In 23 hypothyroid children--15 girls and 8 boys--all of them receiving continuous substitution therapy, skeletal maturity was evaluated by use of a specific point system. The examination of 106 X-rays of the left hand revealed a clear-cut carpal retardation more than that of RUS (radius, ulna, short bones) in both sexes. It could be demonstrated a higher stage of development for RUS than carpal bone maturity in relation to height, independent of duration of therapy. A closer correspondence between RUS development and height may be assumed.

The evaluation of 2758 X-rays of the hands of 1047 patients with mongolism showed some morphological and metric aberrations: We discovered pseudoepiphyses in 82,4%, a brachymesophalangia in 67%, a dysmesophalangia in 4,6% and clinodactyly in 56,4%. The length of the hands of the patients with mongolism is in all ages shorter than in controls; there is also a distinct acromicria. These alterations are interpreted as a sign of the cerebral affection as well as a general aberration of the mesenchyma.
Since the measurement of intracortical resorptive spaces by histologic methods is difficult and very few data are available in normal humans, we have measured their lengths and widths and calculated the intracortical porosity in metacarpals and phalanges of 79 normal women and 69 normal men, using fine-detail radiographs of the hands and a computerized semi-automatic image analysis system (Zeiss MOP-3), this being the first study of this kind. Several methodological problems were solved satisfactorily, and the results of this study could serve as a data bank for further investigations concerned with intracortical resorption. Significant differences were found between age and sex versus several intracortical resorptive parameters; also significant correlations were found with age in some cases. Normal intracortical porosity was found to be about three times greater in the proximal phalanges than in the metacarpals. It is concluded that this methodology could be used for further studies of intracortical resorption in osteoporosis and other metabolic bone diseases.

BACKGROUND: Bone geometry (BG) and size (BS) are important factors in determining bone fragility. Previous studies have suggested that more than half of BG and BS variation is genetically determined. The possible chromosomal locations of genes involved in BS and BG determination have not been explored. We evaluated the extent and mode of inheritance of the radiographic hand BS index (BSI) and the metacarpal cortical index (MCI), and tested the hypothesis of linkage between these traits and the 11q 12-13 chromosomal region. MATERIAL/METHODS: Hand radiographs and blood samples were collected from 1190 individuals belonging to 349 Chuvasha nuclear families (Russian Federation). Segregation analysis was conducted on a total sample. Transmission disequilibrium testing (TDT) and model-based linkage analyses (MBLA) were performed on a sub-sample of 163 families. RESULTS: The hypothesis of a major gene effect was confirmed for both studied traits. The best-fitting models were Mendelian, with an additive type of inheritance. The inferred major gene explained 50% of the CI and 40% of the BSI variation. The TDT and MBLA results did not permit confirmation of hypotheses about linkage between hand BSI and the 11q 12-13 chromosomal region, but a possible linkage between CI and that region cannot be ruled out. CONCLUSIONS: We support the hypothesis of a major gene effect in the heritability of BSI and MCI. We provide suggestive evidence for possible linkage disequilibrium between MCI and the 11q12-13 chromosomal segment (marker D11S1983), but not for a linkage between BSI and this
