Discovery of macrocyclic hydroxamic acids containing biphenylmethyl derivatives at P1', a series of selective TNF-alpha converting enzyme inhibitors with potent cellular activity in the inhibition of TNF-alpha release.

SAR exploration at P1' using an anti-succinate-based macrocyclic hydroxamic acid as a template led to the identification of several bulky biphenylmethyl P1' derivatives which confer potent porcine TACE and anti-TNF-alpha cellular activities with high selectivity versus most of the MMPs screened. Our studies demonstrate for the first time that TACE has a larger S1' pocket in comparison to MMPs and that potent and selective TACE inhibitors can be achieved by incorporation of sterically bulky P1' residues.

Synthesis and biological evaluation of novel macrocyclic ligands with pendent donor groups as potential yttrium chelators for radioimmunotherapy with improved complex formation kinetics.

Novel 1,4,7-triazacyclononane-N,N',N' '-triacetic acid (NOTA) based octadentate ligands [2-(4,7-biscarboxymethyl[1,4,7]triazacyclononan-1-ylethyl)carbonylmethylamino]acetic acid tetrahydrochloride (1) and [3-(4,7-biscarboxymethyl[1,4,7]triazacyclononan-1-yl-propyl)carbonylmethylamino]acetic acid tetrahydrochloride (2) with pendent donor groups as potential yttrium chelators for radioimmunotherapy (RIT) have been prepared via a convenient and high-yield cyclization route. The complexation kinetics of the novel chelates with Y(III) was investigated and compared to that of 1,4,7,10-tetraazacyclododecane-N,N',N' ',N" '-tetraacetic acid (DOTA), a macrocyclic chelating agent well recognized as forming very stable complexes with yttrium but also limited in usage because of slow Y(III) complex formation rates. The in vitro stability of the corresponding (88)Y-labeled complexes in human serum was assessed by measuring the release of (88)Y from the complexes over 14 days. The in vivo biodistribution of (86)Y-labeled 1 in mice was evaluated and compared to that of the (86)Y-DOTA complex. Formation of the Y complex of 1 was significantly more rapid than that of either 2 or DOTA. Serum stability of the (88)Y complex formed with 1 was equivalent to the DOTA complex, while the complex formed with 2 proved to be significantly unstable. The results obtained from a biodistribution study indicate that the (86)Y-1 complex possesses in vivo stability comparable to the analogous DOTA complex.

Structure-activity study on a novel series of macrocyclic inhibitors of the hepatitis C virus NS3 protease leading to the discovery of BILN 2061.

From the discovery of competitive hexapeptide inhibitors, potent and selective HCV NS3 protease macrocyclic inhibitors have been identified. Structure-activity relationship studies were performed focusing on optimizing the N-terminal carbamate and the aromatic substituent on the (4R)-hydroxyproline moiety. Inhibitors meeting the potency criteria in the cell-based assay and with improved oral bioavailability in rats were identified. BILN 2061 was selected as the best compound, the first NS3 protease inhibitor reported with antiviral activity in man.

Synthesis of C2-C3'N-linked macrocyclic taxoids. Novel docetaxel analogues with high tubulin activity.

Novel C2-C3'N-linked macrocyclic taxoids 4 bearing an aromatic ring at position C2 were synthesized. These compounds, tethered between N3' and the C2-aromatic ring at the ortho, meta, or para position, were constructed by ring-closing metathesis. The para-substituted derivatives were unable to stabilize microtubules, whereas the ortho- and meta-substituted compounds show significant activity in cold-induced microtubule disassembly assay. The meta derivative 4c is the first C2-C3'-linked cyclic analogue to be equipotent to paclitaxel in this assay and to show significant cytotoxicity. Computational studies of the conformational behavior of these compounds indicate that they can adopt several conformations including mainly the "T-shaped" forms. Docking experiments have shown that the "T-shaped" form is preferred for a good interaction of these compounds with the beta-tubulin binding pocket.

Improved in vivo stability of actinium-225 macrocyclic complexes.

The favorable nuclear properties of actinium-225 ((225)Ac) have led to proposal of this isotope for use in radioimmunotherapy. In an effort to reduce the toxicity of free (225)Ac, a series of ligands were evaluated for stability in vivo. Loss of (225)Ac from acyclic chelating agents resulted in high liver uptake and poor whole body clearance. The macrocyclic ligands c-DOTA, PEPA, and HEHA were evaluated, and (225)Ac-HEHA showed exceptional stability in vivo. (225)Ac chelated with EDTA, DTPA, DOTA, or PEPA permitted substantial accumulation of the radionuclide to the liver, while the (225)Ac-HEHA complex was essentially excreted within minutes of administration. The preparation of the ligands and radiolabeled complexes and the biodistribution results will be discussed.

Biaryl-based macrocyclic and polymeric chiral (salophen)Ni(II) complexes: synthesis and spectroscopic study.

Polymeric/oligomeric and macrocyclic (salophen)Ni(II) complexes have been synthesized starting from both an achiral biphenol dialdehyde and an optically active BINOL dialdehyde. It was found that these polysalophens contain nonplanar coordination of Ni(II) units that are paramagnetic. This is different from the previously reported (salophen)Ni(II) complexes which are square planar and diamagnetic. The nonplanar (salophen)Ni(II) units make the new polymeric Ni(II) complexes different from the helical structure proposed for chiral biaryl-based polymers containing square-planar (salophen)Ni(II) units. The copolymerization of the chiral binaphthyl monomer with the achiral biphenyl monomer demonstrates that the chirality of the binaphthyl unit is not propagated along the biphenyl polymer chain.

Applications of phthalimide photochemistry to macrocyclic polyether, polythioether, and polyamide synthesis.

Irradiation of phthalimides which contain N-linked omega-trimethylsilylmethyl-substituted polyether, polythioether, and polysulfonamide chains results in efficient production of the corresponding macrocyclic polyether, polythioether, and polysulfonamide products. These photocyclization reactions follow sequential single electron transfer (SET)-desilylation pathways. Only in the cases of phthalimides, bearing mixed ether-thioether N-substituents, do these excited-state cyclization reactions proceed with lower degrees of regioselectivity. This is a result of competitive desilylation and alpha-to-sulfur deprotonation reactions of the zwitterionic diradical intermediates formed by initial SET.

Supramolecular assemblies between macrocyclic porphyrin hexamers and star-shaped porphyrin arrays.

The syntheses of eight new star-shaped D(3)-symmetric arrays in which three 15-(pyrid-4-yl)porphyrin subunits are attached to the 1, 3, and 5 positions of a benzene core through linkers consisting of collinear repetitive phenylethynyl units have been carried out using Pd(0)-catalyzed coupling reactions. By the same procedure, an analogous 10-(4-pyridin-yl)porphyrin hexamer in which all positions of the benzene core are substituted has been obtained. Likewise, the preparation of suitably sized cyclic porphyrin hexamers, in which all six or at least three alternate porphyrin rings are complexed with Zn(II) ions, is described in detail. In solution, such cyclic porphyrin hexamers form supramolecular assemblies with the star-shaped polyporphyrins in which the latter are held in the interior of the macrocycle through coordination of the apical pyridine rings with the Zn(II) ions. The suggested structures are supported by (1)H NMR spectroscopic and MALDI-TOF mass spectrometric measurements. They agree with the high values of the binding constants of the corresponding supramolecules, which range between K = 1.1 x 10(10) and 1.4 x10(9) M(-1).

Role of conformational effects on the regioselectivity of macrocyclic INOC reactions: two new asymmetric total syntheses of (+)-brefeldin A.

We have gained some insight into the role of conformational effects on the regioselectivity of the macrocyclic intramolecular nitrile oxide cycloaddition observed in our (+)-brefeldin A synthesis. During the course of this regiochemical study, we have developed two novel stereoselective and regioselective schemes for total synthesis of (+)-brefeldin A (i.e. intramolecular nitrile oxide cycloaddition-isomerization and intermolecular nitrile oxide cycloaddition-ring closing metathesis strategies).

Silver complexes with unsaturated thiacrown ethers: inclusion behavior of conformationally restricted macrocycles.

The reactions of 18- and 21-membered unsaturated thiacrown ethers, 18-UT-6 and 21-UT-7, with CF(3)COOAg in acetone afforded novel silver(I) complexes Ag(I)(18-UT-6)(CF(3)COO) and Ag(I)(2)(21-UT-7)(CF(3)COO)(2), respectively. The crystal structure of Ag(I)(18-UT-6)(CF(3)COO) shows that the silver atom occupies the cavity of the 18-UT-6 and the geometry around the silver atom has a distorted five-coordinate square pyramidal arrangement. The crystal structure of Ag(I)(2)(21-UT-7)(CF(3)COO)(2) shows that the two silver atoms and all sulfur atoms are nearly coplanar and the two trifluoroacetate groups are located at the opposite sides of the plane. The stoichiometry for the complexation of 15-UT-5, 18-UT-6, and 21-UT-7 with CF(3)COOAg in solution was examined by (1)H NMR measurement. The titration plots of 15-UT-5 and 21-UT-7 show a distinct inflection point at 1:1 and 2:1 metal/macrocycle ratios, respectively, whereas the plot of 18-UT-6 gradually changes at the range of 1:1 to 2:1. From these results, 15-UT-5 and 21-UT-7 were found to show inclusion selectivity for number of silver ions, respectively, whereas 18-UT-6 showed low selectivity for the inclusion number of metals. Comparison of the oxidation and reduction potentials of the silver(I) complexes with those of free macrocycles and CF(3)COOAg revealed that unsaturated thiacrown ethers become difficult to be oxidized by complexation with CF(3)COOAg, and CF(3)COOAg becomes difficult to be reduced by complexation with unsaturated thiacrown ethers.

Synthesis of new tren-based tris-macrocycles. Anion cluster assembling inside the cavity generated by a bowl-shaped receptor.

The synthesis of three new tris-macrocycles, containing three [12]aneN(4) (L1), [12]aneN(3)O (L2), or [14]aneN(4) (L3) moieties appended to a tren unit, is reported. The crystal structure of the [(Na(ClO(4))(6)) subset L1(2)H(13)]Na(6)Cl(2)(ClO(4))(12) compound shows the anionic cluster [Na(ClO(4))(6)](5)(-) assembled inside the cavity defined by two bowl-shaped polyammonium receptors, held by multiple charge-charge and hydrogen bond interactions.

Solid-phase synthesis of an A-B loop mimetic of the Cepsilon3 domain of human IgE: macrocyclization by Sonogashira coupling.

The solid-phase synthesis of a cyclic peptide containing the 21-residue epitope found in the A-B loop of the Cepsilon3 domain of human immunoglobulin E has been carried out. The key macrocyclization step to form the 65-membered ring is achieved in approximately 15% yield via an "on-resin" Sonogashira coupling reaction which concomitantly installs a diphenylacetylene amino acid conformational constraint within the loop.

Synthesis and characterization of cationic iodonium macrocycles.

A synthetic strategy for the high-yield preparation of iodonium containing macrocycles such as rhomboids, a square, and a pentagon is described, with the long-term objective of preparing iodonium compounds for potential molecular electronics applications. Two cationic rhomboid shaped molecules were prepared for the first time (55-70%) by the treatment of compounds 11 and 12a or 12b with Me3SiOTf. The reaction of dication 8 with 6 in the presence of Me3SiOTf gave an iodonium containing molecular square in 70% yield. In addition, a pentagon-shaped macromolecule was prepared in 60% yield. These iodonium-containing charged macromolecules were characterized by multinuclear NMR, mass spectrometry, and physical means.

Novel crystal structure, cis-trans isomerization, and host property of meta-substituted macrocyclic azobenzenes with the shortest linkers.

Azobenzenophanes, in which two (2), three (3), and four (4) azobenzene units are connected cyclically by methylene linkers at the meta positions, were synthesized. The effect of ring strain on the structure and the photochemical and the thermal isomerization of azobenzene were investigated. Complex formation with guest species such as alkali metal cations and solvent molecules was observed. X-ray crystal analyses revealed the crystal structure of all three isomers of 2. Compound 2(t,c) was distorted due to the ring strain and 2(c,c) was not deformed. Both 3 and 4 formed supramolecular channel structures in which an azobenzenophane molecule holds a solvent molecule in the molecular cavity. Upon exposure to light, 2, 3, and 4 exhibited stepwise trans-cis photoisomerization in solutions. The quantum yields for all isomerization steps of 2 were determined, and these values suggested that interactions between chromophores such as energy transfer and/or steric distortion affect the isomerization process. The lifetime of 2(c,c) (19.7 days) was longer than that of 2(t,c) (6.1 days) at room temperature. The relative stability of these isomers was explained by the isokinetic (or compensation) relationship between deltaH++ and deltaS++ values and by the effect of the ring strain. The relative energies of three isomers of 2 were estimated by HF/6-31G** calculations, and these values indicated that 2(t,c) has the largest ring strain. Complexes (1:1 and 2:1) between macrocycles and alkali metal cations (Li+, Na+, K+, Rb+, Cs+) were observed by ESIMS. The cation selectivity was shifted upon photoirradiation especially in 4.

Synthesis and evaluation of macrocyclic transition state analogue inhibitors for alpha-chymotrypsin.

Lactams 1 and 2 are readily formed from acyclic precursors in the presence of trypsin and chymotrypsin, respectively, identifying the macrocyclic ring system as a potential motif for constrained transition state analogue inhibitors of the serine peptidases. Ketone 3 was synthesized and shown to be a modest inhibitor of chymotrypsin (Ki = 220 microM), albeit 4-fold more potent than the acyclic hydroxy acid 25 (Ki = 1.5 mM as a mixture of epimers). A precursor (31) to the amino boronic acid 4 was also prepared; although this derivative was a potent inhibitor of chymotrypsin (Ki = 130 nM) by virtue of the boronic acid moiety, it showed no advantage over the des-amino analogue 32 (Ki = 120 nM), which is not capable of cyclizing.

Conformational preferences and self-template macrocyclization of squaramide-based foldable modules.

Secondary squaramides have considerable potential as hydrogen bond donors and acceptors. In CHCl(3) both, anti- and syn-squaramide rotamers are observed by NMR. The energetic barrier for anti/syn mutual interconversion determined by complete band shape analysis is approximately 63 kJ mol(-1). As in proline derivatives, a low rotational barrier is crucial for the design of foldable modules. In this paper, folding based on the low rotational barrier of squaramides is driven by donor atoms (N or O) located in the gamma position of an alkyl chain of a secondary squaramide. We demonstrate that the resulting minimal module exists as a folded conformer through the formation of a nine-membered ring stabilized by intramolecular hydrogen bonding. Molecular mechanics calculations and NMR studies support the existence of these folded conformers. The intramolecularly hydrogen bonded conformers are clearly visible even in CHCl(3)-EtOH mixtures. Folding occurs even in pure ethanol. As an indirect test, we studied the effectiveness of macrocyclization reactions in pure ethanol that require an effective templating effect to take place. The high yields obtained support the dominant role of a folded conformer even in this solvent.
