[The ultrastructural characteristics of the bronchial mucosa in chronic inflammation in participants in the cleanup of the aftermath of the accident at the Chernobyl Atomic Electric Power Station].

Bronchial biopsies from persons exposed to radiation after the Chernobyl accident with clinically verified chronic bronchitis were studied by electron microscopy. The results demonstrate transformed alterative, compensatory-adaptive and inflammatory reactions in bronchial mucosa of contingent. Nevertheless, their integral significance does not alter the nosologic form, and some structural equivalents have dynamic expression.

[Efficacy of antioxidant preparations used for correction of impairment of oxidative homeostasis in Chernobyl liquidators].

The influence of two antioxidative preparations (Vetoron, Morevit) on the parameters of antioxidative system and lipoperoxidative processes in blood of men, who took part in the cleaning up after Chernobyl NPP accident, under the clinical control in the hospital of Scientific Center for Radiation Medicine AMS of Ukraine was studied. It was shown that Morevit (extract of protein-hydrocarboneus concentrate of Mytilis galloprovincialis) is more effective as promoter of normalization lipoperoxidative processes disorders and restoration of antioxidative enzymes activity. The main principles of antioxidative therapy were formulated.

[Bone marrow hematopoiesis in those who worked in the cleanup of the aftermath of the accident at the Chernobyl Atomic Electric Power Station with a history of acute radiation sickness (based on myelogram data)].

On the basis of myelograms an evaluation was done of medullary hemopoiesis in 15 subjects who had been taken ill with acute radiation sickness (ARS) as a result of Chernobyl accident in 1986. 6-12 years following the exposure to ionizing radiation the bone marrow of ARS convalescents still shows a tendency toward hypocellularity (the number of myelokaryocytes in the examinus ranged from 40 to 80 x 10(9)/l), high index of neutrophil maturation (0.99) was recordable, which fact was related to depletion of granulocyte reserve and increase among the elements of granulopoiesis of share of proliferating cells, together with a rise in the number of lymphoid elements (19%).

[The characteristics of the slow electrical activity of the brain in those who worked in the cleanup of the aftermath of the accident at the Chernobyl Atomic Electric Power Station].

We studied the slow electrical activity of the brain in people subjected to irradiation as a result of the Chernobyl accident and in healthy donors. The mean constant potential, which is an index of energy metabolism in different regions of the cerebral cortex, is reliably lower in eliminators, as compared to the donors, thus suggesting a postradiation energy deficiency of the brain. The structure of correlations between the constant potential indices is different: the number of significant correlations in the group of eliminators is lower by 30% than in the group of donors, thus suggesting disturbed intercentral relations in the CNS under the conditions of postradiation energy deficiency of the brain.

Changes in the auditory system related to cerebral circulation in the Ukraine population after the Chernobyl disaster.

The hearing and cerebral circulation was evaluated in 100 liquidators ranging from 23 to 50 years of age 1 year and 6 to 8 years after the disaster. Also, the hearing state data was compared from 80 liquidators of the same age dwelling in clean and radionuclide-contaminated areas for 5 to 8 years. The control group consisted of 20 young healthy individuals having no history of contact with radiation. Subjective audiometry, AEP and REG was used. Testing performed indicate that under radiation, more pronounced changes occur in the central part of the acoustic analyser rather than in its receptor system. More impaired were the persons residing the radionuclide-contaminated areas. The liquidators of the ChFS accident who live in clean areas presented with better state of the cerebral circulation and bioelectric brain activity. But over the time, the REG data 6-8 years after the disaster became considerably worse, as compared to the data obtained 1 year after the disaster. It should be emphasized that individual irradiated during and after the Chernobyl disaster require a constant dynamic observation and their identified changes require periodical management. Withdrawal of the liquidators from the contaminated to the clean areas is one of the preventive measures against the hearing and cerebral circulation disorders.

Specific p53 gene mutations in urinary bladder epithelium after the Chernobyl accident.

After the Chernobyl accident, the incidence of urinary bladder cancers in the Ukraine population increased gradually from 26.2 to 36.1 per 100,000 between 1986 and 1996. Urinary bladder epithelium biopsied from 45 male patients with benign prostatic hyperplasia living in radiocontaminated areas of Ukraine demonstrated frequent severe urothelial dysplasia, carcinoma in situ, and a single invasive transitional cell carcinoma, combined with irradiation cystitis in 42 cases (93%). No neoplastic changes (carcinoma in situ or transitional cell carcinoma) were found in 10 patients from clean areas (areas without radiocontamination). DNA was extracted from the altered urothelium of selected paraffin-embedded specimens that showed obviously abnormal histology (3 cases) or intense p53 immunoreactivity (15 cases), and mutational analysis of exons 5-8 of the p53 gene was performed by PCR-single-strand conformational polymorphism analysis followed by DNA sequencing. Nine of 17 patients (53%) had one or more mutations in the altered urothelium. Urine sediment samples were also collected from the patients at 4-27 months after biopsy and analyzed by PCR-single-strand conformational polymorphism analysis or yeast functional assay, and identical or additional p53 mutations were found in four of five cases. Interestingly, a relative hot spot at codon 245 was found in five of nine (56%) cases with mutations, and 11 of the 13 mutations determined (73%) were G:C to A:T transitions at CpG dinucleotides, reported to be relatively infrequent (approximately 18%) in human urinary bladder cancers. Therefore, the frequent and specific p53 mutations found in these male patients may alert us to a future elevated occurrence of urinary bladder cancers in the radiocontaminated areas.

[The functional status of the thyroid system in those irradiated as a consequence of the accident at the Chernobyl Atomic Electric Power Station (based on screening study data)].

A screening study was made of functional indices for the status of the thyroid system in 1097 persons who took part in the elimination of the effects of the Chernobyl accident (PChAEE) during the "iodine" and "non-iodine" periods of irradiation. In 94.6% of the "iodine" period PChAEE, the basal levels of the thyroid-stimulating hormone (TSH) were in the normal range, 4.79% displayed a rise in the parameter. 90.06% of the "non-iodine" period had normal range TSH, 8.77% exhibited augmented levels. 94.68% of the "iodine" period PChAEE showed normal range for the basal levels of free thyroxin, in 3.72% there was a rise, in 1.6% a decrease in the values. Among the examinees of the "non-iodine" period, the levels of free thyroxin were within normal range in 92%, above norm in 5%, decreased in 3%. The level of antibodies to thyroglobulin in 86.38% of the "iodine" period examinees was in the normal range, in 90.99% of the "non-iodine" period it was also normal. There were no statistically significant differences suggesting effects of radiation factors of the "iodine" period of irradiation on the functional indices for the hypophysis-thyroid system during the first 7 years following irradiation.

[The laboratory diagnosis of autoimmune thyroid diseases in those who worked in the cleanup of the aftermath of the accident at the Chernobyl Atomic Electric Power Station].

Serum antibodies to thyroid tissue antigens were tested by enzyme immunoassay and the thyroid hormonal status was analyzed in 80 liquidators of the Chernobyl accident consequences. Antibodies to thyroglobulin in concentration of 160.7 +/- 10.5 IU/ml were detected in 11 (13.7%) subjects 11-12 years after the accident. Antibodies to thyroperoxidase were detected in this cohort: the reaction was positive in 2.5% cases and weakly positive in 5%. Groups of liquidators with changed thyroid functional activity in the T3-T4-TTH system were identified. Immunochemical screening for thyroid hormones and antibodies is an affective method for the diagnosis of autoimmune thyroid diseases in liquidators.

[Analysis of risk factors for the formation of internal exposure dose to the rural population and effectiveness of preventive actions in agriculture in the long-term after the Chernobyl Accident].

The regularities in the formation of internal exposure doses to the population living in the Chernobyl affected areas are described. The factors responsible for changes in doses are identified and their significance is assessed. The effects of countermeasures on the radiation situation variations are estimated. The need for an addressed application of countermeasures is shown with taking into account types of rural settlements based on the radioecological factors responsible for dose formation in the population.

[ Delayed effect of prenatal irradiation on the critical organs functional state in children after Chernobyl accident].

Some indexes of a metabolism in 186 children exposed in utero through the Chernobyl accident were investigated ten years after the irradiation. All the children (basic and control groups) live in the territory with contamination less than 15 Ci/km2. It was shown, that the activity of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase and the level of bilirubin (conjugated and unconjugated) differed from the corresponding indexes in the control group (p < 0.05). These change depended on sex. The correlation between the levels of the total bilirubin, activity of the ACT, ALT in serum and absorbed doze to the thyroid of fetus was found (correlation coefficient > -0.8, p < 0.05). The linear regression equations for these three indexes are straight line: the total bilirubin, ACT and ALT decreased with the increase of the absorbed dose to the thyroid of fetus (p < 0.05). The relationship between the all indexes in serum and effective doze and levels of cesium accumulation was not detected.

[Embryology of ovules from the second-year-old cones of scotch pine after acute irradiation in the zone of Chernobyl nuclear plant].

The embryological processes after acute irradiation in the zone of Chernobyl (radiation level 4-5 Yy) were studied in ovules from the second-year-old cones of scotch pine. Four types of anomalies were detected in ovules of all experimental pine trees: undeveloped ovules in layer of strobiles; a substitution of female gametophit by tissues originaled from tapetal or nucellar cells; death of macrogametophytes connected with the interruption of divisions during the cenocitic stage in full-fertile pine trees. These anomalies appear during the normal development of microgametophytes.

[Sensitivity of soil bacteria isolated from the alienated zone around the Chernobyl Nuclear Power Plant to various stress factors].

Seventy strains of chemoorganotrophic bacteria isolated by our group in 1993-1994 from soil sampled in the zone around the Chernobyl Nuclear Power Plant (ChNPP) were studied with respect to their sensitivity to various stress factors damaging DNA. Bacillus subtilis, B. cereus (both spores and vegetative cells), Methylobacterium extorquens, M. mesophilicum, and unidentified pigmented bacteria were found to be the most resistant to ultraviolet (UV) radiation, exhibiting LD90 values of 40 to more than 211 J/m2. The same bacteria, as well as Bacillus polymyxa, were tolerant to hydrogen peroxide (lethal concentrations of H2O2 ranged from 0.3 to 1.0 M); i.e., UV-resistant strains were also tolerant to hydrogen peroxide and vice versa. Fluorescent pseudomonads were the most sensitive to both UV radiation and H2O2, showing LD90 from 6 to 18 J/m2 and a lethal concentration of H2O2 lower than 0.1 M. All of the soil samples collected in the alienated zone around the ChNPP, where the radioactivity of the soil had decreased from 1000 to 2 microCi/kg soil over the period from 1987 to 1995, contained not only resistant bacteria but also a small number of bacteria sensitive to UV radiation and H2O2.

Parameters of peroxidation and proteolysis in the organism of the liquidators of Chernobyl accident consequences.

The specificity of lung irradiation caused by ionizing radiation is influence on mucous membranes of respiratory ways, alveolar epithelium and capillaries of a small circle of the blood circulation. Under diseases of bronchus-lung system the lipid peroxidation (LPO) processes activation is observed. The radiating influence strengthening effect. In results in imbalance aggravation in system "LPO-antioxidants", and long expressing of LPO intensification is the important mechanism of the inflammation chronization. The sharp increase of proteolytic activity and inhibitor activity decrease is found out in the patients-liquidators. Noticed imbalance results in the further change of permeability of membranes and correlates with an index of endoscopy inflammation changes and index of irreversible changes in lung tissue. Thus, the direct connection between LPO intensity and imbalance degree of proteinase-inhibitor system of blood at the patients with chronic bronchitic taking part in Chernobyl accident liquidation is revealed.

[The clinical course and histological characteristics of medullary hematopoiesis in acute leukemia in those who worked in the cleanup of the aftermath of the accident at the Chernobyl Atomic Electric Power Station].

The course is analyzed of myeloblastic leukemia in twelve participants in the elimination of the aftermath of the Chernobyl Atomic Power Plant breakdown. The iliac trepanobiopsies bone-marrow hemopoiesis is studied. A gross depression of normal hemopoiesis has been revealed together with a fibrous transformation of the bone marrow, which fact suggests a profound damage to the hemopoietic micro-surroundings, being a cause of ineffective hemopoiesis, of grave course of the illness, and of failure to respond to cytostatic therapy.

[The immunohistochemical determination of p53 and of proliferating cell nuclear antigen in the epithelial nuclei of benign prostatic hyperplasia following the accident at the Chernobyl Atomic Electric Power Station].

With the purpose of studying into the morphogenesis and proliferous activity of the prostatic epithelium under a long-term exposure to low doses of ionizing radiation there have been conducted comparative histological and immunohistochemical (expression of p53 and proliferous cellular nuclear antigen-PCNA) investigations designed to study benign prostatic hyperplasia in patients living in those Ukraine territories affected by radionuclide contamination (group III), residents of Kiev (group II), and patients having been operated on before the Chernobyl accident, having constituted the control group I. It has been found out that the incidence of prostatic intraepithelial neoplasia (PIN), the level of nuclear expression of proteins p53 (in the PIN epithelium) and PCNA (in the epithelium of both benign prostatic hyperplasia and PIN) of patients in groups II and III are by far higher as compared with those in group I. The stroma of benign prostatic hyperplasia in patients of groups II and III was clearly different from that in the control group in that the former was characterized by apparent phenomena of hyalinosis, sclerosis, fibrosis, and extensive inflammatory infiltration, which changes can be explained by a long-term systematic exposure of prostatic tissue to low doses of ionizing radiation.

[Biological effects in natural populations of small rodents in radiation contaminated areas. The frequency of bone marrow polyploid cells of bank voles in different years following Chernobyl accident].

On the basis of the metaphase analysis results, the peculiarities of dynamics of the genome mutation frequency (polyploid cells) were studied in bone marrow of bank voles inhabiting the areas with different radiocontamination level due to the Chernobyl accident (8-1526 kBq/m2 for 137Cs) in 1986-1991. Unexpectedly high frequencies of polypoid cells exceeding the pre-accidental level by a factor of 10(1)-10(3) were recorded in all populations studied. Relationship between the frequency of the parameter studied and the concentration of radionuclides incorporated in animal carcasses was proved. A statistically significant rise in the frequency of genome mutations with the time was revealed up to 1991, i.e. approximately to 12th post-accidental animal generation.
