An aromatic hydroxylation assay for hydroxyl radicals utilizing high-performance liquid chromatography (HPLC). Use to investigate the effect of EDTA on the Fenton reaction.

A highly sensitive HPLC method for the separation of hydroxylation products derived from the attack of hydroxyl radical upon phenol is described. Catechol and hydroquinone are the major hydroxylation products formed, with little resorcinol. The effect of EDTA upon hydroxyl radical generation from an iron (II)-H2O2 system is shown to depend upon the order of addition of chelator and metal ion to the reaction mixture, the ratio [iron salt]/[chelator] and the presence or absence of a phosphate buffer. Reasons for these different effects are discussed.

Role of serum total sialic acid in differentiating cholangiocarcinoma from hepatocellular carcinoma.

AIM: This study was designed to evaluate the clinical application of serum total sialic acid (TSA) in the diagnosis of cholangiocarcinoma (CCA). METHODS: Serum TSA was determined by periodate-resorcinol microassay in 69 patients with CCA, 59 patients with hepatocellular carcinoma (HCC), 37 patients with cirrhosis, 61 patients with chronic hepatitis and 50 healthy blood donors. RESULTS: The mean serum TSA concentration in CCA (2.41+/-0.70 mmol/L) was significantly higher than those of HCC, cirrhosis, chronic hepatitis and healthy blood donors (1.41+/-0.37 mmol/L, 1.13+/-0.31 mmol/L, 1.16+/-0.26 mmol/L, and 1.10+/-0.14 mmol/L, respectively; P<0.001). Based on ROC curve analysis, a cut-off point of 1.75 mmol/L discriminated between CCA and HCC with a sensitivity, specificity and accuracy of 82.6 %, 83.1 %, and 82.8 %, respectively. CONCLUSION: Based on our results, serum TSA would be a useful marker for the differential diagnosis of CCA from HCC.

Toxicity of rubber chemicals towards three-day chicken embryos.

Three-day chicken embryos were exposed to 80 different rubber chemicals dissolved in either acetone or water. The following classes of chemicals were studied: thiurams, dithiocarbamates, thioureas, benzothiazoles, benzenesulfohydrazide, dithiodimorpholine, amines, acrylates, guanidines, resorcinol, phthalates, adipates, phosphates, oils, peroxides, heavy metal salts, and sodium nitrite. Dose-response curves and median lethal doses (LD50) or median effective doses (ED50) were calculated for mortality in 2 d after the treatment, for total mortality in 11 d after the treatment, and for the total effect, including deaths and malformations 11 d after the treatment, when the test was finished. Sixteen of the chemicals had no effect on the embryos when injected into the air chamber. Incomplete and irregular dose-response curves for the total effect were obtained with 13 of the chemicals. For them, the ED50 could be calculated. Among the remaining 51 chemicals, the most potent were the dithiocarbamates and cadmium and copper acetates, with the total effect ED50 from 2.4 to 160 nmol/egg. Other chemicals that had an ED50 smaller than 100 nmol/egg were thiurams, cyclohexylthiophthalimide, acrolein, and dithiodimorpholine. The majority of the chemicals had an ED50 between 100 nmol/egg and 10 mumol/egg. The least potent were sodium nitrite and methylmethacrylate with an ED50 of 22 mumol/egg. All the 64 embryotoxic chemicals caused malformations with only one exception (dibutylthiourea). The maximum observed frequencies of malformed embryos varied from 3 to 100% of the treated embryos.

A new type of chemical modification of glycopeptides antibiotics: aminomethylated derivatives of eremomycin and their antibacterial activity.

A series of derivatives of eremomycin aminomethylated at the 7d position of the resorcinol ring of the amino acid No. 7 was prepared by interaction of eremomycin with formaldehyde and various primary and secondary amines and ammonia. The most active compound obtained was 7d-decylaminomethyl derivative, whose minimal inhibitory concentrations for clinical isolates of staphylococci are 2 approximately 8 times lower than those of the parent antibiotic. 7d-Decylaminomethyl derivative was also active against vancomycin-resistant VanA enterococci (8 microg/ml) and Neisseria gonorrhoeae (16 microg/ml).

Hydrophobic vancomycin derivatives with improved ADME properties: discovery of telavancin (TD-6424).

Novel derivatives of N-decylaminoethylvancomycin (2), containing appended hydrophilic groups were synthesized and their antibacterial activity and ADME properties were evaluated. The compounds were prepared by reacting amines with the C-terminus (C-) of 2 using PyBOP mediated amide formation, or with the resorcinol-like (R-) position of 2 using a Mannich aminomethylation reaction. These analogs retained the antibacterial activity of 2 against methicillin-resistant staphylococci and vancomycin-resistant enterococci. Compounds with a negatively charged auxiliary group also exhibited improved ADME properties relative to 2. In particular, R-phosphonomethylaminomethyl derivative 21 displayed good in vitro antibacterial activity, high urinary recovery and low distribution to liver and kidney tissues. Based on these results, 21 was advanced into development as TD-6424, and is currently in human clinical trials. The generic name telavancin has recently been approved for compound 21.

[Experimental anatomo-pathologic study in the myocardium of animals with arterial hypertension caused via a nitric oxide synthesis blocker].

INTRODUCTION: Cardiac hypertrophy and reactive (including perivascular) and reparative fibrosis in L-NAME model are similar to that of hypertensive and cardiomyopathic patients. THE AIM: To qualitatively evaluate the reactive (including perivascular) and reparative fibrosis, on days 21 and 35, occurring in hearts submitted to the L-NAME model, using special staining. MATERIAL AND METHODS: We utilized 33 normotensive Wistar rats. L-NAME was administered orally--in a concentration of 75 mg x 100 ml-1 in drinking water. Six rats were submitted during 21 days and an other 15 rats during 35 days. The arterial pressure was obtained on days 12, 20 and 34 using hydraulic plethysmography. On days 21 and 35 during the experiment the animals were anesthetized and submitted to cardiotomy. The hearts were fixed in Bouin fixative during 48 hours and processed using routine methods, emblocked in paraplast and cut in slices 4 to 7 microns thick. The special staining utilized were: Gomori's trichrome (aniline blue) or Masson trichrome, picro-sirius red-polarization, alcian blue technique (pH 0.5 and pH 2.5), periodic acid-Schiff technique (with and without amylases) and Weigert's resorcinol fuchsin solution (with and without oxon). RESULTS: Our results demonstrated an increase in the arterial pressure in animals submitted to the model. On day 21 of submission we observed modest to extensive infarct areas in the right and left ventricular myocardium. On day 35 the wide reparative areas were from old infarct areas. In some cases at day 35 the lesions reached the totality of the right ventricle in several histological slices. The right ventricle was much more affected than the left one. In both groups perivascular fibrosis was observed, nevertheless, on day 21 it was very reduced. DISCUSSION: The degree of direct influence of NO or hypertension produced by NO on hypertension and cardiac lesions during L-NAME model is discussed nowadays. Nevertheless, the fact is that NO deficit has great influence in several cases of cardiac lesions occurring in hypertension and also in cardiomyopathies.

[Branch-specific detection of phenols and assessment of ground water solubility].

There are about 500 technically relevant phenolic compounds such as cresols, chlorophenols or nitrophenols. It is most preferable to determine phenols as single compounds via gas chromatography. Further, phenols can also be assayed as photometrically as an overall parameter (Phenolindex): however, no conclusions about specific compounds can be drawn from this type of test. Also this method is not as reliable for an hazard assessment as gas chromatography. First, not all phenols, for instance resorcinol or 1-naphthol can be determined with this method. Second, phenolic groups in humic substances, which do not constitute a threat for groundwater, are determined alongside environmentally relevant phenols using this method. In most cases, it is possible to deduce which phenols can be expected in the groundwater of contaminated sites from the type of industrial usage, such as chlorophenols and pulp bleaching or nitrophenols and the production of explosives. Phenols are formed during coal combustion for instance at cokemanufactures or gasworks. They are important raw materials for the chemical industry from which resins, surfactants, pharmaceuticals, pigments, explosives, and stabilizers are produced. During the 80's phenol, cresols, nonylphenols, anisidines, aminophenols, dihydroxybenzenes, and naphthols were manufactured in amounts exceeding 10,000 t/a. Also, phenolic compounds are used as additives in many areas for example as solvents in the electric industry, in sawmills, papermanufacture, electroplating of metal sheets, as photographic developers, as textile dyes, or for the tanning of hydes. Due to the formation and use of phenols at industrial sites, groundwater contaminations are possible via infiltration through the unsaturated zone. Especially at gasworks and ammunition factories, groundwater contaminations with phenols have become known. In the vicinity of railway tracks and associated facillities contaminations due to the use of pesticides or mineral oils are possible. Input of phenols on agricultural lands can be caused by pesticides, sewage sludge or manure. The groundwater downstream of landfills often contains phenol, chlorophenols, cresols, and xylenols. The formation of phenol from other organic contaminants as benzene in groundwater has been reported. The potential for mobilization of phenols in the saturated zone can be estimated from their physical and chemical properties. Especially low molecular weight phenols are easily mobilized due to their high solubility in water and low potential for accumulation. These compounds are: phenol, cresols, xylenols, chlorophenols, hydroxybenzenes, nitrophenols, anisidines, aminophenols, anisol, 2-phenoxyethanol, and thiophenol. The stability of phenols under laboratory conditions varies. The complete mineralization depends mainly on the experimental set-up, i.e. nutrients, temperature, and type of inocula. The anaerobic degradation of phenols is generally slower than the aerobic. Phenol is readily biodegradable under both aerobic and anaerobic conditions. In general, the biodegradability depends on the type, number and position of substitutes. Phenols with nitro-, alkyl-, or chlorosubstitutes are more recalcitrant than phenol itself. Our biodegradability test show that the decomposition of alkylphenols is determined by the length and branching of the alcyllic chain. Phenols with high contamination potential are chlorophenols, xylenols, and nitrophenols. These compounds are both mobile and recalcitrant in the saturated zone. Phenolic compounds of a medium contamination risk are dichlorophenols, trichlorophenols, cresols, and phenol because they are mobile but less stable in groundwater. These compounds are known contaminants in the groundwater at gasworks, landfills, and ammunition factories. Aminophenols, anisidines, tert-butylphenols, ethylphenols, hydroxybenzenes, and 2-phenoxyethanol also constite a potential hazard for groundwater; however, no contaminations with these compounds are known. Poisoning due to oral uptake of phenol contaminated ground- or drinking water have not been reported, which might be due to the pungent odor and taste phenolic compounds have even at very low concentrations (mg/L). Because of the taste problem, the German drinking water standard for phenols is 0.5 microgram/L. In Berlin, groundwater with phenol concentrations higher than 30 micrograms/L of alkylphenols or 2 micrograms/L of chlorophenols is considered contaminated.

Enzymic determination of fructose in seminal plasma by initial rate analysis.

We describe a spectrophotometric assay for fructose in seminal plasma. The method is based on reduction of fructose by a commercially available preparation of sorbitol dehydrogenase (EC 1.1.1.14), with the concomitant oxidation of NADH. The initial rate of NADH oxidation, which is proportional to the fructose content of seminal plasma, can be measured either with a recording spectrophotometer or by conventional two-point kinetic assay. The method was as accurate, precise, and sensitive as, and more specific and rapid than, currently used colorimetric (resorcinol) methods for fructose determination. Values (mmol/L) for fructose in seminal plasma from several species are: man, 9 +/- 2 (SD); cynamolgus monkey (Macaca fasicicularis)., 108 +/- 19; bull, 30 +/- 1; and rabbit, 13 +/- 4. These values agree with previously published results. We believe the method is appropriate for both research and clinical use.

Aortic valve preservation in acute type A dissection: mid-term results.

BACKGROUND AND AIM OF THE STUDY: The study aim was to evaluate the long-term effectiveness of a strategy for managing the aortic valve, aortic root and ascending aorta according to the pathology in acute aortic type A dissection. Results after surgery for acute type A dissection with preservation of the aortic valve were reviewed. METHODS: The patient group included 57 hospital survivors operated on according to a surgical strategy of aortic valve resuspension and supracoronary ascending aortic graft implantation. Reinforcement of the aortic stumps with gelatin-resorcinol-formaldehyde glue was performed in all patients. Aortic valve function in all survivors was investigated by echocardiographic follow up at 30 days, 6 and 12 months after surgery, and yearly thereafter. RESULTS: During the follow up period, nine patients (16%) died without reoperation. Actuarial probability of freedom from reoperation for aortic valve failure in the complete series was estimated as 100% after both 30 days and 12 months. Postoperatively, one patient underwent reoperation 14 months for aortic regurgitation, and three patients for aortic regurgitation with sinus of Valsalva dilatation between 48 and 88 months. The hospital mortality rate at reoperation was 50% (n = 2). CONCLUSION: Valve-sparing surgery is possible and can be recommended for the majority of patients with acute type A aortic dissection.

Testicular migration: remodeling of connective tissue and muscle cells in human gubernaculum testis.

PURPOSE: We present the main morphological modifications in the human gubernaculum during testicular migration in humans. MATERIALS AND METHODS: We obtained 12 gubernacula from fresh, macroscopically normal human fetuses at 15 to 29 weeks of gestation. Collagen was evidenced using trichrome and Sirius red staining procedures, while Weigert's resorcinol-fuchsin and anti-human elastin antibody were used to reveal elastic system fibers. Smooth muscle cells were detected by anti-human smooth muscle alpha-actin antibody. RESULTS: When the testes were still located in the abdomen at 15 to 16 weeks of gestation, collagen fibers were sparse and embedded in a loose extracellular matrix. The amount of fibers then gradually increased with age and at 28 weeks of gestation the gubernaculum was mostly collagenous in composition. Elastic fibers had a similar growth pattern, although they were located mainly at the distal end of the gubernaculum. Fibroblasts largely predominated over other cell types and decreased in number with gestational age, whereas smooth muscle cells were restricted to the walls of blood vessels. Striated muscle cells were detected at the scrotal end of the gubernaculum, where they were disposed as isolated and scattered bundles running in various directions. Like fibroblasts, their number also decreased with age. CONCLUSIONS: During testicular migration gubernacular connective tissue undergoes extensive remodeling and ultimately becomes an essentially fibrous structure rich in collagen and elastic fibers. Such changes should decrease the size of the gubernaculum and, thus, contribute to other forces that cause the testes to move toward the scrotum. In fact, because of the lack of smooth muscle cells, and the amount and organization of striated muscle cells, active contraction of the gubernaculum is less likely to be an important factor in testicular descent.

Growth phase dependent substrate utilization by Pseudomonas strain PH1.

Pseudomonas strain PH1 can utilize nitro-, chloro-, and aminophenols and has been used in this study. The enzymes of the two-pathway viz., phenol, and meta-aminophenol (MAP) were analyzed under different growth conditions. The enzymes responsible for phenol to catechol conversion followed by the ring cleavage enzyme for catechol; and also the enzymes responsible for MAP oxidation and hydroxylation of resorcinol, were studied. Enzyme and respirometric assays were carried out with cells harvested from log phase and stationary phase from medium with different carbon sources and nitrogen levels. It was observed that the first step for utilization of both the substrates requires the same physiological state of the cells; whereas, the subsequent step follows independent approach to intermediates, based on cellular physiology.

[Influence of the mixed disinfectant of ClO2 and Cl2 on formation of chloroform].

The study on influence of mixed solution of ClO2 and Cl2 on formation of chloroform from 9 THMs precursors, such as fulvic acid (FA), resorcinol, phloroglucinol and cresorcinol etc. under conditions of different ClO2/Cl2 ratio, content of precursors, reaction pH, reaction time and reaction temperature etc. was studied. Results showed that as the mass percentage of ClO2 in the mixed disinfectant was increased, the amount of chloroform formed was reduced; such as when mass percentages of ClO2 were 25%, 50% and 75%, CHCl3 formed reduced 55%-65%, 70%-86% and 80%-93% respectively; until ClO2 was 90%, reduction of CHCl3 reached 95%-99%. In order to control the formation of chloroform thoroughly, mass percentage of ClO2 must be higher than 90%. This provided important scientific base for the necessity of using highly pure ClO2 in disinfection and developing generator which produced high quality ClO2.

[Determination of extraction separation of Pd(II), U(VI) and Mo(VI) in PEG-Na2SO4-PAR system using spectrophotometry].

In polyethylene glycol-2000(PEG)-sodium sulfate (Na2SO4)-4-(2-Pyridylazo) resorcinol (PAR) system the extraction separation behaviour of the complexes of Pd(II), U(VI), Mo(VI) and La(III) was investigated. The results indicated that the complexes of Pd(II) and U(VI) with PAR were almost completely extracted by the PEG phase, La(III) was partly extracted, while Mo(VI) was not extracted from the aqueous solution of pH 4.5-7.5. So Mo(VI) was quantitatively separated from the mixed solution of Mo(VI), Pd(II) and U(VI) ions. The extraction mechanism for PEG phase was studied preliminarily.

[Induction of SOS-response of cells exposed to autoregulatory factors of microorganisms].

Among examined microbial growth regulators of alkyl hydroxybenzene group (hexylresorcinol, methylresorcinol, and hydroxyethylphenol), only hexylresorcinol induces cellular SOS response, demonstrating a dose-dependent increase of the induction factor in the SOS chromotest with the Escherichia coli PQ37 strain. At the highest of used concentrations (100 micrograms/ml), hydroxyethylphenol and nonalkylated resorcinol were shown to exert a weak toxic effect, reducing the activity of constitutive alkaline phosphatase, but did not induce SOS response. Nontoxic methylresorcinol did not induce genome damage, which can trigger SOS functions. It is concluded that substitutions in phenolic ring affect genotoxic activity of alkylresorcinols.

Common phenotypic expression of gangliosides GM3 and GD3 in normal human tissues and neoplastic skin lesions.

The expression of gangliosides in non-malignant tissues (epidermis and pigmented nevus) and neoplastic lesions (melanoma, squamous cell carcinoma [SCC] and basal cell carcinoma [BCS]) of the human skin was analyzed immunohistochemically and biochemically to characterize the features associated with malignancy. Immunohistochemical staining with an anti-II3NeuAc-LacCer (GM3) monoclonal antibody (M2590 mAb) and an anti-II3(NeuAc)2-LacCer (GD3) mAb (R24) showed the expression of the gangliosides GM3 and GD3 to vary among the different tissues. M2590 clearly stained epidermal keratinocytes and the tumor cells of BCC and SCC, and strongly stained melanocytes and melanoma cells. In contrast, R24 did not stain epidermal keratinocytes and only faintly stained SCC cells, while it clearly stained BCC cells, and intensely stained melanocytes and melanoma cells. GM3 showed a similar level of staining among the tissue specimens, while the level of GD3 staining was quite variable among the tumor specimens. Biochemical analysis by thin-layer chromatography (TLC) with resorcinol staining and TLC immunostaining with either M2590 or R24 showed both GM3 and GD3 to be commonly expressed by both the normal and malignant skin tissues, including SCC. There was no close correlation between the intensity of immunohistochemical staining and the biochemically detected amounts of these gangliosides. This may have been partly due to the so-called cryptic expression of cell membrane gangliosides. Our results thus suggest that analysis of the tumor-associated expression of gangliosides requires several methods, since the sensitivity of the methods used may have a considerable effect on the diagnostic value of gangliosides as skin cancer markers.

[Peroxidase oxidation of phenols].

Partially purified preparations of horseradish peroxidase were able to catalyze the effective transformation of such phenol compounds as phenol, o-chlorophenol, 2,4,6-trichlorophenol, pentachlorophenol (giving rise to the formation of polymer products insoluble in water), resorcinol, and thymol (giving rise to the formation of low-molecular-weight products). The following conditions were found to be optimal for peroxidase oxidation and provide the maximum extent of elimination of phenol compounds: temperature, 15-25 and 25-30 degrees C for phenol and chlorophenol compounds, respectively; molar ratio H2O2/phenol, 1:1; and transformation time, 1-3 h. Although effective transformation was observed within a broad range of pH, the efficiency of the process slightly increased at a pH from 6.0 to 7.5. It was suggested to carry out multiple peroxidase oxidations of phenols using partially purified peroxidase enclosed in a dialysis membrane bag placed into a solution of a phenol compound containing hydrogen peroxide.

Serum sialic acid (TSA/LSA) and carcinoembryonic antigen (CEA) levels in cancer patients undergoing radiotherapy.

The main aim of this study was to evaluate the response of total Sialic Acid (TSA) and "Lipid-bound" Sialic Acid (LSA) compared to Carcinoembryonic Antigen (CEA), in 284 patients undergoing radiotherapy. Serial measurements of TSA by the enzymatic method (Boehringer-Mannheim Kit), LSA by the resorcinol-HC1 (Katopodis and Stock) and CEA by EIA (Abbott Kit) were performed in a total of 1017 blood sera. We statistically estimated the four greater groups of cancer patients [bladder (69), lung (58), uterus (31) and breast (29)]. Diagnostic marker sensitivities (% true positives) estimated from the 0-time-values--before initiation of radiotherapy--in relation to the established cut-off levels were in decreasing order: TSA 89.3% (80 mg/dL). LSA 88.8% (20 mg/dL) and CEA 26.75% (5 ng/mL). The overall tumor marker response to treatment, after its completion, estimated as % of patients with final blood serum levels of these markers, was in decreasing order: LSA 85.6%, TSA 81.3%, and CEA 65.8%. These data show that a) the diagnostic sensitivity of Sialic Acid (LSA/TSA) is more than 3 times higher than that of CEA and b) the response of Sialic Acid (LSA/TSA) to treatment is about 15% higher than that of CEA. In conclusion, this study confirms the high diagnostic sensitivity of Sialic Acid as a tumor marker and suggests that, with marginal superiority of Sialic Acid, all three markers are sufficiently responsive to be employed as adjunctive means in monitoring cancer patients underdoing radiotherapy.

Benzimidazolyl quinolinyl mercaptotriazoles as potential antimicrobial and antiviral agents.

Condensation of ethylaceto acetate (EAA) with resorcinol in concentrated H2SO4 afforded 7-hydroxy-4-methyl coumarin (1), which on reaction with thiosemicarbazide in anhydrous pyridine yielded 7-hydroxy-4-methyl-quinolinyl [1,5-c]-mercaptotriazole (2). Reaction of 2 with formaldehyde solution and amino acid in ethanol yielded 7-hydroxy-4-methyl-8-(N-methyl-aminoacid)-quinolinyl [1,5-c]-2''-mercaptotriazole (3a-e). Interaction of 3 with o-phenylenediamine in pyridine yielded 7-hydroxy-4-methyl-8-(aminobenzimidazolyl)-quinolinyl [1,5-c]-2''-mercaptotriazole derivatives (4a-e). The latter compounds were evaluated for their antiviral and antimicrobial activities.
