Nondisjunction rates of mouse specific chromosomes involved in heterozygous Rb rearrangements measured by chromosome painting of spermatocytes II. I. The effects of the number of trivalents.

Dual-colour FISH painting with alternative fluorescent chromosome-specific probes allowed us to distinguish chromosomes 1, 4, 6 and 14. The purpose was to check whether nondisjunction rates of specific chromosomes involved in heterozygous Robertsonian fusions are independent of the number of trivalents, or an epistatic effect among Rb chromosomes takes place affecting nondisjunction rates. Probes were used on DAPI-stained metaphases of spermatocytes II of laboratory strains of mice with reconstructed karyotypes heterozygous for one, two, three or four Robertsonian metacentrics in an all-acrocentric background. The existence of such epistatic interactions was not verified.

The impact of chromosome sorting and painting on the comparative analysis of primate genomes.

Chromosome sorting by flow cytometry is the main source of chromosome-specific DNA for the production of painting probes. These probes have been used for cross-species in situ hybridization in the construction of comparative maps, in the study of karyotype evolution and phylogenetics, in delineating territories in interphase nuclei, and in the analysis of chromosome breakpoints. We review here the contributions that this technology has made to the analysis of primate genomes.

Phylogenetic inferences of Atelinae (Platyrrhini) based on multi-directional chromosome painting in Brachyteles arachnoides, Ateles paniscus paniscus and Ateles b. marginatus.

We performed multi-directional chromosome painting in a comparative cytogenetic study of the three Atelinae species Brachyteles arachnoides, Ateles paniscus paniscus and Ateles belzebuth marginatus, in order to reconstruct phylogenetic relationships within this Platyrrhini subfamily. Comparative chromosome maps between these species were established by multi-color fluorescence in situ hybridization (FISH) employing human, Saguinus oedipus and Lagothrix lagothricha chromosome-specific probes. The three species included in this study and four previously analyzed species from all four Atelinae genera were subjected to a phylogenetic analysis on the basis of a data matrix comprised of 82 discrete chromosome characters. The results confirmed that Atelinae represent a monophyletic clade with a putative ancestral karyotype of 2n = 62 chromosomes. Phylogenetic analysis revealed an evolutionary branching sequence [Alouatta [Brachyteles [Lagothrix and Ateles]]] in Atelinae and [Ateles belzebuth marginatus [Ateles paniscus paniscus [Ateles belzebuth hybridus and Ateles geoffroyi]]] in genus Ateles. The chromosomal data support a re-evaluation of the taxonomic status of Ateles b. hybridus.

Investigation of marmoset hybrids (Cebuella pygmaea x Callithrix jacchus) and related Callitrichinae (Platyrrhini) by cross-species chromosome painting and comparative genomic hybridization.

We report on the cytogenetics of twin offspring from an interspecies cross in marmosets (Callitrichinae, Platyrrhini), resulting from a pairing between a female Common marmoset (Callithrix jacchus, 2n = 46) and a male Pygmy marmoset (Cebuella pygmaea, 2n = 44). We analyzed their karyotypes by multi-directional chromosome painting employing human, Saguinus oedipus and Lagothrix lagothricha chromosome-specific probes. Both hybrid individuals had a karyotype with a diploid chromosome number of 2n = 45. As a complementary tool, interspecies comparative genomic hybridization (iCGH) was performed in order to screen for genomic imbalances between the hybrids and their parental species, and between Callithrix argentata and S. oedipus, respectively. These genomic imbalances were confined to centromeric and telomeric heterochromatin, while euchromatic chromosome regions appeared balanced in all species investigated. When comparing marmosets and tamarins, sequence divergence of centromeric heterochromatin was already clearly noticeable. In the C. argentata and C. pygmaea genomes numerous subtelomeric regions were affected by amplification of different repetitive sequences. Cross-species FISH with a microdissection-derived C. pygmaea repetitive probe revealed species specificity of this repetitive sequence at the molecular cytogenetic level of resolution.

Further insights into the ancestral murine karyotype: the contribution of the Otomys-Mus comparison using chromosome painting.

The African vlei rat, Otomys irroratus, comprises several distinct chromosomal races that may be grouped into two major cytogenetic clades. Recognition of these clades is underpinned by a complex chromosomal rearrangement involving three different autosomes in the unfused state. We have used unidirectional fluorescence in situ hybridization (FISH) of mouse chromosome-specific painting probes to molecularly define the components of this rearrangement as well as to establish the chromosomal homologies between the mouse and the vlei rat genomes. This has allowed for the detection of 41 autosomal segments of conserved synteny. Nine mouse chromosomes were conserved in toto (MMU3, 4, 6, 7, 11, 12, 14, 18, 19) with a further seven (MMU2, 5, 8, 9, 10, 13, 16) showing homology to two discrete regions in the vlei rat genome. Two mouse autosomes (MMU15, 17) correspond to three regions in O. irroratus with MMU1 being the most fragmented showing five sites of hybridization in this species. By mapping these data to published sequence-based phylogenies we are able to confirm most of the published putative ancestral murine chromosomal states. Our data further indicate that MMU15a+ MMU13b+MMU10b+MMU17b was present in the murine ancestral karyotype suggesting an ancestral 2n = 52 rather than the 2n = 54 previously postulated.

Experimental lead paint poisoning in nonhuman primates. III. Pathologic findings.

Necropsies were performed on 25 rhesus monkeys, three cebus monkeys and three baboons which had been fed leaded paint or lead acetate at various doses up to 666 days. The 31 test primates and six controls ranged in age from five days to about eight years. In addition, the brains of 13 subadult squirrel monkeys fed lead oxide and two controls were studied grossly and microscopically. Lead content of liver, kidney and brain correlated with clinical outcome and typical histologic changes. Neuropathologic lesions, most severe in the young, occurred in 28 of 43 test primates despite a paucity of neurological signs. Brain lesions were similar to those occurring in human lead encephalopathy and included degenerative and proliferative changes of small vessels, ring hemorrhages, edema, perivascular hyalin droplets, rosette-like deposits of proteinaceous exudates, focal loss of myelin, astrogliosis and necrosis of hippocampal neurons.

Dual Y-chromosome painting and immunofluorescence staining of archival human liver transplant biopsies.

Combining fluorescence in situ hybridization (FISH) and indirect immunofluorescence staining of protein markers provides a highly specific method for identifying chromosomes in phenotypically defined cells and tissues. We developed a technique enabling dual chromosome painting and immunofluorescence staining of archival formalin-fixed, paraffin-embedded material, and used this to phenotype chimeric cells in female-to-male human liver transplants.

Impact of a lead-safe training program on workers conducting renovation, painting, and maintenance activities.

OBJECTIVE: An important source of lead exposure is lead-based paint that is disturbed when unsafe work practices are used during renovation, remodeling, and maintenance activities. This study explores the success of a pilot lead-safe skills training program for home improvement contractors and their employees (including renovators, remodelers, and painters) and small property owners. METHODS: The study evaluates whether attendees at eight-hour lead-safe work practices training courses learned and retained information about lead exposure; developed and retained positive attitudes toward lead-safe work practices; and developed lasting, positive behavioral intentions to use lead-safe work practice skills and techniques. A questionnaire was administered immediately before, immediately following, and several months following the training program. Coded data from the questionnaires were analyzed using SPSS software. RESULTS: Respondents showed statistically significant changes from before to after the training program, and the changes were maintained over time. Knowledge improved, and attitudes and behavioral intentions changed in a favorable direction. CONCLUSION: These results suggest that lead-safe training can be successful and can create lasting changes in lead-safe knowledge, attitudes, and behaviors.

Respiratory and systemic reaction following exposure to heated electrostatic polyester paint.

A 39 year old nonatopic man developed episodes of cough, dyspnoea, sweating and shivers within 2-3 weeks of starting a new job in a factory where metallic boards were treated with an electrostatic powder paint, made of an epoxy resin and a carboxylated polyester containing polyethylene terephthalate and polybutylene terephthalate. The subject sprayed the metallic boards which were then heated in 200 degrees C ovens. The subject was first seen in an emergency room after being at work for 4 h. The physical examination revealed bilateral wheezing with fever (39 degrees C), hypoxaemia (arterial oxygen tension (PaO2) 58 torr (7.7 kPa), leucocytosis (white blood count cells.mm-3 17,000 (17 x 10(9) cells.l-1) and severe airway obstruction (forced expiratory volume in one second (FEV1)/forced vital capacity, (FVC) 1.3/2.4 l, improving to 2.2/3.8 l after bronchodilator; predicted values = 3.4/4.1 l). The subjects condition improved after being treated with oral steroids. His spirometry was normal two weeks later, although he showed mild bronchial hyperresponsiveness to methacholine with the (provocative concentration producing a 20% fall in FEV1 (PC20) being 1.7 mg.ml-1). The subject underwent specific inhalation challenges at the workplace 4 months later. After being exposed at work for 4 h, he developed a significant fall in FEV1 (-40%), fever, leucocytosis, and a fall in diffusing capacity. Lung function tests were back to normal two weeks later. Exposing the subject to heated granulated polyester for one hour in a hospital laboratory produced a fall in FEV1 of 41%, fever, leucocytosis and a fall in diffusing capacity.(ABSTRACT TRUNCATED AT 250 WORDS)

On painting and philosophy: Sir Henry Raeburn.

This article considers the 18 century European Enlightenment as an example of the cross-fertilization between philosophy propounding rationalist-empirical approach, and arts. The example of Sir Henry Raeburn, a Scottish portraitist, is given to illustrate the influence of the concepts of perception developed by Thomas Reid, the founder of the Scottish school of common sense philosophy, on the style of painting.

Science, alchemy and light: paintings by Joseph Wright of Derby.

This article considers two paintings by the English painter, Joseph Wright of Derby (1734-97), 'An Experiment on a Bird in the Air Pump' and 'The Alchemist in Search of the Philosopher's Stone' as painterly reflections on the early industrial culture. Particularly interesting in the "Experiment on a Bird" is the broad spectrum of spectator's reactions to the experiment; this can be related to the contemporary debate on the contextualization of science.

Solvent constituents in paint, glue and thinner for plastic miniature hobby.

Solvent-containing products (8 paint, 29 glue and 4 thinner preparations) for plastic miniature hobby were analyzed for solvent constituents by capillary gas-chromatography. Acetone and methyl isobutyl ketone were two most extensively used solvents both in terms of frequency and concentration. Other popular solvents included ethyl alcohol and two esters of ethyl acetate and butyl acetate. In contrast, aromatics, chlorinated hydrocarbons and ethylene glycol derivatives were seldomly detected. The present findings were discussed in comparison with the observation on the materials abused for sniffing in the past, as well as constituents in the products for industrial application.

Analysis of FISH-painted chromosomes in individuals living near the Semipalatinsk nuclear test site.

The Semipalatinsk nuclear test site (STS) is located in the Republic of Kazakhstan. A total of 498 nuclear weapons tests were conducted in this area between 1949 and 1989. The radiation exposure to people who lived close to the STS resulted mostly from the above-ground explosions. Blood samples for chromosome analysis were obtained from 10 subjects who were born before the first explosion in August 1949 and lived continuously in the village of Dolon. The individual calculated effective doses were about 3 Sv. Chromosomes 2, 4 and 8 were painted by means of the FISH technique. In total, 22,240 cells were analyzed. The mean frequency of translocations in the subjects who were irradiated during childhood (2.4/1000 cells) did not differ from the control value (3.1 translocations/1000 cells). It is assumed, therefore, that the calculated physical dose is too high. A significantly increased level of complex cells was determined, however, and this was assumed to have been induced in circulating lymphocytes. The reason for this may be the incorporation of radionuclides from fallout which were not distributed homogeneously within the body, but accumulated instead in tissues that were well supplied with peripheral blood.

Stability of chromosome aberrations in the blood lymphocytes of astronauts measured after space flight by FISH chromosome painting.

Follow-up measurements of chromosome aberrations in the blood lymphocytes of astronauts were performed by FISH chromosome painting at various intervals from 5 months to more than 5 years after space flight and compared to preflight baseline measurements. For five of the six astronauts studied, the analysis of individual time courses for translocations revealed a temporal decline of yields with half-lives ranging from 10 to 58 months. The yield of exchanges remained unchanged for the sixth astronaut during an observation period of 5 months after flight. These results may indicate complications with the use of stable aberrations for retrospective dose reconstruction, and the differences in the decay time may reflect individual variability in risk from space radiation exposure.

Justice and fairness in the Kennedy Krieger Institute lead paint study: the ethics of public health research on less expensive, less effective interventions.

The Kennedy Krieger lead paint study stirred controversial questions about whether research designed to develop less expensive interventions that are not as effective as existing treatments can be ethically warranted. Critics questioned the social value of such research and alleged that it sanctions a double standard, exploits participants, and is complicit in perpetuating the social injustice. In response, we demonstrate the propriety of conducting research on interventions that can be extended to the population in need by stipulating the limited conditions in which it is ethically warranted and providing fair terms of participation. We contend that the failure to conduct such research causes greater harm, because it deprives disadvantaged populations of the benefits of imminent incremental improvements in their health conditions.

Longitudinal changes in blood lead level in children and their relationship to season, age, and exposure to paint or plaster.

Children screened for lead poisoning in the Brownsville district of New York City in either summer or winter were followed with blood lead tests for approximately six months to one year from screening to measure longitudinal changes in blood lead level and to identify some determinants of the changes. Only minimal evidence was found of the hypothesized summer rise in blood lead level, while the predominant trend seemed to be for blood lead levels to display statistical regression to the mean. In children found to have low to intermediate blood lead levels (less than 55 microgram/100ml) at screening, variables which were found to predict a rise in blood lead level of 10 microgram/100ml or greater from winter to summer were under age three and/or exposure to paint or plaster.

Personality and preference for painting style.

40 university students were asked to choose between the cubist or surrealist from among 15 pairs of slides of paintings according to their aesthetic preference. Their choices were correlated with scores on Rotter's internal vs external locus of control scale. The data support the hypothesis that there is a positive relationship between preference for cubism over surrealism and internal locus control and vice versa.
