Routine identification and determination of 11 barbiturates in biological samples.

A very easy, reliable and specific gas-chromatographic method for identification and dosage of 11 barbiturates in plasma is presented. Methylene unit values determined on two types of column (one polar, one non-polar) allows very accurate identification with a minimum risk of error due to fluctuations in operating conditions. Sensitivity of the method is 0.5 microgram/ml plasma with a flame ionization detector. Derivatization procedure is complete, without any degradation phenomenon as tested by mass spectrometry. Such a method can be easily used as routine procedure for toxicological or pharmacological applications.

Determination of phospholipids in biological samples by an improved densitometric method on thin-layer chromatograms.

The quantitative determination of the different phospholipids present in samples of bile, liver or plasma has been performed by densitometric scanning of high-performance thin-layer chromatography plates. These have been developed as follows: pre-treatment with sulphuric acid-ethanol mixture, staining with ethanol-modified molybdenum blue reagent and post-treatments with water and ethanol added with a small quantity of sulphuric acid. The sequential treatment, which introduces some modifications in a previously described procedure, allowed a linear and stable colour response for phosphorous-containing lipids on uniform and colourless background. The calibration of each plate with standard mixture was required for obtaining results in good agreement with other routine methods of determination.

A rapid and sensitive ELISA for rainbow trout maturational gonadotropin (tGtH II): validation on biological samples; in vivo and in vitro responses to GnRH.

A rapid and sensitive heterologous enzyme-linked immunosorbent assay (ELISA) was developed to measure rainbow trout maturational gonadotropin. Purified salmon maturational gonadotropin (sGtH II) was used as reference hormone. Optimization of the procedure was performed by using an anti-beta sGtH serum. Two procedures were developed: an equilibrium assay (which did not involve a preincubation step) which lasted for 8 hr and a nonequilibrium assay (which involved a preincubation step) which lasted for 26 hr. The nonequilibrium assay gave the best sensitivity (70 pg/ml sample). GtH II measurements on in vivo and in vitro samples from GnRH analogs or sGnRH experiments showed that the ELISA procedure could be used over a wide range of concentrations. The method was validated by comparing GtH II concentrations measured by both RIA and ELISA.
