Endoproteolytic activity in mammalian brain membranes cleaves 5-hydroxytryptamine-moduline into dipeptides.

This work was intended to determine which enzymatic activities from crude synaptosomal mammalian brain membranes could qualify for the status of 5-hydroxytryptamine-moduline (5-HT-moduline, LSAL, Leu-Ser-Ala-Leu) inactivating enzymes. An enzymatic assay for 5-HT-moduline metabolism was developed using [3H]5-HT-moduline measurement and high performance liquid chromatography (HPLC) technique to identify and quantify 5-HT-moduline metabolites. 5-HT-moduline metabolism displayed all characteristics of metalloprotease activity: sensitivity to divalent ion chelators, reactivation by Zn2+ ions and a pH optimum in the 7-8 range. Bestatin, an aminopeptidase inhibitor, allowed the identification of two enzymatic activities responsible for this metabolism: a bestatin-sensitive aminopeptidase and an endoprotease cleaving 5-HT-moduline into LS (Leu-Ser) and AL (Ala-Leu) dipeptides. This latter enzyme was shown to have a Km of 37.1 +/- 3.6 microM and a Vmax of 5.5 micromol min(-1) l(-1) per mg of protein. Moreover, this enzyme was insensitive to peptidyl dipeptidase A (angiotensin converting enzyme, EC 3.4.15.1), endothelin converting enzyme and neutral endopeptidase (neprylisin, EC 3.4.24.11) inhibitors and displayed some specificity among 5-HT-moduline-analogues and in particular recognized only tetrapeptides. These results, together with the isolation of the LS and AL metabolites [Rousselle, J.C., Massot, O., Delepierre, M., Zifa, E., Rousseau, B., Fillion, G., 1996. Isolation and characterization of an endogenous peptide from rat brain interacting specifically with the serotonergic 1B receptor subtypes. J. Biol. Chem. 271, 726-735] during the purification process of 5-HT-moduline are strong arguments for the physiological implication of this endoprotease in 5-HT-moduline metabolism.

Dipeptide synthesis by an isolated adenylate-forming domain of non-ribosomal peptide synthetases (NRPS).

A deletion mutant of tyrocidine synthetase 1 (DeltaDeltaTY1), comprising the adenylation domain of TY1 as an independent functional adenylate-forming unit, was used to investigate the ability of the adenylation domain in non-ribosomal peptide synthetases to catalyse peptide bond formation from the aminoacyl adenylate intermediate. The results demonstrate that only one substrate amino acid needs to be activated as an aminoacyl adenylate. In view of the potential exploitation of peptide synthetases for enzymatic synthesis of dipeptides of choice, it is important to note that this does not necessarily require a dimodular construct or an intermediate acyl transfer step.

Influence of the structure of cyclodextrins and amino acid sequence of dipeptides and tripeptides on the pH-dependent reversal of the migration order in capillary electrophoresis.

The pH-dependent reversal of the migration order in cyclodextrin (CD)-mediated capillary electrophoresis (CE) enantioseparations of dipeptides and tripeptides has been studied between pH 2.5 and 3.5 using beta-CD and several of its neutral derivatives. The occurrence of the phenomenon depended on both the structure of the CD and the amino acid composition and sequence of the peptides. While an inversion was observed for several peptides when native beta-CD, dimethyl-beta-cyclodextrin or trimethyl-beta-cyclodextrin were added to the run buffer, no alteration of the order occurred in the presence of permethyl-beta-cyclodextrin or hydroxypropyl-beta-cyclodextrin. Most peptides that displayed a change of the migration behavior upon increasing the buffer pH contained Phe at the C-terminus. An ionizable carboxyl group in the peptide structure was a prerequisite. As seen with other uncommon migration effects in CE, the pH-dependent reversal of the migration order occurred in the pH region of the pKa values of the peptide carboxyl functions.

Low temperature high-performance liquid chromatography of cis-trans proline dipeptides.

Isomeric dipeptides which contain proline at the C-terminus and undergo on-column cis-trans isomerization were separated by reversed-phase chromatography at subambient column temperature. Chromatography at low temperatures offers a convenient method for measuring the relative concentrations of the isomers and, thus, the calculation of the equilibrium constants for such cis-trans isomerization at various conditions of solvent, pH, and temperature. Pure fractions of the cis and trans isomers were collected and used in kinetic studies of the isomerization. High-performance liquid chromatography at temperatures near the freezing point of the eluent is a potentially useful tool for the study of a wide range of biochemical molecules.

Determination of the chiral purity of dipeptide isosteres containing a reduced peptide bond by gas chromatographic analysis.

The stereochemical purity of psi (CH2-NH) dipeptides has been determined using gas chromatography-mass spectrometry. Different structures were found due to the derivatization procedures. A selective preparation of the linear bistrifluoroacetylated derivative and the monotrifluoroacetylated lactam makes it possible to monitor the chiral purity of the pseudodipeptides synthesized. Racemization occurring during peptide hydrolysis can be differentiated from racemization during the synthesis by using deuterium labelling. The method allows the optimization of the synthesis protocols and will be useful for further monitoring of the chiral purity of the pseudopeptides synthesized.

Methionine dipeptide stationary phases for the resolution of enantiomers.

N-Trifluoroacetyl-L-methionyl-L-methionine cyclohexyl ester as well as the disulfide and disulfone derivatives of this phase have been synthesized. The methionine phase exhibits improved properties as a stationary phase for the separation of amino acid enantiomers as compared to other dipeptide stationary phases and can be used at temperatures ranging from 70 to 150 degrees. The sulfoxide phase, however, was incapable of resolving the enantiomeric derivatives. Physical properties of the sulfone compound limit its use as an effective resolving agent.

Chiral high-performance liquid chromatography of aromatic cyclic dipeptides using cyclodextrin stationary phases.

A series of enantiomers of cyclic and linear dipeptides containing aromatic amino acids was prepared and chromatographed on beta- and gamma-cyclodextrin (CD) columns. The retention times, separation factor alpha and resolution values were calculated. The relevance of the distance of the chiral center from the phenyl ring for chiral resolution was studied. A model was developed using X-ray crystallographic data for an inclusion complex of beta-CD and the enantiomers of cyclic (Phe-Gly).

Enantioseparation of dipeptides by capillary electrochromatography on a teicoplanin aglycone chiral stationary phase.

This work deals with investigations on the enantioseparation of glycyl-dipeptides by capillary electrochromatography (CEC) on a capillary packed with teicoplanin aglycone immobilized on 3.5 microm silica gel. The results were compared to those obtained with micro-HPLC using the same chiral stationary phase. Polar organic and reversed-phase mode were checked, whereby the latter showed better results. Out of 12 glycyldipetides investigated, all compounds showed baseline separation with Rs values up to 20. Plate numbers were in the range of 10,000-300,000/m. The choice of organic modifier was found to be crucial. While methanol increased retention time, acetonitrile reduced it. A ternary mixture of ethanol-acetonitrile-aqueous triethylamine acetate solution pH 4.1 was found to be a useful compromise, providing excellent resolution with retention times less than 25 min. Efficiency and resolution were generally found to be higher in CEC than with micro-HPLC.

Chiral separations using polymeric dipeptide surfactants: effect of number of chiral centers and steric factors.

Two polymeric dipeptide chiral surfactants (PDCSs), poly sodium N-undecanoyl isoleucyl-valinate (SUILV) with three chiral centers and poly sodium N-undecanoyl leucyl-valinate (SULV) with two chiral centers, have been evaluated and compared as chiral pseudo-stationary phases in electrokinetic capillary chromatography. The performance of these surfactants, in terms of enantioselectivity was examined using anionic, cationic and neutral analytes. Analyses of the data suggest that the enantiomeric resolutions of the analytes with these two PDCSs are dependent upon steric factors rather than number of stereogenic centers.

Synthetic analogs for oxovanadium(IV/V)-glutathione interaction: an NMR, EPR, synthetic and structural study of oxovanadium(IV/V) compounds with sulfhydryl-containing pseudopeptides and dipeptides.

The reaction of [VO(CH3COO)2(phen)] (phen = 1,10-phenanthroline) with the sulfhydryl-containing pseudopeptides (scp), N-(2-mercaptopropionyl)glycine (H3mpg), N-(2-mercaptopropionyl)cysteine (H4m2pc), N-(3-mercaptopropionyl)cysteine (H4m3pc) and the dipeptides glycylglycine (H2glygly) and glycyl-L-alanine (H2glyala), in the presence of triethylamine, results in the formation of the compounds Et3NH[VO(mpg)(phen)] (1), (Et3NH)2[VO(m2pc)] (4), [(Et3NH)2[VO(m3pc) (5), [VO(glygly)(phen)] x 2CH3OH (2 x 2CH3OH) and [VO(glyala)(phen)] x CH3OH (3 x CH3OH). Evidence for the molecular connectivity in 2 x CH3OH was established by X-ray crystallography, showing the vanadium(IV) atom ligated to a tridentate glygly2- ligand at the N(amine), N(peptide) and O(carboxylato) atoms. Combination of the correlation plot of the EPR parameters gz versus Az, together with the additivity relationship supported the prediction of the equatorial donor atom sets of the V(IV)O2+ center at various pH values for the V(IV)O2+-glutathione system considered in this study. Model NMR studies (interaction of vanadium(V) with the scp H3mpg) showed that there is a possibility of vanadium(V) ligation to glutathione.

Dipeptide vinyl sulfones suitable for intracellular inhibition of dipeptidyl peptidase I.

In granules of hematopoetic cells, dipeptidyl peptidase I (DPPI) processes inactive proenzymes into active enzymes, e.g., lymphocyte progranzyme A. Our goal was to develop irreversible inhibitors of intracellular DPPI. First, we identified inhibitors with aqueous stability. Then we determined which inhibitors were nontoxic, could enter cells and inactivate intracellular DPPI. We screened nine dipeptide vinyl sulfone (VS) inhibitors (kobs/[I] > 72 M-1 s-1) and found six that were nontoxic. Four affected intracellular DPPI at < 25 microM. These compounds contained only uncharged amino acid residues; the two less reactive compounds contained charged Glu residues. The best one, Leu-Phe-VS-CH3, inactivated DPPI in cells with an ID50 of approximately 5 microM. This inhibitor was not the best inhibitor of purified DPPI. Longer aqueous stabilities were important predictors of cellular efficacy. Leu-Phe-VS-CH3 had a half life of 97 min at the pH of the extracellular medium (7.5) and 1302 min at pH 5.5 (the intracellular environment of DPPI). This VS had no direct effect on granzyme activities. In contrast, the diazomethyl ketone inhibitor Gly-Phe-CHN2 inhibited chymase activity. Several good intracellular DPPI VS inhibitors lacked reactivity with cathepsins B, H and L. In conclusion, we have identified DPPI inhibitors suitable for cellular applications.

Reciprocal effect of optical isomerism of EW-dipeptides on immune response.

Dipeptides of the EW-sequence, consisting of D-amino acids, were shown to inhibit spleen colony formation both after in vitro treatment of bone marrow and after the peptide injection to the donor 2-48 h prior to bone marrow taking. The inhibiting effect of D-(EW) peptides can be eliminated by injection of thymic cells to the recipient. L-(EW) peptides have no influence on colony forming activity of intact bone marrow cells but stimulate regeneration of colony formation by irradiated (1 Gy) bone marrow. Unlike L-(EW), the D-(EW) peptides suppress the development of hemopoietic stem cells into the S-phase. Supposedly, D-(EW) represent a new generation of immunoactive peptides possessing of inhibiting activity.

Synthesis of ZAlaPheOMe, the precursor of bitter dipeptide in the two-phase ethyl acetate-water system catalysed by thermolysin.

Enzymatic synthesis of ZAlaPheOMe, the precursor of a bitter dipeptide due to a very strong hydrophobic character of this compound was carried out in a two-liquid-phase ethyl acetate-0.1 moll(-1) Tris-HCl buffer system. The selected system and process conditions (60 degrees C, pH 7.0) ensured high activity and stability of thermolysin, a catalyst of this reaction. Owing to an almost complete extraction of the product to organic phase it was possible to obtain conversion degrees close to unity. The reaction phase was the aqueous phase in which almost whole catalyst mass was accumulated. The amount of protein adsorbed on the interface corresponded to 4-5% of protein dissolved in the aqueous phase. In the description of the enzymatic process rate the equation suitable for the mechanism of tetrahedral complex formation at a random order sequence of substrate bonding was used. The values of constants k=0.222 s(-1), K(mA)=11.5 mmoll(-1), K(mB)=149.3 mmoll(-1) were obtained.

Carboxypeptidase A-catalyzed dipeptide synthesis in organic media.

The carboxypeptidase A-catalyzed syntheses of dipeptides from L-amino acids (Phe, Tyr, Trp, Leu and Ile) were studied in various water-miscible (acetone, acetonitrile, ethanol, methanol and 1,4-dioxane) organic solvents. The highest yield (43%) was achieved in acetonitrile with L-Phe as substrate, after a 24-h incubation. The optimal conditions of Phe-Phe synthesis in acetonitrile were determined. For maximal conversion 1.2 mM L-Phe, 1.4 mg ml-1 enzyme and about 10% water are needed in buffered aqueous acetonitrile (pH 5.5) at 30 degrees C.

Dipeptide model prodrugs for the intestinal oligopeptide transporter. Affinity for and transport via hPepT1 in the human intestinal Caco-2 cell line.

The human intestinal di/tri-peptide carrier, hPepT1, has been suggested as a drug delivery target via increasing the intestinal transport of low permeability compounds by designing peptidomimetic prodrugs. Model ester prodrugs using the stabilized dipeptides D-Glu-Ala and D-Asp-Ala as pro-moieties for benzyl alcohol have been shown to maintain affinity for hPepT1. The primary aim of the present study was to investigate if modifications of the benzyl alcohol model drug influence the corresponding D-Glu-Ala and D-Asp-Ala model prodrugs' affinity for hPepT1 in Caco-2 cells. A second aim was to investigate the transepithelial transport and hydrolysis parameters for D-Asp(BnO)-Ala and D-Glu(BnO)-Ala across Caco-2 cell monolayers. In the present study, all investigated D-Asp-Ala and D-Glu-Ala model prodrugs retained various degrees of affinity for hPepT1 in Caco-2 cells. These affinities are used to establish a QSAR of our benzyl alcohol modified model prodrugs, aided at elucidating the observed differences in model prodrug affinity for hPepT1; additionally, these data suggest that the hydrophobicity of the side-chain model drug is the major determinant in the compounds affinity for hPepT1. Transepithelial transport studies performed using Caco-2 cells of D-Asp(BnO)-Ala and D-Glu(BnO)-Ala showed that the K(m) for transepithelial transport was not significantly different for the two compounds. The maximal transport rate of the carrier-mediated flux component does not differ between the two model prodrugs either. The transepithelial transport of D-Asp(BnO)-Ala and D-Glu(BnO)-Ala follows simple kinetics, and the release of benzyl alcohol is pH-dependent, but unaffected by 1 mM of the esterase inhibitor Paraoxon in 80% human plasma and Caco-2 cell homogenate.

Adamantylamide dipeptide stimulates hematopoiesis and increases survival in irradiated mice.

It has been demonstrated that the synthetic immunostimulatory compound, adamantylamide dipeptide (AdDP) produces hematopoiesis-stimulating effects in mice exposed to sublethal doses of ionizing radiation and increases survival in experimental animals irradiated with a lethal dose. These findings might suggest contingent extension of clinical indications for the administration of AdDP for the conditions of hematopoietic suppression, especially in oncology.

Stimulatory effect of zinc-chelating dipeptide on deoxyribonucleic acid synthesis in osteoblastic MC3T3-E1 cells.

Whether deoxyribonucleic acid (DNA) synthesis in osteoblastic MC3T3-E1 cells is stimulated by zinc, an activator of bone formation, was investigated in vitro. After subculture for 3 days, the cells were cultured for up to 3 days (72 h) with zinc sulfate or zinc-chelated dipeptide (beta-alanyl-L-histidinato zinc; AHZ) in the range of 10(-7) to 10(-5) M. The culture with zinc compounds (10(-5) M) produced a significant increase of cell number, DNA content, and protein concentration in the cells, as reported previously. The culture with zinc compounds (10(-6) and 10(-5) M) clearly stimulated DNA synthesis in the homogenate, when it was estimated by the incorporation of [3H]deoxythymidine 5'-triphosphate into the DNA in the homogenate of cells. The AHZ effect was greater than that of zinc sulfate. The culture together with cycloheximide (19(-6) M) completely abolished the zinc compounds (10(-5) M)-induced increase of DNA synthesis in the cells, suggesting that the zinc compound effect is based on a newly synthesized protein component. Moreover, when zinc sulfate (10(-7) and 10(-6) M) or AHZ (10(-8) to 10(-5) M) was added into the reaction mixture with the homogenate of cells cultured without zinc compounds, the DNA synthesis was clearly increased. The effect of addition of zinc compounds (10(-6) M) on the DNA synthesis was completely inhibited by the presence of staurosporine (10(-8) M), an inhibitor of protein kinase C, or okadaic acid (10(-7) M), an inhibitor of protein phosphatase. The present study demonstrates that zinc compounds have a stimulatory effect on DNA synthesis in osteoblastic cells.

Synthesis and activity of pyrrolidinyl- and thiazolidinyl-dipeptide derivatives as inhibitors of the Tc80 prolyl oligopeptidase from Trypanosoma cruzi.

Pyrrolidinyl- and thiazolidinyl- dipeptide derivatives, featuring either a vinyl sulfone-, a 2-ketobenzothiazole-, a nitrile-, or a benzimidazole group at the C-terminus, were designed and synthesized as potential inhibitors of the prolyl-specific Tc80 proteinase from Trypanosoma cruzi, the agent of Chagas' disease. These compounds were evaluated in vitro towards the target enzyme which was classified as a serine protease belonging to the prolyl oligopeptidase family (EC 3.4.21.26). A peptidyl nitrile and two peptidyl alpha-ketobenzothiazoles were shown to be potent reversible and competitive inhibitors of Tc 80 proteinase, with K(i) values in the range 38-219 nM, and compared advantageously with some known mammalian prolyl oligopeptidase inhibitors.
