Components of the ether-insoluble resin glycoside fraction from the seed of Cuscuta australis.

An ether-insoluble resin glycoside fraction was obtained from the seeds of Cuscuta australis. Identification and characterization of alkaline hydrolysis products by means of GC, FABMS, and 1H, 13C, and 2D NMR revealed the material to be composed of three new glycosidic acids, cuscutic acids A1-A3 (1-3); triglycosides with (11S)-jalapinolic or (11S)-convolvulinolic acid as the aglycon; and acetic, isobutyric, (2S)-2-methylbutyric, tiglic, and (2R,3R)-nilic acids. The resin glycoside is considered to be a complex mixture of glycosidic ester-type oligomers (up to heptamers) with a core consisting of a number of the above cuscutic acids each acylated with one or two carboxylic acid moieties.

Choline kinase in Cuscuta reflexa.

1. Choline kinase is a mitochondrial enzyme in Cuscuta reflexa. It can be solubilized from the particles by treatment with 350mm-sodium chloride, or by freezing and thawing. 2. Choline kinase of C. reflexa was purified by starting from the crude mitochondrial fraction. A 33-52% recovery of the enzyme, on the basis of the activity in the original homogenate, in 1200-2250-fold enrichment, was effected. 3. The purified preparation of choline kinase had a sigmoid saturation curve with respect to choline, with a Hill number of 2.3, and was inhibited by ADP (competitive in nature and allosteric in binding, with a Hill number of 2.7) and by phosphorylcholine (non-competitive and non-allosteric). The kinetic characteristics of the enzyme were consistent with the K type allosteric model of Monod et al. (1965). 4. The enzyme was desensitized, with respect to choline regulation, by prolonged storage in the cold, was activated significantly on warming before assay and was inactivated by high concentrations of sodium chloride. 5. The significance of allostery in choline kinase in relation to the intracellular regulation of phospholipid synthesis is discussed.

Structural features of an immunostimulating and antioxidant acidic polysaccharide from the seeds of Cuscuta chinensis.

Three crude polysaccharide fractions, named CS-A, CS-B and CS-C, were prepared from the seeds of Cuscuta chinensis by hot water extraction and diluted alkali extraction subsequently, then EtOH precipitation, and tested for lymphocyte proliferation activity. CS-A showed a stimulating effect on concanavalin A or lipopolysaccharide induced mitogenic activity of lymphocytes. An acidic polysaccharide (CS-A-3beta) was purified from CS-A by anion exchange and gel filtration chromatography. The polysaccharide showed potent stimulating effects on lymphocyte proliferation and antibody production, but did not significantly influence the serum IgG level. Its structural features were elucidated by methylation analysis, partial acid hydrolysis, 1D and 2D NMR and ESI-mass spectroscopy. The data obtained indicated that CS-A-3beta had a backbone consisting of alpha-D-1,4-linked GalpA residues and beta-L-1,2-linked Rhap residues with branches at C-4 of Rhap residues and C-3 of GalpA residues, composed of arabinogalactan and glucobiose. This structure is typical for a pectic polysaccharide of the rhamnogalacturonan type. In addition, the effect of CS-A, CS-B, CS-C and CS-A-3beta on hydrogen peroxide induced cell lesion in rat pheochromocytoma line PC 12 was investigated. The results indicated that, besides its immunostimulating activity, CS-A-3beta had a protective effect against free radical-induced cell toxicity.

Resource choice in Cuscuta europaea.

Individual stems with growing tips of dodder (Cuscuta europaea) were transplanted onto host plants (hawthorn) of varying nutritional status. The parasite was more likely to coil on ("accept") hosts of high nutritional status and grow away from ("reject") hosts of poor quality. Dodder exhibits this acceptance/rejection response before taking up any food from the host, so it is possible to dissociate active choice from the passive effects of growth and mortality. Probability of acceptance is gauged to expected reward.

Acquisition order and resource value in Cuscuta attenuata.

Field observations showed that in its native Texas grasslands, dodder (Cuscuta attenuata) grows more vigorously in patches of mixed host species than in monospecific host patches. Field experiments with naturally occurring host individuals demonstrated that the parasite achieved greater volume when simultaneously infesting two hosts of differing species rather than two hosts of the same species, and that this effect depends on the order in which the parasite encounters those hosts. Sodium, acting as an osmoticum, is implicated as the factor responsible for experimentally produced as well as observed field patterns.

Alpha-glucosidase inhibitory constituents from Cuscuta reflexa.

Two new compounds, 7'-(3',4'-dihydroxyphenyl)-N-[(4-methoxyphenyl)ethyl]propenamide (4), and 7'-(4'-hydroxy,3'-methoxyphenyl)-N-[(4-butylphenyl)ethyl]propenamide (5) have been isolated from Cuscuta reflexa along with five known compounds, 6,7-dimethoxy-2H-1-benzopyran-2-one (1), 3-(3,4-dihydroxyphenyl)-2-propen-1-ethanoate (2), 6,7,8-trimethoxy-2H-1-benzopyran-2-one (3), 3-(4-O-beta-D-glucopyranoside-3,5-dimethoxyphenyl)-2-propen-1-ol (6), 2-(3-hydroxy-4-methoxyphenyl)-3,5-dihydroxy-7-O-beta-D-glucopyranoside-4H-1-benzopyrane-4-one (7), reported for the first time from this species. Structures of these compounds were determined by spectral analysis. These compounds showed strong inhibitory activity against alpha-glucosidase.

[Flavonoids of Cuscuta australis R. Br].

Six flavonoids were isolated from the seed of Cuscuta australis and four of them were identified as kaempferol, quercetin, astragalin and hyperoside. Hyperoside was obtained from this plant for the first time. In comparison with the flavonoids in C. chinensis, it is found that quercetin and its glycoside are the main flavonoids in C. australis. This result suggests that the flavonoids can be used to distinguish these two medicinal materials.

[Identification of seeds of Cuscuta australis and C. chinensis by TLC and HPLC].

Identification of seeds of Cuscuta australis R. Br. and C. chinensis Lam. was carried out by TLC and HPLC. Polyamide membrane was used as stationary phase, MeOH-HOAc-H2O and CHCl3-MeOH-HOAc were used as mobile phase for TLC. For HPLC, Hypersil-ODS column was used; the mobile phase was MeOH-0.025 M H3PO4; the flow rate was 1.0 ml.min-1; detection wavelength was 360 nm; and column temperature was 40 degrees C. Both methods represented significant identification characteristics, and were simple, accurate and reproducible.

Chemical characterization and immunological activities of an acidic polysaccharide isolated from the seeds of Cuscuta chinensis Lam.

AIM: To study chemical characterization and immunological activities of an acidic polysaccharide, CHC-1, isolated and purified from the seeds of Cuscuta chinensis Lam. METHODS: Both chemical and spectral methods were used to investigate the chemical characterization of CHC-1. Effect of CHC-1 on the proliferation rates of T- and B-lymphocytes both in vivo and in vitro, and antibody production in vivo was measured at various concentrations of CHC-1. RESULTS: The molecular weight of CHC-1 was estimated to be more than 1.0 x 10(6). The analytical results of sugar components indicated that CHC-1 was composed of Rha, Ara, Gal, and GalA in a molar ratio of 0.8:1.0:1.5:0.3. Methylation analysis and 1H, 13C NMR further identified the linkages of the residues of CHC-1. CHC-1 0.1 g/L promoted remarkably the proliferation of T-cells and B-cells in vitro. CHC-1 25 mg/kg, 50 mg/kg caused an evident increase in spleen weight, lymphocyte proliferation, antibody production, etc. But its effect on IgG levels was not significant. CONCLUSION: CHC-1 is a highly branched heteropolysaccharide and possessed immune enhancement activities.

[Changes in DNA methylation in alfalfa plants infected with Cuscuta and tissue differences in DNA methylation of the parasite plants].

The tissue-specific differences in the 5-methylcytosine (m5C) content in total DNA of the parasite plant Cuscuta reflexa have been found: DNA from apical parts of the plant is less methylated (m5C = 4,2 mol %) as compared to the DNA from haustoria and posthaustorial regions (m5C = 5,4 mol %). The base compositions of total DNA preparations from C. reflexa grown on various hosts are similar. The m5C amount in stem DNA of the alfalfa plant infected with C. reflexa is by approximately 25% higher than that in the non-infected plant DNA. The GC content in alfalfa DNA does not change as a result of infection. Thus, the parasite induces the hypermethylation of DNA in the host plant. It is assumed that the changes in DNA methylation induced by the parasite plant may play a regulatory role and may cause changes in transcription and replication of host DNA.

[Effects of semen Cuscutae, rhizoma Curculiginis, radix Morindae officinalis on human spermatozoan's motility and membrane function in vitro].

OBJECTIVE: To observe the effects of Semen Cuscutae (SC), Rhizoma Curculiginis (RC) and Radix Morindae Officinalis (RMO) on human sperm motility in vitro and cytomembrane function. METHODS: Compare the sperm motility of the above-mentioned medicinal plants with modified Tyrode's solution (MTS) and MTS added caffeine in vitro. Human spermatozoa were incubated with decoctions of medicinal plant at various concentrations in 37 degrees C for 30 minutes. The motility of sperm was evaluated by the sperm capillary penetrating test, sperm speed test and sperm activity index calculations. The spermatozoa membrane function was evaluated by spermatozoal hypo-osmotic swelling test together with eosin staining methods. RESULTS: The sperm motility improved markedly and sperm membrane function became more stabilized after incubation, the SC had best effect, RC took second place, RMO was relatively poor. CONCLUSIONS: SC decoction is an effective promoting preparation for sperm motility and the membrane function stabilized. Therefore, SC may be beneficial in treating male sterility and may raise success rate in artificial insemination.

New ELISA kits using C3 binding glycoprotein from Cuscuta europea detect mainly IgM CIC in rheumatoid arthritis and progressive systemic sclerosis, but not in systemic lupus erythematosus.

Elevated levels of circulating immune complexes (CIC), containing IgG, IgM or IgA antibodies were detected in the sera of patients with autoimmune diseases. This might indicate a different biological meaning of the three isotypes of immunoglobulin (Ig) in the CIC. Each CIC assay detected only certain classes and subclasses of Ig in CIC material or fixed complement protein. In this study, a new method based on C3binding glycoprotein named CIF-ELISA and a well-known method ANTI-C3 ELISA, were used for quantitative assessment of IgM-CIC, IgG-CIC and IgA-CIC levels in human sera. A modified CIF-ELISA and ANTI-C3 ELISA for simultaneous detection of CIC, containing IgG, IgM and IgA, (stCIC), were also performed. The assays were evaluated on the same specially prepared samples: 55 normal sera, 99 sera from rheumatoid arthritis (RA), 88 sera from systemic lupus erythematosus (SLE), and 27 sera from progressive systemic sclerosis (PSS). We found that the sensitivity of the tests used varied depending on the diseases studied. CIF-ELISA displayed higher sensitivity of IgM-CIC when compared to ANTI-C3 ELISA in RA patients (40.0 and 20.95%, respectively) and PSS (44.43 and 37.04%, respectively). Results for the sensitivity of IgA-CIC were in adverse direction in the RA group (14.28 and 19.05%) and PSS (14.81 and 25.93%) by both methods. It was also established that the concordance of IgM-CIC positives by both methods was 48.84% in RA and 46.67% in PSS, while in SLE it was 18.78%. These results are most probably due to the different assay abilities to detect antibody isotype of the CIC material and help to explain what specific role each Ig isotype in CIC has in the course of the disease.

Anti-steroidogenic activity of methanolic extract of Cuscuta reflexa roxb. stem and Corchorus olitorius Linn. seed in mouse ovary.

Methanolic extract (ME) of both C. reflexa stem and C. olitorius seed arrested the normal oestrus cycle of adult female mouse and significantly decreased the weight of ovaries and uterus. The cholesterol and ascorbic acid contents in ovaries were significantly increased in the treated mice. Two key enzymes, delta5-3beta-hydroxysteroid dehydrogenase and glucose-6-phosphate dehydrogenase, were decreased significantly in ME of both C. reflexa stem and C. olitorius seed after 17 days of treatment. High level of substrates and low level of enzymes indicate the inhibition of steroidogenesis in treated mice and may be due to the presence of flavonoids.
