Differential effects of body fatness and body fat distribution on risk factor for cardiovascular disease in women. Impact of weight loss.

This study examines the role of obesity and body fat distribution (ie, waist-to-hip ratio [WHR]) on cardiovascular disease (CVD) risk factors in 50 nondiabetic, obese (body mass index [BMI], 31 +/- 2 kg/m2, mean +/- SD), 45 +/- 10-year-old women. The data obtained at baseline and after weight loss were analyzed after dividing subjects by WHR into upper-body (WHR > 0.80) and lower-body (WHR < or = 0.80) groups and by median-split BMI into more obese (BMI > or = 31) and less obese (BMI < 31) groups. At baseline, the upper-body obese women, when compared with lower-body obese women, had higher plasma triglycerides (TGs) (175 +/- 85 versus 111 +/- 47 mg/dL, respectively; P < .001) and lower high-density lipoprotein cholesterol (HDL-C) (44 +/- 10 versus 54 +/- 11, respectively; P < .01) but similar total and low-density lipoprotein cholesterol levels and blood pressure. There were no significant differences in these CVD risk factors at baseline by BMI split. Although weight loss (-9 +/- 5 kg) lowered blood pressure and TGs irrespective of WHR or BMI, only upper-body obese women raised HDL-C. Moreover, the magnitude of the changes was greatest in women with an upper-body fat distribution. In women with WHR > 0.80, HDL-C increased by 11%, to 49 mg/dL (P < .001), and TGs decreased by 24%, to 134 mg/dL (P < .001).(ABSTRACT TRUNCATED AT 250 WORDS)

Trafficking of mutant carboxypeptidase E to secretory granules in a beta-cell line derived from Cpe(fat)/Cpe(fat) mice.

We have reinvestigated the stability and intracellular routing of mutant carboxypeptidase E in NIT3 cells, a pancreatic beta-cell line derived from the Cpe(fat)/Cpe(fat) mouse. Pulse-chase experiments demonstrated that this protein has a half-life of approximately 3 h in these cells and that up to 45% of the proCPE(202) can escape degradation by the proteosome. In double-label immunofluorescence microscopy, a portion of the mutant CPE did not colocalize with calnexin, an endoplasmic reticulum marker, but was found in prohormone convertase 2-containing secretory granules, demonstrating that it had escaped degradation and arrived at a post-Golgi compartment. The mutant CPE as well as prohormone convertase 2 were secreted into the medium in a stimulated manner by treatment with the physiological secretagogue, glucagon-like peptide-1, consistent with its presence in granules of the regulated secretory pathway. The presence of mutant carboxypeptidase E in granules supports a potential role for its involvement as a sorting/retention receptor in the trafficking of proinsulin to the regulated secretory pathway.

The validity of estimating total body fat and fat-free mass from skinfold thickness in adults with growth hormone deficiency.

The regression equations of Durnin and Womersley for estimating total body fat (TBF) and fat-free mass (FFM) from skinfold thickness were validated for adult GH-deficient (GHD) patients by comparing the values of TBF and FFM from the prediction equations with the directly measured values from dual energy x-ray absorptiometry. Twenty-seven male and 24 female patients (aged 21-61 yr) were studied. GHD was isolated in 5 cases and was part of a spectrum of hypopituitarism due to a variety of causes in 46 cases. The mean period of GHD was 6.9 +/- 4.6 yr. All patients were receiving stable replacement therapy. The validation statistics showed no significant differences (P > 0.05) between measured and predicted values of TBF and FFM in either males (24.5 vs. 24.9 and 65.5 vs. 65.8 kg) or females (24.6 vs. 26.3 and 44.7 vs. 43.9 kg). Mean differences were smaller in males (0.4 and 0.2 kg) than females (1.7 and -0.8 kg); they were less than 1% in males and less than 2% in females. Therefore, the Durnin and Womersley equations are suitable for general use with GHD patients. Using TBF (kilograms) from dual energy x-ray absorptiometry as the dependent variable and the log of the sum of skinfold thickness as the independent variable, linear regression equations were formulated to predict TBF in GHD patients. The lowest SE of estimate was 3.8 kg in males and 4.6 kg in females. To determine their general applicability, these equations will need to be cross-validated.

Subcutaneous adipose tissue metabolism at menopause: importance of body fatness and regional fat distribution.

The aim of this study was to examine the contribution of menopause per se on sc adipose tissue (AT) metabolism in 16 women classified on the basis of their menopausal status: 8 postmenopausal (mean +/- SD age, 57 +/- 6 yr) vs. 8 premenopausal individuals (37 +/- 5 yr). These 2 groups were matched for sc abdominal adipose cell size (within 0.02 microg lipid/cell) and visceral AT accumulation (within 15 cm2), measured by computed tomography. Fasting plasma glucose and insulin levels as well as their responses to an oral glucose load were similar regardless of the women's hormonal status. Subcutaneous abdominal and femoral AT lipoprotein lipase activities as well as fat cell lipolysis were determined in both groups. Epinephrine induced antilipolysis at low concentrations and lipolysis at higher doses in both adipose sites and groups. The maximal lipolytic response to epinephrine or to isoproterenol (beta-adrenergic agonist) as well as the maximal antilipolytic effect of either the catecholamine or UK-14304 (alpha2-adrenergic agonist) assessed in sc adipocytes were similar in pre- and postmenopausal women. In addition, neither the beta- nor the alpha2-adrenoceptor sensitivity of sc adipose cells differed according to subjects' age. Finally, maximal lipolysis promoted by postadrenoceptor agents and AT-lipoprotein lipase activity did not vary among adipose regions or between groups. Taken together, these results suggest that menopause per se does not influence sc AT metabolism once the variation related to adipose cell size and total body fatness is taken into account.

Suppression by licorice flavonoids of abdominal fat accumulation and body weight gain in high-fat diet-induced obese C57BL/6J mice.

We applied licorice flavonoid oil (LFO) to high-fat diet-induced obese C57BL/6J mice and investigated its effect. LFO contains hydrophobic flavonoids obtained from licorice by extraction with ethanol. The oil is a mixture of medium-chain triglycerides, having glabridin, a major flavonoid of licorice, concentrated to 1.2% (w/w). Obese mice were fed on a high-fat diet containing LFO at 0 (control), 0.5%, 1.0%, or 2.0% for 8 weeks. Compared with mice in the control group, those in the 1% and 2% LFO groups efficiently reduced the weight of abdominal white adipose tissues and body weight gain. A histological examination revealed that the adipocytes became smaller and the fatty degenerative state of the hepatocytes was improved in the 2% LFO group. A DNA microarray analysis of the liver showed up-regulation of those genes for beta-oxidation and down-regulation of those for fatty acid synthesis in the 2% LFO group. These findings suggest that LFO prevented and ameliorated diet-induced obesity via the regulation of lipid metabolism-related gene expression in the liver.

Fat-suppression failure artifacts simulating pathology on frequency-selective fat-suppression MR images of the head and neck.

PURPOSE: To describe fat-suppression failure artifacts and to caution against their misinterpretation. METHOD: Magnetic-susceptibility artifacts were studied in a phantom model and the results were compared to MR images obtained in clinical cases. FINDINGS: Artifacts manifested themselves as regions of focal fat-suppression failure and appeared as bright signals without geometric distortions at magnetic-susceptibility interfaces along the static field (z) direction. The location and extent of these artifacts were independent of either frequency or phase-encoding direction and are different from those observed in gradient-echo images. CONCLUSIONS: In representative clinical MR exams, these artifacts were identified in the high nasopharynx and low orbit and should not be misinterpreted as pathology.

A comparative study of CT densitometry of vertebral cancellous bone, abdominal aorta, paravertebral fat and the relative fat volume.

Fluctuations are observed in CT densitometry of cancellous bone of the lumbar vertebra in relation to age and sex and also per age/sex group. An investigation was carried out in 119 patients to possibly detect comparable fluctuations in CT measurements of other tissues. The spongiosa of the fourth lumbar vertebra, the abdominal aorta, paravertebral fat and the ratio fat volume/total irradiated volume were compared in the slice plane through the middle of the fourth lumbar vertebra. Calibration was performed using a water phantom. A correlation between the mentioned parameters could not be demonstrated. The relation between Bone Mineral Density (BMD) and age and sex showed the same values as known from the literature, being representative for a normal population. Using the aorta as a reference (which in fact is related to water) a similar curve is found as in Cann and Cann et al. The density of the aorta showed a constant value, varying between 38 and 50 HU with a mean of 45 HU. Extreme values were only found with streak artefacts. It seems useful to use the aorta values as a reference to measure the relative value of vertebral cancellous bone.

Size, fatness and relative fat distribution of males of contrasting maturity status during adolescence and as adults.

The somatic characteristics of boys of contrasting biological maturity status during adolescence are compared from 13-18 years and at 30 years of age. Within the mixed longitudinal Leuven Growth Study of Belgian Boys, 173 boys were followed annually from 13-18 years and were subsequently measured at 30 years of age. Age at peak height velocity (PHV) was estimated for 149 boys and the sample was then divided into three contrasting maturity categories based on the age at PHV: early (PHV < 13.37 years), average (PHV between 13.85 and 14.80 years) and late (PHV > 15.27 years) maturers. Using ANOVA for repeated measures and one-way ANOVA, differences in 18 somatic dimensions and five ratios of body proportions and subcutaneous fat distribution among the three maturity groups were tested from 13-18 years and at 30 years of age. There are consistent differences among boys of contrasting breadths, circumferences, and skinfolds on the trunk. There are no differences in skinfolds on the extremities. None of the differences in somatic dimensions and ratios among the three contrasting maturity groups are significant at 30 years of age except those for subscapular skinfold and the trunk/extremity skinfold ratio. Thus, during adolescence and in adulthood, late maturing boys have a distribution of subcutaneous fat that is associated with lower risk for several adult degenerative diseases.

Effect of infrared analyzer homogenization efficiency on repeatability of uncorrected fat A and fat B signals.

Poor repeatability by infrared milk analyzers may be caused by inefficient homogenization as a result of light scattering and the Christiansen effect. The objectives of this study were to identify instruments with good and poor homogenization efficiency and to determine if a difference exists in repeatability performance between instruments with good vs poor homogenization efficiency. Unhomogenized and homogenized portions of the same milk were tested 20 times consecutively on 22 instruments. An instrument was considered to have poor homogenization efficiency if the mean difference in the uncorrected signal between unhomogenized and homogenized portions of the same milk was > or = 1.43% of the fat test (i.e., > or = 0.05% at 3.5% fat). Instruments were evaluated for repeatability by calculating the sample standard deviation and the range of the latter 19 uncorrected readings for unhomogenized and homogenized milks. When repeatability was evaluated as a function of homogenization efficiency, there was a significant (p = 0.001) correlation between poor homogenization efficiency and poor repeatability when testing unhomogenized milk but not when testing homogenized milk. Improved homogenizer performance within infrared milk analyzers is needed to improve the repeatability of raw milk testing.

Differences in the coalescence kinetics of fat emulsions in dependence on the amount of fat and age.

The destabilizing effect of calcium ions on emulsions was studied as a function of the age of the emulsions and the degree of emulsion dilution (2%, 0.2% an 0.02% fat). Particle size measurements were performed both in the Coulter counter and a laser diffraction device equipped with PIDS technology. The data of both instruments showed a good correlation. zeta-Potential was determined by laser doppler anemometrie. The physical stability of the emulsions in 6 mmolar Calcium Chloride decreased with increasing dilution--despite the diminished rate of collision in diluted systems. In addition, and increased electrolyte sensitivity was observed with increasing age of the emulsions--despite enhanced electrostatic stabilization by an increased zeta-potential. Both effects were attributed to an increased binding of calcium ions per surface area of the droplets, i.e. increased ratio calcium ions to surface by dilution and increased binding by the increased charge of aged emulsions.

[Determination of trace elements in dietary fats and emulsifiers by non-flame atomic absorption spectrometry (AAS). 2. Determination of mercury in dietary fats and emulsifiers].

Of the trace elements, mercury holds an exceptional position as to its chemico-physical and toxic properties. In the light of the special analytical problem, the methods used (cold-steam AAS in combination with the addition method) are discussed. The experimental conditions required for the digestion and the cold-steam AAS of samples of emulsifiers and dietary fats are indicated. The accuracy and the reproducibility of the procedure are dealt with, and the amounts of mercury found in products of the oil and margarine industries are reported.

Dietary fats and diabetes mellitus: is there a good fat?

As knowledge of the fatty acid functions has increased, so has the complexity of making dietary fat recommendations to people with type 2 diabetes. Oleic acid seems to offer a slight advantage over linoleic acid in reducing plasma glucose, insulin levels, total cholesterol, low-density lipoproteins (LDLs), and triglycerides, but may also have atherogenic properties through another mechanism. A diet containing a higher proportion of polyunsaturated fatty acids (PUFAs) may require a concomitant increase in antioxidant intake because PUFAs oxidize easily and are then converted to oxidized LDL, which is more atherogenic. In addition to raising total and LDL cholesterol, long chain saturated free fatty acids may interact with plasma glucose to increase insulin secretion. Omega-3 fatty acids decrease triglycerides and reduce the risk of fatal cardiac arrhythmias. Glycemic control does not appear to be adversely affected by omega-3 fatty acids at amounts of up to 3 g/d.

Effects of fat level, feeding period, and source of fat on lipid fluidity and physical state of rabbit plasma lipoproteins.

Elevating fat content from 5 to 20% of diet by weight or extending the feeding period from 6 months to more than 1 year did not substantially alter the fluidity of rabbit plasma lipoprotein lipid domains. Dietary fatty acid saturation was not adequate as a predictor of lipoprotein fluidity. Rabbits fed corn oil, high in polyunsaturated fatty acid content, did not have more fluid lipoproteins than rabbits fed cocoa butter which contains a high level of saturated long chain fatty acids. Order parameters calculated from fluorescence depolarization measurements with diphenylhexatriene (DPH) showed that very low density lipoprotein (VLDL) lipids were in highly fluid or 'liquid' states at or below body temperature. Order parameter data showed transitions from ordered phase to isotropic liquid in low density lipoproteins (LDL) that were heretofore unnoted with DPH fluorescence depolarization measurements. The transition temperature was inversely related to the LDL triglyceride content, indicating probe intercalation between the fatty acyl chains of the core triacylglycerols in VLDL and LDL.

Total fat, fat distribution and blood pressure according to eating frequency in children living in southern Italy: the ARCA project.

The relationships between eating frequency and adiposity and cardiovascular risk factors have been poorly investigated in children. To evaluate this issue, anthropometric indices and blood pressure (BP) were measured in 3668 children, aged 6-11 years. Meals/snack frequency, assessed by a questionnaire, was 'Low' in 332 ( or=5/day) children. Body mass index (BMI) and waist circumference significantly decreased going up across meals/snacks categories (BMI=20.5+/-0.2, 19.7+/-0.1 and 18.8+/-0.1 kg/m2; waist=68.0+/-0.6, 66.6+/-0.3, 63.9+/-0.2 cm, for Low, Intermediate and High groups, respectively; M+/-s.e.; P<0.001 by ANCOVA trend analysis adjusted by age, sex, birth weight, physical activity, parental overweight and education level). BP decreased from Low to High group but differences were no longer significant after adjustment for BMI. In our sample of school children, the daily eating frequency was inversely associated with the degree of both total adiposity and central fat deposition.

Regulation of pyruvate dehydrogenase and pyruvate dehydrogenase phosphate phosphatase activity in rat epididymal fat-pads. Effects of starvation, alloxan-diabetes and high-fat diet.

1. Pyruvate dehydrogenase phosphate phosphatase activity in rat epididymal fat-pads was measured by using pig heart pyruvate dehydrogenase [32P]phosphate. About 80% was found to be extramitochondrial and therefore probably not directly concerned with the regulation of pyruvate dehydrogenase activity. The extramitochondrial activity was sensitive to activation by Ca2+, but perhaps less sensitive than the mitochondrial activity.
