Oncostatic properties of the complex of bivalent copper with 3-mercapto-2-hydroxypropyl ether of dextran (C-79).

The oncostatic properties of C-79 preparation were examined on the mice with: sarcoma Sa-180, melanoma B-16, Ehrlich carcinoma and leukemia P-388. The preparation was observed to prolong the life of animals with sarcoma Sa-180 and to modulate the morphology of tumors in them. The preparation had no effect on three remaining investigation models. The investigations carried out on mice of AKR strain have revealed that C-79 significantly suppresses the occurrence of spontaneous leukemia, prolonging the mouse survival-time.

Radioprotective activity of the complex of bivalent copper with 3-mercapto-2-hydroxypropyl ether of dextran (C-79).

The radioprotective activity of C-79 preparation was determined in the course of investigations carried out on mice. The activity was measured by the survival time of lethally irradiated animals and by means of the number of colonies (CFUs) on the spleens of irradiated animals. The endogenous and exogenous CFUs were determined. It was stated that the preparation, when introduced into animals several days before the irradiation with an appropriate dose of X rays, protected the homeopoetic cells of the examined animals against the irradiation effects. It was also revealed that the preparation significantly prolonged the survival time of the lethally irradiated mice. The protection activity of the preparation depended on the dose and the strategy of administration.

[Increase in the permeability of the lymphocyte plasma membrane for uni- and bivalent cations and low-molecular metabolites following exposure to mitogenic polyanions].

The paper is concerned with a study of the molecular mechanisms responsible for activation of B lymphocyte division by polyanions, polyacrylic acid (PAA) and dextran sulfate (DS). The mitogenic doses of PAA and DS were discovered to provoke an early increase in lymphocyte plasma membrane permeability. The cell membrane permeability for K+, Ca2+ and for labeled thymidine and uridine was measured in murine spleen cultures in vitro The K+ outflow from the cells was recorded according to variation in K+ concentration in the extracellular medium with the aid of a selective valinomycin electrode. The intensity of cell penetration by exogenous 45Ca, 3H-uridine or 3H-thymidine was determined by radioindicator analysis of the cytoplasma isotope pool. One to 2 min after adding the mitogenic doses of PAA or DS, the K+ outflow from lymphocytes markedly increased. It has been proved that this effect is not connected with inhibition of Na+, K+-ATPase of the plasma membrane. The increased membrane permeability for 45Ca and 3H-uridine was recorded 30-40 min after lymphocyte activation with the polyanion, that for 3H-thymidine was seen later (after 4-6 h). It is assumed that the differences between the time of recording high accumulation in the cytoplasm of 45Ca, 3H-uridine and 3H-thymidine and the time of recording high outflow of K+ from lymphocytes are determined by the differences in the sensitivity of the methods applied.

[Effect of monovalent and bivalent cations and anions on the properties of ATPase from rabbit small intestine mucosa].

The effects of monovalent cations and anions on the ATPase activity of rabbit small intestine mucosa membranes was studied. It was shown that the small intestine mucosa contains the ATPase activity sensitive to NaCl and NaHCO3. This activity, in contrast to the HCO3--ATPase activity, is inhibited by ethacrinic acid, is practically insensitive to thyocyanate, has a pH optimum of 6.2, is unstable upon storage and is not inhibited by 2 mM EDTA. The catalytic and regulatory properties of HCO3--ATPase and NaCl/NaHCO3-stimulated ATPase were compared; their possible involvement in secretion in small intestine epithelium is discussed.

[Effect of bivalent cations on the temperature of phase transition in lipid membranes].

A theory is developed which explains the temperature shift of phase transition of bilayer delta Tc resulting from the interaction of charged polar heads of lipid molecules. It has been shown that adsorption of monovalent cations on the strongly charged membrane increases the transition temperature by about 15 degrees, this value being independent of the ionic strength and dielectric permeability epsilon of the medium. When divalent cations are adsorbed the temperature shift of transition is larger, the value delta Tc depending on medium epsilon, where the polar heads are located. If the membrane is in the electrolyte solution delta Tc approximately equal to 19 degrees; when a dehydrated contact with low dielectric permeability is initiated between the membranes, the transition temperature in this zone is to increase by about 160 degrees, which well agrees with the experimental data.

[Biochemical and immunochemical characteristics of a new oral, chemical cholera bivalent vaccine and results of a trial of the preparation on volunteers].

Oral cholera vaccine contains 45% of O-antigen (serovars Ogawa and Inaba in equal parts) and at least 9 serologically active proteins; of these, toxoid (about 60% of the total amount of protein) and 5 enzymes have been identified: neuraminidase, proteinase, ribonuclease, phospholipase and ATPase. The safety, absence of reactogenicity and definite immunological effectiveness of the preparation in the primary immunization of volunteers have been shown.

[Chemiluminescence of human blood serum fractions activated by bivalent iron ions].

Chemiluminescence of separate human serum fractions induced by ferrous iron ions was studied. After removal of apoB-containing lipoproteins from serum by ultracentrifugation the bottom fraction demonstrated a more intense chemiluminescence and the whole serum luminescence could not be expressed as a simple sum of its individual components. Among various ultra-weak luminescence parameters the intensity of the slow flash measured at the 15th minute after Fe2+ introduction correlated with the level of malonic dialdehyde in apoB-containing lipoproteins.

Cytogenetics of the South American akodont rodents (Cricetidae). IX. Chiasmatic sex bivalent in male meiosis of Akodon mollis.

Akodon mollis (Rodentia, Cricetidae) has a large subterminal Y chromosome measuring 5.24 percent of the haploid set. The long arm of the Y has a pattern of homology with the short arm of the X, as indicated by the similar distribution of G bands and by the formation of a chiasma in diakinesis and metaphase I of teste cells. Conversely, the short arm of the Y and the long arm of the X are nonhomologous and do not pair at meiosis.

[Effect of bivalent copper ions on conformation of polyriboadenylic acid].

Ultraviolet differential spectra (UDS) and CD-spectra of poly (A) in the presence of Cu2+ ions are studied at different degrees of phosphate groups screening. The binding of cu2+ ions with bases of phosphates is shown to lead to disordering or to ordering respectively, of the mutual orientation of nucleotides. The UDS components due to conformation variations in poly (A) and association between Cu2+ ions and its bases are calculated. Comparison of the latter UDS-component with the differential spectrum of a protonated adenosine-monophosphate permits to assume that Cu2+ ions are in a coordination bond with N (1) adenine. The calculation of the concentration dependence of the binding constants indicates binding cooperativity and weak dependence on the screening degree of poly (A) phosphate groups.

[Spectroscopic study of the effect of binding of bivalent manganese ions on the conformation of polyriboadenylic acid].

UV-differential spectra and CD spectra of poly(A) were studied in the presence of manganese ions at 10(-3) and 0.1 M Na+ in solution. It was shown that at at both low and high ionic strengths Mn2+ ions in low concentrations form bindings with phosphates of the polymer, increasing the degree of its helicity. At higher concentrations Mn2+ ions start to bind with poly(A) bases leading to a conformational helix-random coil transition. With manganese contents exceeding 10(-3) M, aggregates are formed, in which the mutual orientation of bases is characterized by a high degree of order. The calculation of concentration dependences of Mn2+ - poly(A) base binding constants shows that the process is cooperative. As the ionic strength increases from 10(-3) to 0.1 M Na+, the constant of the poly(A) - Mn2+ complex formation decreases by about an order of magnitude.

[Effect of bivalent cations and fusogenic factors on the interaction of liposomes with planar phospholipid bilayers].

Using liposomes containing amphotericin B as an ionophoric marker the influence of some factors on fusion of the liposomes with planar phospholipid membranes was studied. Such liposomes increase the planar bilayer conductance and this fact can serve as quantitative characteristic of membranes fusion. The dependence of planar membrane conductance on Ca2+ concentration and phosphatidyl serine contents in liposomes was determined. It is shown that fusogenic factors such as lysophosphatidyl choline, retinol and osmolysis promote the fusion of vesicules with planar bilayer.

[Effects of certain biologically active compounds and bivalent cations on lactate and malate dehydrogenase activities in cerebral cellular fractions].

It is shown that acetylcholine, noradrenaline and serotonin have no effect on the lactate (LDG) and malate dehydrogenase (MDG) activities in mitochondria, mitoblasts and supernatant fluid of the rat brain tissue. A combined effect of noradrenaline and serotonin with acetylcholine increases the activity of MDG in the mitochondria but does not change that of LDG. In the mitoblast MDG was less active. After treatment with some detergents noradrenaline reduces that LDG activity in the mitochondria and supernatant fluid and did not change the MDG one. Serotonin enhances the LDG and decreased the MDG activity. In the presence of Mg2+ an increase in the LDG activity in supernatant fluid is accompanied by a decrease in the mitochondria while the MDG activity is unchanged. Ca2+ activates MDG in the mitochondria and did not affect the LDG activity.

[Effects of some bivalent and trivalent ions and of EGTA on the process of activation of the ampullar receptors].

In order to define the role of Ca++ in labyrinthine receptors the endolymph in isolated canal preparations was replaced with modified solutions containing Ca-substitutes(Sr++, Ba++), Ca-antagonists(La+++, Cd++, Mg++) and Ca-chelating agents(EGTA). The response of the receptors was evaluated by recording the slow and propagated potentials from the ampullar nerve. All the tested solutions proved able to depress or cancel the mechanically evoked responses of ampullar receptors. The presence of Ca++ in the endolymph is therefore essential to grant the mechanosensitivity of the hair cells. The receptor blockade produced by Ca-withdrawal develops much earlier under continuous mechanical stimulation. This suggests that Ca++ might be bound in some structures and released during cupula deflections.
