OBJECTIVE: To study neuroprotective effects of astragulus membraneaceus on a neonatal rat hippocampus of hypoxia-ischemia brain damage (HIBD). METHOD: The neonatal hypoxia-ischemia model was established with 7-day-old rat pups. Brain injury was examined by neuron death rate in the hippocampal CA1 area. Caspase-3 (cysteinyl aspartate-specific proteinase) mRNA expression in ipsilateral hippocampal was measured by half-quantitative reverse transcription and polymerization chain reaction (RT-PCR). 90-day-old rats were used in tri-equal-arm maze to observe discrimination learning ability. Sham, model and astragulus-membraneaceus treated groups were set up. RESULT: In model group, caspase-3 mRNA showed an increase at 6h, with maximum arrivimg at 24 h - 48 h after HI. In astragulus-membraneaceus treated group, neurons death rate and caspase-3 mRNA were significantly reduced by astragulus membraneaceus, and discrimination learning ability of developed rats were improved obviously. CONCLUSION: Astragulus membraneaceus has a strong protective effect on neuronal damage in the immature rat hippocampus, which is ralated reducing caspase-3 expression.
OBJECTIVE: To study the influence of Qilong capsule (QLC) on the hemoreology in acute stress blood stasis model rats. METHOD: The model of acute stress blood stasis rats were induced by putting the rats into ice-water between hyodermic epinephrine twice of 1 mg x kg(-1). With the models, the effect of QLC on hemoreology such as whole blood viscosity, whole blood reduction viscosity, plasma viscosity, haematocrit (Hct), erythrocyte deformation and erythrocyte aggregation were observed. RESULT: QLC 0.6, 0.3, 0.15 g x kg(-1) could significantly reduce the increase of whole blood viscosity at high-middle-low shear rate, reduce the whole blood reduction viscosity at middle-low shear rate, reduce Hct and erythrocyte aggregation, and increase the erythrocyte deformation in acute stress blood stasis rats (P < 0.05, P < 0.01, compared with vehicle). QLC 0.6, 0.3 g x kg(-1) could also reduce plasma viscosity (P < 0.05, P < 0.01, compared with vehicle). CONCLUSION: QLC can significantly improve some indexes of hemoreology in acute stress blood stasis rats.
OBJECTIVE: To design DNA microarray and investigate the molecular anti-tumor mechanism of herbs of traditional Chinese medicine. METHOD: cDNA microarrays consisting of 56 probes representing 24 human cell cycle genes were constructed, Four anti-hepatocarcinoma herbs including Radix Linderae, Hebra Artemisiae Annuae, Radix Amebiae, Radix Astragli, were chosen. Effects of herbs on SMMC-7721 cell cycle were observed by flow cytometry assay. Effects of herbs on cell cycle gene expression in SMMC-7721 cells were analyzed by comparing hybridization of Dig-Labeled cDNAs from herb-treated cells and cDNAs from untreated cells. RESULT: Expressions of cell cycle geneswere changed in different degrees after herbs treated. Some genes were down-regulated and some genes were up-regulated. The changes in gene expression agreed with the results of flow cytometry assay. CONCLUSION: The results suggest that these herbs may have effects on cell cycle and DNA damage checkpoint genes which may be the mechanism of the herbs, and DNA microarray can be used to investigate the biological function of extracts of traditional Chinese medicine.
OBJECTIVE: To investigate the effect of Shenluotong on the expression of transforming growth factor-beta1 (TGF-beta1) and extracellular matrix (ECM) in Ang II-induced MCs. METHOD: Fibronectin (FN) and collagen type IV (Col IV) of extracellular matrix were detected by enzyme-linked immunosorbent assay; the expression of TGF-beta1 mRNA were measured by semi-quantitative reverse transcription-polymerase chain reaction. RESULT: A positive correlation between TGF-beta1 and ECM were found in the present study. FN, Col IV and TGF-beta1 mRNA were inhibited by Shenluotong significantly. CONCLUSION: Shenluotong can decrease the accumulation of ECM and inhibit the expression of TGF-beta1, suggesting further that shenluotong can be used to prevent and treat various glomerular diseases and delay glomerular sclerosis.
OBJECTIVE: To study the mechanism of injury of cortical nerve cell in the newborn with hypoxia/ischemia brain damage (HIBD), and the neuroprotective effect of Radix Astragali (RA). METHODS: Neonatal HIBD model rats were established and divided into the sham group, the model group and the RA group. Brain of rats obtained at different time points after HIBD to conduct histopathological examination, neuron death rate count, as well as determination of caspase-3 (cysteinyl aspartate-specific proteinase-3) protein mRNA expression in cerebral cortex by immunohistochemistry, semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) respectively. RESULTS: In the model group, caspase-3 mRNA and protein showed an increase at 6 hrs, reached the peak at 24 hrs, and decreased at 48 hrs after HIBD, on the 5th and 7th day restored to baseline level. After being treated by RA, the neuron death rate of ligated side was obviously reduced, caspase-3 mRNA and protein expression peak value decreased by 45% (mRNA) and 40% - 43% (protein). CONCLUSION: RA shows markedly neuron protection in immature brain cortex after HIBD, which is related with the inhibition on caspase-3 expression.

OBJECTIVE: To compare the effect of Astragulus injection (AGI) and interleukin-2 (IL-2) in enhancing anti-tumor metastasis action of dendrite cells (DCs) based vaccine. METHODS: C57BL/6 mice's myelogenic DCs were prepared and pre-sensitized by Mut1, a MHC class I-restricted tumor antigen polypeptide of Lewis lung cancer. Then the DCs were used to treat mice with metastatic lung cancer in combination with AGI or IL-2. Change of proportion of T-lymphocyte cell subsets in splenic cell was analyzed by flow cytometry, and the serum contents of IL-2 and IL-4 of the tumor bearing mice were detected by ELISA. RESULTS: After being treated with tumor antigen polypeptide sensitized DCs plus AGI or IL-2, the tubercle of lung cancer decreased, proportion of subsets CD4+T and CD8+T in mice's splenic cell increased, and serum IL-2/IL-4 ratio also increased obviously. During the observed period, the tumor developing rate in the immune mice treated with DCs combined treatment, either with IL-2 or with AGI, was lower than that in mice treated with DCs alone. CONCLUSION: Both AGI and IL-2 can enhance the anti-tumor metastasis action of DCs, effectively promote the immune response of tumor bearing host, therefore have obviously inhibitory effect on lung cancer metastasis in vivo. Their immune protective function in normal animals is even more evident.

OBJECTIVE: To evaluate the effects of traditional Chinese complex prescription of Radix Astragali and Rhizoma Ligustici Chuanxiong on urinary albumin excretion (UAE) and vascular endothelial dysfunction in type 2 diabetic patients with microalbuminuria. METHODS: Twenty-one type 2 diabetic patients with microalbuminuria were involved in this before-after study by individual informed consent. Each of the eligible subjects was given the decoction of Radix Astragali and Rhizoma Ligustici Chuanxiong per os 150 ml q.d. for six months. The following examinations were performed at baseline and after treatment: (1) high-resolution ultrasonography to measure the diameter changes of brachial artery in response to reactive hyperemia (endothelium-dependent) and on administration of glyceryl trinitrate (endothelium-independent); (2) high resolution ultrasonography to measure combined intima-media thickness (IMT) of common carotid arteries (CCA); (3) fasting plasma plasminogen activator inhibitor type 1 (PAI-1) activity, C reactive protein (CRP) and malonic aldehyde(MDA) concentration. RESULTS: The patients had impaired endothelial dependent vasodilation (EDV), elevated plasma PAI-1 activity and increased CRP and MDA concentration at baseline. After six months treatment with Radix Astragali and Rhizoma Ligustici Chuanxiong, their urinary albumin-to- creatinine ratio decreased from (86.5 +/- 53.9) microg/mg to (55.05 +/- 51.67) microg/mg (P=0.002). The EDV was improved at the end of the treatment (baseline: 7.49 +/- 2.98%, after treatment: 12.73 +/- 5.36%, P=0.001). Meanwhile, the activity of PAI-1 and the levels of MDA and CRP were significantly decreased CPAI-1: (83.49 +/- 5.11) X 10(-2) AU/ml vs. (79.7 +/- 7.8) x 10(-2) AU/ml, P=0.015; MDA: (3.20 +/- 1.13) nmol/L vs. (2.09 +/- 0.71) nmol/L, P=0.000; CRP: (7.04 +/- 2.64) mg/ L vs. (1.58 +/- 0.69) mg/L, P=0.000]. But no significant changes of the CCA IMT and endothelial independent vasodilation (EIV) were observed. Partial correlated analysis showed that MDA concentration was negatively correlated with EDV (r=-0.3736, P = 0.018). Correlated analysis also showed that CRP was negatively correlated with EDV (r=-0.348, P=0.028). CONCLUSION: Radix Astragali and Rhizoma Ligustici Chuanxiong compound medication may decrease urinary albumin excretion and improve endothelial dysfunction in type 2 diabetic patients with microalbuminuria. The mechanism may relate with the therapeutic effects of Radix Astragali and Rhizoma Ligustici Chuanxiong on anti-inflammation, anti-oxidation and alleviation of the hypo-fibrinolytic/pro-thrombotic state.
A randomized double blind placebo controlled trial to determine the efficacy and safety of combined-herbs (SH) given with zidovudine (ZDV) and zalcitabine (ddC) for the treatment of HIV infection in Thai adults was conducted in 3 hospitals in northern Thailand during 2002 to 2003. The eligible subjects were HIV-infected Thai adults who had never received anti-retrovirals, had a Karnofski Performance Score (KPS) of > or = 70, and had no opportunistic infections. The subjects were randomized to receive either a combination of ZDV 200 mg three times per day, ddC 0.75 mg three times per day, and SH 2.5 g three times per day or a combination of ZDV 200 mg three times per day, ddC 0.75 mg three times per day, and placebo 2.5 g three times per day for 24 weeks. The main outcome measures were HIV-RNA, CD4 cells, and blood chemistry profiles prior to the treatment and then every 4 weeks for 24 weeks. The baseline characteristics of 60 evaluable subjects, 40 in the SH group and 20 in the placebo group, were not significantly different. HIV RNA at week 4 and thereafter was significantly decreased from the baseline value in both groups (p<0.001). However, the decline in HIV RNA in the SH group was significantly more than that in the placebo group. The CD4 cells in the SH group at week 12 and thereafter were significantly increased from the baseline value. Serious adverse events in the two groups were not observed. It is concluded that an addition of SH herbs to two nucleoside reverse transcriptase inhibitors has greater antiviral activity than antiretrovirals only. The SH herbs may be an alternative for the third anti-retroviral agent in the triple drug regimen for the treatment of HIV infected patients in countries with limited resources.
OBJECTIVE: To determine the effects of astragali injection on tubular and its possible mechanisms. METHODS: Sixty patients with primary nephritic syndrome (PNS) were randomly divided into astragali group (n = 30) and control group (n = 30). The surm albumin (sALB) and urinary excretion of retinol-binding protein (RBP), N-Acety-P-beta-Glucosaminidase (NAG), beta2- Microglobulin (beta2MG) were measured before and after the treatment. RESULTS: After the one month treatment, sALB was significantly higher, and ubeta2MG, uNAG, uRBP, and 24 huP were significantly lower in astragali group (P <0.01, P < 0.05). CONCLUSION: Astragali injection on tubular plays a protective role in PNS.
OBJECTIVE: To study the effects of Ligustrazine (Lig) and Radix Astragali (Rad) on activities of myosin adenosine triphosphatase (mATPase) of soleus muscle and atrophy in tail-suspended rat. METHOD: Weightlessness was simulated by tail suspension in female rats. The activities of mATPase of intrafusal and extrafusal fibres in soleus muscle were detected by method of Ca2(+)-ATPase. RESULT: 1) Compared with tail-suspended (TS) group, the percentage of type II fibres of SOL in both Ligustrazine (Lig) group and Rad group decreased distinctly. Furthermore, the percentage of type I and type II fibres of SOL in Rad group showed no difference with control (CON) group. 2) The type I CSA of Lig group was markedly larger than that in TS group, and there was no difference as compared with that of CON group. CSA of type I, II and average CSA of Rad group were remarkably larger than those in TS group, and there were no difference as compared with those of CON group. 3) mATPase activities of intrafusal fibres in both Lig group and Rad group were approximate to that of CON group. CONCLUSION: Lig and Rad are both able to effectively prevent muscle atrophy caused by tail suspension, restrain the slow-twitch muscle transform to fast-twitch muscle and control the increase of mATPase activities caused by weightlessness.
The investigation on the growth, yield and benefit of rape inter- and mixed cropped with milk vetch showed that comparing with sowing rape under zero tillage, the physiological and ecological characters of transplanted rape, including its height, leaf size, root diameter, opening degree and yield were obviously improved. Under zero tillage, mixed cropping had an obvious advantage than inter- and single cropping. After tillage, the yield of transplanted rape under mixed cropping was 11.9% more than that under single cropping with zero tillage. The benefit of transplanting rape under mixed cropping with tillage was the biggest, followed by single cropping rape with zero tillage. It could be concluded that the patterns of transplanting rape under mixed cropping after tillage and zero tillage had the best physiological and ecological effects, not only increasing rape yield and income, but also improving soil fertility.
