[The role of biogenic amines in the mechanism of uterine hemorrhage in uterine myoma].

Function of the adrenosympathetic system has been assessed in 103 patients with uterine myoma. Levels of biogenic amines (adrenaline, noradrenaline, histamin) were determined in uterine tissues using spectrofluorometry. It was found out that storage of vasoactive mediators in the uterine tissue represents a mechanism of uterine bleedings in patients with uterine myoma. Another determinant is stress experienced by patients with menometrorrhagia.

[Granular biogenic amine-containing cells of the rat endometrium as components of the system of mononuclear phagocytes].

To 10 non-inbred white mature rats 24 h before sacrifice 7.0 ml of Indian ink colloid solution has been injected intraperitoneally. Histochemical reactions of Falk-Hillarp (catecholamines, serotonin) have been performed on nonfixed cryostat slices of the uterus, those of Cross, Even, Rost (histamine) against non-specific esterase and acid phosphatase. Before carrying out these reactions, the slices have been examined under luminescent microscope LUMAM-I3 by means of luminescent and usual illumination in order to reveal cells, possessing autoluminiscence and containing phagocyted Indian ink particles. Presence of autoluminescence, phagocyted material, catecholamines and serotonin, histamine, nonspecific esterase, acid phosphotase, prostaglandin E2 in the same granular cells of the rat endometrium have been stated. All cellular properties revealed are specific for macrophages. A conclusion is made that granular biogenic amine-containing cells of endometrium can be considered as belonging to the system of mononuclear phagocytes.

[Blastomogenic activity of biogenic methoxyindoles].

A considerable blastogenic activity of biogenic methoxyindoles--melatonin, 5-methoxytryptamine (5-MOT) and their common metabolite--5-methoxyindolyl-3-acetic acid (5-MIAA) was established by prolonged s.c. injection in C57BL/6 mice. However, the blastogenic effect of 5-MOT decreased by 26% when its further metabolism to 5-MIAA was blocked for some time and the synthesis of its carcinogenic metabolite was inhibited. These results showed that the blastogenic effect of 5-MOT is not direct; and it is mediated by its transformation to 5-MIAA in the body.

[Effect of antidepressants on the balance of biogenic amines and their metabolites in the rat brain].

A study was made of changes in the balance of neuromediator monoamines and metabolistes in rat brain, induced by a single administration of pyrazidol, imipramine and norzimelidine. It was revealed that pyrazidal in a dose of 50 mg/kg increased the level of noradrenaline (NA) and serotonin (5-HT), particularly in the hypothalamus and raphe-nucleus of rat brain. In the same brain regions, imipramine (30 mg/kg) raised the NA content and lowered the concentration of 5-HT and dopamine (DA) metabolites. Norzimelidine (60 mg/kg) elevated the levels of NA and DA in the hypothalamus and reduced the 5-HT content in the raphe nucleus of rat brain. It is suggested that the topography of changes in the balance of biogenic amines in the brain induced by different antidepressants reflects the pharmacological features of their action.

[Mechanism of mobilization of biogenic amines from rat mast cells in radiation protected conditions].

A study was made of the role of calcium ions and some other factors in secretion of biogenic amines from rat mast cells. The data obtained indicate that the radioprotective preparations of indolyl alkylamine and imidazole series can mobilize endogenous radioprotectors. The process of the mediator release from mast cells under the effect of sulfur-containing radioprotective agents is indirect. The molecular mechanisms of mast cell exocytosis are discussed.

Metabolism of biogenic aldehydes in human blood: effects of ethanol, acetaldehyde and disulfiram.

The metabolism of biogenic aldehydes was measured in different fractions of human blood. Isolated erythrocytes, leukocytes, platelets and plasma were incubated with the aldehydes derived from dopamine and serotonin (DOPAL and 5-HIAL, respectively). The disappearance of the aldehydes and the formation of the acid and alcohol metabolites were analysed using HPLC with electrochemical detection. When erythrocytes were incubated with 1 microM DOPAL or 5-HIAL only the acid metabolites were formed, whereas both acid and alcohol metabolites were formed in incubations with leukocytes or platelets. No metabolites were formed when the aldehydes were incubated with plasma. In incubations with erythrocytes, the oxidation of the aldehydes was totally inhibited in the presence of 50 microM disulfiram, whereas the formation of both acid and alcohol metabolites was inhibited in incubations with platelets. Disulfiram did not affect the metabolism of DOPAL or 5-HIAL in incubations with leukocytes. The oxidation of the aldehydes was inhibited by 50-70% in the presence of a 500-fold excess of acetaldehyde. Ethanol at a concentration of 10 mM did not affect the metabolism.

[Biogenic amines and the nervous system of Opisthorchis felineus].

Dopamine (0.08-0.25 micrograms/g of the tissue raw weight) and serotonin (0.13-0.31 micrograms/g of the tissue) were determined by spectrofluorimetry in the Opisthorchis tissue homogenate. Histochemical analysis (with glyoxylic acid) demonstrated distribution of biogenous amines in Trematode's nervous system. Fluorescence characteristic of catecholamines was found in 3 pairs of symmetrically situated cells in the anterior part of the worm and in a small number of relatively small assymmetric neurons in its posterior part. Similar fluorescence was observed in cephalic ganglia commissure, anterior and posterior nervous trunks, commissures and multiple fine fibrae. Flourescence characteristic of indolamine was seen in 5 pairs of symmetric neurons situated in the anterior part of the worm. Processes of these cells were observed from a close distance.

[Experimental therapy of thermal burns with biogenic peptide-containing preparations].

The authors studied the therapeutic activity of biogenic peptide-containing preparations derived from microbial cell membrane fractions, solcoseryl, and plasma alcohol extract (PAE) in experimental thermal trauma. Treatment of the burnt animals with solcoseryl and PAE reduced mortality to 40-42%, accelerated the healing of the burn wound, restored the behavior status, and increased the intensity of oxygen consumption. This is evidence of the promising character of further studies of the biological activity and therapeutic properties of these preparations.

[Equipment to find odour compounds, produced by bacteria in drinking water. I. Comparison of a bacteria produced odour compound with a chemical reference substance and a simple method for getting and concentrating biogenic odour compounds (author's transl)].

An increase in using surface water and bank filtration processes as raw water for drinking water production sometimes gave rise to problems of bad taste and odour. Some of these odourous compounds may be produced by bacteria, especially Streptomyces species. They are able to pass active carbon filters and therefore are a problem as well in raw water as in treated water. Biogenic odourous compounds often are soluble in water, that means they are present without being fixed to the bacteria cell. Human nose is able to percieve even dilutions of about 10(-9). If a bacterial produced substance is compared with an equal smelling chemical substance such as Isoborneol, also diltuions of 10(-9) may be smelt. Gas-chromatography only detects concentrations up to 10(-5), so the nose is much more efficient.

The effect of beta blockade on the reaction of pressure and mesenteric flow to biogenic amines in normal and X-irradiated rats.

A beta-blocker, propranolol, was infused to control and irradiated rats the 1st and 3rd day after 2-kR X-ray irradiation. Blood pressure and mesenteric blood flow were recorded during infusion of biogenic amines (noradrenaline, dopamine, serotonin, acetylcholine and histamine). After beta-blockade, there was intensified increase in pressure and decrease in flow with increased overshoot after infusion of noradrenaline, no effect on the increase in pressure and flow after dopamine, but an enhanced increase in pressure and decrease in flow after serotonin. No effect on the decrease in pressure and flow was seen after acetylcholine, and a greater decrease in pressure and flow, after histamine. Irradiation altered the vascular reactivity, mainly for noradrenaline, serotonin and histamine.

[Biogenic amine metabolism in homeotherms exposed to pesticides of different chemical nature].

Some aspects of the biogenic amines -- serotonin and histamine metabolism in rats and guinea pigs following introduction to them of pesticides of different chemical nature, viz. gamma-isomer of hexachlorcyclohexane (lindane) and dimethyldithiocarbamate zinc (zyram) were studied. These pesticides were found to dissimilarly affect the passage with urine of the principal serotonin metabolite -- 5-oxyindol-acetic acid. A short-term (2 days) introduction to rats of large doses (34 mg/kg) of lindane and a protracted (90 days) action of low doses (1.7 mg/kg) produce changes in the content of serotonin and of 5-oxyindol-acetic acid in the brain, liver and kidneys. In the blood of guinea pigs receiving for a lenthy time (120 days) small amounts (4 mg/kg) of zyram shifts in the histamine-histaminase-histaminopexy system were revealed, this pointing to the ability of this pesticide to provoke the state of sensitization.

Role of some biogenic amines in the pathomechanism of glomerulonephritis. II. Adrenaline, noradrenaline and serotonin in acute serum sickness in rabbits.

In acute serum sickness induced with one intravenous dose of bovine serum albumin (BSA) in 40 rabbits the patterns of excretion of adrenaline (A), noradrenaline (NA), serotonin (5HT) and 5-hydroxyindole-acetic acid (5HIAA) were studied in 24-hour urine, and the 5HT level was determined at intervals of three days. The urinary levels of biogenic amines were determined daily. Some rabbits immunized with BSA received also additionally 5HT, reserpine or parachlorphenylalanine (PCPA). Administration of BSA to rabbits caused a significant increase in the excretion of A and NA and a less evident increase in 5HT level in the blood. The greatest correlation with the course of the immune reaction was shown by the increase in NA excretion observed on the 2nd and 3rd days after BSA administration, and then between the 5th and the 12th days of the experiment. Daily subcutaneous injections of 5HT during 15 days caused a significant rise of its level in the blood and urine, and an increase of 5HIAA excretion. After reserpine or PCPA administration a significant decrease of the levels of all these amines was observed. Taking into account the results of histological examination of the kidneys, that is intensification of the inflammatory changes after 5HT administration and evident inhibition of the inflammatory process after administration of reserpine and PCPA it must be accepted that the studied amines have an important role in the pathomechanism of glomerulonephritis in acute serum sickness.

[Metabolism of various biogenic amines in acute viral neuroinfections].

The metabolism of biogenic amines was examined in 62 patients with various acute viral neuroinfections. The control group consisted of 57 persons. Depending on the process character and disease period variations of the levels of serotonin, 5-hydroxyindolylacetic acid, coeruloplasmin and histamine were discovered. A comparison of the results obtained with the clinical course of the diseases revealed a certain correlation, especially in patients with acute meningoencephalitis.

[Effect of the nonspecific biogenic stimulators pentoxyl and mumie on metabolic processes].

Unspecific biogenic stimulants (pentoxyl and mummie) accelerated metabolism of nucleic acids and protein in rat liver tissue. After the treatment with the stimulants the rate of lipolysis exceeded that of lipogenesis. Increase in content of lactate was similar if glycogen and glucose-6-phosphate were used as substrates of glycolysis, but it was stimulated 2-3-fold, when glucose was used; the phenomenon appears to be due to activation of hexokinase. As shown by polarographic measurements mitochondrial respiration was increased in all the metabolic states, but increased doses caused an inhibition of phosphorylation apparently due to functional overstrain of mitochondria. Increased doses of the stimulants accelerated also some other metabolic processes studied, but the effects were not dose-dependent. Pentoxyl and mummie apparently increased processes of protein and nuclei acid metabolism and stimulated the energy-providing reactions.

Pulmonary extraction of biogenic amines during septic shock.

The effect of live Escherichia coli on the pulmonary extraction of the biogenic amines 14C 5-hydroxytryptamine, (5-HT) and 3H-epinephrine was investigated. The labeled isotopes were injected into a central venous catheter and collected from an aortic catheter. One hundred per cent of the labeled epinephrine was recovered in the control and septic state. Only 32.8 +/- 3.6% SEM of the 5-hydroxytryptamine was recovered before sepsis and 42.5 +/- 4.9% SEM after sepsis. During sepsis, mean arterial pressure fell to 58 mm Hg from 121 mm Hg. Pulmonary shunt increased from .7 +/- .05 SEM to .33 +/- .09 SEM.

[Role of biogenic amines in the pathogenesis of intellectual disorders in children with minimal psychoorganic syndromes].

Memory, attention and emotional reactivity in comparison to indices of catecholamine and serotonin metabolism were studied in 25 children from 7 to 10 years with the minimal psychoorganic syndrome during aminalon treatment. According to the excretion indices the activity equilibrium of catecholamines and serotonin were changed and were accompanied by disturbances of memory, attention and emotional reactivity. An improvement of these indices during treatment corresponded to a normalization of the activity in these systems. Questions concerning the signficance of disturbed metabolism of biogenic amines in the pathogenesis of the psychoorganic syndrome are discussed.

[Alteration of biogenic amines, serotonin, histamine and polyamines, in cases of diarrhea induced by various cathartics (author's transl)].

Effects of various cathartics on charcoal transport, permeability of blood vessels and biogenic amines (serotonin (5-HT), histamine (His), polyamines) in the small intestine of mice were investigated. Diarrhea was induced in mice by oral administration of magnesium sulfate, mannitol, dioctyl sodium sulfosuccinate (DSS), castor oil and pilocarpine. In diarrhea following ingestion of magnesium sulfate and mannitol, vascular permeability in the intestinal membrane accompanied release of His from the small intestine, but such was not so when diarrhea was induced by DSS and castor oil. In diarrhea induced by various cathartics, 5-HT and His were apparently not related to intestinal motility. Thus, alteration in polyamine metabolism in the small intestine may exert an influence on transport of substances in cases of diarrhea in mice.
