Acid-base catalysis of chiral Pd complexes: development of novel catalytic asymmetric reactions and their application to synthesis of drug candidates.

Using the unique character of the chiral Pd complexes 1 and 2, highly efficient catalytic asymmetric reactions have been developed. In contrast to conventional Pd(0)-catalyzed reactions, these complexes function as an acid-base catalyst. Thus active methine and methylene compounds were activated to form chiral palladium enolates, which underwent enantioselective carbon-carbon bond-forming reactions such as Michael reaction and Mannich-type reaction with up to 99% ee. Interestingly, these palladium enolates acted cooperatively with a strong protic acid, formed concomitantly during the formation of the enolates to activate electrophiles, thereby promoting the C-C bond-forming reaction. This palladium enolate chemistry was also applicable to electrophilic enantioselective fluorination reactions, and various carbonyl compounds including beta-ketoesters, beta-ketophosphonates, tert-butoxycarbonyl lactone/lactams, cyanoesters, and oxindole derivatives could be fluorinated in a highly enantioselective manner (up to 99% ee). Using this method, the catalytic enantioselective synthesis of BMS-204352, a promising anti-stroke agent, was achieved. In addition, the direct enantioselective conjugate addition of aromatic and aliphatic amines to alpha,beta-unsaturated carbonyl compound was successfully demonstrated. In this reaction, combined use of the Pd complex 2 having basic character and the amine salt was the key to success, allowing controlled generation of the nucleophilic free amine. This aza-Michael reaction was successfully applied to asymmetric synthesis of the CETP inhibitor torcetrapib.

Evaluation of the adaptation interface of one-piece implant-supported superstructures obtained in Ni-Cr-Ti and Pd-Ag alloys.

Several formulations of alternative alloys have been proposed for the substitution of gold-based alloys used in Dentistry. Recently, a Ni-Cr-Ti-based alloy has been introduced. The purpose of this work was to verify the marginal adaptation obtained with one-piece superstructures for implant-supported prostheses obtained in Ni-Cr-Ti alloy, compared to a semi-noble alloy Pd-Ag. Eight superstructures for each alloy were produced over 4 implants in the anterior region of the mandible. The superstructures were placed in a torquemeter and the fixation screw of implant #1 was tightened with a 20 Ncm load with the others loosened (Sheffield test). The unfitness (in mm) was measured using a three-dimensional optical measurer in each implant, in the buccal and lingual aspects. The obtained data were submitted to statistical analysis by the analysis of variance and Tukey's test at 5% level. Significant differences were found for the factors material (p< or =0.05), with Ni-Cr-Ti pieces better than Pd-Ag ones, and implants (p< or =0.01). There were no significant differences for the factor position and interactions among factors (p>0.05). Based on the analysis of the data, it is possible to conclude that the Ni-Cr-Ti alloy makes possible the obtainment of one-piece implant-supported superstructures with a smaller misfit compared to the one obtained in Pd-Ag alloy, traditionally indicated for this situation. Additional tests may verify the superiority of the Ni-Cr-Ti alloy.

The use of periodic acid-thiocarbohydrazide-silver protein-physical development (PA-TCH-SP-PD) procedure for the histochemical detection of neutral carbohydrates in the circumventricular organs of the rat.

A periodic acid-thiocarbohydrazide-silver protein-physical development (PA-TCH-SP-PD) procedure was applied to the rat circumventricular organs (CVOs), which are known to be specialized structures in the brain. In the rat CVOs, vivid PA-TCH-SP-PD reactions were obtained in the apical surface and cytoplasm of the ependymal cells of the subcommissural organ (SCO) and the epithelial cells in the choroid plexuses (CPs) examined, and similar positive reactions were detected in the vascular walls and perivascular connective tissues in all the CVOs tested. Further, varying intensities of PA-TCH-SP-PD reactions were noted in the neuronal and glial networks of the organum vasculosum laminae terminalis (OVLT), subfornical organ (SFO) and area postrema (AP). The results obtained in the present study indicate that in the rat CVOs the histologic structures mentioned contain varying amounts of neutral carbohydrates and possible histophysiological significances of these carbohydrates in these organs have been discussed with references to their functions.

Implantation of Spheramine in advanced Parkinson's disease (PD).

Evaluation of the safety and efficacy of unilateral stereotactic implantation of cultured human retinal pigment epithelial (hRPE) cells attached to microcarriers (Spheramine) in patients with advanced PD in an open label pilot study. Six patients with advanced PD (3 males; 3 females; mean age 52.2 years; mean duration of PD 10.2 years; mean Hoehn and Yahr stage "off" 3.75) were assessed at baseline and post-operatively using the modified CAPIT. Each patient underwent MRI-guided stereotactic transplantation of 325,000 hRPE cells attached to microcarriers in 5 tracts, 5 mm apart in the post-commissural putamen contralateral to the most affected side. Immunosuppression was not used. The UPDRS Motor (UPDR-M) score in the practically defined "off" state was the primary outcome measure. At 6 months post-op, the mean UPDRS-M (off) score improved to 35 (34%) from a pre-op baseline mean of 52 (p <.001). Secondary outcome measures improved including the total UPDRS (33%), Timed Motor Tests (on, 14%; off, 23%), PDQ39 QOL (30%), and Schwab and England score (on, 11%; off, 30%). Bilateral improvements have been observed in motor symptoms, with the greatest effect seen contralateral to the implants. Three of six patients currently have lower Dyskinesia Rating Scale scores than at baseline, while the scores of the other three are unchanged from baseline values. No "off-state" dyskinesias have been observed. Thus Spheramine implantation therapy appears to be safe and well tolerated for 6 months post-implantation.

Participation of angiotensin receptors in acute hypoxia in mice. II. Effects of angiotensin II nonpeptide receptor ligands losartan (DuP-753) and PD-123319.

The effects of intracerebroventricularly (i.c.v.) administered angiotensin nonpeptide receptor ligands losartan (DuP-753) and PD-123319 and their interactions with Ang II were examined on acute anoxic hypoxia in male mice. Results showed that losartan and PD-123319 exerted antihypoxic effects including increased latency to convulsive seizures and survival time and pretreatment with both drugs enhanced Ang II-increased survival time. Taken together, the results suggest that the antihypoxic effect of losartan and PD-123319 is most likely due to action on brain AT1 and AT2 receptors with both drugs behaving as partial agonists.

ITDI is the preferred treatment for iron supplementation in rHuEpo-treated anemic PD patients.

In recombinant human erythropoietin (rHuEpo)-treated anemic peritoneal dialysis (PD) patients, oral iron therapy, despite its limited efficacy, remains an accepted and convenient method of iron supplementation. In this study, we compared infusion of total dose iron (ITDI) to oral iron for the treatment of anemia in PD patients. Thirteen patients with serum transferrin saturation (TSAT) < 25% received ITDI and 11 patients with TSAT between 25% to 35% received oral iron. Doses of rHuEpo were adjusted monthly to maintain target hematocrit at 35%. Both groups had similar baseline mean hematocrits (31.0 +/- 0.9% vs. 33.0 +/- 1.0%), and comparable mean baseline weekly rHuEpo dose (7886 +/- 1449 units/week vs. 6370 +/- 1553 units/week). At the end of the 6-month study, the ITDI group had a 16% improvement in mean hematocrit compared to baseline. The final mean dose of weekly rHuEpo was also reduced by 59% in the ITDI group. During the same period, compared to baseline, the oral group had a 4.8% fall in mean hematocrit despite 51% higher requirement of rHuEpo doses. There was no adverse reaction to intravenous iron. We conclude that ITDI is the preferred treatment for iron supplementation in rHuEpo-treated anemic PD patients.

Impact of Pd-mordenite pretreatment on the heterogeneity of Heck catalysis.

In Heck reactions with tributylamine as the base and in toluene, Pd(NH3)4(2+)-mordenite (0.4 wt% Pd) and Pd0-mordenite (0.4 and 4 wt% Pd) are not only active and selective, but also truly heterogeneous catalysts, while oxidized PdII species in an all-oxygen environment, i.e. ionic Pd2+ or PdO, are prone to leaching.

[Self-rating scales in schizophrenia: assessment of the practical applicability of the Paranoid-Depression Scale (PD-S), the Frankfurt Self-feeling Scale (FBS) and of two visual analogue scales].

AIM: To assess the practical feasibility of the Paranoid-Depressivity Scale (PD-S, Paranoid-Depresivitäts-Skala), the Frankfurt Self-feeling Scale (FBS, Frankfurter Befindlichkeitsskala), and two visual analogy scales of: sense of illness (WAC) and self-feeling (WAS). METHOD: 210 patients with schizophrenia of various clinical courses. All patients in the study group were required to complete each scale twice, at 48 hr intervals. For statistical analysis, two sets of data were singled out (1) the relationship between refusal/inability to complete the scale repeatedly and the selected clinical variables; (2) observations made by the doctor, while the patient was completing the scales. RESULTS: Statistical analysis revealed, that clinical factors like restlessness, autism, maladaptation, recurrence/remission and lower educational status imply a significantly lower readiness of the patient towards completion of self-rating scales. The visual analogy scales were those more readily--and easily--completed by the patients; their interpretation, however, is difficult. Given this, the authors of this paper decided to shorten the questionnaire scales, which reducing the whole to aspects of vital diagnostic relevance. Whether this operation would influence the scales' diagnostic value, required ex-post analysis, which was subsequently performed. Summary analysis revealed that the application of the shortening of the PD-S and FBS had no significant impact on these scales' reliability and validity indices; furthermore, it resulted in a significant decline in the number of ambiguities and thus improved the comprehensiveness of the questionnaire's structure. Also, a significant increase in concordance between the self-rating results and the clinicians' diagnoses was observed. CONCLUSIONS: Visual analogy scales are the more readily and easily applied in practice; a comprehensive interpretation is, however, virtually impossible. On the other hand, the PD-S and FBS are the more difficult for the patients to complete, due to their length, but their interpretation does not constitute a major problem. Ex-post analysis reveals, that shortening the complex questionnaires to their core aspects does not affect their psychometric value negatively; in fact--the contrary. The latter statement, however, requires verification in prospective studies.

[Determination of extraction separation of Pd(II), U(VI) and Mo(VI) in PEG-Na2SO4-PAR system using spectrophotometry].

In polyethylene glycol-2000(PEG)-sodium sulfate (Na2SO4)-4-(2-Pyridylazo) resorcinol (PAR) system the extraction separation behaviour of the complexes of Pd(II), U(VI), Mo(VI) and La(III) was investigated. The results indicated that the complexes of Pd(II) and U(VI) with PAR were almost completely extracted by the PEG phase, La(III) was partly extracted, while Mo(VI) was not extracted from the aqueous solution of pH 4.5-7.5. So Mo(VI) was quantitatively separated from the mixed solution of Mo(VI), Pd(II) and U(VI) ions. The extraction mechanism for PEG phase was studied preliminarily.

Specific inhibition of cyclin-dependent kinase 4/6 by PD 0332991 and associated antitumor activity in human tumor xenografts.

PD 0332991 is a highly specific inhibitor of cyclin-dependent kinase 4 (Cdk4) (IC50, 0.011 micromol/L) and Cdk6 (IC50, 0.016 micromol/L), having no activity against a panel of 36 additional protein kinases. It is a potent antiproliferative agent against retinoblastoma (Rb)-positive tumor cells in vitro, inducing an exclusive G1 arrest, with a concomitant reduction of phospho-Ser780/Ser795 on the Rb protein. Oral administration of PD 0332991 to mice bearing the Colo-205 human colon carcinoma produces marked tumor regression. Therapeutic doses of PD 0332991 cause elimination of phospho-Rb and the proliferative marker Ki-67 in tumor tissue and down-regulation of genes under the transcriptional control of E2F. The results indicate that inhibition of Cdk4/6 alone is sufficient to cause tumor regression and a net reduction in tumor burden in some tumors.

[Soluble PD-1 can augment anti-tumor immunity induced by HSP70-peptide complex in tumor-bearing mice].

AIM: To explore the effect of soluble PD-1 (sPD-1) on the anti-tumor immunity induced by HSP70-peptide. METHODS: BALB/c mice were immunized with HSP70-peptide, then the cytotoxic activity of splenocytes was detected by MTT colorimetry. Expression of PD-1 and its ligand on splenocytes was determined by semi-quantitative RT-PCR. For some mice, sPD-1 plasmid was injected alone or in combination with HSP70-peptide and then the tumor weight and cytotoxic activity of splenocytes were measured. RESULTS: sPD-1 alone had anti-tumor effect. HSP70-peptide complex not only induced tumor-specific cytotoxic T lymphocytes(CTLs), but also promoted expression of PD-1 and its ligand. The combination of sPD-1 with HSP70-peptide complex induced much more stronger anti-tumor effect as compared with sPD-1 or HSP70-peptide alone. sPD-1 transfection in mice could enhance the cytotoxic activity of splenocytes from immunized mice. CONCLUSION: sPD-1 enhanced the anti-tumor efficacy of HSP70-peptide.

Influence of the therapy with pergolide mesylate plus L-DOPA and with L-DOPA alone on serum cGMP level in PD patients.

The aim of our study was to investigate serum cGMP level in patients treated with L-DOPA alone and L-DOPA with pergolide mesylate (PM) and to compare the obtained values with age-matched healthy subjects. PD patients treated with L-DOPA or with PM + L-DOPA had significantly higher cGMP levels in serum as compared to control. Moreover, cGMP level was significantly higher after treatment with PM + L-DOPA then L-DOPA alone. We did not observed significant differences in UPDRS scores between both PD groups. Our data suggest that changes in serum cGMP level in PD patients are related with PM + L-DOPA or L-DOPA alone therapy.

Antimicrobial-resistant gram-positive bacteria in PD peritonitis and the newer antibiotics used to treat them.

The incidence of resistant gram-positive bacteria in nosocomial and, more recently, community-acquired infections is increasing. Staphylococci, because of their natural habitat on the skin, have always been the leading cause of peritonitis in patients receiving peritoneal dialysis (PD). These organisms have demonstrated a remarkable ability to develop resistance to antibiotics, first with penicillin, then antistaphylococcal penicillins (methicillin-resistant Staphylococcus aureus), and more recently, strains expressing resistance to vancomycin (vancomycin-intermediate and vancomycin-resistant S. aureus) have emerged. Enterococci are normal inhabitants of the gastrointestinal tract and occasionally cause PD peritonitis. In the past 15 years, vancomycin-resistant enterococci have emerged as significant pathogens in many areas. In the past 5 years, novel antibiotics that have activity on gram-positive bacteria, including vancomycin-resistant strains, have become available. The problem of resistant gram-positive bacteria in PD peritonitis, their therapy, and the role of these newer agents, quinupristin/dalfopristin, linezolid, and daptomycin, are reviewed.

Effect of alpha thalassaemia, G-6-PD deficiency and Hb F on the nature of sickle cell anaemia in south-western Saudi Arabia.

This study was conducted on sickle cell anaemia (SCA) patients from the south-western province (SWP) of Saudi Arabia to determine the effect of thalassaemias, glucose-6-phosphate dehydrogenase (G-6-PD) deficiency and Hb F level on the clinical presentation of sickle cell anaemia. The results showed that associated alpha-thalassaemia improved the haematological parameter values while associated G-6-PD deficiency and high Hb F level did not play a significant role in amelioration of the disease in these patients. Hb S/beta(0) -thalassaemia cases showed a severe anaemia similar to the SCA patients without alpha-thalassaemia. However, considerable improvement of the haematological parameters were found in patients with S/beta(0)-thalassaemia and associated alpha-thalassaemia. This paper reveals that alpha-thalassaemia may partially ameliorate the clinical manifestations of SCA in Saudi patients from the SWP, while high Hb F level and G-6-PD deficiency do not modify SCA to any statistically significant extent.

Prevalence of G-6-PD deficiency in a malaria endemic tribal area of Orissa (India).

A total of 207 men belonging to Bhuyan, Juanga and Munda tribes and 108 non-tribal residents of Banspal block area of Keonjhar district of Orissa were screened for glucose-6-phosphate dehydrogenase (G-6-PD) deficiency, abnormal haemoglobin and malaria parasite. In the tribal group, G-6-PD enzyme deficiency was observed in 13.52 per cent as compared to 3.70 per cent in non-tribals. No abnormal haemoglobin was detected in the studied population, while 15.87 per cent of individuals were positive for Plasmodium falciparum. An extremely weak association was observed between malaria and G-6-PD deficiency.

[Double-blind controlled comparison of placebo and paracetamol in patients with G-6-PD deficiency].

In this study are reported 17 patients with G-6-PD deficiency. These subjects have been studied with double-blind trial between placebo and Paracetamol, looking for the possible hemolysis induced from drug in these children with G-6-PD deficiency. Hemolysis had been valued by various hematological parameters (Hb, erythrocyte count, reticulocytosis, bilirubinemia, haptoglobin, hemopexin, and survival of erythrocytes marked with Cr). The results (8 cases with placebo and 9 cases with paracetamol) demonstrate that paracetamol have not induced hemolysis in these G-6-PD deficient subjects.

In vitro study on the corrosion behavior of three commercial Ag-Pd-Cu-Au alloys in Ringer's and 0.1%Na2S solutions.

The corrosion resistance of three commercial Ag-Pd-Cu-Au alloys was estimated in Ringer's and 0.1% Na2S solutions by electrochemical techniques and surface analyses. In Ringer's solution, the three alloys showed high corrosion resistance and there was no significant difference in the anodic polarization characteristics of the three alloys. In the 0.1% Na2S solution, the Alloy A which had the lowest noble metal content (Au + Pd) exhibited the highest anodic reactivity with the largest amount of corrosion product on the alloy surface. It was determined that the Ag-rich phase of Ag-Pd-Cu-Au alloy was preferentially attacked to form Ag2S corrosion product. The polarization resistance data showed that the corrosion rate for Alloy A in 0.1% Na2S solution was determined to be 500 times higher than that in Ringer's solution. The corrosion rate of the alloy in the freely corroded condition can be estimated quantitatively and precisely by measuring the polarization resistance.
