Identification and characterization of a complex chromosomal replication origin in Schizosaccharomyces pombe.

In the budding yeast, S. cerevisiae, two-dimensional (2D) gel electrophoresis techniques permit mapping of DNA replication origins to short stretches of DNA (+/- 300 bp). In contrast, in mammalian cells and Drosophila, 2D gel techniques do not permit precise origin localization; the results have been interpreted to suggest that replication initiates in broad zones (several kbp or more). However, alternative techniques (replication timing, nascent strand polarity analysis, nascent strand size analysis) suggest that mammalian origins can be mapped to short DNA stretches, just like S. cerevisiae origins. Because the fission yeast, Schizosaccharomyces pombe, resembles higher organisms in several ways to a greater extent than does S. cerevisiae, we thought that S. pombe replication origins might prove to resemble--and thus be helpful models for--animal cell origins. An attempt to test this possibility using 2D gel techniques resulted in identification of a replication origin near the ura4 gene on chromosome III of S. pombe. The 2D gel patterns produced by this S. pombe origin indeed resemble the patterns produced by animal cell origins and show that the S. pombe origin cannot be precisely located. The data suggest an initiation zone of 3-5 kbp. Some aspects of the 2D gel patterns detected at the S. pombe origin cannot be explained by the rationale of initiation in broad zones, suggesting that future biochemical and genetic studies of this complex origin are likely to provide information useful in helping to understand the apparent conflict between the 2D gel mapping techniques and other mapping techniques at animal cell origins.

[A preparation of short DNA chains synthesized in vivo after introduction into DNA from live cells of trioxsalen crosslinks is not enriched by replication initiation regions].

The experiments undertaken in order to verify the trioxsalen-crosslinking method suggested by Russev and Vassilev for isolation of eukaryotic replication origins are described. It was found that the preparation of viral DNA isolated by the above mentioned method from CV-1 cells lytically infected with SV40 was not enriched in sequences including SV40 replication origin. The hybridization pattern of DNA preparation isolated by the trioxsalen-crosslinking procedure from chicken erythroblastosis cells with the cloned fragments of globin gene domain was found to be identical to those of the total DNA probe. The DNA fraction enriched in replication origins was isolated from the same cells with the aid of nascent DNA strand extrusion method by Zannis-Hadjopoulos et al. The hybridization pattern of this DNA fraction with the cloned fragments of chicken alpha-globin gene domain was different from those of total DNA. Taking together, the results of our experiments demonstrate that trioxsalen-crosslinking procedure does not lead to the isolation of replication origins from the objects studied in the present investigation.

Intractable seizures of frontal lobe origin: clinical characteristics, localizing signs, and results of surgery.

PURPOSE: We analyzed the clinical characteristics of seizures of frontal lobe (FL) origin with particular emphasis on establishing different categories and determining if these categories had any localizing or lateralizing value. In addition, results of surgery are reported. METHODS: Seizure characteristics were established by historical review and electroencephalographic/videotape analysis of 449 seizures in 26 adult patients with refractory seizures of FL origin. RESULTS: No outstanding risk factor was identified for seizures of FL origin. Seizures were frequent (7.1 per week), brief (mean duration, 48.3 seconds), and had a nocturnal preponderance in 58% of the patients. Status epilepticus was reported in 54%, and generalized convulsions as a prominent seizure type were reported in 26% of patients. The most common reported aura was a nonspecific sensation, often localized to the head (35%). Early forced head and eye deviation was not a consistent lateralizing sign, whereas late head and eye deviation always occurred contralateral to the site of seizure origin. Early asymmetric tonic posturing occurred consistently contralateral to the side of seizure origin. Clinical seizure patterns did not consistently localize to specific regions of the frontal lobe, although there were some noticeable trends: focal clonic seizures were associated with seizure origin in the frontal convexity; tonic seizures were most often associated with origin in the supplementary motor area but also occurred with origin in other parts of the frontal lobe; seizures resembling typical temporal lobe seizures with oroalimentary automatisms were observed with seizure origin in the orbitofrontal region; and seizures with hyperactive, frenetic automatisms were not associated with any specific region within the frontal lobes. Eighty percent of patients had favorable seizure outcome after surgery (class I/II). CONCLUSION: Although certain clinical features are characteristic for seizures of frontal lobe origin and some have lateralizing value, they do not localize to specific areas within the FL. After careful presurgical evaluation, both lesional and nonlesional patients benefit from epilepsy surgery.

Electrophoretic protein pattern of human parotid saliva in Sjögren's syndrome and sialosis.

The electrophoretic pattern of the basic proteins in parotid saliva was examined in 12 control subjects and 45 patients suffering from xerostomia and parotid swelling of diverse origins. The protein pattern in the control subjects did not differ from the observed in sialotic patients. Characteristic quantitative and qualitative alterations in protein composition were observed in Sjögren's syndrome. The concentration of the protein fractions with a low Rp value showed a considerable increase, while one or two additional protein bands with high Rp value could be detected in these patients. The appearance of these additional protein bands may prove to be a valuable laboratory indicator in the diagnostic work. If they really indicate the beginning of an autoimmune process, the disease could be diagnosed much earlier than by the methods available today.

[ Radiological anatomy of the breast].

The macroscopic anatomy of the breast is often not well understood; it is the reason why the radiologic images formation of this organ needs to be explained. This work is based upon anatomical-radiological confrontations, which allow to separate the two main compartments of the gland, the fibroglandular tissue and the adipose tissue, and to study the limits of these two components. Their different proportions are at the origin of different patterns leading to a simple classification of non-pathologic breast X-rays. This classification makes easier the diagnosis of abnormalities, especially of tumoral origin.

Thymus independence of hybrid resistance against a panel of T-cell lymphomas of H-2b origin.

Adult thymectomy, followed by whole-body irradiation and reconstitution with fetal liver, was performed to study the T-cell dependence of F1 hybrid resistance to a panel of lymphomas of H-2b origin. Previously, the pattern of hybrid resistance against the same lymphomas was found to correlate with the pattern of NK-activity in a spectrum of F1 hybrids (Kiessling et al., 1975). We now show that hybrid resistance against three lymphomas of C57BL/6 origin, P-52-127-166, RBL-5 and EL-4 and against YLD, of C57L origin, is expressed in the absence of thymus. In another series of experiments, the effectors responsible for hybrid resistance to the transplanted lymphoma EL-4 were studied by reconstituting thymectomized and non-thymectomized C57BL mice with syngeneic bone marrow from NK-deficient beige mutant or wild-type C57BL donors. While the recipients of beige bone marrow had a clearly reduced tumor resistance, thymectomy did not decrease resistance further. This study supports the hypothesis that resistance to these lymphomas in F1 hybrids as well as in syngeneic mice is mainly mediated by natural killer cells.

X chromosome inactivation in carriers of Barth syndrome.

Barth syndrome (BTHS) is a rare X-linked recessive disorder characterized by cardiac and skeletal myopathy, neutropenia, and short stature. A gene for BTHS, G4.5, was recently cloned and encodes several novel proteins, named "tafazzins." Unique mutations have been found. No correlation between the location or type of mutation and the phenotype of BTHS has been found. Female carriers of BTHS seem to be healthy. This could be due to a selection against cells that have the mutant allele on the active X chromosome. We therefore analyzed X chromosome inactivation in 16 obligate carriers of BTHS, from six families, using PCR in the androgen-receptor locus. An extremely skewed X-inactivation pattern (>=95:5), not found in 148 female controls, was found in six carriers. The skewed pattern in two carriers from one family was confirmed in DNA from cultured fibroblasts. Five carriers from two families had a skewed pattern (80:20-<95:5), a pattern that was found in only 11 of 148 female controls. Of the 11 carriers with a skewed pattern, the parental origin of the inactive X chromosome was maternal in all seven cases for which this could be determined. In two families, carriers with an extremely skewed pattern and carriers with a random pattern were found. The skewed X inactivation in 11 of 16 carriers is probably the result of a selection against cells with the mutated gene on the active X chromosome. Since BTHS also shows great clinical variation within families, additional factors are likely to influence the expression of the phenotype. Such factors may also influence the selection mechanism in carriers.

Positive electron affinity of fullerenes: Its effect and origin.

The universal variation pattern of the total energy of various fullerenes including single-walled carbon nanotubes with respect to their extra charge is revealed by the density-functional-theory calculations. The parabolic energy-charge curve with its lowest energy value corresponding to a negatively charged fullerene indicates that these carbon materials have positive electron affinity and are not in the most stable state. The positive electron affinity seems to originate from the pi-electrons and is found to be related to the aggregation property of fullerenes.

Anatomic variability in coronary arterial distribution with regard to the arterial switch procedure.

BACKGROUND: We investigated the coronary arterial origins and course and the position of the great arteries in hearts with discordant ventriculoarterial connections. At the same time, we sought to evaluate the practicality of alphanumeric classifications in accounting for surgically relevant features of the coronary arteries. METHODS AND RESULTS: We studied 200 postmortem hearts, noting the patterns of coronary arterial branching, the vertical and horizontal location of the arterial orifices within the aortic sinuses, the course of the proximal coronary arteries in relation to the aortic wall, and the relations of the great arteries and their respective commissures. All hearts examined had concordant atrioventricular and discordant ventriculoarterial connections. We found 7 of the 8 predicted patterns for sinusal origin of the 3 major coronary arteries and identified 5 different positions of the arterial trunks relative to each other. A correlation was found between less frequent relationships of the arterial trunks and unusual patterns of coronary arterial branching, as well as with mismatch between the valvar commissures. CONCLUSIONS: The surgically relevant features of the coronary arteries in hearts with discordant ventriculoarterial connections are best described rather than classified. Correlations exist between certain, less frequent relations of the great arteries and unusual patterns of branching of the coronary arteries. The presence of unusual great arterial positions should alert the surgeon to potentially complicated arrangements of the origin and distribution of the coronary arteries.

Do neurons process information by relative intervals in spike trains?

We suggest the possibility that neurons process information in terms of the relative duration of clusters of adjacent and successive inter-action potential intervals ("bytes" of intervals). If this concept is plausible, as is supported by research from several laboratories which have specifically addressed this possibility, one should be able to see evidence for such patterning in the published illustrations from studies in which this concept was not considered. We present some of this evidence here, along with some illustrations from the original publications. Byte patterns are evident in these examples, even though they went unrecognized by authors and readers alike. It is true that interval patterns are not obvious in all published illustrations of spike trains, and we suggest that this can be explained by one or more of the following: (1) some neurons may operate with an interval-pattern code while others do not, (2) a given neuron may use an interval-pattern code only under certain conditions, and (3) even when such a code exists, it may be difficult to detect for identifiable technical reasons. Therefore, we believe that the relative-internal-pattern concept is a valid scientific hypothesis which merits specific testing of its validity and range of applicability.

Learning by selection in the trion model of cortical organization.

The basic issue of whether mammalian learning in cortex proceeds via a selection principle, as stressed by Edelman, versus an instructional one is of major importance. We present here a realization of selection learning in the trion model, which is based on the Mountcastle columnar organizational principle of cortex. We suggest that mammalian cortex starts out with an a priori connectivity between minicolumns that is highly structured in time and in space, competing between excitation and inhibition. This provides a "naive" repertoire of spatial-temporal firing patterns that stimuli and internal processing map onto. These patterns can be learned with small modifications to the connectivity strengths determined by a Hebbian learning rule. As various patterns are learned, the repertoire changes somewhat in order to respond properly to various stimuli, but the majority of all possible stimuli still map onto spatial-temporal firing patterns of the original repertoire. In order to show that the example presented here is showing true selectivity and is not an artifact of more stimuli evolving into the learned pattern, we develop a selectivity measure. We suggest that some form of instructional learning (in which connectivities are finely tuned) is present for difficult tasks requiring many trials, whereas very rapid learning involves selectional learning. Both types of learning must be considered to understand behavior.

Establishment and maintenance of DNA methylation patterns in mouse Ndn: implications for maintenance of imprinting in target genes of the imprinting center.

Ndn is located on chromosome 7C, an imprinted region of the mouse genome. Imprinting of Ndn and adjacent paternally expressed genes is regulated by a regional imprinting control element known as the imprinting center (IC). An IC also controls imprint resetting of target genes in the region of conserved synteny on human chromosome 15q11-q13, which is deleted or rearranged in the neurodevelopmental disorder Prader-Willi syndrome. Epigenetic modifications such as DNA methylation, which occur in gametes and can be stably propagated, are presumed to establish and maintain the imprint in target genes of the IC. While most DNA becomes substantially demethylated by the blastocyst stage, some imprinted genes have regions that escape global demethylation and may maintain the imprint. We have now analyzed the methylation of 39 CpG dinucleotide sequences in the 5' end of Ndn by sodium bisulfite sequencing in gametes and in preimplantation and adult tissues. While sperm DNA is completely unmethylated across this region, oocyte DNA is partially methylated. A distinctive but unstable maternal methylation pattern persists until the morula stage and is lost in the blastocyst stage, where low levels of methylation are present on most DNA strands of either parental origin. The methylation pattern is then substantially remodeled, and fewer than half of maternally derived DNA strands in adult brain resemble the oocyte pattern. We postulate that for Ndn, DNA methylation may initially preserve a gametic imprint during preimplantation development, but other epigenetic events may maintain the imprint later in embryonic development.

Cyclic esotropia and the treatment of over-elevation in adduction and V-pattern.

AIM: To describe the development and treatment of V-pattern and bilateral over-elevation in adduction in patients with cyclic esotropia. METHODS: Three patients with cyclic esotropia are described in retrospect. All patients underwent bilateral medial recession, while one patient additionally had a weakening procedure of both inferior oblique muscles. RESULTS: All patients developed a V-pattern and bilateral over-elevation in adduction. After strabismus surgery, normal eye alignment with some signs of binocularity was reached in all three patients. Moreover, the over-elevation in adduction and V-pattern completely resolved. CONCLUSIONS: V-pattern and bilateral over-elevation in adduction was seen in all patients with cyclic esotropia in the stage that fusion was disrupted. Possible explanations for the origin of these patterns are discussed. Strabismus surgery aimed to correct the horizontal esotropic angle restored fusion and eliminated the vertical incommitancies in lateral gaze, and the V-pattern.

Metaphyseal pattern: uniqueness of this structure in growing bones originating from cartilaginous anlage. A microradiographic study.

Microradiographic examination of metaphyses in long and short tubular growing bones allowed detection of a repetitive, clearly defined pattern of three adjacent zones; the latter are successively formed by the mineralization of cartilaginous longitudinal intercolumnar septa and by the subsequent apposition of other mineralized tissues concurrently with resorption. Consequently, each zone of the metaphysis includes mineralized tissues of various compositions and ages, identifiable by their different mineralization densities. Microradiography of pieces of the growing skeleton in several animal species shows that the same organization is not only present in long and short tubular bones but also in many other such as the pelvis and scapula, cuboid bones like the calcaneum and talus, and cartilaginous bones at the base of the skull. This suggests that there is no difference between the osteogenesis pattern of these bones and tubular ones. The problem of identifying the factors generating such metaphyseal organization is raised.

Major outer membrane proteins of Escherichia coli strains of human origin.

The major outer membrane protein patterns of 45 Escherichia coli strains of human origin were compared with that of E. coli K12 by sodium dodecyl sulphate-polyacrylamide gel electrophoresis. Preparations of the former strains contained between two and five major bands in the molecular weight range between 30 000 and 42 000. The patterns were very heterogeneous with respect to the numbers and electrophoresis mobilities of the major outer membrane protein bands. In all cases the fastest moving band was characterized as a protein similar to the ompA protein of strain K12 as it was partially degraded by trypsin and reacted specifically with antiserum against the purified omp A protein in a gel immuno-radioassay. All the other major outer membrane proteins are related to the ompC and ompF proteins (the porins) of strain K12 as they were peptidoglycan-associated and reacted with antisera against the purified ompC and/or ompF proteins.

HCV genotype 4 in Belgium: three distinct patterns among patients from European and African origin.

BACKGROUND: Considered uncommon in western countries some years ago, hepatitis C virus of genotype 4 is now spreading in some areas of Europe. This is assumed to be due to immigration from a region of high prevalence for this genotype and to propagation among drug users. In the south of Belgium, genotype 4 currently accounts for 10% of hepatitis C virus patients and its prevalence is increasing with time. OBJECTIVE: To better define the genotype 4 carriers' characteristics. METHODS: In a database comprising 1726 viraemic hepatitis C virus patients, the files of 85 genotype 4 carriers were reviewed. RESULTS: Beside the African (58%) and European drug user (15%) subgroups classically described, a third subgroup consisting of European nondrug users (26%) was identified as peculiar: these patients were older, had been mostly contaminated sporadically, presented a great diversity of subtypes, and were mainly of Italian origin. In this subgroup, contamination was supposed to be ancient, having occurred probably in Italy before immigration into Belgium. By contrast, European drug users were infected with only two subtypes (4c/4d and 4), an observation in favour of recent spread. Africans had a great diversity of subtypes, were young, and were mostly contaminated sporadically in their home countries. Despite their epidemiological differences, the clinical management, and in particular the rates of eligibility for treatment, were similar for these three groups. CONCLUSIONS: Three different patterns of genotype 4 carriers were observed, corresponding to three different spreading profiles. They did not induce, however, different clinical management.

Modelling the qualitative response of fungal mycelia to heterogeneous environments

Many species of fungi form a mycelium, an indeterminate system of protoplasm-filled, apically extending, branching tubes (hyphae). At the macroscopic level, mycelia produce organisational patterns that are found in many other indeterminate systems (e.g. nervous and vascular systems). Therefore they provide an experimentally and observationally accessible model system for investigating the dynamic origins of phenotypic patterns in such systems. Moreover, mycelial fungi form a vital link in the ecosystem allowing for the redistribution and reutilisation of nutrients and minerals over a far wider spatial domain than would be possible without their presence. In vivo these networks grow and function in heterogeneous environments and therefore to gain any true understanding of their form and function such heterogeneity must be taken into account. In this paper we develop a model based on a system of partial differential equations for the interaction of the fungal mycelium with a heterogeneous environment. Using this model we are able to test the hypothesis that mycelia react to their environment in a global manner and by close comparison with experimental results, we are also able to highlight two specific mechanisms in this global response which are central to the macroscopic distribution of biomass: uptake of nutrients in excess of local needs and subsequent internal redistribution (translocation) of this excess.
