[Clinical study of butane gas abuse: in comparison with toluene-based solvent and marihuana].

We reported 2 cases of patients who abused butane gas, toluene-based solvent and marihuana. They showed different signs in the each substance, respectively. Butane gas was easier to make visual hallucinations and distorted perception of body form, and was less potent and addictive than toluene-based solvent. Spontaneous laughter and the most amotivational state were characterized by marihuana intoxication. Alteration of auditory perception that simple music sounded wonderful was also experienced. Furthermore, the above symptoms were thought to change by the order of taking the substance. Therefore, it is needed to examine the order of the use of drugs and clarify differences of symptoms in abuse among drugs, respectively.

[Experimental poisoning with gaseous hydrocarbons: changes in concentrations of propane and butane in the lung and adipose tissue in relation to the time of death].

Postmortal changes of propane, butane and iso-butane concentration in the lung and fat of mice exposed to lethal intoxication with commercial liquid gas are studied. The hydrocarbon tissue concentrations after exposure were determined by gas liquid chromatography. The hydrocarbons progressively decreased in the lung and disappeared, depending on the degree of putrefaction; even in case of remarkable putrefaction, it was still possible to detect them in the fat tissue.

Pulmonary infiltrates following butane 'fire-breathing'.

Rapidly progressive bilateral pulmonary infiltrates occurred in a 19-year-old man following an unusual hydrocarbon abuse. The acute illness was the result of a "trick" known as "fire-breathing." Fire-breathing involves filling the oral cavity with butane gas, from an ordinary butane cigarette/cigar lighter, and exhalation of the volatile vapors over an open flame producing a flame-throwing effect. Because of the pulmonary toxic reaction, this activity could have a serious or even fatal outcome.

Perfluoro-n-butane. A gas for a maximum duration retinal tamponade.

The behavior of the gas perfluoro-n-butane (C4F10) in the rabbit vitreous suggests that the gas might be useful as a long-term intravitreal tamponade in retinal detachment surgery. Perfluoro-n-butane was found to expand more and remain in the eye longer than any gas previously reported. A 0.3-mL bubble expanded five times and displaced the entire rabbit vitreous in three days. The volume of the bubble began to shrink after one week, but still filled more than half the eye at three weeks. The time for total disappearance was more than three months.

Analysis of Silica Surface Heterogeneity Using Butane and Butene Adsorption Data

Adsorption isotherms for butane and butene measured on silica samples at different temperatures are used as a data base for describing the chemical and physical changes in the systems. The stable numerical method for solving the adsorption integral equation (SAIEUS) is used to deconvolute adsorption data. The effect of lateral interactions between adsorbed molecules on the results obtained is underlined. The adsorption properties of the initial and heat-treated silica samples are discussed in terms of their butane and butene adsorption energy distributions. The analysis of the results obtained reveals the complex nature of the silica surface due to its microporosity and the presence of surface hydroxyl groups. Copyright 1997Academic Press

A High-Resolution Microwave Study of the Conformations of Butan-2-ol in a Supersonic Expansion.

The microwave Fourier transform spectrum of the chiral, secondary alcohol butan-2-ol has been recorded in the range 6-18 GHz. The spectrum is relatively dense, due to the large number of species formed in the supersonic expansion used. Three conformational isomers of the butan-2-ol molecule have been identified and their spectra assigned using a semirigid, asymmetric-rotor model. They correspond to the three configurations that the central C-C bond can adopt. The basis of our assignment is a series of ab initio calculations, which have been performed using the GAUSSIAN 94 package, good agreement being observed between theoretical and experimental values of the rotational constants. First-order centrifugal distortion coefficients have also been extracted. It has proven possible, by examination of the relative strengths of a-, b-, and c-type transitions, to infer some further information about the position of the hydroxyl hydrogen atom within each conformer. Valence isomerization of 2-phospha-4-silabicyclo[1.1.0]butane: a high-level ab initio study.

The rearrangements for 2-phospha-4-silabicyclo[1.1.0]butane, analogous to the valence isomerization of the hydrocarbons bicyclobutane, 1,3-butadiene, and cyclobutene, were studied at the (U)QCISD(T)/6-311+G**//(U)QCISD/6-31G* level of theory. The monocyclic 1,2-dihydro-1,2-phosphasiletes are shown to be the thermodynamically preferred product, in contrast to the isomerization of the hydrocarbons, which favors the 1,3-butadiene structure. Furthermore, an unprecedented direct isomerization pathway to the 1,2-dihydro-1,2-phosphasiletes was identified. This pathway is competitive with the isomerization via the open-chain butadienes and becomes favorable when electron-donating substituents are present on silicon. Figure 2-Phospha-4-silabicyclo[1.1.0]butane can isomerize directly into the more stable P,Si-cyclobutene via an unprecedented [sigma2s+sigma2a] process, which becomes favorable over the isomerization via the P,Si-butadiene when electron-donating substituents are present on silicon.

A series of (2R,3S)-2-(2,4-difluorophenyl)-3-(5-[2-[4-aryl-piperazin-1-yl]-ethyl]-tetrazol-2-yl)-1-[1,2,4]-triazol-1-yl-butan-2-ol (11a-n) and (2R,3S)-2-(2,4-difluorophenyl)-3-(5-[2-[4-aryl-piperazin-1-yl]-ethyl]-tetrazole-1-yl)-1-[1,2,4]-triazol-1-yl-butan-2-ol (12a-n) has been synthesized. The antifungal activity of compounds was evaluated by in vitro agar diffusion and broth dilution assay. Compounds 11d and its positional isomer 12d having 3-trifluoromethyl substitution on the phenyl ring of piperazine demonstrated significant antifungal activity against variety of fungal cultures (Candida spp. C. neoformans and Aspergillus spp.). The compound 12d showed MIC value of 0.12 microg/mL for C. albicans, C. albicans V-01-191A-261 (resistant strain); 0.25 microg/mL for C. tropicalis, C. parapsilosis ATCC 22019 and C. krusei and MIC value of 0.5 microg/mL for C. glabrata, C. krusei ATCC 6258, which is comparable to itraconazole and better than fluconazole. Further, compound 11d showed significant activity (MIC; 0.25-0.5 microg/mL) against Candida spp. and strong anticryptococcal activity (MIC; 0.25 microg/mL) against C. neoformans.

N-Acridin-9-yl-butane-1,4-diamine derivatives: high-affinity ligands of the alpha2delta subunit of voltage gated calcium channels.

A series of N-acridin-9-yl-butane-1,4-diamines were found to be high-affinity ligands of the alpha(2)delta subunit of voltage gated calcium channels. The SAR studies of butane-1,4-diamine side chain resulted in the identification of compound 10 (IC(50)=9 nM), which is more potent than gabapentin (IC(50)=27 nM). Partial saturation of the acridine ring was also pursued and provided a compound with higher binding affinity than 1.

Probing molecular conformations with electron momentum spectroscopy: the case of n-butane.

High-resolution (e,2e) measurements of the valence electronic structure and momentum-space electron density distributions of n-butane have been exhaustively reanalyzed in order to cope with the presence of two stable structures in the gas phase, namely the all-staggered and gauche conformers. The measurements are compared to a series of Boltzmann-weighted simulations based on the momentum-space form of Kohn-Sham (B3LYP) orbital densities, and to ionization spectra obtained from high-level [ADC(3)] one-particle Green's Function calculations. Indubitable improvements in the quality of the simulated (e,2e) ionization spectra and electron momentum profiles are seen when the contributions of the gauche form of n-butane are included. Both the one-electron binding energies and momentum distributions consistently image the distortions and topological changes that molecular orbitals undergo due to torsion of the carbon backbone, and thereby exhibit variations which can be traced experimentally. With regard to the intimate relation of (e,2e) cross sections with orbital densities, electron momentum spectroscopy can therefore be viewed as a very powerful, but up to now largely unexploited, conformational probe. The study also emphasizes the influence of thermal agitation in photoionization experiments of all kind.

Oxidative activation of n-butane on sulfated zirconia.

Catalytic activation and conversion of light alkanes by sulfated zirconia is unequivocally shown to be initiated by producing small concentrations of olefins. This occurs via stoichiometric oxidative dehydrogenation of butane by SO3 or pyrosulfate groups to butene (present mostly as alkoxy groups), water, and SO2. Thermal desorption and in situ IR spectroscopy have been used to determine all three reaction products. The concentration of butene formed determines both the catalytic activity of sulfated zirconia as well as the deactivation via formation of oligomers. The thermodynamics of the oxidative dehydrogenation of n-butane by different SZ surface structures has been examined by density functional (DFT) calculations. The calculations show that pyrosulfate or re-adsorbed SO3 species have the highest oxidizing ability.

Potential bis-alkylating agents for cancer chemotherapy. Approaches to the synthesis of 2-sulfonyl-1,4-bis(methanesulfonoxy)butanes.

Three general methods for the synthesis of derivatives of the bis-alkylating agent, myleran [1,4-bis(methane-sulfonoxy)butane (1)], are presented. These derivatives contain a lipophilic group attached to the 2 position of 1,4-bis(methanesulfonoxy)butane by means of a sulfonyl function. As examples of the scope of the methods, the synthesis of seven such derivatives (12a-g) is presented. All seven were inactive against L1210 and P388 leukemia in the mouse.

Pt3Ru6 clusters supported on gamma-Al2O3: synthesis from Pt3Ru6(CO)21(mu3-H)(mu-H)3, structural characterization, and catalysis of ethylene hydrogenation and n-butane hydrogenolysis.

The supported clusters Pt-Ru/gamma-Al2O3 were prepared by adsorption of the bimetallic precursor Pt3Ru6(CO)21(mu3-H)(mu-H)3 from CH2Cl2 solution onto gamma-Al2O3 followed by decarbonylation in He at 300 degrees C. The resultant supported clusters were characterized by infrared (IR) and extended X-ray absorption fine structure (EXAFS) spectroscopies and as catalysts for ethylene hydrogenation and n-butane hydrogenolysis. After adsorption, the nu(CO) peaks characterizing the precursor shifted to lower wavenumbers, and some of the hydroxyl bands of the support disappeared or changed, indicating that the CO ligands of the precursor interacted with support hydroxyl groups. The EXAFS results show that the metal core of the precursor remained essentially unchanged upon adsorption, but there were distortions of the metal core indicated by changes in the metal-metal distances. After decarbonylation of the supported clusters, the EXAFS data indicated that Pt and Ru atoms interacted with support oxygen atoms and that about half of the Pt-Ru bonds were maintained, with the composition of the metal frame remaining almost unchanged. The decarbonylated supported bimetallic clusters reported here are the first having essentially the same metal core composition as that of a precursor metal carbonyl, and they appear to be the best-defined supported bimetallic clusters. The material was found to be an active catalyst for ethylene hydrogenation and n-butane hydrogenolysis under conditions mild enough to prevent substantial cluster disruption.

Studies on the generation of enolate anions from butane-2,3-diacetal protected glycolic acid derivatives and subsequent highly diastereoselective coupling reactions with aldehydes and acid chlorides.

Highly diastereoselective coupling reactions of enolates derived from butane-2,3-diacetal protected glycolic acids 1 and 2 and their alkylated derivatives with aldehydes are reported together with their efficient acid-catalysed deprotection to yield enantiopure anti-2,3-dihydroxyesters. A procedure to provide the corresponding syn-2,3-dihydroxyesters is also described in two cases, proceeding via an acylation-reduction sequence. An usual double addition reaction of butane-2,3-diacetal protected glycolic acid to small aliphatic acid chlorides provides a synthetically useful, densely-functionalised lactone after acidic deprotection.

Inactivation of glutamate dehydrogenase and glutamate synthase from Bacillus megaterium by phenylglyoxal, butane-2,3-dione and pyridoxal 5'-phosphate.

Reaction of phenylglyoxal with glutamate dehydrogenase (EC 1.4.1.4), but not with glutamate synthase (EC 2.6.1.53), from Bacillus megaterium resulted in complete loss of enzyme activity. NADPH alone or together with 2-oxoglutarate provided substantial protection from inactivation by phenylglyoxal. Some 2mol of [14C]Phenylglyoxal was incorporated/mol of subunit of glutamate dehydrogenase. Addition of 1mM-NADPH decreased incorporation by 0.7mol. The Ki for phenylglyoxal was 6.7mM and Ks for competition with NADPH was 0.5mM. Complete inactivation of glutamate dehydrogenase by butane-2,3-dione was estimated by extrapolation to result from the loss of 3 of the 19 arginine residues/subunit. NADPH, but not NADH, provided almost complete protection against inactivation. Butane-2,3-dione had only a slight inactivating effect on glutamate synthase. The data suggest that an essential arginine residue may be involved in the binding of NADPH to glutamate dehydrogenase. The enzymes were inactivated by pyridoxal 5'-phosphate and this inactivation increased 3--4-fold in the borate buffer. NADPH completely prevented inactivation by pyridoxal 5'-phosphate.

Effect of solvation and confinement on the trans-gauche isomerization reaction in n-butane.

The effect of solvation and confinement on the conformational equilibria and kinetics of n-butane is examined using molecular dynamics simulations of the bulk and confined fluids and compared to appropriately chosen reference states. Clear evidence for a solvent shift of the preferred conformation in bulk n-butane is found. At a temperature of 292 K and a density of 6.05 nm-3 a small solvent shift in favor of gauche is observed (similar to previously reported values), and the shift increases substantially with an increase in density to 8.28 nm-3. The rate of torsional interconversion from the trans to the gauche state, calculated using the relaxation function method, was found to increase with increasing temperature and density. The rate constants kTG and kGT have an Arrhenius temperature dependence yielding activation energies significantly lower than the trans-gauche and gauche-trans barrier heights in the torsional potential for a free molecule, depending on the density. In the confined phase, we considered the same densities as simulated in the bulk phase, and for four different values of the physical pore width (approximately 1.5-4.0 nm). At the high density, we find that the position of the trans-gauche equilibrium is displaced towards excess trans compared with the bulk phase, reflecting the confinement and interactions of the molecules with the pore wall. The isomerization rate is found to decrease with decreasing pore width. Again, we find that the kinetics obeys an Arrhenius rate law and the activation energy for the trans-gauche and gauche-trans interconversions is slightly smaller than that of the bulk fluid at the same density.

Comparative mutagenicity of 4-(carbethoxynitrosamino)-4-(3-pyridyl)butanal and 4-(carbethoxynitrosamino)-1-(3-pyridyl)-1-butanone, model compounds for alpha-hydroxylation of N'-nitrosonornicotine.

4-(Carbethoxynitrosamino)-1-(3-pyridyl)butanal, a stable precursor to the putative diazohydroxide formed by 5'-hydroxylation of the tobacco-specific nitrosamine, N'-nitrosonornicotine, was synthesized in six steps from nicotinaldehyde. Its mutagenicity toward S. typhimurium was compared to that of 4-(carbethoxynitrosamino)-1-(3-pyridyl)-1-butanone, a precursor to the diazohydroxide formed by 2'-hydroxylation of N'-nitrosonornicotine. At equimolar doses, 4-(carbethoxynitrosamino)-1-(3-pyridyl)-1-butanone was a potent mutagen, but 4-(carbethoxynitrosamino)-1-(3-pyridyl)butanal was inactive toward strains TA 100 and TA 1535. The results of this study indicate that the putative diazohydroxide formed by 2'-hydroxylation of N'-nitrosonornicotine has higher inherent mutagenicity toward S. typhimurium than does the corresponding diazohydroxide formed by 5'-hydroxylation.
