The concept of limbal stem cells (LSC) opened new possibilities for the treatment of severe caustic damage of the anterior segment. These possibilities include transplantation of LSC, perforating keratoplasty, possibly transplantation of the amniotic membrane. In three patients with severe caustic damage of the anterior segment of the eye the authors implemented transplantation of LSC, perforating keratoplasty and in one patient also transplantation of the amniotic membrane. Where only one eye was damaged LSC for transplantation were obtained from the sound eye (autotransplantation, in case of damage of both eyes LSC were obtained in one case from a close relative in the second case from a cadaverous eye from a corneal bank (Allotransplantation). In the patient with autotransplantation of LSC, perforating keratoplasty and transplantation of the amniotic membrane the result was very good. The corneal disc remained clear 1.5 years after operation, the conjunctival sac was free from synbleropharons and vision improved from movement of the hand to 0.5. In the two patients with allotransplantation of LCS and perforating keratoplasty, LCS did not become incorporated despite immunosuppressive treatment and the corneal disc became turbid. Restored activity of LCS in severe caustic damage of the anterior segment opened new therapeutic opportunities.
Using 2 x 10(-5) mol/ml beta-NF (beta-naphthoflavone) to induce FL cells to express cytochrome P450 isozymes at a reasonable level, we have studied the mutagenesis of two procarcinogens, aflatoxin B1 and benzo (alpha)pyrene, at four different doses. Except for their top doses, all other doses induced mutation in target gene SupF carried by plasmid pWB1 at the frequency of 10(-4), which is higher than the spontaneous mutation frequency (7.61 x 10(-6)). Aflatoxin B1 demonstrated normal effect-dose relationship. Meanwhile no mutant was detected in their correspondent negative controls. All the results suggest that shuttle vector pWB1 can be conveniently used for detection of procarcinogens and study of their mutagenesis mechanisms in human cells.

OBJECTIVE: The aim of this experiment is to find proper cell carrier for skin tissue engineering. METHODS: Various concentration of fibroblasts were seeded onto HA-ECM and cultured in vitro. The performance of cells' growth, array, adhesion and collagen secretion on HA-ECM was observed with light microscope and transmission electron microscope. RESULTS: The fusiform fibroblasts oriented radiantly or longitudinally and closely packed onto the HA-ECM, they attached firmly and proliferated to confluence on the stromal surface of HA-ECM. CONCLUSION: The optimal cell concentration is 3.5 x 10(6)/ml, HA-ECM is ideal carrier for fibroblasts because of its excellent scaffold and diffusion characteristics. Futhermore, it has the bioactive molecules such as fibronectin and laminin which play an important role in fibroblasts attachment and proliferation on HA-ECM.

OBJECTIVES: The aim of the study was to compare the reliability of chorionicity determination in twin pregnancy using different ultrasonographic methods as compared with post-partum placental assessment. STUDY DESIGN: The study included 262 spontaneous twin pregnancies delivered at Kutno Hospital between 1989 and 1996. All twin gestations were diagnosed by sonography by the 28-th week's gestation. The ultrasonographic and pathologic correlation between the chorionic and amniotic type was assessed in this group. RESULTS: The ultrasonographic evaluation of the 262 spontaneous twin gestations demonstrated 181 (69.1%) dichorionic and 81 (30.9%) monochorionic twin pregnancies. Three of monochorionic twins were monoamniotic. In all 262 transvaginal and abdominal ultrasonography correctly predicted the chorionic and amniotic type as determined by the pathologic findings. CONCLUSIONS: Ultrasonography before the 28-th week can accurately determine the chorionic and amniotic type in twin pregnancies. The determination of chorionicity during the first half of pregnancy should be based on the various signs. In the second half of pregnancy, the obstetrician should rely on fetal sex in combination with the number of placentas, and characteristics of the dividing membrane.
The elemental ionic distribution in the epithelial layer (EL) and in connective tissue (CT = compact layer + fibroblast layer) of the human amniotic membrane has been studied in reference samples, after conservation in a physiological fluid (Hanks' solution) and after addition of 2 mM MgCl2 in Hanks' solution. Particle induced X-ray emission and Rutherford backscattering spectrometry techniques were used to provide quantitative measurements. In physiological fluid, with regard to reference samples, the monovalent ions (Na+, K +, Cl-) concentrations were identical on both layers. This data indicates that the connective tissue, in particular the compact layer, acts as a buffer which fix minerals. Mg2+ and Ca2+ levels were higher in EL than in CT. The addition of MgCl2 in Hanks' solution induced a decrease of the monovalent ion concentrations in both layers except Na+ level in EL which remained constant, an increase of the Mg2+ level in both layers, while the Ca2+ remained constant. These data indicate the possible role of connective tissue in pregnancies complicated by poly or oligohydramnios.
