful preparation of a hindered epoxide from O-isopropylidene-protected 4-aminocyclohexenol required three steps consisting principally of controlled bromohydration and base-promoted closure and N-alkylation. It was found that a catalytic amount of SnCl(4) not only maintained the catalytic cycle but also effected clean arylation to form a fused BC ring system. Several tactics that ultimately proved to be unsatisfactory are also discussed in an effort to set important boundary limits on arene-epoxide coupling. The requisite enantiopure 4-aminocyclohexenol was available via an asymmetric cycloaddition of diene to camphor-based chloronitroso. The total synthesis of (+)-narciclasine was realized in nine steps with an overall yield of 19%.

A delicate balance of energetics. Subtleties associated with alpha-ketol-based bridge migration to afford 9-keto-10beta-p-methoxybenzyloxytaxanes.

The feasibility of the title reaction has been pursued for the purpose of advancing a concise total synthesis of Taxol. Of the two closely related series examined, the first featured an exo-methylene group at C4. The second consisted of an alpha-epoxide at that site. Strikingly, the olefinic construct proved inert to attempted alpha-ketol rearrangement. In contrast, the oxiranyl derivative isomerized smoothly. The reaction sequence associated with arrival at taxane 18 is short (15 steps from a D-camphor derivative) and notably efficient. The thermodynamic issues that are raised by this investigation have been clarified by an assessment of molecular mechanics-derived (MM3) steric energy calculations.

Stereoselective hydrocoupling of cinnamic acid esters by electroreduction: application to asymmetric synthesis of hydrodimers.

The electroreduction of Ar-substituted methyl cinnamates in acetonitrile gave all-trans cyclized hydrodimers stereoselectively (58 approximately 90% de). In all cases, small amounts (<10% yield) of meso hydrodimers were also formed. The electrolysis was performed conveniently using an undivided cell at a constant current. The transition states for the hydrocoupling were calculated with semiempirical methods. The all-trans cyclized hydrodimers were transformed to C(2)-symmetric dl-3,4-diaryladipic acids and trans-3,4-diarylcyclopentanones. The chiral auxiliary [(1R)-exo]-3-exo-(diphenylmethyl)borneol, prepared from (1R)-(+)-camphor, was highly effective for the stereoselective hydrocoupling of its cinnamates by electroreduction. From the resulting hydrodimers, (3R,4R)-3,4-diaryladipic acid esters and (3R,4R)-3,4-diarylhexane-1,6-diols were synthesized in 87-95% ee.

Selective synthesis of either enantiomer of alpha-amino acids by switching the regiochemistry of the tricyclic iminolactones prepared from a single chiral source.

Preparation of l-alpha-amino acids was easily accomplished simply by exchanging the position of the lactone group of our recently reported chiral template 1 from C2 to C3. The new chiral template 7 was prepared in 54% overall yield over five steps from (1R)-(+)-camphor. Alkylation of iminolactone 7 afforded the alpha-monosubstituted products in good yields and excellent diastereoselectivities (>98%). Hydrolysis of the alkylated iminolactones furnished the desired l-alpha-amino acids in good yields and ee with nearly quantitative recovery of chiral auxiliary 4.

Chiral Lewis acid-catalyzed asymmetric Baylis-Hillman reactions.

An effective chiral Lewis acid-catalyzed asymmetric Baylis-Hillman reaction is described. Good to high enantioselectivities were obtained using 3 mol % chiral catalyst. Novel camphor-derived dimerized ligands were prepared from the condensation of (+)-ketopinic acid with the corresponding diamines and hydrazine under acidic conditions. When alpha-naphthyl acrylate was used as a Michael acceptor, the reaction is complete within 20 min with high stereoselectivity and in reasonable chemical yields.

Diastereoselective electrochemical carboxylation of chiral alpha-bromocarboxylic acid derivatives: an easy access to unsymmetrical alkylmalonic ester derivatives.

The diastereoselective electrochemical carboxylation of chiral N-(2-bromoacyl)oxazolidin-2-ones has been studied. This reaction was carried out by cathodic reduction of the C-Br bond, in the presence of carbon dioxide, followed by treatment with diazomethane. The yields and the diastereomeric ratio of the two epimeric alkylmalonic acid derivatives are strongly affected by various factors: solvent-supporting electrolyte system, temperature, electrode material, electrolysis conditions, oxazolidinone moiety. The higher yields (88%) were obtained starting from N-(2-bromopropionyl)-4R-phenyloxazolidin-2-one 1a, but with poor diastereoselectivity (61:39). The two epimers were easily separated by flash chromatography. The best results were achieved using a different chiral auxiliary: Oppolzer's camphor sultam. Starting from 1j a good yield in carboxylated product was obtained (80%) with excellent diastereoselectivity (98:2). These chiral alkylmalonic acid derivatives are valuable building blocks in the synthesis of molecules with biological activity and of chiral propane-1,3-diols derivatives.

A practical total synthesis of hapalosin, a 12-membered cyclic depsipeptide with multidrug resistance-reversing activity, by employing improved segment coupling and macrolactonization.

A practical total synthesis of hapalosin, a compound with multidrug resistance-reversing activity, has been carried out using an unprecedented macrolactonization strategy. One of the features of the new approach is the straightforward and fully stereocontrolled access to the key gamma-amino beta-hydroxy carboxylic acid subunit via an efficient acetate aldol addition reaction with N-methyl alpha-aminoaldehydes, which relies on a camphor-derived chiral lithium acetate enolate reagent. The scope of this aldol reaction is investigated and its potential application to the synthesis of other structurally related, biologically relevant compounds illustrated. Remarkably, the chiral tether in the resulting gamma-amino aldol adducts sterically protect the carbonyl group, thus avoiding intramolecular cyclization during the amino group deprotection and the subsequent segment coupling event. After successful segment coupling and smooth, clean release of the chiral auxiliary, a new macrolactonization protocol, based on the principle of double activation of both reactive sites, is applied, which leads to the 12-membered macrolactone hapalosin in unprecedented chemical efficiency.

Synthesis of carbocyclic hydantocidins via regioselective and diastereoselective phosphine-catalyzed [3 + 2]-cycloadditions to 5-methylenehydantoins.

The phosphine-catalyzed [3 + 2]-cycloaddition of 5-methylenehydantoins 4 with the ylides 5, derived from addition of tributylphosphine to the 2-butynoic acid derivatives, 6a-d, gives spiro-heterocyclic products. The camphor sultam derivative 6b gives optically active products. Noteable was that the ylides derived from ethyl 2-butynoate and the 3-(2-butynoyl)-1,3-oxazolidin-2-one derivatives 6c and 6d gave spiro-heterocyclic products with reverse regioselectivities. The N,N-dibenzylprotected cycloadduct has been converted to carbocyclic hydantocidin and 6,7-diepi-carbocyclic hydantocidin.

Facile preparation and functionalization of chiral stabilized ylides from common chiral auxiliaries using triphenyl- phosphoranylideneketene (the Bestmann ylide) and their use in Wittig reactions.

Camphor-derived lactams and other related chiral controllers have been found to react with the Bestmann ylide (triphenylphosphoranylideneketene) upon heating in toluene. The resulting parent ylides provide convenient access to a structurally diverse set of chiral stabilized ylides via functionalization. The utility of these chiral ylides for Wittig reactions has been briefly investigated and the effects of alpha-substitution noted.
