Antibody to an artificial disaccharide antigen cross-reactive with Neisseria gonorrhoeae lipopolysaccharide.

An artificial antigen was prepared from 4-O-beta-I-galactopyranosyl-D-glucose (lactose) and 8-ethoxycarbonyloctanol. Covalent attachment to bovine serum albumin provided an antigen that elicited antilactose antibody in rabbits and goat. These antibodies were active against Neisseria gonorrhoeae lipopolysaccharide in passive hemagglutination tests. The same antibody agglutinated cells of Streptococcus faecalis, strain N, and precipitated the lactose-containing cell wall diheteroglycan of this organism. Fractionation of rabbit and goat antibody raised against the synthetic antigen of S. faecalis vaccine provided two antibody fractions only one of which, eluted from the immunoadsorbent by galactose, was active against N. gonorrhoeae lipopolysaccharide.

In vitro inhibition of rat intestinal surface hydrolysis of disaccharides and dipeptides by guaran.

The effect of guaran on intestinal surface hydrolases was studied in everted jejunal segments by measuring the hydrolysis of maltose and phenylalanylglycine. On the basis of kinetic experiments, guaran was found to inhibit competitively the hydrolysis of these substrates only if the cleavage was performed with intact tissue. Hydrolysis in mucosal homogenates was not affected by guaran, indicating that this polysaccharide may increase the diffusion barrier overlaying the mucosa and thus may retard final digestion of carbohydrates and peptides.

The effect of disaccharides on the plaque-forming potential of Streptoccoccus mutans.

The comparative and combined effects of sucrose, maltose, and lactose as factors on the plaque-forming potential of Streptococcus mutans were assessed. With increasing additions of maltose to sucrose-supplemented medium there was decreasing plaque formation. Lactose additions slightly increased plaque formation, but when combined with maltose they significantly enhanced the maltose inhibition of plaque formation.

Disaccharides as endomannosidase inhibitors: syntheses of alpha-homomannojirimycin and beta-homomannojirimycin linked to D-glucose and D-mannose.

4-O-(alpha-D-Glucopyranosyl)-alphaHMJ (Glcalpha1,4HMJ), 4-O-(alpha-mannopyranosyl)-alphaHMJ (Manalpha1,4alphaHMJ), 4-0-(alpha-glucopyranosyl)-betaHMJ (Glcalpha1,4betaHMJ), and 4-0-(alpha-mannopyranosyl)-betaHMJ (Manalpha1, 4betaHMJ) were synthesized as endomannosidase inhibitors which are potentially useful both for probing the pathways of N-linked glycoprotein processing and for the chemotherapy of some viral diseases.
