[Analysis of the antigenic properties of hemagglutinins of isolates of the influenza A virus (H1N1), isolated in 1986 in the Byelorussian SSR].

The influenza epidemic occurring in Minsk in the autumn of 1986 was caused by at least two drift variants of influenza A virus (H1N1 subtype) differing in two antigenic determinants of hemagglutinin from the viruses isolated in previous years (A/USSR/90/77, A/Dunedin/6/83). One of the epidemic drift variants was found to be identical in the antigenic properties of hemagglutinin with the reference A/Taiwan/1/86 virus.

Investigations of excitability of isolated and non-isolated neurons from the terminal ganglion of Periplaneta americana by current/voltage clamp and intracellular perfusion.

It was shown by means of current and voltage clamp measurements in combination with the intracellular perfusion method that the isolated motoneuron nerve cell bodies of the cockroach Periplaneta americana show overshoot action potentials. A regenerative Na current was found to be responsible for the excitability of these membranes; however, the results are not in agreement with a de novo synthesis of these channels.

Studies on bovine herpesviruses. Part 1. Isolation and characterization of viruses isolated from the genital tract of cattle.

Herpesviruses, previously isolated from cattle (Theodoridis, 1978), were further studied and provisionally placed in the bovid herpesvirus 4 (BHV-4) group. Major differences were found between IBR-IPV (BHV-1) and BHV-4 virus strains. In MDBK cells, all BHV-4 strains started growing at the edges of the culture, the process progressing slowly until destruction of the cells was complete by the 10th day. BHV-4 strains failed to induce neutralizing antibodies in cattle, goats and rabbits. Only the addition of mineral oil adjuvant induced neutralizing and complement fixing antibodies in goats. BHV-1 strains, in contrast, produced very potent antisera in all these systems. Cross-neutralization tests indicated the existence of 2 distinct serological groups representing BHV-1 and BHV-4. The BHV-4 strains appear to be interrelated and they could not be grouped. A BHV-1 strain showed fixation of complement with the antisera of 6 BHV-4 strains. Electron micrographs showed an accumulation of nucleocapsids in the cytoplasm and an early release of virus particles due to cell destruction. Variation in incubation temperature had a significant effect on the particle formation. At lower temperatures, the number of enveloped particles in the cytoplasm increased. On the basis of the characteristics uncovered in this study, it is possible that all the BHV-4 strains represent one and the same virus which has undergone certain biological changes, thus illustrating a phenomenon which appears to be a characteristic of the herpesviruses.

Structural study of circulating thymic factor: a peptide isolated from pig serum. I. Isolation and purification.

A circulating thymic factor (FTS) has been characterized by a bioassay based on its ability to render theta-negative rosette-forming cells theta-positive and azathioprine-sensitive. FTS was sequentially purified and finally isolated from 1000 liters of pig serum by ultrafiltration, gel filtration, and ion exchange chromatography. Its amino acid composition and apparent molecular weight estimated by Sephadex G-25 chromatography, indicate that FTS is a nonapeptide of composition lysine, aspartic acid (or asparagine), serine 2, glutamic acid (or glutamine) 2, glycine 2, and alanine.

[Isolation, identification and properties of the calcium component of the action potential in isolated neurons of the spinal ganglia of the rat].

Electrophysiological properties of isolated dorsal root ganglion neurons from adult rats were studied by intracellular microelectrodes. All investigated cells were separated into two groups according to the duration of their action potential and the shape of its falling phase. The calcium component of the action potential could be separated by placing the cells into sodium-free, calcium- and TEA-containing medium. The presence of this component was revealed in all studied neurons.

A study of procine enteroviruses isolated from swine in the Toronto area. I. Isolation and serological grouping of viruses.

A number of enteric viruses isolated from swine in the Connaught Medical Research Laboratories and the Mimico Reformatory herds were grouped serologically and compared with previously described porcine enteroviruses. Four of five antigenic types defined were related to other porcine enteroviruses. The fifth was unrelated to any viruses with which it was compared in this study.

[Changes in liver metabolism following a standardized skin burn and intraperitoneal injection of a burn toxin isolated from burned mice skin. Studies on rat liver using isolation perfusion].

Parameters of liver metabolism were studied in the isolated perfused rat liver 5 days after a standard skin burn or an i.p. injection of a specific cutaneous burn toxin. Decreased values of the energy quotient ATP/ADP compared with normal controls were found. The absolute content of adenonucleotides was unchanged. The excretion of glucose and urea was also decreased in both groups and glucose synthesis after a lactate load was only slightly stimulated. The results show a direct influence of a burn toxin on hepatic metabolism suggesting a possible disturbance of oxydative phosphorylation.

[Taxonomy and ecology of brucellosis pathogens isolated from Muridae in the northern foothills of the Caucasus mountains. I. Cultural and biochemical properties of Brucella isolated from Muridae].

The results of the study of 65 Brucella strains isolated from myomorphous rodents in the Northern Caucasus are presented. The study was made with the aim of finding out additional characteristics for the identification of these strains. Using the main tests recommended by the FAO/WHO Subcommittee on the Taxonomy of Brucella, as well as some additional tests, we have revealed that the strains under study are very similar to B. suis. At the same time their capacity for agglutination with anti-melitensis monospecific serum, their high sensitivity to pyronin B, safranine T and gentian violet, their low urease activity and their oxidizing activity in respect to L-alanine, L-asparagine, L-glutamic acid, L-arginine, DL-ornithine, DL-citrullin and L-livin make it possible to consider them the fifth independent biotype of B. suis.

[Taxonomic position and ecology of the causative agent of brucellosis isolated from murine rodents in regions of the northern foothills of the Greater Caucasus. II. The ecological and pathogenetic characteristics of Brucella strains isolated from murine rodents].

To establish the possibility of the circulation of brucellae among the population of myomorphous rodents, the mass survey of these rodents was carried out in the mountainous and foothill regions of the Caucasus. In the area of the northern foothills of the Caucasian Mountains 23 Brucella cultures were isolated from 2715 rodents under examination; these cultures proved to be similar to B. suis 1330 in respect to their pathogenicity and the character of pathomorphological changes induced in guinea pigs. Experiments on white mice demonstrated the possibility of brucellar contamination through sexual route and through feces.

Isolated atrioventricular discordance. Report of two surgical cases with isolated ventricular inversion.

Isolated ventricular inversion is a term used for a congenital heart malformation with the segmental arrangement atrioventricular (AV) discordance and ventriculoarterial concordance. It describes a condition which from a physiological point of view resembles complete transposition of the great arteries. We have recently seen two patients with this anomaly. Both underwent intracardiac repair by means of a Mustard operation at 10 years and 10 months of age, respectively. Associated lesions in the first patient were an AV septal defect with two AV orifices (partial AV canal, ostium primum defect), partial anomalous pulmonary venous return, and anomalies in the systemic venous drainage. A perimembranous ventricular septal defect complicated the condition in the second patient.

Isolation of the homogeneous constituents from non-diffusible sugar-peptide (NSP) fraction originating from bovine plasma. III. The method of isolation and partial characterization of the four peptide constituents.

The purification of four different peptides from the NSP fraction of bovine plasma, using: organic solvents extraction, column Sephadex G-50 filtration, thin-layer Sephadex G-25 electrophoresis in 5 M guanidine hydrochloride and thin-layer Bio-Gel P-6 electrophoresis methods was described. The molecular weights of the isolated peptides and their amino acid composition were determined. The dose-dependent effect two of the four peptides on 3H-thymidine incorporation into BHK-21 and BHK/RSV cells was described. The results support previous observations that the NSP fraction contains a peptide growth prometing factor and a potent peptide growth inhibitor. The possible physiological role of the peptide growth inhibitor was discussed.

Isolation and partial characterization of yeast mannan hydrolysing enzymes from bacterial isolates.

A range of aerobic, mesophilic bacterial strains capable of producing extracellular alpha-mannanase and alpha-mannosidase enzymes were isolated from various natural habitats using yeast cell wall material as the selective medium. Five of them were capable of producing extracellular alpha-mannosidase and alpha-mannanase on yeast mannan as the sole carbon substrate, and they were tentatively identified to the genus level as Arthrobacter, Streptomyces, Pseudomonas, Bacillus and Acinetobacter. Among these strains, Arthrobacter M-02 showed the highest biosynthetic activity of the enzymes. The crude enzyme preparations from the strains were active at pH 4 to 9, reaching a maximum activity between 5.5 and 6.8. The crude Arthrobacter M-02 alpha-mannosidase was more thermostable than that of the other strains, but the alpha-mannanase of the strain was thermolabile.

[Evaluation of concomitant use of cyclosporin and percutaneous isolated liver perfusion under complete venous isolation and charcoal hemoperfusion].

It is difficult to administer an effective dose of cyclosporin A (CsA), a potent inhibitor of P-glycoprotein, to prevent multiple drug resistance due to its side effect. We herein evaluated the efficacy of concomitant use of this agent and complete hepatic venous isolation and charcoal hemoperfusion (HVI.CHP). Dogs were divided into two groups; group I (n = 4), intraarterial infusion of only adriamycin (ADM) and group II (n = 4), intraarterial infusion of CsA and ADM. In both groups, ADM was intraarterially administered for 10 minutes under HVI.CHP. In addition, in group II, CsA infusion (0.3 mg/min.kg) was initiated 20 min prior to the start of ADM infusion and maintained for 30 min. The AUC (micrograms.min/ml) of ADM were 21.2 +/- 8.6 (mean +/- SD) and 28.4 +/- 10.3 in groups I and II, respectively, at prefilter (hepatic venous level) and 8.1 +/- 4.6 and 4.8 +/- 3.8, respectively, at postfilter, showing an effective drug elimination in both groups. The Cmax (micrograms/ml) were 14.1 +/- 2.2 at prefilter, 2.4 +/- 0.5 at postfilter, and 3.4 +/- 1.2 in systemic level. These results indicated that HVI.CHP allowed the high-dose CsA infusion required for P-gp inhibition in the liver and could reduce extraregional CsA leakage.

The isolation and characterization of some vibrio parahaemolyticus strains isolated from salted herring and roe.

390 food samples (frozen, Kippered herring, Salted roe, fresh carp and salted ham) were investigated for the presence of Vibrio parahaemolyticus. 8 V. parahaemolyticus strains were isolated from herring (6.2%) and roe samples (1.3%). Some biochemical characteristics are discussed. In food borne outbreaks, presence of V. parahaemolyticus must be investigated, especially when the epidemiologically incriminated food is fish or fish products.
