Enumeration and confirmation of Bacillus cereus in foods: collaborative study.

A collaborative study was conducted in 15 laboratories to evaluate 2 different techniques for enumerating Bacillus cereus in foods. A direct plating technique using mannitol-egg yolk-poly-myxin agar and a most probable number (MPN) technique using trypticase-soy-polymyxin broth were compared for the enumeration of high and low populations of B. cereus in mashed potatoes. The collaborative results showed that the overall mean recovery obtained with the low population level was essentially the same by both techniques. However, the overall mean recovery was significantly higher by the direct plating technique at the high population level. A statistical evaluation of the data also showed that the direct plating technique had better repeatability and reproducibility than did the MPN technique at both the high and low population levels. These results suggest that the MPN technique is suitable for examining foods containing low populations of B. cereus, but that the direct plating technique is preferable for foods that contain a high population of this organism. The confirmatory technique used in the proposed method is reliable for presumptive identification of isolates as B. cereus. The method has been adopted as official first action.

Effect of different drinks on the absorption of non-heme iron from composite meals.

A study was made on the effect of various drinks on the absorption on non-heme iron. The drinks were taken with standard meals composed of a hamburger, string beans and mashed potatoes. In each series identical meals were served to the same subject either with water or with the drink under study, labelling the meals with two different radio-iron isotopes. A reduction in iron absorption was seen when serving tea (62 per cent) or coffee (35 per cent) with the meals. Orange juice increased the iron absorption (85 per cent). Pure alcohol and wine increased only slightly the percentage absorbed. Wine often has a high iron content, which increased significantly the amount of iron absorbed (three times). Milk and beer have no significant effect. Coca-Cola increased only slightly the absorption. The present studies clearly shows that the choice of drink drunk with a meal can markedly affect the absorption of non-heme iron.

Amounts of twelve elements required to induce selenium-vitamin E deficiency in ducklings.

Mortality and myopathy of selenium-vitamin E (Se-E) deficiency was produced, in a concentration-dependent pattern, during a 4-week study of 750 ducklings fed a commercial duck starter mash that contained adequate amounts of Se and E, and supplemented with multiple amounts of Ag (50 to 3,000 mg/kg of feed, as acetate), Zn (3,000 to 6,000 mg/kg, as sulfate), Cd (10 to 500 mg/kg, as sulfate), Te (25-500 mg/kg, as tetrachloride), Co (100 to 1,000 mg/kg, as chloride), Cu (500 to 1,500 mg/kg, as sulfate), Hg (200 to 400 mg/kg, as chloride), and Sn (1,000 mg/kg, as chloride). Also, feeding supplements of Pb (500 mg/kg, as acetate), As (600 mg/kg, as sodium arsenilate), Fe (5,000 mg/kg, as sulfate), and S (5,000 mg/kg, as sodium sulfite) produced a low-to-medium frequency of lesions of Se-E deficiency. In ducklings with muscle lesions, the gizzard was most often affected (84.2%), followed in decreasing order by skeletal muscles (69.7%), intestine (34.9%), and heart (23.0%). The frequency of skeletal muscle lesions was high in birds fed Ag, and myocardial necrosis was frequent in ducklings fed Te and Hg. Ducklings affected with myopathy were reluctant to stand. Subcutaneous edema, with or without hemorrhages, and pale areas of myonecrosis in gizzard, skeletal muscles, intestine, and heart were seen at necropsy. Birds fed Te and Hg often had hydropericardium and hemorrhagic myocardial necrosis. Seemingly, addition of many elements to a Se-E adequate commercial diet will increase the requirement for Se-E. In our duckling model, minimal amounts shown to induce Se-E deficiency were 50 mg of Ag/kg, 3,000 mg of Zn/kg, 10 mg of Cd/kg, 25 mg of Te/kg, 1200 mg of Co/kg, 500 mg of Cu/kg, 200 mg of Hg/kg, 1,000 mg of Sn/kg, 500 mg of Pb/kg, 600 mg of As/kg, 5,000 mg of Fe/kg, and 5,000 mg of S/kg.

Altered growth patterns and depressed pituitary growth hormone content in young rats: effects of pre- and postnatal thiouracil administration.

The important influence of postnatal thyroid status on development is well recognized, while the effect of prenatal thyroid status on postnatal growth is less understood. The present study was undertaken to assess the effects of pre- and postnatal thiouracil administration on postnatal growth and pituitary grown hormone (GH) content in 25 day old rats. Pups were born to Sprague-Dawley rats fed lab chow mash containing 0.25% thiouracil according to the following schedules: a) during gestation only; b) during lactation only (25 days postpartum); and c) through gestation and lactation. Mothers of euthyroid controls were fed diet without thiouracil for the length of the experiment. Pups were weighed daily through day 21 and decapitated on day 25. Pituitary GH content was measured by densiometric comparison of disc gel column electrophoresed pituitary homogenates with similarly treated rat GH standard. Incorporation of thiouracil into the diet of pregnant and/or lactating rats significantly depressed the body weight and pituitary GH levels in their 25 day old pups. Furthermore, analysis of the daily mean body weight of each litter revealed four significantly different patterns of growth. While previous investigators have reported that prenatal thyroid restriction does not significantly alter the developmental processes of young rodents, the present study indicates that prenatal thyroid status is an important influence on normal postnatal growth.

Liquid chromatography with fluorescence detection of lasalocid sodium in feeds and premixes.

Lasalocid sodium is extracted directly from feeds and premixes with the mobile phase solution, an aliquot is chromatographed, and the concentration (peak height) of lasalocid sodium is measured by high pressure liquid chromatography with fluorescence detection. Chromatography and detection are completed in about 7 min. The results agree well with those found by the official AOAC fluorometric method, 42.A08--42.A13. The method offers an alternative to the AOAC method and has the capability of processing almost twice as many samples. The average recovery of 4 levels (0.0008, 0.0040, 0.0080, and 0.0120%) of lasalocid sodium from feeds is 100%. Replicate assays of typical poultry mashes and pellets containing about 0.0080% lasalocid sodium yielded coefficients of variation of 2.68--3.35%.

Robenidine resistance in Eimeria tenella.

Robenidine protected chickens against cecal coccidiosis infections initiated by a strain of the parasite that had no previous exposure to drugs. No cross resistance was found with 13 strains resistant to other anticoccidials. A strain of Eimeria tenella that was serially propagated in chickens fed mash containing robenidine became resistant to the chemical. No cross resistance was detected when this experimental strain was tested against 12 other anticoccidials.

Long-term effects of diazepam on plasma lipids and atheroma in roosters fed in atherogenic diet.

Long-term effects of diazepam on plasma lipids and atheroma in roosters fed an atherogenic diet have been studied. Our data indicate that long-term administration (8 months) of diazepam (Valium) 0.2 mg/kg orally lowered the plasma cholesterol level when compared to the atherogenic group, but this was not statistically significant (P greater than 0.05). However, at the dose of 0.4 mg/kg of Valium the plasma cholesterol was significantly lowered when compared to the controls on an atherogenic diet (P less than 0.05). Both doses of Valium significantly decreased the incidence and severity of lesions on the endothelial surface of the aorta and coronary artery in roosters fed an atherogenic diet. No changes in plasma triglyceride values were noted. Administration of 0.4 mg/kg of Valium to birds receiving the atherogenic diet produced a small but significant increase of mean, systolic and diastolic blood pressures as compared to the birds receiving an atherogenic diet but no Valium. A similar increase in blood pressure was also noted in the control group on plain mash diet receiving 0.2 mg/kg of Valium. Microscopic examination of the coronary arteries showed that birds on an atherogenic regimen receiving Valium had less atherosclerosis than those not receiving Valium.

Induction of lesions of selenium-vitamin E deficiency in ducklings fed silver, copper, cobalt, tellurium, cadmium, or zinc: protection by selenium or vitamin E supplements.

In 3 experiments, 684 newly hatched White Pekin ducklings were fed (for 15 to 28 days) a commercial starter mash that was adequate in selenium and vitamin E (Se-E) content, either alone or with supplements of Ag (3,000 mg/kg of feed, as acetate), Cu (1,500 mg/kg, as sulfate), Co (200 or 500 mg/kg, as chloride), Te (500 mg/kg, as tetrachloride), Cd (100 or 500 mg/kg, as sulfate), Zn (3,000 or 6,000 mg/kg, as sulfate), or V (100 mg/kg, as vanadate). The ducklings fed Ag, Cu, Co, Te, Cd, and Zn frequently developed lesions characteristic of Se-E deficiency, such as necrosis of skeletal and cardiac muscle and of smooth muscle of the gizzard and intestine. Complete protection from the muscle lesions produced by Cu, Co, Te, Cd, and Zn supplements was provided by vitamin E (200 IU of alpha-tocopherol acetate/kg) and Se (2 mg/kg, as selenite). Ducklings fed Ag were protected by supplements of vitamin E and partial protection was achieved by Se addition. The birds fed excessive Zn developed pancreatic necrosis and fibrosis that was not prevented by supplements of Se or vitamin E. Terminally, blood glutathione peroxidase activity was low and hepatic Se concentration was increased in the ducklings fed Ag. However, neither blood glutathione peroxidase activity nor hepatic Se concentrations was consistently abnormal in ducklings fed other trace elements, although lesions of Se-E deficiency were often present in these animals.

Circadian variation of 3-hydroxy-3-methylglutaryl coenzyme A reductase activity in swine liver and ileum.

The temporal variation of HMG-CoA reductase activity in the liver and intestine of swine was investigated. The thin-layer chromatographic method widely used in the assay of the reductase was successfully applied to the porcine enzymes. Parallel circadian rhythms were demonstrated in both hepatic and ileal reductases from mash-fed animals. Peak activity occurred approximately 6 hr after feeding, 2.7-fold over the basal level in the liver, and 1.6-fold in the ileum. A milk-cholesterol diet caused a marked depression of both rhythms (90% in liver, 50% in ileum); however, the hourly variation in activity persisted in both organs. Cholestyramine was found to elevate hepatic activity (2.7-fold throughout the rhythm) without affecting that of the intestine. Clofibrate had no effect on either enzyme at any time during the cycle despite a 34% reduction in serum cholesterol concentrations.

[Radio-immunoassay of antidiuretic hormone. Choice of a technic of extraction, Normal and pathological values (author's transl)].

The low serum concentration of ADH requires extraction prior to assay. Various methods are compared (direct extraction by acetone, adsorption on talcum powder. Florisil, silicilic acid, Fuller's earth, QUSO, carbon-dextran, adsormone, then elution with hydrochloric acetone). The technic finally used was extraction with Florisil. The antibody used was commercially accessible. The separation of bound from free hormone required a second antibody fixed on Sephadex (DASP). The usual values found in 31 normal subjects from 22 to 89 years were: 7,4 . 4,1 pmol/l (M 2 sigma) 1 pmol/l = 1,08 pg/ml. These assays were carried out on tissue extracts prepared by mashing tumour fragments or lymph nodes removed from a patient with the Schwartz Bartter syndrome. They showed the existence in these tissues of high concentrations of ADH immunologically identical with that present in the serum of normal subjects.

Acceptability of potatoes groun in West Texas.

This study indicated that methods used to prepare fresh potatoes were, in decreasing order: Baking, mashing, boiling, and frying. The two varieties of potatoes commonly grown in West Texas were acceptable and compared favorably with similar varieties available in a local supermarket. However, the potatoes grown in West Texas had lower specific gravity. The Norgold Russet variety was judged by a sensory panel to be more acceptable than the Viking. Baked potatoes had superior texture but poorer color than boiled potatoes. Storing potatoes under home-like conditions for twenty-eight days resulted in lower acceptability, weight loss, and lower specific gravity. Some Norgold Russet potatoes were rotten after twenty-eight days' storage. Specific gravity on the day of cooking was correlated with texture of cooked potatoes. Supermarket shoppers' comments suggest that research is needed on the acceptability of West Texas potatoes baked in microwave ovens.

Sample preparation of carbadox, furazolidone, nitrofurazone, and ethopabate in medicated feeds for high pressure liquid chromatography.

Medicated feeds (pelleted or mash) containing guarantees of carbadox, furazolidone, nitrofurazone, and ethopabate are pretreated with water, extracted with 95% dimethylformamide overnight at room temperature, cleaned up on a column of alumina, and injected into a high pressure liquid chromatograph for quantitative measurement. Carbadox, nitrofurazone, and furazolidone can be separated; chromatograms show excellent baseline resolution, and results are in good agreement with colorimetric methods. The same extraction and cleanup can be used to improve colorimetric methods for furazolidone and nitrofurazone.

[Carcass disposal in Hungary (author's transl)].

In Hungary more than half of the entire livestock is kept in industrial breeding plants i.e. under mass breeding conditions. So long as the industrial capacity of the plants remains insufficient to process the volume of carcasses, the animal breeding plants are compelled to find their own solutions for the disposal and/or rendering of the carcasses and waste. On some state farms the carcasses are processed into so-called meat-mash which must be utilised as fodder within 48 hours after the death of the animal. High-quality meat-meal can only be obtained from fresh carcasses. Carcasses are processed for reasons of economy, but the methods of carcass disposal are subject to stringent requirements imposed by the public health service, veterinary hygiene inspectorates and by the environment protection authorities. The carcasses or slaughter-house waste destined for processing must not contain any pathogens likely to be injurious to man or animal or to threaten the health of the persons concerned with the carcass processing. For this reason the carcasses must be kept in boilers for 2 to 4 hours at a pressure of 4 atü. The formulation of the factory rules and the technology of processing must make due allowance for these stipulations.

Transit of a meal through the stomach, small intestine, and colon in normal subjects and its role in the pathogenesis of diarrhea.

A method for measuring the transit time of a meal, containing sausages, mashed potato, baked beans, and a pineapple custard dessert, through the gastrointestinal tract was evaluated in 14 healthy volunteers. Gastric emptying was determined by incorporating a radioactive marker in the meal and counting over the surface of the stomach using a crystal scintillation detector. Small intestinal transit time was determined by measuring breath hydrogen excretion and by estimating the radioactivity over the cecum. Finally, whole gut transit time was measured by incorporating radiopaque plastic markers or carmine red in the meal and estimating the appearance of these markers in the stool. Our results showed that measurements of small intestinal transit time were reproducible and in the majority of subjects the increase in hydrogen excretion occurred at the same time as the increase in radioactive counts over the surface of the cecum. The passage of the first marker in the stool coincided with the appearance of carmine red. There were no significant correlations between small intestinal transit time and whole gut transit time or the half time for gastric emptying. Incorporation of 10, 25, and 40 g lactulose into our standard meal in place of sucrose increased the rate of transit through the small intestine but did not significantly alter the rate of gastric emptying or the whole gut transit time. Total stool weight for 48 hr after ingestion of the meal was inversely related to whole gut transit time but not to small intestinal transit time suggesting that the tendency to develop diarrhea in response to a meal containing unabsorbable carbohydrate depends more on the lack of colonic accommodation than on the rate of small intestinal transit. Finally, there was no significant correlation between the measurements of small intestinal transit time after a drink of lactulose and the transit time of a meal in the same subjects.

Fatty acid composition of porcine muscle and adipose tissue lipids as affected by anatomical location and cod liver oil supplementation of the diet.

In pigs fed a standard pig mash the contents of polyunsaturated fatty acids (PUFAs) of both the n-6 and n-3 series were significantly higher in the dark red mm adductores compared to the light coloured m longissimus lumborum. Perirenal fat had a higher concentration of saturated fatty acids (14:0, 16:0, 18:0) than backfat, and a lower concentration of monounsaturated fatty acids, such as 16:1n-7 and 18:1n-9. Daily supplementation of 50 ml cod liver oil, rich in n-3 PUFAs, during the fourth and third week before slaughter led to a 1.4 to 1.7 times increase in the contents of n-3 PUFAs in muscles and fat depots. There was no difference between the incorporation of n-3 PUFAs in dark and light muscles. Perirenal fat contained more 20:5n-3 (EPA) and 22:6n-3 (DHA), but less 20:1n-9 (eicosenoic acid) than the backfat, after cod liver oil supplementation rich in these 3 fatty acids. Supplementation of cod liver oil reduced the n-6/n-3 fatty acid ratio in all anatomical locations examined.

[Use of Jerusalem artichokes in diet therapy of patients with type II diabetes mellitus].

The purpose of present study was to evaluate the therapeutic and preventive actions of topinambur included in controlled diet for patients with II type diabetes. 35 patients aged 46-58 years suffering diabetes mellitus during 3-10 years have been under study. All patients receiving the diet N9 were randomly divided on two groups with (experimental group) and without (control group) consumption of topinambur. The mashed topinambur (100 g/day for second breakfast) and cookies with topinambur (50 g/day) were used in experimental diet during 4 weeks. It was shown that experimental diet containing topinambur caused slight increasing of postprandial hyperglycemia. In patients eating experimental diet with reduced caloric contents an increasing of serum blood triglyceride level was observed. The results do not give background to recommend topinambur in dietary treatment of non-insulin-dependent diabetes mellitus.

Studies on high concentration ethanol fermentation of raw ground corn by Saccharomyces sp. H0.

In this study, high concentration alcoholic fermentation coordinated with saccharification from raw ground corn was performed at 30 degrees C, pH 4-5, in batch culture using Saccharomyces sp. H0 which was constructed by hybridization in this laboratory and the glucoamylase preparation produced on a large scale in China. The optimal conditions for high concentration ethanol production were that the amount of the glucoamylase preparation was 300 u/g of raw ground corn, the inoculum size of the yeast was 2.7 x 10(6) cells/ml, the fermentation temperature was 30 degrees C, the starting concentration of raw ground corn was 33.0% (w/v), and the fermentation period was 70 hr. At the end of fermentation, 17.5% (v/v) ethanol was yielded, with 0.19% reducing sugar, 3.5% total sugar and 16.0% cell viability remaining in the fermented mash. In the case of 36.0% (w/v) of the initial raw ground corn concentration, the strain could produce 18.0% (v/v) ethanol, leaving 0.81% reducing sugar and 5.1% total sugar in the media. We found that the glucoamylase preparation which is usually employed for saccharification of cooked starch could be used for efficient saccharification of raw ground corn starch.

The glycaemic index of spaghetti and gastric emptying in non-insulin-dependent diabetic patients.

OBJECTIVE: In the dietary treatment of non-insulin-dependent diabetes mellitus (NIDDM) great interest has been focused on foods with low glycaemic indices. Spaghetti has a low glycaemic index, but in NIDDM the underlying mechanism has not been elucidated. We investigated whether the low glycaemic index in spaghetti was the result of a retarded gastric emptying. DESIGN: Randomized study. SETTING: Department of Endocrinology & Metabolism, Aarhus County Hospital. SUBJECTS: Eight NIDDM in-patients participated without drop-outs. INTERVENTIONS: Paracetamol was used as a marker of the gastric emptying. On three different occasions the patients ingested 40 g of carbohydrate as: (1) spaghetti without paracetamol, (2) spaghetti containing 1.5 g paracetamol, and (3) mashed potatoes with 1.5 g paracetamol added. During the 4 h observation periods blood samples were drawn frequently. The spaghettis were industrially manufactured. RESULTS: The spaghetti meals induced similar glucose and insulin responses. The potato meal induced significantly higher glucose and paracetamol areas [607 +/- 108 vs 284 +/- 54 mmol/l*240 min (P = 0.02), and 18,668 +/- 999 vs 4979 +/- 369 mumol/l*240 min (P < 0.02)] as well as lower emptying index (time(peak)/peak concentration ratio of paracetamol) [2.1 +/- 0.3 vs 0.4 +/- 0.1 (P = 0.001)], as compared with the spaghetti meal. CONCLUSIONS: This study shows that the ingestion of spaghetti was associated with a slower gastrointestinal accessibility of the carbohydrates, which accounts for the low glycaemic index of spaghetti in NIDDM subjects.
