Influence of puromycin and chloramphenicol on aminoketone synthesis ( delta-aminolevulinic acid synthesis) induction by allylisopropylacetamide in rat liver.

In vitro induction of aminoketone synthesis (AKS) by allylisopropylacetamide (AIA) in liver slices from 30 day-old male rats was reversibly inhibited by puromycin (50 micrograms/ml incubation mixture). Normal AKS was initially stimulated and thereafter blocked. Chloramphenicol (350 micrograms/ml) inhibited AIA mediated induction of AKS only partially and only when added 1 h before AIA to the incubation mixture. In vivo puromycin (3 i.p. injections of 15 mg/kg in 1 h intervals) did not influence AKS or AIA mediated AKS induction in newborn rats, whereas chloramphenicol (500 mg/kg s.c. 30 min prior to AIA) inhibited AIA mediated AKS induction in newborn rats. In 40 day-old rats pretreatment with chloramphenicol completely inhibited AKS induction by 200 mg/kg AIA.

Detection of delta-aminolevulinic acid, porphobilinogen and porphyrins related to heme biosynthesis by high-performance liquid chromatography.

Two new high-performance liquid chromatographic methods are described for the quantitative determination of porphyrins and their precursors. In our method, sub-nanomole quantities of porphyrins, delta-aminolevulinic acid and porphobilinogen derivatized with ophthalaldehyde were injected onto a C18 reversed-phase column and eluted with 0.1 M monobasic sodium phosphate-methanol-tetrahydrofuran (4:6:3) and detected with a spectrofluorometer. A second reversed-phase system using methanol-tetrahydrofuran-22 mM acetate buffer (15:6:11) was also developed.

Haem synthesis during cytochrome P-450 induction in higher plants. 5-Aminolaevulinic acid synthesis through a five-carbon pathway in Helianthus tuberosus tuber tissues aged in the dark.

Chlorophyll and haem synthesis in illuminated Jerusalem artichoke tuber tissues were very efficiently inhibited by gabaculine (3-amino-2,3-dihydrobenzoic acid). This inhibition seems to be due specifically to a blockade of the pathway for 5-aminolaevulinate biosynthesis which used glutamate as a substrate (the so-called C5 pathway) since we could not detect any inhibition of protein synthesis in the treated tissues and there was no effect of gabaculine on the glycine-dependent yeast 5-aminolaevulinate synthase used as a model. In dark-aged artichoke tissues, gabaculine also effectively blocked cytochrome P-450 induction, peroxidase activity and 5-aminolaevulinic acid synthesis, thus suggesting the involvement of a C5 pathway in cytoplasmic and microsomal haemoprotein synthesis in this higher plant. Allylglycine and (2-amino-ethyloxyvinyl)glycine, two olefinic glycine analogues which are potential suicide inhibitors of pyridoxal phosphate enzymes, were also demonstrated to be effective blockers of chlorophyll synthesis in artichoke tuber and Euglena cells exposed to light.

Chemically induced porphyria: prevention by prior treatment with phenobarbital.

The excessive induction of hepatic delta-aminolevulinic acid synthetase in rats after the administration of porphyria-inducing compounds is prevented by prior treatment with phenobarbital. Studies in vivo and in vitro indicate that phenobarbital prevents the induction of chemical porphyria by increasing the rate of detoxification of inducers by way of drug-metabolizing enzymes of the hepatic endoplasmic reticulum.

Statistical investigations into the measured DALA--discharge in the urine of infants.

Using a sample of the size 353 on day-nursery children, the authors show that the DALA-U increase, which is due to the absorption of lead and other substances, does not display any age or sex dependence. A dependence upon the day nursery's location is assumed. The authors elaborate on the statistical problems (normal distribution) and the boundary value problems associated with the various DALA-U values measured. Large sample sizes are required to find a final clarification of the relationships.

Optimized liquid-chromatographic method for fluorometric determination of urinary delta-aminolevulinic acid in workers exposed to lead.

We developed a fluorometric method for determining delta-aminolevulinic acid (ALA) in urine of lead workers. A high-performance liquid chromatograph (HPLC) equipped with a fluorescence HPLC monitor is used. The detection limit for aqueous ALA is 20 micrograms/L (15 pmol of ALA in the 100-microL sample). The working linear range of urinary ALA concentration was 0.1 to 100 mg/L. In 25 lead-exposed workers, ALA values by the present method for urine correlated well with those obtained by a conventional colorimetric method (r = 0.996). The advantage of the present method for micro-determination of urinary ALA is its high sensitivity.
