[Optimal measurement technic for the vasoconstrictor effect of corticosteroids. Comparative studies with reflection photometry and flow measurement].

In 20 healthy male test subjects, a non-specific ointment base was applied to the back of the hand over 16 hours. Two days later, the same ointment base with 0.25% triamcinolone acetonide was once more applied to the back of the hand over 16 hours. The light reflection of the skin was measured by reflection photometry and the heat transport coefficient was measured fluvographically under the condition of the tourniquet test. The results show that the vasoconstrictor effect of triamcinolone acetonide is best detected by the reflection photometric measurements. In fluvographic measurement, the heat transport coefficient is most affected in maximal reactive hyperemia when the tourniquet has been removed. Application of the base led to a reduction of the blood flow and the water content of the skin after 16 hours, whereas the water content of the skin was unaffected in the presence of triamcinolone acetonide.

Lithium test-dose methodology using flame photometry: problems and alternatives.

Two published methods of assessing test-dose 24-hour serum lithium concentrations as measured by flame emission spectrophotometry were compared in 17 patients. The first method was unsuccessful in predicting required maintenance dosage in 53% of patients studied. By applying the second methodology, a strong positive correlation (r = 0.69) was obtained between the reciprocal of the maintenance dose required to achieve a steady-state serum lithium concentration of 1.0 mEq/L and the test-dose 24-hour serum lithium level. Problems relating to the use of flame photometry to measure serum lithium concentrations less than 0.05 mEq/L are discussed.

Sodium and potassium measurements: direct potentiometry and flame photometry.

Data from the 1979 and part of the 1980 College of American Pathologists (CAP) Chemistry Survey Program have been reviewed in order to compare the direct potentiometric measurement of sodium and potassium with flame-emission photometric measurements. As may have been predicted from earlier intralaboratory evaluations, the results of the CAP interlaboratory comparison confirm that flame-emission photometry of the two electrolytes yields lower mean concentrations. Reasons for this difference lie in the physicochemical aspects of the sample and methods. In Survey specimens, the magnitude of difference was rarely one of clinical significance, but the documented increase in the use of direct electrode measurements in laboratories warrants informing clinicians of potentially significant differences in selected patients.

Multiwavelength photometry of thermochromic indicator solutions for temperature determination in multicuvettes.

If buffer/indicator systems are used as optical thermometers in multicuvettes, the temperature resolution is limited by errors of optical measurements produced mainly by variations of pathlength and blank transmittance of the wells. Theoretical and practical approaches show that, in multicuvettes, a between-well temperature resolution of < 0.05 degrees C can be achieved by multiwavelength photometry with use of the Tris/cresol red indicator system. Using up to three absorbances (A0, A1, A2) measured in the same well at different wavelengths for calculation of differences, quotients, and quotients of differences, we found the optimum temperature signal to be In(A1/A2), with equal-ranking absorbances changing with temperature in opposite directions. We have used the method successfully to measure the temperature profiles and temporal dynamics of temperatures at all positions in two types of multicuvettes under controlled conditions, or not controlled thermostatically.

[The quantitative determination of the protein content of milk by means of violet spectral photometry. 7. The milk analyzer MAG 100 (Carl Zeiss Jena GmbH)--a possibility for simultaneous measurement of protein and fat content in raw milk at the dairy farm].

A new analysing apparatus is introduced, which allows to measure protein and fat contents of milk in the dairy farm. The instrument works on the basis of ultraviolet spectral photometry. Before measuring the milk sample must be diluted with a hydrous detergent solution in the ratio 1:100 and then homogenized. Accuracy and precision of the analytic results are influenced by detergent type, the water quality of detergent solution, the detergent foam, the air content and the pH of detergent solution, the reliability of homogenizer and thermostating system, the stability of photometer and by the error of dosage. Also a milk standard was developed, which is required for daily calibration of the apparatus.

Measurement of plasma levels of tricylic psychoactive drugs and their metabolites by UV reflectance photometry of thin layer chromatograms.

A method has been developed for the determination of perazine, clozapine, imipramine and amitriptyline and their demethylated metabolites in plasma. Other metabolites measured were perazine sulfoxide and the N-oxides of clozapine and perazine, the latter two following their reduction to the parent drugs with ascorbic acid. 10-Hydroxynortriptyline was identified as an amitriptyline metabolite in plasma. The general procedure included extraction of alkalinized plasma samples (3 - 6 g) with benzene or toluene and thin layer chromatography of the extracts, followed by reflectance photometry of the plates at appropriate wave lengths in ultraviolet light. Spots of questionable identity were further characterized by two-dimensional chromatography and by colour reactions. Therecoveries of compounds added in therapeutic concentrations were between 70 and 98 %. The limits of detectability were 5 - 10 ng/g plasma.

Minimum enzyme unit for Na+/K+-ATPase is the alpha beta-protomer. Determination by low-angle laser light scattering photometry coupled with high-performance gel chromatography for substantially simultaneous measurement of ATPase activity and molecular weight.

The oligomeric state of canine renal NA+/K+ -ATPase solubilized by octaethylene glycol n-dodecyl ether (C12E8) was studied by means of low-angle laser light scattering photometry coupled with high-performance gel chromatography (HPGC). At around 0 degree C the solubilized enzyme was separated into the (alpha beta)2-diprotomeric and alpha beta-protomeric protein components with Mr values of 302,000 +/- 10,000 and 156,000 +/- 4,000, respectively, in approximately equal quantities. As the temperature of chromatography was increased toward 20 degrees C, the two protein components converged into a single major component. The Mr of this component depended on the monovalent cation included in the elution buffer, and was 255,000 or 300,000 in the presence of 0.1 M NaCl or 0.1 M KCl, respectively. A computer simulation technique showed that the solubilized enzyme was in a dissociation-association equilibrium of 2 protomers = diprotomer at 20 degrees C, and the difference in apparent Mr of the solubilized enzyme between the two species of monovalent cation was interpreted by an association constant (Ka) in the presence of 0.1 M KCl that was about 50-fold larger than in the presence of 0.1 M NaCl. In order to measure ATPase activity and Mr of the solubilized enzyme simultaneously, a TSKgel G3000SW column had been equilibrated and was eluted with an elution buffer containing 0.30 mg/ml C12E8 and 60 microgram/ml phosphatidylserine (bovine brain) as well as the ligands necessary for the enzyme to exhibit the activity at pH 7.0 and 20 degrees C. The solubilized enzyme was always eluted as a single protein component irrespective of the the amount of the protein applied to the column, ranging between 240 and 10 microgram. The Mr of the protein component, however, decreased from 214,000 and 158,000 with the decrease of the protein amount. The specific ATPase activity, however, remained constant at a level of 64 +/- 4% of that of the membrane-bound enzyme even in the range of protein concentration sufficiently low as to allow the enzyme to exist only in the protomeric form. Thus, the alpha beta-protomer is concluded to be the minimum functional unit for the ATPase activity. The value of Ka obtained from the concentration-dependent dissociation curve was 5 . 10(5) M-1 for the enzyme turning over, and 1.1 . 10(7) M-1 for the enzyme inhibited with ouabain. It was discussed, based on the values of Ka obtained, that the enzyme would exist as the diprotomer or the higher oligomer in the membrane.

Interfacing gradient elution ion-exchange chromatography and low-angle laser light-scattering photometry for analysis of proteins.

Molecular weights (MWs) of different proteins were determined by interfacing gradient elution ion-exchange chromatography and low-angle laser light-scattering photometry (IEC-LALLS). A high-performance strong cation-exchange column was used to elute proteins using fast (5 min) and conventional (15-30 min) gradients. The eluted proteins were characterized on-line by determining their MWs using LALLS. The specific refractive index (RI) increment (dn/dc) and the RI of the solvent used over the gradient range were determined off-line and used to calculate the absolute weight-average MWs. Four proteins, ribonuclease A, alpha-chymotrypsinogen A, trypsinogen and beta-lactoglobulin A (beta-LACT) were studied. Accurate MWs were obtained for all the proteins using fast and conventional gradients, except for beta-LACT, which aggregated as a function of the gradient employed. The degree of aggregation of beta-LACT increased as the rapidity of the gradient was increased over a fixed gradient range. This study indicated that it is possible to separate and characterize proteins rapidly using IEC-LALLS.

Characterization of further association of the trimeric membrane protein porin by low-angle laser-light scattering photometry coupled with high-performance gel chromatography.

Porin (OmpF), a trimeric membrane protein, in an extract of the outer membrane of Escherichia coli gave a twin-peaked elution pattern on Sephacryl S-300HR gel chromatography in the presence of sodium dodecyl sulphate. The species eluting earlier and later were found to be the hexamer and trimer, respectively, from molecular mass determination by low-angle laser-light scattering photometry coupled with TSK-G3000SWXL gel chromatography. As the hexamer was dissociated into the trimer under the conditions of sodium dodecyl sulphate polyacrylamide gel electrophoresis, its presence had been overlooked. The addition of lipopolysaccharide, another component of the outer membrane, and subsequent dialysis induced association of the trimer, the product containing an appreciable amount of the hexamer.

Protein-protein interaction of detergent-solubilized Ca2(+)-ATPase during ATP hydrolysis analyzed by low-angle laser light scattering photometry coupled with high-performance gel chromatography.

Protein-protein interaction of detergent-solubilized Ca2(+)-ATPase was examined, employing low-angle laser light scattering photometry coupled with high-performance gel chromatography. When solubilized with octa(ethylene glycol) mono-n-dodecyl ether (C12E8) and chromatographed in the presence of 0.3 mg/ml C12E8, the Ca2(+)-ATPase emerged as a single peak with an intermediate molecular weight between the monomer and the dimer, showing a dissociation-association equilibrium of the two components. In the presence of 50 micrograms/ml phosphatidylcholine and 0.3 mg/ml C12E8 at 0 degrees C, the Ca2(+)-ATPase (0.8 mg) emerged as the two distinct components with molecular weights of 125,000 +/- 2100 (n = 3) and 211 300 +/- 7300 (n = 3), indicating that there was no rapid interconversion between the monomer and the dimer. Under the latter conditions, addition of ATP induced fusion of two components. The apparent molecular weight of the fused peak shifted from the monomer to the dimer as the amount of protein increased. Addition of ADP or adenosine 5'-(beta, gamma-methylene triphosphate), however, did not induce such fusion of the peaks. The ATP-induced fusion of the peaks was not observed either in 5 mM CaCl2, the conditions in which the rate of ATP hydrolysis was extremely slow. Thus, the solubilized Ca2(+)-ATPase underwent a rapid interconversion between the monomer and the dimer during ATP hydrolysis. These results suggest that the protein-protein interaction during ATP hydrolysis is an intrinsic nature of Ca2(+)-ATPase and that such interaction may be important for Ca2+ transport by Ca2(+)-ATPase in the sarcoplasmic reticulum membranes.

In situ evaluation of immunoglobulin synthesis of synovial plasma cells in rheumatoid arthritis and osteoarthritis by plug photometry.

Synovial membranes of patients with osteoarthritis--a disease with unaltered immunoregulation--and rheumatoid arthritis were immunostained for the different immunoglobulin isotypes using the PAP-method and a two-layer alkaline phosphatase technique. Randomly selected plasma cells with positive immunostaining were measured by plug photometry. The average extinction of the single plasma cell is considered to be indicative of the immunoglobulin synthesis of the measured plasma cell and the isotype under study. When we compared both disease entities, patients with rheumatoid arthritis were characterized (i) by higher average extinction values for all Ig-isotypes except IgG (ii) by a higher rate of high producer plasma cells. From this data, we conclude that immunophotometry is a new in vivo method for discriminating two types of synovitis (i) one without increased Ig-synthesis at the single cell level and (ii) a second one with increased Ig-synthesis.

Determination of cyclamate in low-calorie foods by high-performance liquid chromatography with indirect visible photometry.

A rapid and simple method using reversed-phase high-performance liquid chromatography combined with indirect visible photometry at 433 nm was developed to determine cyclamate in some food samples. Cyclamate was not detected in these chosen samples as its use is banned in Hong Kong. Cyclamate can easily be detected in spiked samples using a mobile phase consisting of 30 mumol dm-3 Methyl Red and 0.02 mol dm-3 phosphate buffer (pH 7.0)-methanol in a volume ratio of 3:2. The column temperature was set at 23 degrees C. The detection limit was 0.14 mmol dm-3 and the relative standard deviation of the peak area response was 0.58% for a solution containing 5.0 mmol dm-3 of cyclamate (n = 8). This method was successfully applied to the analysis of eight spiked food samples and the cyclamate recoveries for these samples ranged from 93 to 99%.

Carprofen inhibition of flare in the dog measured by laser flare photometry.

The purpose of this study was to determine whether oral carprofen (Rimadyl(R)) treatment in dogs could prevent or decrease the breakdown of the blood-aqueous barrier. The topical pilocarpine irritative model was used to induce breakdown and cause flare. Pilocarpine was instilled in both eyes of seven dogs at time zero and again 5 h later. At 7 h, laser flare photometry was used to measure the flare concentration in each eye using the Kowa FC-1000 laser flare cell meter. All treatments were then discontinued. Two days later, carprofen was administered to the same dogs for a total of three doses. After the last dose of carprofen, pilocarpine treatments and flare measurements were repeated. Carprofen pretreatment resulted in a 68% inhibition of flare, which was highly significant (P < 0.01). The pilocarpine group had a mean of 16.1 photon counts per millisecond (PC ms-1) +/- 2.2 SE, and the carprofen group had a mean of 7.0 PC/ms +/- 1.2 SE. These results compare favorably with previous studies measuring increased protein or fluorescein concentrations in the aqueous humor after blood-aqueous barrier breakdown in the dog. These results suggest that carprofen may be effectively used as a systemically administered ocular anti-inflammatory drug. Carprofen has the added benefit of fewer reported side effects.

Tetracycline fluorescence incident photometry: a new technique to quantitate bone formation.

We describe a technique for measuring new bone formation by tetracycline fluorescence incident photometry (TFIP) based on the affinity of tetracycline for newly calcified bone. Unlike traditional fluorescent labeling techniques, which measure appositional bone growth, TFIP measures the total amount of newly labeled bone by quantitating the amount of fluorescent light emitted by tetracycline bound to new bone in response to ultraviolet light excitation. TFIP offers a technically simple and accurate technique for quantitative comparison of new bone formation. It may have broad applications in experimental as well as clinical situation.

The effect of periosteal preservation on osteogenesis in a canine rib autograft model: tetracycline fluorescence incident photometry.

In the first portion of this study, we describe a new technique, tetracycline fluorescence incident photometry (TFIP) for quantifying new bone formation. In this portion, we use routine histology, Microfil vascular injection, and tetracycline incidence fluorescence (TFIP) to determine the effect of periosteal preservation on bone formation in canine rib autografts. We found that periosteal preservation significantly enhanced new bone formation in both cortical and trabecular bone (83-70% of control) when compared with autografts without the periosteum (76-60% of control) (p < 0.05). In addition, periosteal preservation favorably influenced graft revascularization. We found that the periosteum of the transferred rib autografts was made up of 3 distinct layers: (1) the inner (cambial) layer of osteogenic cells, (2) the middle (fibrous) layer of osteogenic reserve cells, and (3) the outer vascular network of arterioles and venules, which communicate with the trabecular vessels internally. This outer vascular network has not been previously described in transferred bone grafts and is responsible for early graft revascularization.

Imaging and photometry of comet C/1999 S4 (LINEAR) before perihelion and after breakup.

We analyzed photometric measurements and images of comet C/LINEAR before perihelion and after its breakup. Results from our photometry data include a lower limit of 0.44 kilometer for the radius of the nucleus before breakup, and a determination that it was depleted in carbon-chain molecules relative to most other comets. Our imaging and modeling results, which include a constraint on the rotational state of the nucleus, indicate that the disintegration likely started on 18 or 19 July 2000. The total mass detectable in the dust tail after the breakup was 3 x 10(8) kilograms, comparable to one of the fragments in the Hubble Space Telescope images; we therefore infer that most of the comet's original mass is hidden in remnants between 1 millimeter and 50 meters in diameter.

Haemoglobin saturation in the rat liver after ischaemia and reperfusion: study using a laser photometry technique and correlation with changes in liver tissue blood flow.

Tissue oxygenation depends on the volume of oxygen consumed by the tissue and the volume of oxygen supply. This is particularly important in the liver after ischaemia and reperfusion, due to the relatively low oxygen saturation of the portal blood flow, the main source of oxygen to the liver. In this study we established a correlation between the postischaemic liver blood flow and tissue haemoglobin saturation, measured by laser Doppler flowmetry and laser surface photometry, with the purpose of investigating the possible role of a postischaemic imbalance of oxygen delivery/uptake in reperfusion injury. The experimental procedure consisted of the temporary interruption of blood flow to the left lateral and medial lobes of the rat liver in vivo, and subsequent reperfusion after defined periods, recording the postischaemic liver blood flow and liver oxyhaemoglobin saturation. Changes were found in the postischaemic liver blood flow and haemoglobin saturation in all the groups when compared to control values, showing a correlation between the length of the period of ischaemia and the magnitude of the alteration in the reperfusion blood flow and oxygenation. These alterations may be considered as a prolongation of the metabolic condition of ischaemia and may be part of an additional tissue damage upon reperfusion.

Sodium measurements in the presence of paraproteins by four direct ISE methods and flame photometry compared.

Sodium was determined by flame photometry and by direct potentiometry using four different systems (Ektachem DT60, Corning 614, Orion 1020 and Nova-1) in serum samples from patients with normal proteins and paraproteins. Flame photometric and direct potentiometric differences (ISE-flame), showed the expected relationship with total protein whether or not the sample contained paraprotein, an exception being the Ektachem DT60 on normal protein. With the exception of the Ektachem DT60 with samples with normal protein levels, correlation between different ISE-flame estimates was good. Differences between direct potentiometric measurements are probably more attributable to efforts made by manufacturers to relate them to flame photometric measurements in 'normal samples', than to the volume occupied by increased protein content. The apparent lack of influence of protein on ISE-flame for the Ektachem DT60 was shown to relate to the original external reference electrode salt bridge used. A second study showed that changing this fluid to an equi-transferrant high salt content caesium chloride based reference fluid partially unmasked the protein effect.
