[Proteomic analysis of cellular responses to low concentration of N-methyl-N'-nitro-N-nitrosoguanidine in human amnion FL cells].

OBJECTIVE: To investigate the protein profile after treatment of low concentration of N- methyl-N'-nitro-N-nitrosoguanidine (MNNG) in human FL cells. METHODS: After MNNG treatment, whole cellular proteins were separated using two-dimensional gel electrophoresis and visualized by silver staining; the digitized images were analyzed with 2D analysis software. The differentially expressed protein spots were identified by matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS). RESULT: More than 60 proteins showed significant changes in MNNG-treated cells compared to control cells (DMSO treatment). There were 18 protein spots detected only after MNNG treatment, while 13 protein spots were detected only in the control cells. Moreover, the levels of another 31 proteins were either increased or decreased in MNNG-treated FL cells. And some of the proteins were identified by MALDI-TOF-MS. CONCLUSION: There are significant alterations of protein profile after MNNG attack.

Amnion autografts, permanent structure.

Autografts of amniotic membrane were used to form tubes or conduits in lambs. These conduits survived for periods up to 9 months. The possibility of a clinical application has been explored.

[Pathogenetic characteristics of late spontaneous miscarriage in ascending infection of amnion: reaction of placenta].

Results of pathomorphological examination of placenta in 262 cases of late spontaneous miscarriages produced by ascending infection of fetal liquids with opportunistic microflora are presented. Sequential involvement in the exudative inflammatory process of the fetal membranes was observed (placenta and umbilical cord) depending on the term of pregnancy and duration of placenta contact with the infected fetal liquids. Exudative funiculitis was observed not infrequently this indicating direct involvement of the fetus in the intrauterine infectious process.

[The effect of diphtheria toxin on proliferation of a transplantable culture of human amnion cells].

Cytoxic effect (CTE) of diphtheria toxin (DT) is preceded by disturbances of proliferation process, this being expressed in reduction of the proliferation index (PI) and of mitotic index (MI) before the appearance of the CTE. The effect of the toxin on the proliferation and mitoses of the cell culture is expressed after the incubation period of about 3 hours. The extent and the rate of reduction of the indices directly depended on the dose of the toxin whose low doses at the early periods stimulated the proliferation. The maximum of all the manifestations of the toxin action coincided with the period of increased proliferation in control. On the basis of investigations carried out a hypothesis was put forward that the CTE of diphtheria toxin developed with the entrance of the cells into mitosis.
