Uterine action after induction of labour with oral prostaglandin E2 tablets compared with intravenous oxytocin.

Uterine action throughout the whole of labour induced by low amniotomy and either oral prostaglandin E2 tablets or intravenous ocytocin was studied in 10 randomly matched pairs of patients. Oral prostaglandin E2 tablets induced fewer contractions per hour, and these contractions were longer and more variable in length than those induced by intravenous oxytocin. There was no difference in basal uterine tone or in the amplitude of contractions. A comparison was made between the outcome of labour induced by low amniotomy and either oral prostaglandin E2 tablets 0-5 mg. hourly in 65 patients, or intravenous ocytocin in 41 patients. This dose of prostaglandin E2 tablets was an inadequate adjunct to low amniotomy in nulliparous patients. Despite this, the induction-delivery interval for the whole prostaglandin group was not significantly greater than that for the oxytocin group. There were no significant differences in fetal heart rate pattern, or in the incidence of gastro-intestinal side-effects between the two groups.

Studies on the possible role of brain histamine in behaviour.

1. The possible role of brain histamine in behavioural performance was studied in rats using thirst-induced water consumption, continuous (Sidman) avoidance, and reinforcement withdrawal test systems.2. Parenteral administration of a variety of antihistamines to rats decreased thirst-induced water consumption; this effect could be antagonized by administration of histamine directly into the brain by a ventricular cannula.3. When intraventricular doses of histamine were administered to rats at weekly intervals, an adaptation was seen in the effects of the amine on continuous avoidance behaviour. With succeeding doses, the initial period of depression of avoidance responding was shortened and the subsequent rebound stimulation disappeared.4. The results support the hypothesis that histamine in the brain is involved in several behavioural phenomena.

Structure-activity relations for the inhibition of 5-hydroxytryptamine uptake by tricyclic antidepressants into synaptosomes from serotoninergic neurones in rat brain homogenates.

1 The inhibitory effects of various analogues of imipramine on [(3)H]-5-hydroxytryptamine (5-HT) uptake into homogenates of rat hypothalamus were examined.2 For structures with a three carbon side chain the tertiary amine derivative was more potent than the compound with a secondary amine function.3 Potency was reduced by increasing or decreasing the length of the three carbon side chain by one carbon atom.4 Substitution of a methyl group in the alpha or beta position in the side chain reduced potency.5 Replacement of the dimethylene bridge in imipramine by a sulphur atom or substitution of a C=C double bond for the exocyclic N-C bond of imipramine both led to a fall in potency.6 3-Chlorimipramine was the most potent inhibitor of [(3)H]-5-hydroxytryptamine uptake of the compounds tested.

Relationship between growth inhibition and mitochondrial function in petite-negative yeasts. II. Effects of central nervous system drugs upon pathogenic and non-pathogenic Candida species.

Six nervous system drugs which inhibited vegetative reproduction of S. cerevisiae arrested also mitotic division of C. utilis, C. albicans and C. tropicalis. Chlorpromazine and chlorpheniramine which proved to be the most effective, affected respiration and cytochrome biosynthesis. Electrophoretic bands with MW congruent to 100 K were faint in silver-stained electrophoregrams of proteins from cells grown in the presence of a sub-inhibitory concentration of chlorpromazine.

Dopamine receptor blockade and the neuroleptics, a crystallographic study.

The X-ray structures of 12 drugs of the tricyclic class having varying pharmacological profiles have been examined in detail in an attempt to rationalize the known structure-activity relations of neuroleptic drugs with respect to their ability to block dopamine receptors in the brain. Further evidence is presented in support of the theory that the neuroleptics are able to block dopamine receptors because of a conformational complementarity between certain portions of these drugs and dopamine.
