Temporal and spatial distribution of interleukin-1 beta in balloon injured porcine coronary arteries.

OBJECTIVE: Interleukin-1 beta (IL-1 beta) is a proinflammatory cytokine with a wide range of biological activities. We determined the distribution of IL-1 beta following percutaneous transluminal coronary angioplasty (PTCA) of porcine arteries, and the presence of caspase-1 (required for the activation of IL-1 beta). METHODS: Oversized balloon angioplasty was performed in Yorkshire White pigs and the vessels excised at selected intervals (1, 6, 18 h, 3, 7, and 14 days) post-PTCA. IL-1 beta and caspase-1 were then identified using reverse transcription polymerase chain reaction (RT-PCR), in situ RT-PCR, and immunohistochemistry. RESULTS: IL-1 beta protein was detected in the inflammatory infiltrate up to 3 days after PTCA. Luminal endothelial cells contained IL-1 beta at 1 h, with peak expression at 3-7 days. Adventitial capillaries were IL-1 beta-positive at all timepoints. IL-1 beta was detected in adventitial cells at 3 days, with reduced levels at 7 and 14 days. At 7 days, neointimal cells were also IL-1 beta positive. No IL-1 beta was detected in non-PTCA control arteries. RT-PCR demonstrated IL-1 beta mRNA expression to be induced at 1 h, and absent at 3 days. In situ RT-PCR revealed this expression to be distributed throughout the arterial layers at 6 h, but localized to the adventitia at 18 h, with a baseline expression in the adventitial layer of non-PTCA controls. Caspase-1 was detected in luminal endothelial cells from 6 h, in adventitial cells from 3 days, and in neointimal cells from 7 days post-PTCA. This expression colocalized with IL-1 beta, indicating the potential for the IL-1 beta present to become activated. CONCLUSIONS: We conclude that IL-1 beta is synthesised, in conjunction with caspase-1, by endothelial, inflammatory, and adventitial cells early (within 3 days) after PTCA, with decreased levels at later timepoints, suggesting that it has a key role to play in the early stages of healing following PTCA.

Centrifugation does not alter spatial distribution of 'BEP4' mRNA in paracentrotus lividus EGG.

Paracentrotus lividus unfertilized eggs were centrifuged in a sucrose gradient, so to split each into two parts: a nucleated light fragment and an anucleated heavy fragment. Northern blot analyses utilizing a bep4 probe as animal marker and H2A histone gene and 12S-mit RNA as controls indicate that the eggs are elongated along the animal-vegetal axis during centrifugation and thereafter split into an animal and a vegetal half. Treatment of the eggs with colchicine before centrifugation abolishes the animal localization of bep4 mRNA.

Diagnostic implications of the spatial distribution of colonic mass lesions (polyps and cancers). A prospective colonoscopic study.

A prospective colonoscopic study of 642 consecutive patients demonstrated that 156 (66%) mass lesions including 131 polyps and 25 cancers were located within 60 cm of the anus and 82 (34%) mass lesions including 68 polyps and 14 cancers were located beyond 60 cm, the working length of the rigid sigmoidoscope. One hundred twenty-one (61%) polyps and 14 (36%) cancers were located above 25 cm but less than 60 cm from the anus, the working length of the flexible sigmoidoscope. Sixty-eight (34%) polyps and 14 (36%) cancers were located beyond the reach of the flexible sigmoidoscope. Although the flexible sigmoidoscope detects more lesions than the rigid sigmoidoscope, 50% of colon cancers located beyond detection by the rigid sigmoidoscope would not have been detected with the flexible sigmoidoscope. Of all mass lesions located beyond 25 cm, 38% would not have been detected with the use of the flexible sigmoidoscope. It appears that total colonoscopy rather than flexible sigmoidoscopy must play an increasingly prominent role in the evaluation of patients with suspected colonic mass lesions.

Diminutive polyps: histopathology, spatial distribution, and clinical significance.

This retrospective study of 329 diminutive colon polyps revealed that 49.2% of the polyps were neoplastic, whereas 49.9% were nonneoplastic. 36.8% of the polyps were metaplastic, and 0.9% had mixed metaplastic and adenomatous components. The metaplastic polyps had a predominance in the distal portion of the large bowel, with 76.9% being located distal from the splenic flexure and 52.9% being located in the rectosigmoid area. Neoplastic polyps were distributed throughout the colon, with 65.4% located from the splenic flexure distally and 34.5% located proximal to the splenic flexure. These findings suggest that a much larger proportion of diminutive polyps are neoplastic and therefore at risk to progress to carcinoma than previously considered.

Spatial distribution of hip capsule structural and material properties.

Contemporary computational models potentially allow the practical incorporation of the effects of a joint capsule on both motion and the loads transmitted to the other parts of the joint. However, the required material properties have not been available for this purpose. To determine these properties we took both hip joints from five fresh-frozen, nondiseased cadavers. Following dissection and potting of the hemi-pelvis, distraction of the intact joint was conducted to measure the structural tangent stiffness of the joint capsule. Anatomical insertion points of the hip capsule were then recorded, and a complete capsulectomy was performed. Once excised, the capsule was sectioned into eight, approximately even sectors, and initial geometrical measurements were recorded for material property calculations. Material properties (i.e., structural tangent stiffness, failure load, ultimate strength, tangent modulus) were calculated using the load-displacement and geometric data collected for each of the sectors. This specimen-to-specimen thickness variability reveals significantly lower (p<0.01) average tangent structural stiffness values in the posterior-inferior portion of the capsule. Explorations of hip stability using numerical models can now be enhanced by incorporation of these experimental capsule data.

Spatial distribution of calcareous dinoflagellate cysts in surface sediments of the Atlantic Ocean between 13 degrees N and 36 degrees S.

To enhance the limited information available about the palaeo-ecological significance of calcareous dinoflagellates, we have studied their lateral distribution in surface sediments of the equatorial and south Atlantic between 13 degrees N and 36 degrees S. Calcareous dinoflagellate cysts appear to be widely distributed throughout the studied area. In the surface sediments, concentrations (cyst per gram dry sediment) of the vegetative stage Thoracosphaera heimii are generally higher than that of the (presumably) calcareous resting cysts. Distribution patterns in surface sediments of Orthopithonella granifera (Fütterer) Keupp and Versteegh, Rhabdothorax spp. Kamptner., Sphaerodinella albatrosiana (Kamptner) Keupp and Versteegh S. albatrosiana praratabulated, Sphaerodinella tuberosa var. 1 (Kamptner) Keupp and Versteegh and S. tuberosa var. 2 and the ratios between these species have been compared with temperature, salinity, density and stratification gradients in the upper water column. Rhabdothorax spp. is characteristically present in sediments of more temperate regions characterized by high seasonality. Dinoflagellates producing these cysts are able to tolerate high nutrient concentrations, and mixing of the water column. S. albatrosiana is abundant in regions characterized by high sea surface temperatures and oligotrophic surface water conditions. In contrast, the distribution of S. tuberosa var. 2 is negatively related to temperature. The other cyst species did not show a characteristic pattern in relation to the studied environmental gradients.The ratio of Sphaerodinella tuberosa var. 2 to Orthopithonella granifera can be used for reconstructing the presence of stratification in the upper 50m of the water column, whereas the ratios of S. tuberosa var. 2 to Sphaerodinella albatrosiana and of O. granifera to Rhabdothorax spp. might be used for palaeotemperature reconstructions. Calcareous dinoflagellate cysts are abundant in oligotrophic areas and may be useful for the reconstruction of palaeoenvironmental conditions.

Hydrodynamical effects on spatial distribution of enteric bacteria in the Jordan River-Lake Kinneret contact zone.

The aim of this study was to determine under what hydrodynamic conditions the change in the number of enteric bacteria in the water of the River Jordan--Lake Kinneret contact zone was due to sedimentation and under what conditions the change was due to dilution. The data were then utilized to build a conceptual model explaining the distribution of biological pollutants (bacteria) in the river-lake contact zone of a shallow tropical lake. The study uses, as an example, the microbial communities of the River Jordan--Lake Kinneret contact zone. The changes in numbers of three groups of bacteria (fecal coliforms, Escherichia coli and Klebsiella pneumoniae) along the jet flow agree well with changes in the concentration of suspended particulate matter, caused by the sedimentation of particles.

Spatial distribution of polybrominated diphenyl ethers and polybrominated biphenyls in lake trout from the Laurentian Great Lakes.

Concentrations of two types of brominated flame-retardants (BFRs); polybrominated diphenyl ethers (PBDEs) and polybrominated biphenyls (PBBs) were determined in a single age class of lake trout (Salvelinus namaycush) collected from the Laurentian Great Lakes in 1997. Mean concentrations of total PBDE were highest in samples from Lake Ontario at 95+/-22 ng/g wet weight (ww) or 434+/-100 ng/g lipid weight (lw) while the lowest concentrations were observed in Lake Erie lake trout (27+/-8.6 ng/g ww, 117+/-37 ng/g lw). In all samples, the predominant PBDE congeners were 2,2',4,4'-tetrabromodiphenyl ether (BDE-47), 2,2',4,4',5-pentabromodiphenyl ether (BDE-99), and 2,2',4,4', 6-pentabromodiphenyl ether (BDE-100), which are the primary components of the commonly used penta-BDE formulation flame retardant. Lake trout collected from Lake Huron had the highest concentrations of PBBs (3.1+/-1.7 ng/g ww, 15+/-8.5 ng/g lw), while the lowest levels were detected in fish from Lake Superior (0.25+/-0.13 ng/g ww, 1.7+/-0.89 ng/g lw). In all lake trout samples, 2,2',4,4',5,5'-hexabromobiphenyl (BB-153), a major constituent of the flame-retardant FireMaster BP-6, was the predominant PBB congener.

Spatial distribution and temporal variation of metals in the vicinity of a municipal solid waste incinerator after a modernization of the flue gas cleaning systems of the facility.

In June 1994 and 1997, the concentrations of a number of elements were determined in soil and herbage samples collected in the vicinity of a municipal solid waste incinerator (MSWI) (Tarragona, Catalonia, Spain). In August 1997, an adaptation to the EU legislation on pollutant emissions from the stack was carried out to the incinerator. In June 1999, soil and herbage samples were collected again at the same sampling points and the levels of arsenic (As), cadmium (Cd), chromium (Cr), lead (Pb), manganese (Mn), mercury (Hg), nickel (Ni) and vanadium (V) were measured by ICP-MS or AAS with graphite furnace. The results are compared with those obtained in the 1994 and 1997 surveys. In the period 1997-1999, the only significant changes in soil levels corresponded to Cd and Pb, with decreases of 21.0% and 53.5%, respectively. In vegetation, only Mn levels showed a significant reduction, which contrasts with the notable increases found in the concentrations of As, Hg and Ni. According to the results of this survey, it seems evident that other metal emission sources in the same area of study are masking the environmental improvements carried out in the MSWI.

Spatial distribution of tectotectal connections in the cat.

In mammals, the paired superior colliculi of the midbrain play a significant role in the generation and guidance of eye movements that enable an animal to orient to novel visual stimuli. In several species including monkey, cat and hamster, the paired colliculi are connected by a commissure. In the cat, many commissural axons arise from tectotectal neurons located in the deep layers in the rostral two-thirds of the colliculus. The role of these tectotectal neurons is unclear, but it is likely that they play some role in eye movement control. In this study, the neuroanatomical tracer Biocytin was used to make small, localized injections into the deep layers of the cat superior colliculus at a variety of different locations in nine animals. The distribution of tectotectal synaptic terminals in the opposite colliculus was then plotted. Regardless of which layers were included the injection site, labelled boutons were most dense in the deep layers in the contralateral colliculus. There was a striking point-to-point organization in the tectotectal projection such that terminals were concentrated at an almost mirror-symmetrical region to the injection site in the rostrocaudal plane. In the majority of cases, however, the focus of terminal boutons was shifted medially by 1-2 mm. These results suggest that tectotectal connections may influence select populations of neurons in the contralateral colliculus. By coupling specific groups of neurons in the two colliculi, their effectiveness in sensory motor processing could be enhanced. At this time it is not clear whether specific commissural terminals contain excitatory or inhibitory neurotransmitters, and our ongoing studies are addressing this question.

Oviductal progesterone concentration and its spatial distribution in cyclic and early pregnant cows.

Changes and local distribution of oviductal progesterone (P(4)) concentration during the estrous cycle and early pregnancy in cows were investigated. Intact reproductive tracts were collected from 16 Holstein cows at an abattoir. Samples were classified in to 4 stages (follicular, postovulatory, luteal and early pregnant,< 20 d) based on visual observation of corpus luteum (CL), uterine characteristics and luteal P(4) concentrations. Oviducts were separated from the uterus at the utero-tubal junction and divided into 4 parts: fimbriae, proximal, medial and distal parts. Luteal tissue samples were also collected. Progesterone levels in oviductal and luteal tissues were determined by radioimmunoassay (RIA). Comparatively higher (P < 0.001) P(4) levels were found in stages with a functioning CL ( luteal phase and early pregnancy) than in those with a regressing CL (follicular phase and post ovulation). The oviduct ipsilateral to the CL bearing ovary during the luteal phase and early pregnancy showed higher ( P < 0.001) P(4) concentrations than the contralateral side. Such a difference was not observed during the follicular phase or post ovulation. The ipsilateral oviduct to the functioning CL at early pregnancy showed higher (P <0.05) P(4) levels than at the luteal phase, while no significant difference in luteal P(4) levels between these 2 stages was observed. Neither were any differences in P(4) concentration within the oviduct observed during any phase of the estrous cycle or during early pregnancy. A positive relationship between luteal and oviductal P(4) concentrations was noted. In conclusion, changes in P(4) levels in the oviduct depend on the location and functional stage of the CL. Localized levels of P(4) in the oviduct may be due to local delivery of P(4) from the CL.

Temporal and spatial distributions of contaminants in sediments of Santa Monica Bay, California.

Contaminant inputs from wastewater discharge, a major source of contamination to Santa Monica Bay (SMB), have declined drastically during the last three decades as a result of improved treatment processes and better source control. To assess the concomitant temporal changes in the SMB sediments, a study was initiated in June 1997, in which 25 box cores were collected using a stratified random sampling design. Five sediment strata corresponding to the time intervals of 1890-1920, 1932-1963, 1965-1979, 1979-1989, and 1989-1997 were identified using (210)Pb dating techniques. Samples from each stratum were analyzed for metals, 1,1,1-Trichloro-2,2-bis(p-chlorophenyl)ethane (DDT) and its metabolites (DDTs), polychlorinated biphenyls (PCBs), and total organic carbon (TOC). Samples from the 1965-1979, 1979-1989, and 1989-1997 strata were also analyzed for polycyclic aromatic hydrocarbons (PAHs) and linear alkylbenzenes (LABs). Sediment metal concentrations increased from 1890-1979 and were similar during the time intervals of 1965-1979, 1979-1989, and 1989-1997, although the mass emissions of trace metals from sewage inputs declined substantially during the same time period. Trace organic contamination in SMB was generally highest in sediments corresponding to deposition during the years of 1965-1979 or 1979-1989 and showed a decline in concentration in the 1989-1997 stratum. Temporal trends of contamination were greatest in sediments collected from areas near the Hyperion Treatment Plant (HTP) outfall system and on the slope of Redondo Canyon. The highest contaminant concentrations were present in sediments near the HTP 7-mile outfall in the 1965-1979 stratum. Elevated trace metal and organic concentrations were still present in the 1989-1997 stratum of most stations, suggesting that sediment contaminants have moved vertically in the sediment column since sludge discharges from the 7-mile outfall (a dominant source of contamination to the bay) ceased in 1987. The widespread distributions of DDTs and PCBs in SMB and highly confined distribution of LABs around the HTP outfall system were indicative of a dispersal mechanism remobilizing historically deposited contaminants to areas relatively remote from the point of discharge.
