Preparation of hexaaza and heptaaza macrocycles functionalized with a single aminoalkyl pendant arm.

A practical and reproducible route for the preparation of 1,4,7,10,13,16,19-heptaazacyclohenicosane (1), 1,4,7,10,13,16-hexaazacyclooctadecane (2), and 1,4,7,10,13,17-hexaazacycloicosane (3) bearing a single N-(2-aminoethyl) pendant arm has been developed. Richman-Atkins cyclization in the presence of caesium carbonate was applied to construct the macrocycle from 3-benzoyl-N1,N5-ditosyl-3-azapentane-1.5-diamine and the appropriate fully N-tosylated N alpha, N omega-bis(2-mesyloxyethyl) tri- or tetra-amine. The benzoyl group was selectively removed with potassium tert-butoxide, and the exposed nitrogen atom was reacted with N-tosylaziridine. The tosyl protections were removed with hydrogen bromide in acetic acid, and the product was converted to a free base with the aid of a strong anion exchange resin (OH- form).

Donor-substituted peralkynylated "radiaannulenes": novel all-carbon macrocycles with an intense intramolecular charge-transfer.

A novel class of planar, highly conjugated all-carbon macrocycles, which we christened "radiaannulenes", have been prepared based on acetylenic scaffolding using tetraethynylethene (TEE) building blocks; these structures are powerful electron acceptors and, upon peripheral substitution with electron-donating N,N-dialkylanilino groups, display intense intramolecular charge-transfer.

Novel chiral N4S2- and N6S3-donor macrocyclic ligands: synthesis, protonation constants, metal-ion binding and asymmetric catalysis in the Henry reaction.

New hydrophobic chiral macrocyclic ligands L1-L3 with chiral diamino and thiophene moieties have been synthesized by the Schiff base condensation approach. Protonation constants of L1 and L2 were determined by potentiometry titration. Metal-ion binding experiments exhibited that L1 and L3 are pronounced in selective recognition, Ag+, Cu2+ and Ca2+ ions among the surveyed metal ions (Cu2+, Co2+, Ni2+, Zn2+, Cd2+, Pb2+, Ag+, Li+, Na+, K+, and Ca2+). L1 was found to spectroscopically detect the presence of Cu2+ and Ca2+ to function as a multiple readout sensor. The detection limit for Ca2+ ions was found to be 9.8 x 10(-5) M in CH2Cl2-MeOH solution. The trimeric chiral ligand L3 has been shown to be an efficient auxiliary in a Zn(II)-mediated enantioselective Henry reaction.

Selective recognition of thymidylylthymidine (TpT) and antitumor effects of a macrocyclic dizinc(II) complex.

A polyamino dizinc(II) complex, [(N-bisdien)Zn2(II)Cl2](ClO4)2, (LZn), has been synthesized as a new nucleobase receptor molecule in aqueous solution at physiological pH, and shows to be highly selective in recognizing deoxythymidine (dT) and thymidylylthymidine (TpT). The strong acidic Zn(II) ions in LZn at the fifth coordination sites interact with a variety of nucleosides. The binding and recognition processes have been studied by potentiometric titration. The X-ray crystal analysis of LZn shows that the two zinc ions are out of the basal plane of the macrocycle, favoring the effective recognition of TpT on the single strand of DNA. In vitro antitumor investigation shows that LZn is a patent inhibitor of tumor cell growth with IC50 values below 10 micromolar.

Rigid optically-active D2 and D3 macrocycles.

The synthesis and characterization of novel optically-active macrocycles, obtained by esterification reaction from a binaphthyl-containing diol and phthalic or terephthalic acids, and possessing overall D2 or D3 symmetry, is described.

Steric control in the oligomerisation of phosphazane dimers; towards new phosphorus-nitrogen macrocycles.

The reaction of [ClP(mu-NtBu)]2 (1) with H2O (1 : 2 equivalents) in the presence of excess Et3N gives the new chain compound [(mu-O)[P(mu-NtBu)2P(H)=O]2] (3), consisting of two P2N2 rings linked by a mu-O atom and terminating in P(V)(H)=O groups. A similar chain species is obtained from the reaction of the lithiate of [(tBuNH)P(mu-NtBu)2P(H)=O] (5) with [ClP(mu-NtBu)2P(NHtBu)] (2), the product being [(mu-O)[P(mu-NtBu)2P(NHtBu)]2] (6). Compounds 3 and 6 are the first examples of O-bridged chain phosphazanes and potential precursors to new phosphorus-nitrogen macrocycles. The syntheses and X-ray structures of 3, 5 and 6 are reported.

Synthesis, structure and electrochemistry of ferrocene-peptide macrocycles.

Redox active cyclopeptides Fc[CSA]2 (5), Fc[Gly-CSA]2 (6), Fc[Ala-CSA]2 (7), Fc[Val-CSA](2) and Fc[Leu-CSA]2 (9) (CSA = cysteamine) which are formed by the reaction of ferrocenedicarboxylic acid with peptide cystamines at high dilutions. These systems exhibit H-bonding involving the amide NH in solution as shown by their temperature dependent NMR spectra. With the exception of 5, the ferrocene macrocycles display intramolecular N...O cross-ring H-bonding in the solid state involving the amino acids proximal to the ferrocene.

Synthesis and characterisation of macrocyclic palladium(II)-sodium(I) complexes: generation of an unusual metal-mediated electron delocalisation.

Reaction of sodium perchlorate-crown ether derivative (LH2) complex [Na2LH2](ClO4)2 (1) with palladium acetate afforded two related compounds of macrocyclic palladium(II)-sodium(I) dimeric tetranuclear complexes, [Pd2Na2L2(mu-OH2)2](ClO4)2(CH2Cl2)3 (2) and [Pd2Na2(L-)2](CH3CN)2(C3H6O)2 (3) and their structures were characterised by IR, NMR, mass and X-ray analysis; the latter was revealed as an unusual metal-mediated electron delocalised complex.

Synthesis and study of a new diamidodipyrromethane macrocycle. An anion receptor with a high sulfate-to-nitrate binding selectivity.

A new 2,6-diamidopyridinedipyrromethane hybrid macrocycle () has been synthesized that shows a high selectivity for dihydrogen phosphate and hydrogen sulfate relative to nitrate in acetonitrile solution as judged from UV-vis spectrophotometric titrations; this leads to the suggestion that this or related systems might find use in nuclear waste remediation applications requiring the selective removal of hydrogensulfate from nitrate-rich waste mixtures.
