Biosynthesis of glycosylated hemoglobins in the monkey.

We have investigated the in vivo biosynthesis of the minor hemoglobin components in the rhesus monkey. The elution profile of rhesus hemolysate on BioRex 70 cation exchange resin was analogous to that of human hemolysate. The rhesus Hb Alc peak was identified with the TBA test, which revealed a carbohydrate content identical to that of human Hb A1c. A rhesus monkey was injected with autologous 55Fe-bound transferrin, and the specific activity of each of the minor and major components was followed for over 70 days. As previously shown in man, rhesus Hb alc accumulated specific activity almost linearly over the erythrocyte life-span, indicative of slow and continuous conversion of Hb A0 to Hb Alc. This study revealed two new findings. (1) The specific activity of rhesus Hb Alb was always significantly less than that of Hb Alc. This result suggested that HB Alb is made by further posttranslational modification of Hb Alc. (2) The first portion of rhesus Hb A0 to be eluted on BioRex 70 contained a significant amount of carbohydrate and lower initial specific radioactivity than did the latter portion. This unexpected heterogeneity in the major hemoglobin component reflects slow, nonenzymatic glycosylation at sites other than at the N-terminus of the beta-chain.

[Activation of coagulation cascade in children during an idiopathic nephrotic syndrome relapse].

UNLABELLED: The objective of this study was to assess concentrations of selected markers of coagulation in children with relapse of idiopathic nephrotic syndrome during a 6-week therapy. Study groups: 22 subjects (32 relapses)--14 males, 8 females (mean age 7.15 +/- 1.5 y.) with no thrombotic complications were included into the study. All children were clinically steroid-sensitive. METHODS: Coagulation markers (platelet count, thrombin time, APTT, INR, fibrinogen 1 + 2 fragments (F1 + 2), thrombin-antithrombin complexes (TAT), serum levels of D-dimer (DD), fibrin monomers (FM) and antithrombin activity (ATIII)) were measured three times: on admission, after 2 and 6 weeks. The control group consisted of 13 healthy children. RESULTS: Serum concentration of TAT or F1 + 2 did not differ between 3 stages (p > 0.05). However, values at 0 and 2 weeks were significantly higher than in control group (p < 0.05). We found no correlation between TAT or F1 + 2 and FBG, ALB, TCH, TG levels. [table: see text] CONCLUSIONS: The coagulation cascade in relapse of NS was activated during first 6 weeks of therapy whereas metabolic disturbances (low ALB, high FGB, TCH, TG, high platelets) normalized. It is speculative whether it was caused by active immunological process but definitely it resulted in "prothrombotic state" in INS patients.

Microalbuminuria, endothelial dysfunction and cardiovascular risk.

Microalbuminuria was originally considered to be an important new risk factor for diabetic nephropathy. More recently, it has been convincingly shown that microalbuminuria is also an independent risk factor for cardiovascular morbidity and mortality in Type 1 and Type 2 diabetic patients. Even in the non-diabetic background population, microalbuminuria is a risk factor for cardiovascular mortality. What is the link between increased loss of albumin in urine and cardiovascular disease and mortality? As microalbuminuria is apparently associated with increased universal vascular sieving of albumin in terms of the transcapillary escape rate of albumin (TER-alb), microalbuminuria may reflect this universal sieving. The pathophysiology of increased TER-alb is unknown, but could be caused by haemodynamics or damage to the functional properties of the vascular wall. A number of studies have provided evidence of endothelial dysfunction in patients with microalbuminuria, which may be the common link accounting for the associations mentioned above. In this context, a number of markers of endothelial cell dysfunction have been found to be increased in patients with microalbuminuria. In addition, a number of functional in vivo tests of endothelial dysfunction have been performed in Type 1 and Type 2 diabetic patients as well as in normal controls. Overall, these studies indicate the existence of a functional vascular dysfunction in Type 1 diabetic patients and normal controls with microalbuminuria, which may be related to dysfunction of endothelial cells.

[Conception and testing of a regional practice guideline using bronchial asthma as an example].

Frequently, there is a gap between publication and implementation of guidelines into clinical practice. To improve adherence with guidelines, experts recommend among other things local adaptation of guidelines. The goal of the present study was to conceptualize and test such an adaptation in the region of Neckar-Alb in Germany by using asthma guidelines as an example. We conceptualized a three-step approach for regional guideline adaptation: 1) Compilation of existing national and international asthma guidelines by regional experts to develop a draft for regional asthma guideline; 2) regional consensus conference to discuss the draft with local physicians; 3) modified Delphi procedure using regional physicians as "experts" to get consent for the regional asthma guideline. We defined consensus as reaching at least a 2/3 consent for each of the statements. As starting point for the process of guideline adaptation, the regional experts developed a first draft of the regional asthma guideline including 132 recommendations. 60 out of 500 regional general practitioners (GP), internists and asthma specialists participated in the consensus process. After the preparations of the regional experts and the regional consensus conference, only four recommendations had to be changed considerably during the Delphi procedure to reach the consensus criterion. If the recommendations remained unchanged, the whole guideline might be disapproved because they were considered to affect the role of GPs by shifting responsibilities from GPs to asthma specialists. As the result of the adaptation, the first regionally adapted asthma guideline for the Neckar-Alb region fosters the principal responsibility of regional GPs in asthma care, thereby surmounting resistance for implementation that former specialist-centered guidelines provoked. Hence, the new guideline meets the requirements for successfully implementing the recommendations into clinical practice.

The targeted expression of interleukin-2 in human hepatocellular carcinoma cells.

To improve the safety and efficiency of human hepatocellular carcinoma (HCC) gene therapy, we explored the use of a liver-specific promoter and a tumor-specific enhancer to achieve regular IL-2 gene expression for treatment of HCC. The human alpha-fetoprotein (AFP) enhancer [E(AFP)] and the albumin promoter [P(ALB)] were amplified from human genomic DNA. We used eukaryotic expression vector pcDNA-3 for the delivery of the IL-2 gene because this plasmid is a non-transient, fast-selection expression vector. A recombinant plasmid was constructed including the selectable marker neoR gene and the human IL-2 gene derived by the E(AFP) - P(ALB). The liver-predominant expression pattern of the IL-2 gene was observed in the medium of the transfected cells. When human HCC cell lines displaying different levels of AFP and non-hepatocyte tumor cell lines were transfected with the recombinant plasmid, IL-2 was expressed highly in AFP and albumin-positive HCC cells, but low in nonhepatocyte tumor cells. Moreover, the expression level of IL-2 gene was positively proportional to the level of AFP expression in the transfected cells.

Use of a variety of biological parameters in distinguishing cirrhotic from malignant ascites.

Twenty-two different protein measurements were taken in the serum and ascitic fluid of fifty consecutive patients in an attempt to investigate which tests are the most reliable for the differential diagnosis of ascites. Serum and ascitic fluid total proteins (TPR), albumin (ALB), lactate (LAC), ferritin (FER), C3 and C4 complement factors, C-reactive protein (CRP), ceruloplasmin (CER), alpha2-macroglobulin (alpha2MG), haptoglobin (HAP), alpha1-antitrypsin (alpha1AT), alpha1-acid glycoprotein (alpha1AG), transferrin (TRF), immunoglobulins IgG, IgA, IgM and cytokines such as interleukin-1alpha (IL-1alpha), interleukin-1beta (IL-1beta), interleukin-2 (IL-2), interleukin-6 (IL-6), interleukin-8 (IL-8) and tumor necrosis factor-alpha (TNF-alpha) were measured to distinguish between malignant and cirrhotic ascites. Correlations and non-parametric Mann-Whitney tests were used for ascitic fluid:serum ratio comparisons between the two groups. Multivariate analyses were used to determine the most significant biochemical ratio predictors for the differential diagnosis and a recursive partitioning model was constructed. Highly positive correlations (r>0.50) were found between the ratios IgA, IgG, IgM, CER, alpha2 MG, HAP, alpha1AT, alpha1AG and TRF. There was evidence that TPR, ALB, LAC, FER, IgG, CER, alpha2MG, alpha1AT, alpha1AG, TRF and IL-8 ascitic fluid:serum ratios are significnatly higher in patients with malignant neoplasms than in cirrhotics. In the recursive partitioning model the most significant parameters were found to be the ratios of albumin and IL-1alpha. The model fitted allowed for 100% correct classification of ascites. In conclusion, we have shown that a simple and very accurate model based on two ascitic fluid: serum measurements is able to differentiate between malignant and non-malignant ascites.

[Influence of electroporation on the biological activities of primary rat hepatocytes].

OBJECTIVE: To investigate the influence of electroporation on the biological activities of primary rat hepatocyte and to optimize the electroporation conditions introducing foreign genes into hepatocytes. METHODS: A single-pulse procedure was performed at low voltage (220-400 V) but high capacitance (500-950 microF). Its influence on hepatocyte activities was detected by Trypan blue exclusion (TBE) and MTT analysis. Besides, ALB, ALT and LDH in the supernatants of hepatocytes were tested by biochemical assay. RESULTS: Little hepatocyte damage and high survival rate (>90%) was found from 36 hours till 9th day of culture. At 36th hour after electroporation, ALB, ALT and LDH in the supernatants of Group B (220V, 950 microF) and C (400 V, 950 microF) were higher than those of control group. Whereas TBE and MTT analysis failed to indicate the significant difference of cell viability between electroporation groups and control group. CONCLUSIONS: This electroporation procedure is one of the optimal choices to introduce foreign genes into primary rat hepatocytes.

[Usefulness of 99mTc-GSA scintigraphy for estimation of residual hepatic functions and postoperative changes of HH15 and LHL15].

We investigated the usefulness of 99mTc-GSA scintigraphy for estimation of residual hepatic functions and postoperative changes of HH15 and LHL15. 99mTc-GSA scintigraphy was performed on 61 patients and following points were studied: 1) correlation between HH15-LHL15 and conventional liver function tests, 2) correlation between HH15-LHL15 and the Child classification, 3) relations of preoperative HH15 and LHL15 to postoperative complications after hepatectomy, 4) early changes of HH15 and LHL15 after hepatectomy, 5) changes of HH15 and LHL15 1 year after hepatectomy, and 6) changes of HH15 and LHL15 1 year after Hassab's operation. Results were as follows. 1) There were significant relations between preoperative HH15 and ICGR15 and Alb. There were significant relations between preoperative LHL15 and ICGR15, T-Bil, Alb, HPT and ChE. 2) There were significant relations between preoperative HH15-LHL15 and the Child classification. 3) In Hr2 group, preoperative LHL15 of patients who contracted postoperative complications had been significantly lower than that of those who did not. 4) There were no early changes of HH15 and LHL15 after hepatectomy. 5) HH15 and LHL15 were significantly improved 1 year after hepatectomy. 6) HH15 were significantly improved 1 year after Hassab's operation. We concluded that 99mTc-GSA scintigraphy is useful for estimation of residual liver function tests and for investigation of changes after surgery from viewpoints different from conventional hepatic function tests.

[Effect of folic acid supplementation on total homocysteine levels in hemodialysis patients].

UNLABELLED: Hyperhomocysteinemia is an independent risk factor for cardiovascular mortality in ESRD, but about 80% of total homocysteine (tHcy) is bound to albumin (alb). We have tried, prospectively, to reduce tHcy levels by using folic acid (f.a.) and vitamin B6 (P.P.) supplementation. All patients on HD, not receiving f.a. or P.P. and all new patients, after their third month on HD, were supplemented with f.a. 5 mg/48 hours p.o and P.P. 40 mg/week. We determined folate, P.P. (RIA), vit. B12, KTV, residual renal function (KRU), PCRn, alb and tHcy levels (HPLC). 80 patients, age 62.6 +/- 13.6 years, time on HD 16.2 +/- 25.1 months, all dialysed with AN69 or PPMA, and bicarbonate, were included. The prevalence of hyperhomocysteinemia was 84.4%, and P.P. deficit was present in 32%, with folate in the low normal range. At the beginning of the study, before supplementation, tHcy was negatively correlating only with folate (r = -0.336) (p = 0.01), and not with P.P., vitamin B12, age, albumin, KTV, KRU or PCRn. 58 patients received six months of supplementation, with normalization of P.P. levels, a significant increase of folate (7.25; I.C = 95% confidence intervals: 6.45, 8.05 vs 61.29; I.C.: 44.47, 78.11) (p < 0.001), and decrease of tHcy (24.1; IC: 21.5, 26.3 vs 19.9; I.C: 17.5, 22.4) (p < 0.05). 33 patients have received 12 months of supplementation, but in spite of a continued increase of folate (100.78; I.C: 74.81, 126.74) (p < 0.001), only 3 have normal levels of tHcy; correlating directly tHcy with albumin (r = 0.56) (p = 0.001), that had increased compared to the beginning of the study (3.39; I.C. 3.29, 3.49 vs 3.50; I.C: 3.37, 3.63) (p < 0.05). CONCLUSION: After f.a. and P.P. supplementation, though initially tHcy is reduced, this response is short lived, and tHcy directly correlates with albumin levels. Good nutrition associated with HD adequacy, in absence of B vitamin deficits, seems to be the best determinant of tHcy levels rather than its removal by dialysis tHcy levels should be interpreted taking into account the serum albumin.

Stress of chronic food restriction attenuates the development of adjuvant arthritis in male Long Evans rats.

OBJECTIVE: To study the effect of the stress of chronic food restriction on the development of adjuvant arthritis in Long Evans male rats. METHODS: Four groups of animals were compared: non-treated control (C) and arthritic (AA) rats, both with free access to food and water and two analogous groups with a 40% food restriction, i.e. non-treated (FR) and arthritic (AA-FR) animals. All rats were killed 22 days following the injection of complete Freund's adjuvant. The parameters measured were: serum levels of albumin (ALB), nitrate, glucose, insulin, corticosterone (CORT), prolactin (PRL) and PRL mRNA in the adenopituitaries. In addition the activity of gamma-glutamyl transpeptidase (GGTP) was measured in the spleen. The pain threshold was determined by the tailflick method. The body weight of the animals was recorded on day 0, 3, 7, 11, 15 and 18 of the disease. RESULTS: Arthritis caused swelling of the hind paw (2.37 +/- 0.15 ml vs 1.1 +/- 0.05ml in controls, p < 0.01) which was prevented in the AA-FR group (1.44 +/- 0.13 ml, not significant against controls). Arthritis increased serum NO and reduced ALB levels; both changes were significantly restored in the FR-AA group. Food restriction did not alter the activation of GGTP, or the decrease of PRL mRNA observed in the AA group. Serum CORT was elevated in rats with food restriction (15.49 +/- 2.1 vs. 3.7 +/- 0.87 microg/dL) and remained enhanced to the same extent in AA and FR-AA groups. The tailflick latency prolonged in the AA group was reduced by food restriction. CONCLUSION: These results show that 40% food restriction associated with elevated CORT levels mitigated the inflammatory parameters activated during AA.

Is interleukin-6 concentration in the dialysate of continuous ambulatory peritoneal dialysis patients a prognostic parameter in peritonitis?

In continuous ambulatory peritoneal dialysis (CAPD) patients, peritonitis is a dangerous complication. Chemical examinations in the dialysate can be successfully used to assess permeability disturbances, hemostatic balance, and (for early detection and follow-up) cellular inflammatory reaction. In 7 CAPD patients (age: 50 +/- 15 years; dialysis duration: 40 +/- 24 months) with peritonitis episodes, and in 17 age-matched CAPD patients (age: 50 +/- 13 years; dialysis duration: 29 +/- 18 months) without peritonitis, we examined daily dialysate cell count (CC) and concentrations of albumin (ALB), immunoglobulin G (IgG), thrombin-antithrombin III complex (TAT), D-dimer (DD), and interleukin-6 (IL-6) after the long dwell (8-10 hours) over an interval of at least 14 days. In CAPD patients with peritonitis episodes, all parameters (CC, ALB, IgG, TAT, DD, IL-6) were significantly increased in the first days [IL-6 mean: 25,190 pg/mL (range: 2560-52,708 pg/mL) vs 66 pg/mL (range: 21-163 pg/mL)]; then, up to day 14 after successful therapy with antibiotics, the levels showed no differences as compared with CAPD patients without peritonitis. In the case of relapse of peritonitis (4 cases), concentration of IL-6 rose again on day 14, 1 day earlier than did the other parameters. Determination of IL-6 in the dialysate is a reliable prognostic parameter for the course of peritonitis (start, end, relapse) in CAPD patients.

[A case of IgG4-lambda type monoclonal immunoglobulin that interfered with determinations for albumin, direct bilirubin and iron in serum].

We report a human IgG4-lambda type M-protein that reacts with reagents of albumin, direct bilirubin and iron. Blood chemistry findings by an automated analyzer were: total protein 8.8 g/dl; albumin(ALB) 9.2 g/dl; direct bilirubin(D-Bil) -6.2 mg/dl; and iron(Fe) 25 micrograms/dl. The white precipitates were formed by the interaction of the patient's serum with reagents of ALB, D-Bil, and Fe. Immunofixation electrophoresis revealed that these precipitates were the IgG4-lambda type M-protein. Western blotting analysis showed that the IgG4 molecules were composed of two 55 kDa gamma 4 chains and two 24 kDa lambda chains.

Brain natriuretic polypeptide in type 2 NIDDM patients with albuminuria.

We investigated plasma brain natriuretic polypeptide (BNP) in Type 2 NIDDM patients with albuminuria. This study involved normal control subjects (Controls), hypertensive patients without diabetes mellitus (HT) and Type 2 NIDDM patients. Diabetic patients were divided into 4 groups by urinary albumin index (Alb-I): group I <30; group II 30-149; group III 150-300; group IV >300 mg/g creatinine. BNP levels in group III or IV were significantly higher than in Controls. The diabetic patients were re-divided into 4 groups (DM: with neither diabetic retinopathy (which was one of indicators of microangiopathy) nor hypertension, DM+HT: without diabetic retinopathy and with hypertension, DM+R: with diabetic retinopathy and without hypertension and DM+R+HT: with both diabetic retinopathy and hypertension). The Alb-I levels in DM+HT and DM+R+HT were significantly higher than the Controls, indicating the development of diabetic nephropathy in those groups. BNP levels in DM+HT, DM+R or DM+R+HT were significantly higher than in Controls. This indicates that BNP levels elevate in diabetic patients with increased albuminuria due to hypertension or microangiopathy deteriorating diabetic nephropathy. Elevated plasm BNP may play a pathophysiological role in the development of diabetic nephropathy.

Differentiation of human hepatoma Hep G2 cells induced by 10-hydroxycamptothecin.

OBJECTIVE: To study the differentiation-inducing effect of hydroxycamptothecin (HCPT) on hepatoma Hep G2 cells. METHODS: 3-[4, 5-dimethylthial-2-yl]-3, 5-diphenyl-tetrazolinium bromide (MTT) colorimetric method was used for determining HepG2 cells viability. Alphafetoprotein (AFP) and albumin (ALB) levels were determined with Elisa Kits. RESULTS: The AFP secretion of HCPT-treated cells was reduced by 44.0%-49.2% while the ALB level was increased markedly. CONCLUSION: Hydroxycamptothecin can induce human hepatoma Hep G2 cells to differentiation.

Relationship between the burden of Pneumocystis carinii, the inflammatory reaction and lung injury in Pneumocystis carinii pneumonia.

OBJECTIVE: To study the relationship between the burden of Pneumocystis carinii (P. carinii) and the inflammatory reaction and biochemical markers in bronchoalveolar lavage fluids (BALF) in a rat model of P. carinii pneumonia (PCP). METHODS: Clean grade 50 male Sprague-Dawley rats were immunosuppressed by a subcutaneous injection of 25 mg cortisone acetate twice a week for 8-12 weeks; the PCP model was successfully induced in 14 rats. The inflammatory reaction and biochemical markers of the activity of lactate dehydrogenase (LDH), alkaline phosphatase (ALP) and type IV collagenase (matrix metalloproteinases, MMP-2, MMP-9) as well as the values of total protein (TP) and albumin (ALB) in BALF between the mild burden group of P. carinii (involved alveoli < 25% per 100 alveoli, Group A) and the moderate to severe burden group (involved alveoli > or = 25% per 100 alveoli, Group B) were measured. The other six clean grade SD rats served as normal control group (Group C). RESULTS: The total white cell count in BALF was higher in Group B [(6.8 +/- 1.7) x 10(6)/L] than in Group A [(3.8 +/- 1.2) x 10(6)/L] (P < 0.01); however, there were no differences in white cell differentiation. Assays of biochemical markers showed that ALB in BALF in Group B (0.893 +/- 0.469 g/L) was increased in comparison with Group A (0.262 +/- 0.169 g/L); it was only 0.026 +/- 0.021 g/L in Group C. The contents of TP and activities of LDH were higher in Group B (TP 1.756 +/- 0.706 g/L, LDH 2580 +/- 550 U/L) than in Group A (TP 0.784 +/- 0.553 g/L, LDH 1410 +/- 620 U/L); the values of TP and LDH were 0.063 +/- 0.020 g/L and 370 +/- 250 U/L respectively in Group C. The activity of Type IV collagenase, including MMP-2 and MMP-9, was higher in Group B than in Group A (P < 0.01) (MMP-2: 1102 +/- 169 grey value vs 459 +/- 274 grey value; MMP-9: 1218 +/- 257 grey value vs 449 +/- 225 grey value). There was no activity of Type IV collagenase in BALF of Group C. No statistically significant difference was observed in ALP between the groups B and A. CONCLUSIONS: These results indicate that there is a significant correlation between the burden of P. carinii in lung tissues and the inflammatory reaction as well as biochemical markers of the resultant activity of lung injury.

[Pathologic grading and clinical features of chronic hepatitis B].

OBJECTIVE: To raise the accuracy of clinical diagnosis of chronic hepatitis B. METHODS: The correlation between the clinical features, biochemical tests (the serum total bilirubin-TBil, albumin-ALB, prothrobin activity-PTA, alanine aminotransferase-ALT, albumin/globulin-A/G, and r-globulin-r-G) and histopathological data in 202 patients with chronic hepatitis B (CHB) were studied. RESULTS: Some of presenting symptoms and signs were obviously associated with histological grade and stage. The grade of necroinflammatory activity of CHB was associated with the rising TBil, ALT, GGT and the declining ALB, A/G and PTA. The coincidence of clinical diagnosis and pathology was highest in mild chronic hepatitis, 63.8%~79.0%; then was in marked chronic hepatitis, 40.0%~62.5%. The coincidence was lowest in moderate chronic hepatitis, 10.0%~28.2%. CONCLUSIONS: Great attention should be paid to the significance of symptoms and signs, meanwhile the standard of clinical diagnosis for moderate chronic hepatitis might be relaxed somewhat.

[Experimental and clinical studies of recombinant human growth hormone treatement of hypoproteinemia of liver cirrhosis patients].

BACKGROUND: To assess the effect of recombinant human growth hormone (rhGH) therapy to liver cirrhosis patients with hypoproteinemia. METHODS: 31 liver cirrhosis patients were treated with rh GH (0.25 IU/kg/day-) and 20 such patients with human albumin (10 g/day) as control all for 10 days. Serum levels of growth hormone (GH), insulin-like growth factor (IGF)-1 and albumin (ALB) were measured by radio-immunoassay before 24h and after 24h and 10 days of rhGH therapy. Serum levels of GH and IGF-1 of 10 age matched healthy subjects were also measured. RESULTS: As compared with healthy subjects, the pretreatment serum GH level of liver cirrhosis patients was high (4.96 +/- 3.97 vs 1.62 +/- 2.26, P <0.05), and the IGF-1 level was markedly lower (36.53 +/- 12.77 vs 56.95 +/- 16.46, P <0.01). After 10 days of rhGH administration, the serum IGF-1 and ALB levels of the treated group increased significantly as compared with the control group (P <0.01). CONCLUSIONS: Exogenous rhGH therapy can overcome the GH resistance of liver cirrhosis patient s and can increase significantly the serum albumin level as compared with the control group who were treated with human albumin only.

[Analysis of renal function on 1,743 people of different ages].

OBJECTIVE: To understand and evaluate the currency and correlation between parameters about renal function. METHODS: We divided 1,743 people whose ages vary from 20-85 years into six groups(A, B, C, D, E, F). Urinary Mini Albumin, RBP, NAG, osmotic pressure, serum BUN, plasma UA and internal creatinine clearance were measured as parameter about renal function. RESULTS: There was no obvious difference between the Group A and Group B(P > 0.05); The mean value of urinary NAG and urinary RBP in the two group was 10.9650 +/- 6.5650 and 0.1885 +/- 0.4709; However, when they were compared with the values of C, D, E group respectively, there were prominent differences (P < 0.05 or P < 0.01). Urinary Alb value of Group A is quite different from other groups. No difference exists between urinary Alb value of Group E and Group F(P > 0.05). Internal creatinine clearance decrease by the rate of 8.5%, 17.8%, 29.9%, 42.9% and 56.3% when Groups B, C, D, E and F was compared with Group A; while urinary osmotic pressure and the value of plasma BUN, Cr and UA has no obvious difference between any two groups(P > 0.05). CONCLUSION: The clearance of internal creatinine decrease with the growing of the age, especially older than 50 years of age. The parameter about tubular injury increase obviously after 40 years old though it does not change prominently before 50 years old, while it tends to be stable after 70 years old. We think it is during period of 40 to 70 years old that the renal function decreases more rapidly.
