Dissociation and reassociation of immobilized porphobilinogen synthase: use of immobilized subunits for enzyme isolation.

The dissociation and association of an immobilized preparation of the octameric enzyme porphobilinogen synthase [5-aminolevulinate hydro-lyase (adding 5-aminolevulinate and cyclizing), EC 4.2.1.24] is described. On treatment of the immobilized preparation with 4 M urea, four subunits per octamer are removed which can be reassociated into a soluble octameric enzyme. The tetrameric bound residual protein can also be reassembled into an octameric structure, with the same initial enzyme activity, by exposing the residual bound protein to a soluble pure enzyme preparation or to a crude liver extract in the presence of urea. The dissociation of the reconstituted bound enzyme releases subunits that again can be reassembled into a soluble octameric pure protein even when the crude liver preparation is used as the donor of the subunits. Thus, a pure enzyme can be isolated in a reassociation-dissociation cycle. The use of immobilized preparations of oligomeric proteins is considered for intra- and interspecies hybridization studies and for the ready preparation of purified enzyme preparations from different species and is suggested as a model for study of the formation of an oligomeric enzyme in the presence of other polypeptides.

Use of immobilized Candida yeast cells for xylitol production from sugarcane bagasse hydrolysate: cell immobilization conditions.

Candida guilliermondii cells were immobilized in Ca-alginate beads and used for xylitol production from concentrated sugarcane bagasse hydrolysate. A full factorial design was employed to determine whether variations in the immobilization conditions would have any effects on the beads, chemical stability and on the xylitol production rates. Duplicate fermentation runs were carried out in 125-mL Erlenmeyer flasks maintained in a rotatory shaker at 30 degrees C and 200 rpm for 72 h. Samples were periodically analyzed to monitor xylose and acetic acid consumption, xylitol production, free cell growth, and bead solubilization. Concentrations of sodium alginate at 20.0 g/L and calcium chloride at 11.0 g/L and bead curing time of 24 h represented the most appropriate immobilization conditions within the range of conditions tested.

Glycosaminoglycan metabolism in experimental osteoarthritis caused by immobilization. The effects of different periods of immobilization and follow-up.

Glycosaminoglycan metabolism at different developmental stages of osteoarthritis provoked by immobilization was studied in rabbits. Tissue concentrations and the specific radioactivity of glycosaminoglycans (GAG) after a long immobilization (IZ) period (12 weeks) without follow-up, and a moderate IZ time (4-7 weeks) with a long follow-up period (average 22 weeks) were compared with the results obtained after a short IZ time (17 days). In tibial weight-bearing cartilage the specific 35S-activity had increased and the galactosamine, glucosamine and uronic acid concentrations had decreased in all the IZ groups examined. In the marginal cartilage, the menisci and the collateral ligament, the specific radioactivity and GAG concentrations had decreased or were normal after a long IZ with or without follow-up, although they were markedly elevated after a short IZ time. THe depletion of GAG correlated roughly with the IZ and follow-up times, except for periodically immobilized rabbits. In long-term "chronic" disease the specific radioactivities and GAG concentrations were depressed on the contralateral, non-IZ sides. The changes in the non-IZ knee are discussed. In studies of experimental osteoarthritis it is important to determine the developmental stage of the disease to be studied.

Immobilization of Norwegian reindeer (Rangifer tarandus tarandus) and Svalbard Reindeer (R. t. platyrhynchus) with medetomidine and medetomidine-ketamine and reversal of immobilization with atipamezole.

The sedative action of medetomidine (-ketamine) was studied in 12 captive Norwegian semidomesticated reindeer (NR), including 4 newborn calves, and in 7 free-living Svalbard reindeer (SR). Medetomidine, with or without ketamine, caused effective, reliable immobilization in NR. Doses of 50-200 micrograms/kg medetomidine alone or 30-125 micrograms/kg medetomidine combined with greater than or equal to 300 micrograms/kg ketamine induced complete immobilization, good muscle relaxation and persistent, deep sedation with little respiratory depression in NR; SR required higher doses. Atipamezole successfully antagonized medetomidine (-ketamine) resulting in rapid and persistent reversal of immobilization in all cases (NR and SR). Both medetomidine and atipamezole had wide safety margins and no conspicuous lasting side effects after reversal.

[Coordination-ionic enzyme immobilization. Dependence of properties of immobilized preparations of penicillinamidohydrolase on the nature of metal ion and stationary ligand].

Penicillinamidohydrolase was immobilized on heteroporous polystyrene supports containing groups of ethylenediamine, diethylenetriamine and triethylenetetramine, using Co(II), Ni(II), Cu(II), Fe(III), Co(III) ions. The effects of substrate concentrations and pH on the activity of the immobilized enzyme were investigated and the kinetic parameters of hydrolysis were determined, using N-phenacetylphenylglycine as substrate. The enzyme immobilization resulted in a slight decrease of Km(app), while the values of V decreased by a factor of two. Preparations possessing high thermal stability were obtained by oxidation of Co(II) to Co(III) in the enzyme sorption complex. High stability was also found for the Fe(III)-containing preparations.

[Use of immobilization for investigation of glyceraldehyde 3-phosphate dehydrogenase. Immobilized tetramers].

Glyceraldehyde-3-phosphate dehydrogenase from yeast and rat skeletal muscle was covalently linked to CNBr-activated Sepharose 4B. When the activation was as high as 4-5 mg of CNBr per ml of Sepharose, the enzymes had their maximal activity and were linked to the carrier only by one of the four subunits. The specific activity of immobilized dehydrogenases makes up to 50-60% of that of soluble preparations, since the rate of the substrate diffusion into Sepharose granules is too low. The Km values for NAD and substrate and the pH dependence of the immobilized enzymes were determined. It was found that the enzymes used as adsorbents for isolation of specific antibodies reveal their maximal activity when CNBr concentration reaches 150-200 mg per ml of gel. No inhibition of activity of the immobilized dehydrogenase from yeast or stabilizing effect of antibodies on the enzyme structure were observed.

[Steroid transformation using immobilized microorganisms. II. Degradation of the sidechain of cholesterol by immobilized cells of Nocardia erythropolis].

The degradation of cholesterol side chain was studied by immobilized cells of Nocardia erythropolis in the presence of an inhibitor of the microbial steroid-skeleton-degradation. Different immobilized preparations such as DEAE-cellulose adsorbates, ionic cross-linked polymer gels, i.e., Al/Coalginates and polyacrylates, respectively, and polyethylene maleic acid anhydride supports have shown a high transformation activity yielding the C22- and C19-degradation products. No side chain degradation activity could be observed after immobilization of the cells in polyacrylamide gel under usual conditions caused by the toxic effect of the acrylamide monomer on the Nocardia cells.

Phospholipid chain immobilization and steroid rotational immobilization in acetylcholine receptor-rich membranes from Torpedo marmorata.

1. The ESR spectra of both phosphatidylcholine and phosphatidylethanolamine spin labels reveal an immobilized lipid component (tau R greater than or equal to 50 ns), in addition to a fluid component (tau R approximately 1 ns), in acetylcholine receptor-rich membranes prepared from Torpedo marmorata electroplax according to the method of Cohen et al. (Cohen, J.B., Weber, M., Huchet, M. and Changeux, J.-P. (1972) FEBS Lett. 26, 43--27). 2. The ESR spectra of the androstanol spin label display a component corresponding to molecules which are immobilized with respect to rotation about the long molecular axis (tau R greater than or equal to 50 ns), in addition to the fluid lipid bilayer component in which the molecules are rotating rapidly about their long axes (tau R approximately 1 ns). This immobilized component is observed throughout the temperature range 2--22 degrees C, at an approximately constant relative intensity of approx. 45% of the total, which is quantitatively the same as previously observed with fatty acid spin labels.

Improved immobilization yields by addition of protecting agents in glutaraldehyde-induced immobilization of protease.

The effects of peptides and polyethylene glycol (PEG) on the immobilization of protease using glutaraldehyde-activated Lewatit R258-K resin were investigated. When soy peptide was added at 0.1 g/l, the activity recovery and effectiveness factor increased by 20% and 38%, respectively. When PEG4000 (0.5 g/l) was used as a protecting agent, the activity recovery and effectiveness factor were improved by 35% and 60%, respectively, leading to an increased specific activity of the immobilized enzyme. The addition of PEG4000 is suggested to improve the cost-effectiveness for the production of immobilized enzymes.

In situ product removal of ketoses by immobilized 3-amino phenyl boronic acid: effect of immobilization method on pH profile.

The use of polymeric derivatives of phenylboronic acid (PBA) as an effective means for specific in situ product removal of ketoses from aldose-containing reaction mixtures, strongly depends on the retention of selective binding of ketoses exhibited by soluble PBA and 3-amino PBA, by their polymeric, water insoluble analogs. In this communication we demonstrate that immobilization chemistry has a strong effect on ketose preferred binding by polymeric PBA derivatives. Our results indicate that for the preparation of an effective and more specific adsorbent, 3-amino PBA should be coupled to the polymeric carrier via alkylamino chemistry and not via the commonly employed amido derivative. Immobilized alkylamino-PBA exhibited selective fructose and xylulose binding throughout glucose and xylose isomerization processes at the pH range of 7.0-8.0.

Immobilized enzyme electrodes for the potentiometric measurement of glucose concentration: immobilization techniques and materials.

Glucose oxidase, catalase, and bovine serum albumin were co-immobilized with glutaraldehyde around a platinum screen or around a single platinum-iridium wire. The potential difference between this dual enzyme electrode and a Ag/AgCL reference electrode was proportional to the logarithm of the glucose concentration over the range from 10 to about 150 mg glucose per 100 ml in buffered solution at pH 7.4 and 37 degrees C. The enzyme electrode responded in serum only if coated with a semipermeable film, such as cellulose acetate, to exclude serum macromolecules. The potentiometric results were similar to those obtained with the two enzymes co-immobilized in polyacrylamide gel around a platinum screen or with only one of the enzymes, glucose oxidase, covalently coupled to a platinum screen. The results so far suggest that glucose for development of a continuous in vivo glucose sensor.

Diffusional limitations in liquid-phase reactions catalyzed by enzymes immobilized on porous supports: sucrose inversion by beta-fructooxidase immobilized on IRA-93 resin.

An experimental procedure has been proposed to study liquid-phase reactions catalyzed by enzymes immobilized on porous supports. The kinetic behavior can be assessed by experimental runs which allow a separate evaluation of the internal diffusion, the surface kinetics, and the fluid-particle mass transfer. The method has been applied in the study of sucrose inversion by beta-fructooxidase immobilized on IRA-93 resin.

Bilirubin removal by the pseudoperoxidase activity of free and immobilized hemoglobin and hemoglobin co-immobilized with glucose oxidase.

We report a process for oxidizing bilirubin. Hemoglobin in the presence of an oxidizing agent can effectively oxidize bilirubin. Hemoglobin can be used either in free form or immobilized. Immobilisation includes microencapsulation, covalent linkage to carriers, intermolecular crosslinking into polyhemoglobin and others. The oxidizing agent can be added in the form of a peroxide. However, no external peroxide is required when hemoglobin is co-immobilized with glucose oxidase. In this case, the oxidation reaction takes place in the presence of glucose which is normally available in the blood stream and the presence of other oxidizing agents becomes unnecessary. In cases where bilirubin is conjugated to large proteins such as albumin, oxidation can be accomplished by using hemoglobin either in free form, adsorbed or bound on a suitable surface. Polyhemoglobin or cross-linked hemoglobins can also be used as oxidation catalysts in this case.

[Immobilization of Penicillium vitale glucose oxidase by aminoethyl cellulose and properties of immobilized enzyme].

Penicillium vitale glucose oxidase modified by oxidation of the carbohydrate component is in a covalent combination with aminoethyl cellulose (AE-cellulose). With the optimal enzyme: carrier ratio the manifested activity of glucose oxidase in the preparation is 4.3 +/- 0.8% of the attached enzyme activity. It is established that thermostability, stability to an inactivating effect of a labilizing fraction and pH-stability with pH alkaline values in the immobilized glucose oxidase are higher than in the soluble enzyme. It is shown that the process of thermal and alkaline inactivation occurs through two stages differing in the rate. The properties of P. vitale glucose oxidase immobilized by the same method by different carriers, aminosilochrome and AE-cellulose are compared.

Shoulder immobilizers. Shoulder immobilization devices.

Currently a myriad of devices are available for immobilization of the injured or postsurgical upper extremity. Some of these devices are straightforward and easily used, but some are more complicated and require more familiarity for their successful application. However, even simple devices have the potential for misapplication and thus prevent their benefit to the patient. This article is the second in a 3-part series. The goals of the series are (1) to present and review several devices on the market used by shoulder surgeons to immobilize the upper extremity, and (2) to discuss proper application and precautions of their use. It is intended that this series will benefit nurses, therapists, and trainers involved in the use of these devices.

Cell growth on immobilized cell growth factor. 6. Enhancement of fibroblast cell growth by immobilized insulin and/or fibronectin.

A cell growth factor protein (insulin) and/or a cell-adhesion protein (fibronectin) were immobilized on surface-hydrolyzed poly(methyl methacrylate) membranes. The growth of mouse fibroblast cells STO was accelerated by the immobilized insulin. This acceleration was enhanced by introduction of spacer arms. Coimmobilization of insulin and fibronectin was very effective for the acceleration of cell growth.

Fourier transform infrared assay of membrane lipids immobilized to silica: leaching and stability of immobilized artificial membrane-bonded phases.

A nondestructive, sensitive assay to monitor the hydrocarbon content of silica-based chromatography particles has been developed. The assay requires a microscope accessory interfaced with a Fourier transform infrared (FTIR) spectrometer. For determining hydrocarbon content, undiluted alkyl-silica-bonded phases were pressed into a thin wafer. Hydrocarbon content was quantitated using the integrated hydrocarbon band intensity between 2995 and 2825 cm-1 [i.e., band area C-H] and the integrated silica oxide band intensity between 1945 and 1780 cm-1 [i.e., band area Si-O]. Plotting the [band area C-H]/[band area Si-O] ratio vs the carbon content determined by elemental analysis gave a correlation coefficient of r = 0.997. The FTIR assay was validated on 5-, 7-, and 12-microns silica particles using three different immobilized artificial membrane (IAM) silica-bonded phases. The utility of the FTIR assay in determining hydrocarbon content was demonstrated by evaluating hydrocarbon leaching from IAM phases exposed to mobile-phase solvents. The ability of organic solvents to leach hydrocarbon from IAM phases containing phosphatidylcholine (PC) as the immobilized ligand was chloroform greater than ethanol approximately methanol greater than ethyl acetate greater than methylene chloride greater than acetonitrile greater than acetone. Acetone and acetonitrile cause very little hydrocarbon leaching from HPLC-IAM.PC columns. When challenged with different mobile phases, IAM.PC columns perfused with mobile phase are more stable than IAM.PC-bonded phases stirred in mobile phases. IAM.PC contains lecithin linked to silica by amide bonds.(ABSTRACT TRUNCATED AT 250 WORDS)
