Proflavine Uptake and Release in Sensitive and Resistant Escherichia coli.

Both Escherichia coli B and a proflavine-resistant mutant, E. coli B/Pr, took up the same amounts of proflavine when suspended in buffer containing the dye. In growth media, however, sensitive cells took up more proflavine than did resistant cells. Adding growth media or any one of several constituents of these media, including amino acids, glycerol, pyruvic acid, and metabolizable sugars, to resistant cells that had taken up proflavine in buffer caused them to lose the dye, but had less or no effect on sensitive cells. Certian salts caused an equal release of proflavine from resistant and sensitive cells. Proflavine released from resistant cells by glucose was not changed chemically. The effects of temperature and metabolic inhibitors suggest that proflavine uptake is a passive process but that its release may be an active one, dependent on metabolism. Glucose had more effect on the proflavine binding of E. coli B grown in a minimal medium than on that of cells grown in a complex medium. E. coli B was less susceptible to proflavine when growing in a minimal medium. The effects of other synthetic media on proflavine susceptibility of E. coli B were also studied. Deoxyribonucleic acid and envelopes from sensitive and resistant cells bound the same amounts of proflavine, and no difference was seen in the site of dye binding when sensitive and resistant cells that had taken up proflavine in buffer were sonically broken and fractionated. The results suggest that sensitive and resistant cells are equally permeable to proflavine but differ in the ease with which metabolites cause them to release bound proflavine. So far, however, these differences do not account completely for the ability of resistant cells to grow in high proflavine concentrations.

Production of a bioflocculant by mixed culture.

Colony groups that form large amounts of slime externally were obtained from activated sludge among phthalate-assimilating microbes. That slime, which R-3 mixed microbes produced externally, had a high level of flocculation activity. R-3 mixed strains, one of such colony groups, efficiently produced a bioflocculant (APR-3) in liquid cultures of production medium, especially that containing starch and glucose (1:1) as carbon sources. Identification of this group of bioflocculant-producing microbes (R-3 mixed strains) showed that it was comprised of four strains belonging to the genera Oerskovia, Acinetobacter, Agrobacterium, and Enterobacter. The bioflocculant (APR-3) was purified electrophoretically to homogeneity by ethanol and CPC precipitation. Its molecular mass is at least 2 x 10(6) Da. APR-3 is an acidic polysaccharide made of glucose, galactose, succinic acid, and pyruvic acid (molar ratio: 5.6:1:0.6:2.5).

Extracellular acidic polysaccharide production by a two-membered bacterial coculture.

A two-membered coculture of strains KYM-7 and KYM-8, identified as Cellulomonas cellulans and Agrobacterium tumefaciens, respectively, produced a large amount of an extracellular polysaccharide, designated APK-78, from starch. Each strain in pure culture produced only very little amount of polysaccharide from starch; the coexistence of the two strains from the early stage of cultivation was indispensable for a large amount of polysaccharide to be produced. The polysaccharide APK-78 was acidic and composed of glucose, galactose, succinic acid, and pyruvic acid with a molar ratio of 8.1:1.0:1.7:1.0, indicating that it is a succinoglycan type of polysaccharide.

Ringer's ethyl pyruvate solution: a novel resuscitation fluid.

Reactive oxygen species (ROS) have been implicated in the pathogenesis of the structural and functional alterations to tissues that are associated with a variety of pathological processes, including: sepsis and septic shock, thermal injury, doxorubicin-induced cardiomyopathy, hemorrhagic shock, and mesenteric ischemia/reperfusion (I/R) injury. Pyruvate (CH3COCOO-), a small molecule that is normally regarded as a key intermediate in the oxidative or anaerobic metabolism of glucose, is also a potent and effective ROS scavenger. Unfortunately, the usefulness of pyruvate as a therapeutic agent is abrogated by its very poor stability in solution. In an effort to take advantage of the ability of pyruvate to scavenge ROS while avoiding the problems associated with the instability of pyruvate in solution, we have developed a novel resuscitation fluid, which consists of a simple derivative of pyruvic acid, ethyl pyruvate, dissolved in a calcium-containing balanced salt solution. We call this solution Ringer's Ethyl Pyruvate Solution (REPS), and have shown in preliminary studies that treatment with REPS can improve outcome in a variety of animal models of critical illness.

Absorption and fluorescence studies on interaction between cationic dyes and Klebsiella K7 capsular polysaccharide.

Interaction of cationic dyes, pinacyanol chloride, acridine orange and phenosafranin, with Klebsiella K7 capsular polysaccharide has been investigated by spectrophotometric and spectrofluorometric measurements. The acidic polysaccharide induce a metachromatic blue shift of the absorption band of pinacyanol chloride from 600 nm to 495 nm, indicating strong metachromasy. Stoichiometry of polyanion and dye cation (1:1.5) in the polymer-dye compound formed by the interaction between pinacyanol chloride dye and K7 polymer indicate that both glucuronic acid and pyruvic acid act as the potential anionic sites for interaction. Both spectrophotometric titration of pinacyanol chloride and spectrofluorometric titration of acridine orange and phenosafranin dyes by the polymer gave quite comparable equivalent weights for the polymer. Dye-polymer interaction studies indicated induction of metachromasy in the cationic dye by the anionic biopolymer, establishing its chromotropic character.

[A case with late-onset MELAS with hallucination and delusion].

We report a 53-year-old male patient with late onset mitochondrial myopathy, encephalopathy, lactic acidosis and stroke-like episodes(MELAS) with hallucination and delusion. The patient manifested various neurological symptoms including perceptive deafness, muscle weakness of limbs with loss of consciousness, sensory abnormalities in hands, feet and a face, abnormal sense of taste, tremor, palsy of upward eye movement and weak deep tendon reflexes prior to the psychotic episode. He was diagnosed as MELAS, because of high serum lactic acid and pyruvic acid, and the point mutation in the mitochondrial DNA 3243. SPECT imaging showed decreased perfusion in occipital cortex and thalamus. These SPECT changes improved after disappearing visual hallucination. Hallucination might be caused by delirium due to stroke-like episode. Dysfunction in the occipital cortex and thalamus might be involved with this perfusion change.

Antitumor effect of geranylamine derivatives on human hepatoma.

In this study, we examined the antitumor activities of isoprenoid derivatives conjugated with substrates of energy metabolism in human hepatoma-bearing athymic mice. Among these compounds, N-geranylpyruvic amide, N-geranyl-p-pyruvaminobenzoic amide, N,N'-digeranylmalic diamide and N,N'-digeranyl-O-acetylmalic diamide had strong antitumor effects. These geranylamine derivatives also inhibited in vitro cell growth. Sugar conjugates of geranylamine, geranic acid and mevalonic acid did not show any antitumor effect in vivo or in vitro. Although the geranylamine derivatives had no impact on the cell cycle distribution at 24 h, a sub-G1 (apoptotic) peak of varying magnitude was seen in DNA histograms of cells treated with the derivatives for 48 h. However, the geranylamine derivatives did not inhibit protein isoprenylation, which has been reported in cancer cells treated with several natural isoprenoids. These results suggest that the geranylamine derivatives conjugated with malic acid and pyruvic acid have a different mechanism of antitumor activity from that of natural isoprenoids.

[Oculopharyngeal muscular dystrophy in a Japanese family].

The first instance of familial oculopharyngeal muscular dystrophy (OPMD) affecting a Japanese family is reported. Three patients (a 62-year-old female, her sisters 66-year-old and 53-year-old) were described with suspicious other 2 cases. A 62-year-old woman (case 1) developed bilateral blepharoptosis at the age of 52. Then she became aware of difficulty in swallowing solid foods, had developed a nasal voice and aspiration of liquids. On admission, neurological examination revealed moderate bilateral blepharoptosis, nasal voice, dysphagia and hyporeflexia of the pharynx. There was mild weakness of the muscles of the temporalis, masseter, face, neck and proximal portions of the upper limbs. The levels of serum creatine phosphokinase, lactic acid and pyruvic acid were normal. Tensilon test was negative. The needle EMG showed a myogenic pattern with no waning phenomenon. Nasopharyngeal fiberscopy, laryngoscopy, esophageal fluoroscopy and hydrodynamic examination showed dyskinesis of the soft palate, retention of saliva in recessus piriformis and streaming into the larynx. Cricopharyngeal myotomy was performed for the purpose of relieving the dysphagia. The muscles were obtained from cricopharyngeus of both sides during surgery, and right deltoid muscle in biopsy. The muscles of sternohyoideus and deltoideus showed myogenic changes, and some fibers with rimmed vacuoles especially in small angulated fibers under the light microscope. Whereas the crycopharyngeus showed dystrophic change, which was apparent in the right side. There were also nemaline rods found in a few fibers undergoing necrosis. ATPase preparations revealed type 1 predominant in cricopharyngeus and type 2 predominant in sternohyoideus. Most atrophic fibers were in type 1 fibers.(ABSTRACT TRUNCATED AT 250 WORDS)

[Placental perfusion. Observations based on examinations made at the Institute of Obstetrics and Gynecological Diseases of the Medical academy in Lublin in the years 1975-1984].

The paper presents the authors' experience resulting from the research into placental perfusion. The use of perfusion technique depended on a working hypothesis. The authors use the method of perfusion of foetal circulation. They analysed vascular resistance on the foetal side of the placenta, glucose consumption, production of lactic acid and pyruvic acid. They also analysed Dolantine, serotonin, papaverin, orciprenalin, phenoterol, theophylline ethylenediamine, isoxysuprin, propranolol, sodium fluoride. Basing on the results of the investigation of substances stimulating the betaadrenergic receptor, the authors came to the conclusion that there exists primary and secondary anoxia syndrome.

Degradation of gallic acid by Aspergillus flavus.

Aspergillus flavus utilized gallic acid as sole carbon source and increased in presence of glucose and sucrose. The enzymes were inducible in nature and during degradation of gallic acid. 4-carboxy, 2-hydroxy cis, cis-muconic acid and pyruvic acid were detected. The estradiol (meta) fission of gallic acid by A. flavus was postulated.

[An experimental study of PGE1 infusion into the liver circulation].

The present study was performed to determine the effects of PGE1 on the liver circulations. Adult mongrel dogs were used for this experimental study. Following the continuous infusion of PGE1 to the hepatic artery (HA) and portal vein (PV), the change of hepatic arterial flow (HAF) and portal venous flow (PVF) was measured by the electromagnetic flow meter, and hepatic tissue flow (HTF) by the hydrogen clearance meter. Hepatic enzymes were also measured at the same time. A single infusion of saline was done for the control. HAF and HTF increased significantly by the continuous infusion of over 0.5ng/Kg/min of PGE1 to HA. PVF and HTF also showed an increase by the continuous infusion of 100ng/Kg/min of PGE1 to PV. Compared with the control group, lactate acid and pyruvate acid levels remained low in the PGE1 infused group. The optimum dose of PGE1 to HA was thought to be 1.0-2.0ng/Kg/min. The results of experimental studies suggest that the direct infusion of PGE1 to the liver may prevent the ischemic change of hepatic metabolism during and after the surgical interventions.

The conversion of L-cystathionine into the cyclic ketimine form by heated rat liver extracts containing cystathionase and transaminase activities.

Rat liver homogenates heated for 10 min at 60 degrees C incubated with L-cystathionine yield cystathionine ketimine which was identified by its typical UV spectrum and by cochromatography with authentic samples on the amino acid analyzer. Alanine and alpha-amino butyric acid have been also detected among the final products. The reaction is due to heat stable gamma-cystathionase and transaminases present in the extracts. Cystathionase produces alpha-keto butyric acid and pyruvic acid which are then used for the transamination of the remaining cystathionine to yield the ketimine. This is the first report indicating the occurrence in a mammalian tissue of an enzymatic system using cystathionine for reactions differing from the traditional transulfuration to cysteine.

[Energetic metabolism of the placenta and vascular blood flow as criteria for the choice of drugs in risk pregnancy--results of our investigations].

Object of this paper was to estimate the vascular resistances of the placenta and the lactate/pyruvate-quotient influenced by 8 and 40 mu/min orciprenaline and 320 micrograms/min papaverine. 38 placentas were tested in extraorganic circulation. The content of lactic acid and pyruvic acid was determined in the perfusion solution flowing out of the umbilical vein. Pressure was registered with cymograph. Infusion of 8 micrograms/min orciprenaline diminishes the lactate/pyruvate-quotient and the vascular resistances. 40 micrograms/min orciprenaline causes spasm of placental vessels in the terminal phase of examination. In this group the processes of oxygenation were limited by the oxygen content in the perfusion solution. Papaverine turned the metabolism of the placenta to an anaerob way and the antispasmodic effect was shortlasting.--Therefore the drug effect results out of a metabolic and a spasmolytic component. In risk pregnancy there has to be considered in addition to the spasmolytic the metabolic effects, too.

[Initial ultrafiltration to the priming solution with preserved blood for cardiopulmonary bypass in infants].

In case of open heart surgery in infants, the initial priming solutions (IPS) of the cardiopulmonary bypasses (CPB) include considerable amount of preserved blood to assure us of the proper hematocrit during the CPB. Aiming at elimination of unfavorable effects of preserved blood on hearts and vessels, the ultrafiltrations (UF) to the IPS before the beginnings of CPB had been carried out in 42 pediatric cases. The IPS amounted to 915 +/- 25 (mean +/- SE) ml. in which 581 +/- 31 ml of preserved blood were included. The 1.5-fold amount infusions over the IPS, consisted of 5% glucose, normal saline and fresh frozen plasma, were added to the IPS, and just the same amount fluid were removed out by the UF. The concentrations of potassium, NH3, lactic acid and pyruvic acid in the IPS decreased significantly after UF (p = 0.001). The potassium concentrations were compared among the blood of patients (PB), the IPS before UF (IPS1), the IPS after UF (IPS2), and the mixed blood drawn 5-10 minutes after the beginnings of CPB (MB) in the cases under the age of 1 year (group-I, n = 26, 4.7 +/- 0.3 kg) and in the elders (group-E, n = 16, 15 +/- 2 kg). The results showed all significant differences but "IPS2 and MB" in group-I, and but "PB and MB" in group-E (ANOVA p = 0.001, p < 0.01 by Newman-Keuls). [Structure of the repeating unit of the intracellular polysaccharide from Mycobacterium convolutum 240].

An acidic extracellular polysaccharide composed of D-glucose, L-fucose, D-glucuronic acid and pyruvic acid residues has been isolated from the Mycobacterium convolutum culture fluid. Activities of superoxide dismutase and catalase in erythrocytes and transaminases in the plasma of carps (Cyprinus carpio L.) exposed to cadmium.

Total superoxide dismutase (SOD, EC 1.15.1.1) and catalase (CAT, EC 1.11.1.6) activities in erythrocytes and the glutamic acid-oxalacetic acid-transaminase (GOT, EC 2.6.1.1) and glutamic acid-pyruvic acid-transaminase (GPT, EC 2.6.1.2) activities in the plasma were measured in experimental groups of carps (Cyprinus carpio L.) exposed to cadmium in a concentration of 20 mg Cd/l water under aquarium conditions for 6, 12, 18 and 24 hours and in control fishes. It was shown that the total activity of SOD in the erythrocytes is significantly decreased after 12, 18 and 24 hours of cadmium exposure. Increased activities of CAT (after 24 hours) in the erythrocytes and GOT and GPT in the plasma were found in cadmium-treated fishes. At the same time the concentration of blood haemoglobin and haematocrit values were significantly diminished. These results indicate that cadmium causes oxidative stress and tissue damage in the exposed fishes.

PRELIMINARY ENZYMATIC EVENTS IN ASPARAGINE-DEPENDENT DENITRIFICATION BY PSEUDOMONAS PERFECTOMARINUS.

Bacteriol. 89:1051-1054. 1965.-The initial events leading to denitrification by Pseudomonas perfectomarinus grown anaerobically in the presence of NO(3) (-) were investigated. Conversion of asparagine to malic acid was demonstrated. The nicotinamide adenine dinucleotide phosphate-linked malic acid dehydrogenase in this bacterium was shown to have a dependence for a divalent cation. A transhydrogenase was demonstrated. Oxaloacetic acid, a likely intermediate in conversion of malic to pyruvic acid, was rapidly decarboxylated by enzymes in cell-free extracts. We were not able to show reversible formation of malic acid with oxaloacetic acid and reduced nicotinamide adenine dinucleotide phosphate, nor of oxaloacetic acid with pyruvic acid and CO(2).
