Flavon- and flavonolglycosides from Achillea pannonica Scheele.

The detailed investigation of a methanolic extract of aerial parts of Achillea pannonica SCHEELE. within a chemotaxonomic study led to the isolation of 6 flavonoid glycosides. Besides rutin, apigenin-7-O-glucopyranoside, luteolin-7-O-glucopyranoside, apigenin-7-O-rutinoside and acacetin-7-O-rutinoside, an unusual flavondiglucoside was isolated. Its structure was established by UV, 1H NMR and 13C NMR spectroscopic methods including 2D-NMR techniques and ESI-MS as luteolin-7,4'-O-beta-diglucoside. This substance is reported for the first time in the genus Achillea. Chemotaxonomic aspects are discussed briefly.

Eupatilin, a pharmacologically active flavone derived from Artemisia plants, induces apoptosis in human gastric cancer (AGS) cells.

Extracts of Artemisia asiatica Nakai (Asteraceae) possess anti-inflammatory and antioxidative activities. Eupatilin (5,7-dihydroxy-3',4', 6-trimethoxyflavone), one of the pharmacologically active ingredients derived from A. asiatica was shown to induce apoptosis in human promyelocytic leukemia (HL-60) cells. In the present study, we examined the ability of eupatilin to induce apoptosis in human gastric cancer (AGS) cells. Eupatilin induced the apoptosis of AGS cells as revealed by a decrease in the ratio of pro-apoptotic Bax and anti-apoptotic Bcl-2, as well as the cleavage of caspase-3 and poly(ADP-ribose)polymerase (PARP). The pro-apoptotic effects of eupatilin were further verified by its perturbation of the mitochondrial transmembrane potential (DeltaPsim). In addition, eupatilin treatment led to an elevated expression of p53 and p21. Eupatilin inhibited the activation of ERK1/2 and Akt, which are important components of cell-survival pathways.

Fundamental, electron transfer mechanism by pyrylium-type ions for the anticancer drugs 5,6-dimethylxanthenone-4-acetic acid (DMXAA) and flavone-8-acetic acid (FAA).

Pyrylium-type salts derived from DMXAA and FAA are proposed to play an important mechanistic role in anticancer action. Electron transfer (ET) processes apparently initiate cell signaling cascades that lead to the observed effects, such as, antivascular influences, cytokine induction, and apoptosis. Possible participation of nitric oxide and serotonin is discussed. Structure-activity relationships involving DMXAA, FAA, acridines, and quinolines support the hypothetical framework, as well as electrochemistry and photochemistry. Similarity is pointed out to the action of plant hormones, e.g. ethylene. Involvement of ET pathways places the cationic salts within the general mechanistic framework for other anticancer agents. Other drug activities of xanthenones are in accord with the ET approach. Insight into fundamental mechanistic aspects should aid in development of improved drugs in this class through rational design.

[Effects of total flavones of fructus Hippophae (TFH) on cardiac function and hemodynamics of anesthetized open-chest dogs with acute heart failure].

This study was carried out employing the model of dog with acute heart failure induced by phenobarbital natricum. It was shown that i.v. TFH 4.8 and 9.7 mg/kg could significantly increase CO, CI, + LVdP/dtmax and LVSP; shorten R-dP/dtmax in 9.7 mg/kg; raise - LVdP/dtmax, reduce LVEDP and T value; decrease MVO2I and TPVR; MAP and HR were not changed significantly. The results suggest that i.v. TFH can strengthen cardiac pump function and myocardial contractility in canine with heart failure; significantly improve myocardial diastolic function and hemodynamic performance; markedly decrease myocardia-used oxygen index and total peripheral vessel resistance, thus proving that TFH is good for heart failure.

[Studies on flavone constituents of Erigeron breviscapus (Vant.) Hand.-Mazz].

OBJECTIVE: To study the chemical constituents of Erigeron breviscapus. METHOD: The constituents were separated and purified by column chromatography with silica gel, and identified by IR, MS, NMR and physical data. RESULT: Five compounds were isolated and identified as 3, 5, 6, 4'-tetrahydroxy-7-methoxy flavonoid(I); 5, 7, 4'-trihydroxy flavonoid(II); 3, 5, 6, 7, 4'-pentahydroxy flavonoid(III); scutellarein (IV) and 5, 7, 4'-trihydroxy flavanone(V). CONCLUSION: Compounds I, III and V were isolated from this plant for the first time.

[Two flavone glycosides from Chinese traditional medicine Amomum villosum].

OBJECTIVE: To study the water soluble constituents from Amomum villosum. METHOD: The constituents were separated and purified with chromatographic methods, identified by NMR, MS, UV and IR. RESULT: Two quercetin glycosides: quercitrin (quercetin-3-O-alpha-L-rhamnoside I) and isoquercitrin (quercetin-3-O-beta-D-glucoside II) were isolated and identified. CONCLUSION: I and II were isolated for the first time from A. villosum.
