Pathological alterations in the ovaries of Culex pipiens induced by fixed oil extracts from Thevetia peruvine, Datura stramonium and Acacia sp.

The fixed oil extracts from Thevetia peruvine, Datura stramonium and Acacia sp. were tested on Culex pipiens larvae. The estimated sublethal concentrations (LC50) were used in the present studies. The reproductive potential of females and the histochemistry of their ovaries were determined. The results indicated that oil treatments of larvae caused drastic changes in reproduction potential of female mosquitoes including the ovarian development in the first gonotrophic cycle, fecundity and fertility of treated females. Histological and histochemical studies revealed clear changes in the oocytes of females derived from treated larvae. Treatment with the oils reduced the vitellogenesis, synthesis of proteins, carbohydrates, lipid contents and DNA and RNA materials. The reproduction was always significant and may reflect the shortage in reproductive potential, the state of being disturbed. Generally, these fixed oils merit further investigations as a potential suppressant of Culex pipiens because their abilities to kill the early stages, inhibit growth and development of ovaries and reduce the reproductive potential of adult females. The three oils have variable activities but they are similar in their mode and site of actions.

The toxicity of Datura stramonium (thorn apple) to horses.

Meal contaminated by Datura stramonium seeds at the rate of 0.5% by weight was fed to two horses. Both horses showed clinical signs of depression, anorexia, weight loss, rapid heart and respiration rates, mydriasis, polyuria, polydipsia and diarrhoea. Both recovered with treatment. Maize screenings contaminated by the seeds had been used in the manufacture of the meal.

Datura stramonium agglutinin-reactive alpha-fetoprotein isoforms in hepatocellular carcinoma and other tumors.

By means of Datura stramonium agglutinin (DSA) affinity electrophoresis, human alpha-fetoprotein (AFP) was resolved into five bands, AFP-D1, D2, D3, D4 and D5, in order of decreasing mobility. AFP-D1, which had no affinity for DSA, comprised more than 84% of the intensity of total AFP bands. The percentage of AFP-D2 increased marginally in hepatitis and liver cirrhosis with or without hepatocellular carcinoma. AFP-D3 increased characteristically in hepatocellular carcinoma and AFP-D4, which had the highest affinity for DSA, increased up to 12% in other tumors, mostly of gastrointestinal origin. AFP-D5 showed no consistent changes among the benign and malignant diseases. The assay of AFP-D3 and D4 proved useful as a highly specific marker of hepatocellular carcinoma and other tumors, respectively.

Studies on lectins. XXXVII. Isolation and characterization of the lectin from Jimson-weed seeds (Datura stramonium L.).

The lectin of Jimson-weed seeds (Datura stramonium L.) was isolated by affinity chromatography on a polysaccharide mixture from mycelium of Aspergillus niger. The lectin yields two bands on disc electrophoresis, it has sedimentation coefficient s20,w = 3.8 S and its apparent molecular weight estimated by thin layer gel chromatography is 120,000. The lectin reduced with mercaptoethanol yields on polyacrylamide gel electrophoresis in the presence of dodecyl sulfate three zones corresponding to subunits of molecular weight 72,000, 45,000 and 25,000. The lectin contains large amounts of cystine, glycine, 6.3% of hydroxyproline residues, 4.5% glucosamine and 28% of neutral sugar, predominantly arabinose. The lectin is nonspecific in human erythrocyte ABO system, it is not inhibited by simple sugars but is inhibited by a partial hydrolysate of chitin-containing mixture of polysaccharides from Aspergillus niger.

Lipids of two Cassia species and Datura stramonium seeds.

The fatty acid constitution of three seed-oils, namely, Cassia alexandrina, Cassia italica and Datura stramonium, were carried out using gas chromatographic analysis. The glyceride structure of these oils was computed using the technique of pancreatic lipase hydrolysis. The phospholipids fraction of these oils were identified using thin layer chromatography, also the fatty acid constitution of the phosphilipids were determined quantitatively by gas liquid chromatography.
