The investigation of the HCN derivative diiminosuccinonitrile as a prebiotic condensing agent. The formation of phosphate esters.

Diiminosuccinonitrile (DISN) is formed readily by the Fe3+ oxidation of diaminomaleonitrile, a tetramer of HCN. DISN effects the phosphorylation of uridine in 13% yield to a mixture of the isomers of UMP when the reaction is performed in dimethylformamide solution. A 4% yield of the UMP isomers is obtained in neutral aqueous solution using 2 times the DISN concentration and 7 times the phosphate concentration used in DMF. DISN did not effect the conversion of adenosine to AMP or 5'-AMP to 5'-ADP in aqueous solution. The cyclization of 3'-AMP and 3'-UMP to the corresponding 2'-3'-cyclic phosphates proceeds in yields as high as 40-50% at 60 degrees C in pH 6 aqueous solutions in the presence of divalent metal ions. Lower yields of the cyclic phosphate are observed when 2'-AMP is the starting material. Substitution of acetate buffer for imidazole buffer results in a decrease in the yield of cyclic phosphate, the extent of which depends on the metal ion used in the reaction. No 3'5'-cyclic AMP was detected as a reaction product with either 5'-AMP or 3'-AMP as the starting material except for a 2.4% yield from 3'-AMP in the presence of Zn2+. BrCN effects the conversion of 3'-AMP to the 2'-3'-cyclic AMP in 37-65% yield depending on the divalent cation used as catalyst. A mechanism has been proposed for these cyclization reactions and their potential significance to the prebiotic synthesis of ribonucleic acid derivatives is discussed.

Physiochemical studies of indocyanine green (ICG): absorbance/concentration relationship, pH tolerance and assay precision in various solvents.

Indocyanine green (ICG) obeyed the Beer-Lambert law within the concentration range 1.25 micrograms/ml-10.0 micrograms/ml in distilled water, methanol, dimethylformamide (DMF), 1.2-propanediol and aqueous buffers (pH 9.0), but only up to 7.5 micrograms/ml in human bile and 0.5% human albumin, and only to 5.0 micrograms/ml in human duodenal fluid. ICG was rapidly (less than 1 h) decomposed to a colorless derivative at pH less than 5 and greater than 11, but remained relatively stable for 48 h at pH 8-10. ICG is an indicator and a weak acid with a pKa of 3.27. In bile stabilized with 25% methanol, the precision of the method (CV) is 5% and the accuracy is 106-127%.

Metabolism of prostaglandin E2 by human HL-60 leukemia cells.

The metabolic fate of prostaglandin E2 (PGE2) was examined in human HL-60 leukemia cells. Cytosolic fractions obtained from dimethylformamide treated HL-60 granulocytes rapidly convert exogonous PGE2 to 15-keto-PGE2. The strict requirement for NAD coupled with other characteristics of the reaction indicate that the enzyme is the NAD-dependent 15-prostaglandin dehydrogenase (15-PGDH). When intact cells are incubated with 3H-PGE2, both 15-keto-PGE2 and its subsequent metabolite, 13,14-dihydro-15-keto-PGE2 are produced. Thus, HL-60 cells express two enzymes of the major prostaglandin catabolic pathway, 15-PGDH and ketoprostaglandin delta 13-reductase.

Analysis of creams. II. Quantitative determination of drugs in creams by titration in non-aqueous solvents.

The possibilities for the determination of active components in creams by acid-base titrations in non-aqueous solvents were investigated. Interference by cream-base components with the titration of weak organic bases and their halides with perchloric acid in acetic acid, and with the titration of weak acids with tetrabutylammonium hydroxide in N,N-dimethylformamide were studied. It appeared to be possible to determine alkaloid halides, salicylic acid, hexachlorophene and methyl salicylate without previous clean-up of the cream samples.

Application of selectively acylated glycosides for the alpha-galactosidase-catalyzed synthesis of disaccharides.

4-Nitrophenyl alpha-D-galactopyranosyl-(1-->3)-6-O-acetyl-alpha-D-galactopyranoside was prepared in a transglycosylation reaction catalyzed by alpha-D-galactosidase from Talaromyces flavus using 4-nitrophenyl alpha-D-galactopyranoside as a glycosyl donor and 4-nitrophenyl 6-O-acetyl-alpha-D-galactopyranoside as an acceptor. 4-Nitrophenyl 6-O-acetyl-alpha-D-galactopyranoside and 4-nitrophenyl 6-O-acetyl-beta-D-galactopyranoside were prepared in a regioselective enzymic transesterification in pyridine-acetone catalyzed by the lipase PS from Burkholderia cepacia. A series of water-miscible organic solvents (acetone, acetonitrile, dimethylformamide, dimethyl sulfoxide, 1,4-dioxane, 2-methoxyethanol, pyridine, 2-methylpropan-2-ol, tetrahydrofuran, propargyl alcohol) were used as co-solvents in this enzymic reaction. Their influence on the activity and stability of the alpha-galactosidase from T. flavus was established. 2-Methylpropan-2-ol and acetone (increasing the solubility of the modified substrate acceptors and displaying the minimum impairment of the activity and stability of the enzyme) were used as co-solvents in transglycosylation reactions.

Antibacterial, antifungal and anticonvulsant evaluation of novel newly synthesized 1-[2-(1H-tetrazol-5-yl)ethyl]-1H-benzo[d][1, 2,3]triazoles.

Several novel 1-[2-(1H-tetrazol-5-yl) ethyl]-1H-benzo[d][1,2,3]triazoles (3a-h) have been synthesized by the condensation of 1-[2-(1H-tetrazol-5-yl)-ethyl]-1H-benzotriazole (2) and appropriate acid chlorides. 1-[2-(1H-tetrazol-5-yl)-ethyl]-1H-benzotriazole (2) was synthesized by reacting 3-(1H-benzo[d][1,2,3]triazol-1-yl)propanenitrile with sodium azide and ammonium chloride in the presence of dimethylformamide. The synthesized compounds were characterized by IR and PMR analysis. The titled compounds were evaluated for their in-vitro antibacterial and antifungal activity by the cup plate method and anticonvulsant activity evaluated by the maximal electroshock induced convulsion method in mice. All synthesized compounds exhibited moderate antibacterial activity against Bacillus subtilis and moderate antifungal activity against Candida albicans. Compounds 5-(2-(1H-benzo[d][1,2,3]triazo-1-yl)ethyl)-1H-tetrazol-1-yl)(4-aminophenyl)methanone 3d and 5-(2-(1 H-benzo[d][1,2,3]triazo-1-yl)ethyl)-1H-tetrazol-1-yl)(2-aminophenyl)methanone 3e elicited excellent anticonvulsant activity.

Synthesis of 2-(allylthio)pyrazines as a novel cancer chemopreventive agent.

2-(Allylthio)pyrazine derivatives were designed as a novel cancer chemopreventive agent that functions through selective inhibtion of cytochrome P-450 and induction of phase II enzymes involved in the detoxification of carcinogens. A practical preparation method of 2-(allylthio) pyrazine derivatives was established by the reaction of 2-mercaptopyrazine and allylbromides in the presence of a catalytic antioxidant, DABCO (1,4-diazabicyclo[2,2,2] octane), in dimethylformamide at below 50 degrees C.

Optimizing industrial enzymes by directed evolution.

Enzymes can be tailored for optimal performance in industrial applications by directing their evolution in vitro. This approach is particularly attractive for engineering industrial enzymes. We have created an efficient para-nitrobenzyl esterase over six generations of random point mutagenesis and recombination coupled with screening for improved variants. The best clones identified after four generations of sequential random mutagenesis and two generations of random recombination display more than 150 times the p-nitrobenzyl esterase activity of wild type towards loracarbef-p-nitrobenzyl ester in 15% dimethylformamide. Although the contributions of individual effective amino acid substitutions to enhanced activity are small (< 2-fold increases), the accumulation of multiple mutations by directed evolution allows significant improvement of the biocatalyst for reactions on substrates and under conditions not already optimized in nature. The positions of the effective amino acid substitutions have been identified in a pNB esterase structural model. None appear to interact directly with the antibiotic substrate, further underscoring the difficulty of predicting their effects in a 'rational' design effort.

Metabolism of benzene in human liver microsomes: individual variations in relation to CYP2E1 expression.

In human liver microsomes the oxidations of benzene, chlorzoxazone, aniline, dimethylformamide, and 4-nitrophenol were significantly correlated with each other and with the level of cytochrome P450 (CYP) 2E1 estimated by immunoblotting. Moreover, benzene oxidation to water-soluble metabolites was suppressed by 0.1 mM diethyldithiocarbamate, supposedly a specific inhibitor of CYP2E1 at this level. None of these metabolic rates correlated with immunochemically determined levels of CYP1A2, 2C9, and 3A4 nor oxidation of 7-ethoxyresorufin, tolbutamide, and nifedipine. Benzene oxidation to water-soluble metabolites was characterized by typical Michaelis-Menten kinetics. The different benzene K(m) values seen in individual human microsomal samples were not correlated with the level or activity of CYP1A2, 2C9, 2E1, and 3A4 but could be due to CYP2E1 microheterogeneity. The lowest K(m) for benzene oxidation could be related to C/D and/or c1/c2 polymorphism of CYP2E1 gene. Covalent binding of benzene reactive metabolites to microsomal proteins was also correlated with the CYP2E1 metabolic rates and immunochemical levels. At high concentrations of benzene covalent binding was inversely related to benzene concentrations (as well as to formation of water-soluble metabolites) in agreement with the view that secondary metabolites, mainly benzoquinone, are responsible for the covalent binding.

Column preconcentration of gold by adsorbing AuCl4- onto methyltrioctyl ammonium chloride-naphthalene and subsequent atomic absorption spectrometric determination.

A sensitive, selective and simple preconcentration method for ultra-trace gold determination has been developed that uses naphthalene-methyltrioctyl ammonium chloride (Aliquat 336s) as an adsorbent. Gold, in the form of AuCl4-, was retained by the adsorbent in the column at a flow rate of 1 ml min(-1). After filtration, the solid mass consisting of the gold complex and naphthalene was dissolved out of the column with 5 ml of N,N-dimethylformamide (DMF), and the metal was then determined by atomic absorption spectrometry. In the initial solution, the calibration graph of absorbance versus gold concentration was found to be linear in the range 0.5-150 ng ml(-1) Au(III) with r=0.997 (n=9), and the 3 s detection limit was 0.428 ng ml(-1). The relative standard deviation for eight replicate measurements of 20 microg of gold was 2.14%. Preconcentration factors of 390 and 650 were achieved using 5 ml and 3 ml of DMF, respectively. The proposed method was successfully applied to the determination of gold in wastewater, processed pool water, slurry pool water, and raw well-water from the Moteh gold mine, and synthetic samples.

Electrochemical determination of the total antioxidant capacity of human plasma.

Electrochemical reduction of oxygen at a glassy carbon electrode in a 0.05 mol L(-1) solution of (C2H5)4NI in dimethylformamide leads to generation of the superoxide anion-radical. This product of reversible one-electron oxygen reduction reacts with antioxidants, a process which is based on protonation of the anion-radical by the antioxidant. Rate constants of this interaction have been calculated. Human plasma antioxidants also react with electrochemically generated superoxide anion-radical. A voltammetric method is proposed for estimation of the total antioxidant capacity (TAC) of plasma on the basis of on this reaction. The TAC of plasma was also determined using constant-current coulometry with electro-generated bromine as the active species. A correlation was observed between TAC data obtained by voltammetry (O2*-, in alpha-tocopherol units) and coulometry (Br2 as titrant). TAC of plasma from patients with purulent infections was determined. Statistically significant differences were found between TAC of patients and control group. Treatment of purulent infections increases the TAC of plasma. So, use of electrochemical methods (voltammetry and coulometry) for determination of TAC can be used for estimation of the effectiveness of treatment.

Ion-exchange behavior of stannic selenoiodate and stannic selenosilicate: analytical application of stannic selenoiodate.

Two new inorganic ion exchangers, stannic selenoiodate and stannic selenosilicate have been synthesized. The ion-exchange capacity of stannic selenoiodate and stannic selenosilicate for K+ was found to be 1.84 and 1.23 meq g(-1), respectively. pH titration studies reveal monofunctional and bifunctional behavior for stannic selenosilicate and stannic selenoiodate, respectively. Distribution coefficients of metal ions in dimethylformamide-HCl and formamide-HCl systems have been determined. Some important and analytically difficult quantitative binary and ternary separations, and selective separations of Ag+, Sn4+, Zr4+, Co2+, and Ni2+ have been achieved on stannic selenoiodate columns. The practical utility of the material has been demonstrated by analyzing the metal ion content of electroplating waste.

Rapid and quantitative extraction method for the determination of chlorophylls and carotenoids in olive oil by high-performance liquid chromatography.

Colour is an organoleptic characteristic of virgin olive oil and an important attribute that affects the consumer perception of quality. Chlorophylls and carotenoids are the main pigments responsible for the colour of virgin olive oil. A simple analytical method for the quantitative determination of chlorophylls and carotenoids in virgin olive oils has been developed. The pigments were isolated from small samples of oil (1.0 g) by solid-phase extraction using diol-phase cartridges (diol-SPE), and the extract was analysed by reverse-phase HPLC with diode-array UV detection. Chromatographic peak resolution, reproducibility (coefficient of variation (C.V.) <4.5%) and recovery (>98.4%) for each component were satisfactory. A comparative study of the proposed method was performed versus classical liquid-liquid extraction (LLE) with N,N'-dimethylformamide and solid-phase extraction using a C18 column (C18-SPE). While 96.4% of the pigments were recovered by LLE, only 51.3% were isolated by C18-SPE in comparison to diol-SPE. Likewise, a higher alteration of pigment composition was observed when such LLE and C18-SPE procedures were used. In this sense, a higher ratio of pheophytin in comparison to that isolated by the diol-SPE procedure was achieved with both extraction procedures, indicating a greater extent of the pheophytinization reaction. Therefore, quantification of pigments from virgin olive oil by diol-SPE followed by RP-HPLC was found to be rapid, simple, required only a small amount of sample, consumed only small amounts of organic solvents, and provided high recoveries, accuracy and precision.

Nanostructured electrochemical DNA biosensors for detection of the effect of berberine on DNA from cancer cells.

Multi walled carbon nanotubes (MWNT) in dimethylformamide (DMF) or aqueous sodium dodecyl sulfate (SDS) solution, colloidal gold nanoparticles (GNP) in phosphate buffer solution (PBS), and a GNP-MWNT mixture in aqueous SDS solution have been investigated for chemical modification of a screen-printed carbon electrode used as the signal transducer of a dsDNA-based biosensor. Differential pulse voltammetry of the DNA redox marker Co[(phen)3]3+ and the guanine moiety anodic oxidation and cyclic voltammetry with K3[Fe(CN)6] as indicator revealed substantial enhancement of the response of the biosensor, particularly when MWNT in SDS solution was used. The biosensor was used in testing of berberine, an isoquinoline plant alkaloid with significant antimicrobial and anticancer activity. Berberine had a very strong, concentration-dependent, effect on the structural stability of DNA from the human cancer cells (U937 cells) whereas non-cancer cells were changed only when berberine concentrations were relatively high 75 and 50 microg mL(-1).

Flame atomic absorption spectrometric determination of trace chromium in various standard samples after preconcentration with 2-(5-bromo-2-pyridylazo)-5-diethylaminophenol.

Chromium can be quantitatively retained as 2-(5-bromo-2-pyridylazo)-5-diethylaminophenol (5-Br-PADAP)-tetra-phenylborate(TPB) complex onto microcrystalline naphthalene in the pH range 4.8-5.9 from a large volume of aqueous solutions of various standard samples. After filtration, the solid mass consisting of the chromium complex and naphthalene was dissolved with 5 mL of dimethylformamide and the metal was determined by air-acetylene FAAS. A detection limit of 4 ng/mL for chromium was established. The interference of a large number of anions and cations has been studied and the optimized conditions developed were utilized for the trace determination of chromium in various standard alloys and biological samples.

Influence of organic solvents on the growth of marine microalgae.

Ninety-six-hour toxicity tests of five organic solvents (methanol, ethanol, acetone, N,N-dimethylformamide [DMF], and dimethyl sulfoxide [DMSO]) on nine species of marine microalgae (Bacillariophyceae; Skeletonema costatum, and Chaetoceros calcitrans, Prasinophyceae; Tetraselmis tetrathele, Haptophyceae; Isochrysis galbana and Pavlova lutheri, Dinophyceae; Prorocentrum minimum, Raphidophyceae; Heterosigma akashiwo, Euglenophyceae; Eutreptiella sp., Chlorophyceae; Dunaliella tertiolecta) were conducted. For D. tertiolecta, T. tetrathele, I. galbana, and P. lutheri, methanol was the least toxic solvent. For S. costatum, C. calcitrans and Eutreptiella sp., DMSO was the least toxic solvent. For Heterosigma akashiwo, DMF was the least toxic solvent. In general, solvent toxicity was ethanol > methanol or acetone > DMF > DMSO. In relation to sensitivity of the microalgae to solvents, D. tertiolecta and T. tetrathele were lower in sensitivity, and Eutreptiella sp. and H. akashiwo were higher in sensitivity than the other species. The no observed effect concentration (NOEC) of acetone, DMF, and DMSO were higher than for ethanol and methanol. Acetone, DMF, and DMSO were suitable for carrier solvents used in toxicity test of water-insoluble chemicals. Excellent regression between logEC(50) and logNOEC was obtained (logNOEC = 1.09 x logEC(50) - 0.84, R(2) = 0.91). This equation indicated that higher toxicity was higher application factor (EC(50)/NOEC) than lower toxicity. All application factors were higher than 0.1.

Purification, characterization, and chemical modification of manganese peroxidase from Bjerkandera adusta UAMH 8258.

Ten strains of Bjerkandera adusta from the University of Alberta Microfungus Collection and Herbarium (UAMH) were compared for manganese peroxidase production. The enzyme from B. adusta UAMH 8258 was chosen for further study. After purification the enzyme showed a molecular weight of 43 kDa on 15% SDS-PAGE, 36.6 kDa on matrix-assisted laser desorption ionization-time of flight mass spectrometry, and an isoelectric point of 3.55. The N-terminal amino acid sequence was determined to be VAXPDGVNTATNAAXXALFA, and the amino acid composition showed no tyrosine residues in the enzyme. Manganese peroxidase exhibited both Mn(II)-dependent (optimum pH 5) and Mn(II)-independent activity (optimum pH 3). The purified enzyme was chemically modified with cyanuric chloride-activated methoxypolyethylene glycol to enhance its surface hydrophobicity. The modified and native enzymes showed similar catalytic properties in the oxidation of Mn(II) and other substrates such as 2,6-dimethoxylphenol, veratryl alcohol, guaiacol, and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonate). However, the modified enzyme showed greater resistance to denaturation by hydrogen peroxide and stability to organic solvents such as acetonitrile, N,N-dimethylformamide, tetrahydrofuran, methanol, and ethanol. The PEG-modified enzyme also showed greater stability to higher temperatures and lower pH than the native enzyme. Thus, chemical modification of manganese peroxidase from B. adusta increases its potential usefulness for applied studies.

Workers' exposures and potential health risks to air toxics in a petrochemical complex assessed by improved methodology.

OBJECTIVE: This study was designed to comprehensively evaluate workers' potential health risks of exposure to 39 air toxics in the Ta-sher Petrochemical Complex. METHODS: Open-Path Fourier Transform Infrared Spectroscopy (OP-FTIR) was used to measure concentrations of air toxics. We used the measured worksite concentrations between 1997 and 1999 at 11 companies in the petrochemical complex, employing 3,100 on-site workers. The 39 measured air toxics included 10 chemicals with acute reference exposure levels (RELa), 19 chemicals with chronic reference exposure levels (RELc), and 3 chemicals classified as Class 1 or 2A human carcinogens by the International Agency for Research on Cancer (IARC). We then used RELa to calculate the hazard index of acute health effects (HI ( A )) for workers in individual plants. We also calculated the hazard index of chronic health effects (HIc) and cancer risks for all workers in the entire petrochemical complex. RESULTS: Workers in five companies had HI ( A ) greater than 1 because of toluene, benzene, methyl ethyl ketone, chloroform and isopropanol exposures. Workers in this petrochemical complex had HIc greater than 1 because of acrylonitrile, 1,3-butadiene, hydrogen cyanide, and n,n-dimethylformamide exposures. Risk of hematopoietic system cancer because of benzene and ethylene oxide exposure, and respiratory system cancer because of 1,3-butadiene exposure was estimated to be 3.1-6.1 x 10(-4) and 5.2-7.1 x 10(-4), respectively. CONCLUSIONS: Our findings indicated that workers in the petrochemical complex might have excess cancer and noncancer risks due to acute or chronic exposures to air toxics from multiple emission sources.
