Neuropathologic verification of Creutzfeldt-Jakob disease in the exhumed American recipient of human pituitary growth hormone: epidemiologic and pathogenetic implications.

Beginning at age 12, a boy with idiopathic hypopituitarism was treated with cadaver pituitary-derived human growth hormone during the period from 1963 to 1969. Fifteen years after the last treatment, the then 32-year-old man developed Creutzfeldt-Jakob disease (CJD). The illness was atypical in showing predominantly cerebellar signs, little mental deterioration, and no abnormal movements or periodic EEG activity. Examination of the embalmed brain, 7 months after interment, revealed the characteristic changes of CJD mainly in the cerebellum and basal ganglia. This case establishes the contamination of at least two American lots of human growth hormone and, together with other cases of iatrogenic disease, suggests that virus enters the brain from the blood, rather than along neural pathways.

[Typing of STR locus in biological objects with pronounced degeneration: analysis of DNA in exhumed remains of victims of war actions].

The authors describe some specific features of enzymatic amplification typing of DNA preparations obtained from degraded tissues of remains of humans, which were brought from regions of war actions in the Chechen Republic. Special attention is paid to the specific artefact of polymerase chain reaction, for the first time detected by the authors in examinations of the above-mentioned objects. This so-called "ladder" effect manifesting by simultaneous nonspecific amplification of many variants of allele fragments of the STR locus on the individual DNA matrix, which can be erroneously interpreted as an evidence of mixed DNA preparation, is apparently characteristic of individual objects with pronounced degradation of biological material. Such phenomena were observed in typing of STR locuses in biological tissues subjected to biological, thermal, and physicohemical degradation. The phenomenon was studied in detail.

[Diagnosis of carbon monoxide poisoning following embalming and exhumation].

Clots of blood, which were found in the cardiac ventricles at an autopsy of an embalmed corpse, were to analyse for carbon monoxide. It was not possible to determine reliably the CO-Hb-content of the blood clots with the known methods. Therefore a new procedure was developed. Carbon monoxide was released by a solution of nitric acid and determined gas chromatographically by a headspace method. In the same vessel the iron-content of the sample was measured after decomposition with atomic absorption spectrometry. This procedure is especially suitable for samples like putrid or heat-clotted blood.
