OBJECTIVE: To study the effects and mechanism of Yishenjiangyafang, a prescription of Chinese traditional herbs used for renal hypertension, on blood pressure and protecting renal function of RPH rats. METHOD: The 5/6 kidney of rat was resected to set up the RPH rat model. Blood pressure, Cr(creatinine), Ccr(creatinine clearance) and BUN(urea nitrogen) were measured dynamically. After eight weeks treatment, plasma content of PAR A II TXB2, 6-keto-PGF1 were measured. At same time, The change of renal pathology was observed. RESULT: Yishenjiangyafang could reduce blood pressure Cr, but had no effect on BUN of RPH rat. The indexes of PAR, A II of each different dosage group of Yishenjiangyafang were decreased. At the same time, it reduced plasma content of TXB2, and increased 6-keto-PGF1 alpha. Glomerulosclerosis and atrophy of renal tubule in Yishenjiangyafang group RPH rats were better than those of the contrast group and the Capten group. CONCLUSION: Yishenjiangyafang can reduce blood pressure of RPH rats and has protective effects on its kidney. Yishenjiangyafang can perform its effects of reducing blood pressure and protecting kidney by influencing the RAS of RPH rats.
OBJECTIVE: TO establish a RP-HPLC method for the determination of calycosin-7-O-beta-D-glucopyranoside in Radix Astragali, and to analyse the calycosin-7-O-beta-D-glucopyranoside content of ten samples of Radix Astragali, collected from different regions. METHOD: A Polaris C18(250 mm x 4.6 mm, 5 microns) column was used and a mixture of methanol-water (30:70) was used as the mobile phase at a flow rate of 1.0 mL.min-1. The column temperature was 25 degrees C and the UV detection wavelength was 254 nm. RESULT: The calibration curve was in good linearity over the range of 0.0106-2.12 micrograms with the regression equation Y = 3035. 97 X - 14.85(r = 0.9999). The average recovery was 95.8% (n = 5, RSD = 1.3%). CONCLUSION: The method is simple, quick, sensitive and reproducible. In all of the samples, the calycosin-7-O-beta-D-glucopyranoside contents differ markedly.
OBJECTIVE: To observe the effects of Shenqifuxin oral liquid(SQFXOL) on plasma kaliuretic peptide (KP), atrial natriuretic polypeptide(ANP), angiotension II (Ang II), endothelin(ET) and the left ventricular remodeling and the myocardial contractility and relaxation of left ventricle in experimental rats with heart failure(HF). METHOD: The SD rat model with HF was produced by constricting abdominal aorta. Hemodynamic parameters including maximum rate of intraventricular pressure rise (+dp/dtmax), left ventricular systolic pressure(LVSP), maximum velocity of contractile element shortening(Vmax), maximum rate of intraventricular pressure down(-dp/dtmax) and left ventricular end diastolic pressure(LVEDP) were measured by the method of the catheterization. Plasma concentrations of KP, ANP, Ang II and ET were determined by radioimmunoassays. The effects of treatment were evaluated by observing and comparing the changes of heart morphological structure, collagen element, heart weight/body weight ratio (HW/BW), left intraventricular area(LVA) and myocardial nuclei number (MNN) per square area. RESULT: In high dose SQFXOL group, the LVSP, -dp/dtmax and Vmax were increased, while LVEDP was decreased, and plasma concentrations of KP, Ang II and ET were decreased. In comparision with those in model group, the difference was significant(P < 0.05 or P < 0.01). Though the +dp/dtmax and the level of ANP were decreased, the difference was insignificant(all P > 0.05). The collagen tissues around myocardial cells were reduced. HW/BW and LVA were lower, and MNN per square area was higher significantly (P < 0.05 or P < 0.01). The indices of +dp/dtmax in all of treatment groups and control group were not considerably different in comparison with those in model group. The levels of plasma ANP in middle dose group and low dose group were significantly lower than those in model group(all P < 0.01). CONCLUSION: SQFXOL can reduce the plasma concentrations of KP, Ang II, ET, and ANP, improve the myocardial contractility and relaxation of left ventricular and inhibitate left ventricular remodeling in rats with HF.
OBJECTIVE: To observe the effect of different drug dosage to activate blood circulation and to nourish Qi on cardiac function and structure of heart failure rats after acute myocardial infarction. METHOD: The congestive heart failure post cardiac infarction rats models subjected to left coronary occlusion were made and given different dosage of drug of Huoxue Yiqi and captopril (positive control) from 4 weeks to 8 weeks after operation. The cardiac function before and after using drugs were observed by impedance methods. RESULT: Treated by different dosage of drug of Huoxue Yiqi and captopil, cardiac function of heart failure rats after acute myocardial infarction improved, SV, CO, CI(P < 0.01); at the same time, prescription of Huoxue Yiqi and Qixue could improve the model rats' heart coefficient, VS SHAM(P > 0.05), but prescription of Yiqi Huoxue and captopril could not improve it. CONCLUSION: Drug to activate blood circulation and to increase qi can treat heart failure rats after acute myocardial infarction, through improving their function and structure. At the same time, more dosage of drug to activate blood circulation and to increase qi(Huoxue/Yiqi drug) may improve the model rats' heart coefficient.

The degree and range of myocardial ischemia, myocardial infarct size and the activity of serum CK, LDH were decreased in acute myocardial infarcion dogs treated with QDYX of 1.0,2.0 mg.kg-1. CONCLUSION: QDYX can decrease cardial oxygen consumption in dogs, thus having protective effect on myocardial ischemia.
OBJECTIVE: To find out the mechanism of Tangfukang on the diabetic nephropathy(DN). METHOD: A Model of streptorotocin diabetic rats was used. The expressions of the AGEs, ICAM-1, IV-C and FN were observed in renal cortex of diabetic rats with immunohistochemical method. The level of the IL-1 beta in blood serum was measured by radioimmunoassay method and the level of TNF was determined in blood serum by ELISA method. Aminoguanidine was selected as the control medicine which could inhibit the expression of monenzymatic glycosylation end products of protein. All the results were compared to analyze the effect of Tangfukang on monenzymatic glycosylation of protein and cytokine expression in early diabetic nephropathy rats. RESULTS: Both Tangfukang and aminoguanidine significantly decreased the expression of AGEs, but Tangfukang was greatly superior to aminoguanidine in suppressing the expression of cytokine like ICAM-1, IL-1 beta, TNF-alpha and the ingredients of extracellular matrix like IV-C, FN. CONCLUSION: It may be the partical mechanism of Tangfukang to suppress the monenzymatic glycosylation of protein and to reduce the expression of cytokine in diabetes. The effect of Tangfukang is superior to that of aminoguanidine.
OBJECTIVE: To observe the effect of Zhimu and Huangqi on increasing heart rate and cardiac hypertrophy, and its bradycardia effect, to evaluate the exercise capacity and response to stimulation and to discuss the use of Zhimu in treating heart failure. METHOD: 1. Rats were treated with isoproterenol 1.25 micrograms.kg-1 i.p., and then the increasing of heart rate and the bradycardia effect were measured after administrated with Zhimu or Huangqi for 3 days. 2. Mice cardiac hypertrophy model was established by s.c. ISO 1 mg.kg-1, 2 times per day for 14 days and heart-weight-index was measured after Zhimu and Huangqi was given orally alone or jointly for 14 days. 3. Abdominal aorta banding operation was done in mice and 3 weeks after operation, they were administrated for 2 weeks, and then run-time(exercise capacity), quiet heart rate, ISO response heart rate after i.p. isoproterenol and heart-weight-index were measured. 4. Cardiac hypertrophy model (inject ISO 14 days) mice were administrated for 12 days, and the mortality and dying time of mice in cold (-20 degrees C) and heat(45 degrees C) stimulative condition were observed. RESULT: Zhimu could cut down the increasing of heart rate induced by ISO, decrease significantly heart-weight-index in cardiac hypertrophy mice, reduce the quiet heart rate and prolong the run time in abdominal banding model. Zhimu combined with Huangqi could also improve the ISO response in abdominal banding model mice, reduce the mortality and delay dying time of mice in stimulative condition. CONCLUSION: Zhimu combined with Huangqi can slow down heart rate, enhance the reserve force of the heart, and improve the response capacity of cardiac hypertrophy mice in stimulative condition.
