Micro- and oligodendroglia, plasma and nucleoli of nerve cells, capillary wall and nuclei of astrocytes become visible when sections of formol fixed human brain are immersed, without any previous treatment, into a physical developer of pH 10.5. The staining is inhibited by the catalytic activity of the tissue elements involved. By means of pretreatments with 1% performic acid and 30% sodium rhodanide dissolved in 0.4% sodium hydroxide, the catalytic activity in the unwanted tissue elements is suppressed, and this results in an elective demonstration of micro- and oligodendroglia. Reducing groups of the tissue or any kind of performed nuclei play no role in this silver staining.
The catalytic activity of the fibrous astrocytes in the reaction of silver ions with dissolved reducing molecules is considerably increased by ethylation and a subsequent treatment with NaJO3. As a consequence, in a special physical developer they produce metallic silver at a considerably higher rate than do other tissue elements, making possible their selective demonstration. In freshly fixed materials myelin too is silverized. This can be avoided by means of pretreatments with performic acid and iodine.
The ESR-spin stabilization approach has been employed to detect and characterize o-semiquinone radicals from the oxidation of epinephrine and related materials (norepinephrine, 3,4-dihydroxynorephedrine, isoproterenol, and adrenalone) in aqueous solutions. Semiquinones were generated by various oxidative procedures--enzymatic oxidation (with horseradish peroxidase/H2O2), chemical oxidation (with Ag2O) and photooxidation--and subsequently kinetically stabilized through complexation with Zn2+ ions. This "spin stabilization" affords high radical concentrations, which has allowed unambiguous identification of the radical intermediates. Where appropriate, spectral assignments have been supported by deuterium substitution experiments and computer simulations of spectra. Two types of free radical have been identified: primary "open chain" semiquinones, formed by one-electron oxidation of the parent catecholamines, and secondary semiquinones, formed subsequent to cyclization reactions. The mechanism of formation of the secondary radicals is discussed, and it is concluded that they are derived from product aminochromes. Thus, oxidation of adrenochrome gives a radical identical to the secondary species observed from oxidation of epinephrine.
We have developed a simple method for detecting high concentrations of chloride in sweat from ambulatory subjects, a measurement useful in the detection of cystic fibrosis. The method is based on the standard approach of stimulating sweat generation through iontophoresis of pilocarpine nitrate into the skin, followed by collection and analysis of the sweat for chloride concentration. The sweat-stimulating reagents are contained in polymeric gel pads, which are used in conjunction with a small battery-powered stimulator. The chloride analysis is subsequently done on the stimulated site by use of a thin test patch that picks up a fixed amount of sweat and changes color if the chloride concentration is higher than a predetermined value. The successful completion of a test is indicated by a fill tab, which changes color when the appropriate amount of sweat has been picked up by the chloride test patch.
