Aldol additions of dihydroxyacetone phosphate to N-Cbz-amino aldehydes catalyzed by L-fuculose-1-phosphate aldolase in emulsion systems: inversion of stereoselectivity as a function of the acceptor aldehyde.

The potential of L-fuculose-1-phosphate aldolase (FucA) as a catalyst for the asymmetric aldol addition of dihydroxyacetone phosphate (DHAP) to N-protected amino aldehydes has been investigated. First, the reaction was studied in both emulsion systems and conventional dimethylformamide (DMF)/H2O (1:4 v/v) mixtures. At 100 mM DHAP, compared with the reactions in the DMF/H2O (1:4) mixture, the use of emulsion systems led to two- to three-fold improvements in the conversions of the FucA-catalyzed reactions. The N-protected aminopolyols thus obtained were converted to iminocyclitols by reductive amination with Pd/C. This reaction was highly diastereoselective with the exception of the reaction of the aldol adduct formed from (S)-N-Cbz-alaninal, which gave a 55:45 mixture of both epimers. From the stereochemical analysis of the resulting iminocyclitols, it was concluded that the stereoselectivity of the FucA-catalyzed reaction depended upon the structure of the N-Cbz-amino aldehyde acceptor. Whereas the enzymatic aldol reaction with both enantiomers of N-Cbz-alaninal exclusively gave the expected 3R,4R configuration, the stereochemistry at the C-4 position of the major aldol adducts produced in the reactions with N-Cbz-glycinal and N-Cbz-3-aminopropanal was inverted to the 3R,4S configuration. The study of the FucA-catalyzed addition of DHAP to phenylacetaldehyde and benzyloxyacetaldehyde revealed that the 4R product was kinetically favored, but rapidly disappeared in favor of the 4S diastereoisomer. Computational models were generated for the situations before and after C-C bond formation in the active site of FucA. Moreover, the lowest-energy conformations of each pair of the resulting epimeric adducts were determined. The data show that the products with a 3R,4S configuration were thermodynamically more stable and, therefore, the major products formed, in agreement with the experimental results.

Competition between superexchange-mediated and sequential electron transfer in a bridged donor-acceptor system.

The temperature- and solvent-dependence of photoinduced electron-transfer reactions in a porphyrin-based donor-bridge-acceptor (DBA) system is studied by fluorescence and transient absorption spectroscopy. Two competing processes occur: sequential and direct superexchange-mediated electron transfer. In a weakly polar solvent (2-methyltetrahydrofuran), only direct electron transfer from the excited donor to the appended acceptor is observed, and this process has weak temperature dependence. In polar solvents (butyronitrile and dimethylformamide), both processes are observed and the sequential electron transfer shows strong temperature dependence. In systems where both electron transfer processes are observed, the long-range superexchange-mediated process is more than two times faster than the sequential process, even though the donor-acceptor distance is significantly larger in the former case.

Electronic structural changes between nickel(II)-semiquinonato and nickel(III)-catecholato states driven by chemical and physical perturbation.

The selective synthesis of tetracoordinate square-planar low-spin nickel(II)-semiquinonato (Ni(II)-SQ) and nickel(III)-catecholato (Ni(III)-Cat) complexes, 1 and 2, respectively, was achieved by using bidentate ligands with modulated nitrogen-donor ability to the nickel ion. The electronic structures of 1 and 2 were revealed by XPS and EPR measurements. The absorption spectra of 1 and 2 in a noncoordinating solvent, dichloromethane (CH2Cl2), are completely different from those in tetrahydrofuran (THF), being a coordinating solvent. As expected from this result, the gradual addition of N,N-dimethylformamide (DMF), which is also a coordinating solvent like THF, into a solution of 1 or 2 in CH2Cl2 leads to color changes from blue (for 1) and brown (for 2) to light green, which is the same color observed for solutions of 1 or 2 in THF. Furthermore, the same color changes are induced by varying the temperature. Such spectral changes are attributable to the transformation from square-planar low-spin Ni(II)-SQ and Ni(III)-Cat complexes to octahedral high-spin Ni(II)-SQ ones, caused by the coordination of two solvent molecules to the nickel ion.

Microporous metal-organic framework constructed from heptanuclear zinc carboxylate secondary building units.

A novel, three-dimensional, noninterpenetrating microporous metal-organic framework (MOF), [Zn7O2(pda)5(H2O)2]5 DMF4 EtOH 6 H2O (1) (H2PDA=p-phenylenediacrylic acid, DMF=N,N-dimethylformamide, EtOH=ethanol), was synthesized by constructing heptanuclear zinc carboxylate secondary building units (SBUs) and by using rigid and linear aromatic carboxylate ligands, PDA. The X-ray crystallographic data reveals that the seven zinc centers of 1 are held together with ten carboxylate groups of the PDA ligands and four water molecules to form a heptametallic SBU, Zn7O4(CO2)10, with dimensions of 9.8 x 9.8 x 13.8 A3. Furthermore, the heptametallic SBUs are interconnected by PDA acting as linkers, thereby generating an extended network with a three-dimensional, noninterpenetrating, intersecting large-channel system with spacing of about 17.3 A. As a microporous framework, polymer 1 shows adsorption behavior that is favorable towards H2O and CH3OH, and substantial H2 uptake. In terms of the heptanuclear zinc carboxylate SBUs, polymer 1 exhibits interesting photoelectronic properties, which would facilitate the exploration of new types of semiconducting materials, especially among MOFs containing multinuclear metal carboxylate SBUs.

Construction of open metal-organic frameworks based on predesigned carboxylate isomers: from achiral to chiral nets.

The four-connected carboxylate ligand N,N,N',N'-tetrakis(4-carboxyphenyl)-1,4-phenylenediamine (TCPPDA) exists as three stereoisomers: a pair of enantiomers (deltaD2- and lambdaD2-TCPPDA) and a diastereomer (C2h-TCPPDA). TCPPDA was predesigned for the construction of isomeric coordination networks. Reactions of M(NO3)2 (M=Cu, Zn, Co) or Nd(NO)3 with TCPPDA under solvothermal conditions gave rise to five novel porous metal-organic frameworks: [Cu2(D2-tcppda)(H2O)2].2 DMSO.6H2O (1), [Cu2(C2h-tcppda)(H2O)2].2DMSO.6H2O (2), [Co3(D2-Htcppda)2].4DEF.5H2O (3), [Nd2(D2-tcppda)(C2h-tcppda)0.5(DMSO)3]3 DMSO5 H2O (4), and [Zn4O(D2-tcppda)1.5].DMF.H2O (5) (DMSO=dimethyl sulfoxide, DEF=diethylformamide, DMF=dimethylformamide). Complexes 1 and 2 are supramolecular isomers, in which all the ligands adopt pseudotetrahedral (both deltaD2- and lambdaD2-TCPPDA) and rectangular (C2h-TCPPDA) geometries, respectively. Both compounds connect paddlewheel secondary building units (SBUs) to form three-dimensional porous networks possessing PtS and NbO nets, respectively. In 3, all ligands possess pseudotetrahedral (both deltaD2- and lambdaD2-TCPPDA) geometry and link hourglass SBUs to form a three-dimensional porous framework. Compound 4 contains all three stereoisomers (C2-, deltaD2-, and lambdaD2-TCPPDA), thus, has both pseudotetrahedral and rectangular geometries. D2-TCPPDA connects the binuclear neodymium units to generate a two-dimensional layer, further linked by C2h-TCPPDA to create a three-dimensional open framework. In 5, all the ligands possess pseudotetrahedral geometry (D2-TCPPDA), as found in 1 and 3. However, all the TCPPDA ligands in 5 appear as either the deltaD2 or the lambdaD2 form, thus, the whole structure is homochiral. Complex 5 crystallizes in the I4(1)32 space group and the octahedral SBU in 5 is connected by the enantiopure TCPPDA to generate a three-dimensional porous network possessing the corundum Al2O3 net. Complexes 1, 2, and 5 possess permanent porosity, and 4 and 5 exhibit strong luminescence at lambdamax=423 and 424 nm, respectively, upon excitation at 268.5 nm.

Electrochemical reduction of G3-factor endoperoxide and its methyl ether: evidence for a competition between concerted and stepwise dissociative electron transfer.

The reduction of the bicyclic G-factor endoperoxides G3 and G3Me was studied in N,N-dimethylformamide using cyclic voltammetry and convolution analysis. Electron transfer leads to irreversible cleavage of the O--O bond. Detailed analysis of the voltammetry curves reveals a non-linear dependence on the transfer coefficient indicating a mechanistic transition from a stepwise mechanism to one with more concerted character with increasing potential. By using quantum calculations to estimate the O--O bond dissociation energies, the experimental data was used to evaluate the standard reduction potentials and other pertinent thermochemical information.

Iridium(III) silyl alkyl and iridacyclic compounds supported by 4,4'-di-tert-butyl-2,2'-bipyridyl.

Treatment of IrCl(3)x H(2)O with one equivalent of 4,4'-di-tert-butyl-2,2'-bipyridyl (dtbpy) in N,N-dimethylformamide (dmf) afforded [IrCl(3)(dmf)(dtbpy)] (1). Alkylation of 1 with Me(3)SiCH(2)MgCl resulted in C--Si cleavage of the Me(3)SiCH(2) group and formation of the Ir(III) silyl dialkyl compound [Ir(CH(2)SiMe(3))(dtbpy)(Me)(SiMe(3))] (2), which reacted with tBuNC to afford [Ir(tBuNC)(CH(2)SiMe(3))(dtbpy)(Me)(SiMe(3))] ([2(tBuNC)]). Reaction of 2 with phenylacetylene afforded dimeric [{Ir(C[triple chemical bond]CPh)(dtbpy)(SiMe(3))}(2)(mu-C[triple chemical bond]CPh)(2)] (3), in which the bridging PhC[triple chemical bond]C(-) ligands are bound to Ir in a mu-sigma:pi fashion. Alkylation of 1 with PhMe(2)CCH(2)MgCl afforded the cyclometalated compound [Ir(dtbpy)(CH(2)CMe(2)C(6)H(4))(2-C(6)H(4)CMe(3))] (4), which features an agostic interaction between the Ir center and the 2-tert-butylphenyl ligand. The cyclic voltammogram of 4 in CH(2)Cl(2) shows a reversible Ir(IV)-Ir(III) couple at about 0.02 V versus ferrocenium/ferrocene. Oxidation of 4 in CH(2)Cl(2) with silver triflate afforded an Ir(IV) species that exhibits an anisotropic electron paramagnetic resonance (EPR) signal in CH(2)Cl(2) glass at 4 K with g( parallel)=2.430 and g( perpendicular)=2.110. Protonation of 4 with HCl and p-toluenesulfonic acid (HOTs) afforded [{Ir(dtbpy)(CH(2)CMe(2)Ph)Cl}(2)(mu-Cl)(2)] (5) and [Ir(dtbpy)(CH(2)CMe(2)Ph)(OTs)(2)] (6), respectively. Reaction of 5 with Li[BEt(3)H] gave the cyclometalated complex [{Ir(dtbpy)(CH(2)CMe(2)C(6)H(4))}(2)(mu-Cl)(2)] (7). Reaction of 4 with tetracyanoethylene in refluxing toluene resulted in electrophilic substitution of the iridacycle by C(2)(CN)(3) with formation of [Ir(dtbpy)(CH(2)CMe(2)C(6)H(3){4-C(2)(CN)(3)})(2-C(6)H(4)CMe(3))] (8). Reaction of 4 with diethyl maleate in refluxing toluene gave the iridafuran compound [Ir(dtbpy)(CH(2)CMe(2)C(6)H(4)){kappa(2)(C,O)-C(CO(2)Et)CH(CO(2)Et)}] (9). Treatment of 9 with 2,6-dimethylphenyl isocyanide (xylNC) led to cleavage of the iridafuran ring and formation of [Ir(dtbpy)(CH(2)CMe(2)C(6)H(4)){C(CO(2)Et)CH(CO(2)Et)}(xylNC)] (10). Protonation of 9 with HBF(4) afforded the dinuclear neophyl complex [(Ir(dtbpy)(CH(2)CMe(2)Ph){kappa(2)(C,O)-C(CO(2)Et)CH(CO(2)Et)})(2)][BF(4)](2) (11). The solid-state structures of complexes 2-5 and 8-11 have been determined.

Recrystallization and shape control of crystals of the organic dye acid green 27 in a mixed solvent.

Recrystallization of the unstructured dye acid green 27 (AG27) in a mixed solvent of alcohol (ethanol or methanol) and water was systematically studied. The results demonstrated that AG27 crystals with uniform sizes and controllable shapes can be produced by simply changing the volume ratio of ethanol (or methanol) and deionized water (DIW). Rodlike and shuttlelike AG27 crystals can be selectively synthesized. The XRD analyses revealed the periodic structures of the organic crystals. Furthermore, crystallization in another mixed solvent of N,N-dimethylformamide (DMF) and DIW results in the formation of longer fibers with high aspect ratio, which further validates the remarkable effects of mixed solvent on the shape of the AG27 crystals. This method of recrystallization in a mixed solvent is expected to facilitate the synthesis of other functional organic crystals with unusual shapes.

Electrochemical, pulsed-field-gradient spin-echo NMR spectroscopic, and ESR spectroscopic study of the diffusivity of molecular probes inside gel-type cross-linked polymers.

The permeability of five gel-type synthetic resins, obtained by polymerization of 1-vinylpyrrolidin-2-one cross-linked with N,N'-methylenebisacrylamide (1, 2, 3, 4, and 5 wt %) and swollen by N,N-dimethylformamide (DMF), has been analyzed. The diffusion of 2,2,6,6-tetramethyl-4-oxo-1-piperidinyloxyl (TEMPONE) was studied by ultramicroelectrode voltammetry. Similar measurements were performed for solutions of non-cross-linked poly(vinylpyrrolidone) in DMF. To provide information on the rotational mobility of TEMPONE and the translational mobility of DMF, electron spin resonance (ESR) spectroscopic and pulsed-field-gradient spin-echo nuclear magnetic resonance (PGSE-NMR) spectroscopic experiments, respectively, were carried out. Comparative analysis of the results obtained by electrochemical, ESR spectroscopic, and PGSE-NMR spectroscopic measurements showed that diffusivity inside the polymer framework is significantly affected by the extent of cross-linking, the size of the diffusing probe, and the presence of electrolytes.

[Bromination of 3-amino-1,2,4-benzothiadiazine-1,1-dioxide].

The bromination of 3-amino-1,2,4-benzothiadiazine-1,1-dioxide (1) in N,N-dimethylformamide has been studied. Depending on the operating conditions the reaction produces 7-bromo-3-amino-1,2,4-benzothiadiazine-1,1-dioxide (II) or 5,7-diabromo-3-amino-1,2,4-benzothiadiazine-1,1-dioxide (III). Derivatives halogenated at the nitrogen were not isolated in any of the experiments.

5-aryl-imidazo[2,1-c][1,4]benzodiazepine derivatives as tricyclic constrained analogues of diazepam and Ro5-4864. Synthesis and binding properties at peripheral and central benzodiazepine receptors.

Four series of 5-aryl-imidazo[2,-c][1,4]benzodiazepine derivatives 1a-f, 2a-f, 3a-f, and 4a-f were synthesized and tested for their affinity at both the peripheral and central benzodiazepine receptors. Among the four series, only N-10 and C-11 sites were changed, mainly [N(CH3)-CO], [N=CH], [NH-CO], [NH-CH2], and in each series the halogen site was varied at the positions C-7, C-2', and C-4'. In particular, 10-methyl-benzodiazepinones 1a and 1b were designed as tricyclic constrained analogues of diazepam and Ro5-4864. All the tested compounds did not show significant binding activity at central benzodiazepine receptors, but relatively good PBzR binding affinities were found for 10-methyl-benzodiazepinone 1c and benzodiazepines 2b, c. Benzodiazepinones 3a-f were prepared by cyclization with 1,1'-carbonyldiimidazole of the corresponding 2-(aryl-imidazol-1-yl-methyl)-arylamines, obtained from the suitable (2-amino-aryl)-aryl-methanols with 1,1'-carbonyldiimidazole in different conditions. N-Alkylation of 3a-f to 1a-f was achieved using dimethylformamide-dimethylacetal. Reduction of 3a-f to 4a-f was accomplished with lithium aluminum hydride or borane and oxidation of 4a-f to 2a-f was performed with manganese (IV) oxide.

On the dissociation of aromatic radical anions in solution.

A new theoretical formulation is given for the reaction rate and path for the important reaction class of aromatic radical anion dissociation in solution [Ar-X]-.-->Ar. + X-, and is illustrated for the case of the cyanochlorobenzene radical anion [CN-phi-CI]-. in dimethylformamide. Among the theory's novel features is the inclusion of the conical intersection aspect of this ground electronic state problem, which is key in allowing the reaction to occur and which has a significant impact on the reaction barrier height. Reasonable agreement with the experimental rate is found.

Immunochemical detection of peptides and proteins on press-blots after direct tissue gel isoelectric focusing.

A sensitive method is described for the detection of tissue peptides and proteins. They are separated by tissue isoelectric focusing using thin large-pore polyacrylamide gels, containing detergent and dimethylformamide, and are fixed with either glutaraldehyde or formaldehyde in gelatin-coated nitrocellulose membranes using press-blotting. The fixed peptide and protein antigens are visualized by immunoperoxidase staining. The spectrum of fixed tissue constituents may also be used to test antiserum reactivity and specificity in immunocytochemical staining procedures. Isoelectric focusing of 2 microL homogenates of the neurointermediate lobe of the pituitary allowed the immunodetection of peptides and proteins of various sizes and the determination of isoelectric points. However, direct application onto gels of small pieces of frozen tissue sections, sliced in a cryostat, appeared to be more efficient. By direct tissue isoelectric focusing of brain tissue, peptides were effectively eluted and separated from sections up to 100 microns thickness. This allowed the detection of small peptides with a detection limit of approximately 10 pg/section.

On the efficiency of methylene blue versus persulfate catalysis of polyacrylamide gels, as investigated by capillary zone electrophoresis.

The efficiency of a novel method of photopolymerization, based on photoinitiating the reaction with methylene blue (MB), in presence of a redox couple (sodium toluenesulfinate and diphenyliodonium chloride), vs, the conventional persulfate--N,N,N',N'-tetramethylethylenediamine redox couple was investigated as a function of different effectors in solution. Oxygen dissolved in the gelling mixture strongly quenches persulfate catalysis, while leaving essentially unaltered the process initiated by photopolymerization. On the contrary, the presence of 8 M urea substantially accelerates a persulfate-driven reaction, boosting the conversion of monomers to near completion (> 98%) while leaving the photopolymerization process largely unaffected. Polyacrylamide polymerization has also been performed in a number of hydroorganic solvents (all in a 50:50 v/v ratio): dimethyl sulfoxide (DMSO), tetramethylurea, formamide and dimethylformamide. In all cases, the persulfate-catalyzed reaction was strongly quenched and even completely inhibited (in DMSO), whereas the photopolymerization process was essentially unaffected by any of these organic solvents. The reaction kinetics of the methylene blue-driven reaction could not be ameliorated when admixing an anionic dye (e.g., eosin Y) to the cationic MB, even when amply changing their molar ratios. Thus, it appears that photocatalysis with MB (and the redox couple sodium toluene-sulfinate and diphenyliodonium chloride) is a unique process, proceeding at optimum rate under the most adverse conditions, completely insensitive to any kind of positive and negative effectors and able to ensure at least 95% monomer conversion under the standard conditions of 1 h reaction time at room temperature.(ABSTRACT TRUNCATED AT 250 WORDS)

Separation of (Z)- and (E)-isomers of thioxanthene and dibenz[b,e]oxepin derivatives with calixarenes and resorcinarenes as additives in nonaqueous capillary electrophoresis.

Five acidic calix[4]arenes with carboxylic or sulfonic groups at either the upper or lower rim of the cavity and one resorc[4]arene were investigated to separate three thioxanthenes (flupentixol, clopenthixol, chlorprothixene) and a dibenz[b,e]oxepin derivative (doxepin) with cis-/trans-isomerism by nonaqueous capillary electrophoresis (NACE). Partial filling of the capillary with the UV-absorbing selectors led to a low detection limit and an advantageous signal-to-noise ratio (S/N). A sufficient electrophoretic mobility of the calixarenes towards the anode was necessary to outweigh the oppositely directed electroosmotic flow (EOF). This depended from the functional groups, the dissociation and the hydrodynamic radius of the cyclophanes. In contrast, the resorcinarene was useable only by addition of sodium dodecyl sulfate (SDS) because only the complex of the two selectors had an anodic apparent electrophoretic mobility. p-Sulfonyl-calix[4]arene (ss-a1) was the most capable selector for all E/Z-isomers with maximal alpha-values ranging from 1.056 for doxepin to 1.224 for chlorprothixene. This was due to the sufficient migration in reversed direction to the EOF even at low pH* values of 3.0. Otherwise, electrostatic as well as hydrophobic interactions with the positively charged isomers seem to contribute to a superior recognition. Increasing the concentration up to 15 mM ss-a1 and using acidic media (pH* 5.0) led to high separation efficiency. Changing the organic solvent provides a powerful tool to improve selectivity with N,N-dimethylformamide-methanol (DMF-MeOH)-mixtures for thioxanthenes. Further electrophoretic parameters were optimized, such as the concentration of the electrolytes, the addition of SDS, the kind of electrolytes and the voltage. Distinct differences in selectivities were found between the derivatives with thioxanthene and dibenzo[b,e]oxepin ring system, respectively. Further, the different basic side chain was responsible for the different selectivity at higher pH* values. In contrast, the substitution at position 2 of the thioxanthenes played a secondary role. Based on the studies of single parameters a method for a simultaneous separation of the four pairs of isomers within 13 min was developed.

Absence of formation of benzo[a]pyrene/DNA adducts in the cuttlefish (Sepia officinalis, Mollusca: Cephalopoda).

Benzo[a]pyrene (B[a]P) injected intramuscularly into the base of the arms of cuttlefish was released continuously from the injection site and removed from the organism. Only a portion of the compound accumulated in the body. Twenty-four hr after its injection, 75% of B[a]P applied in olive oil was removed from the cuttlefish, and 1.2% was found in the body outside the head, the site of injection. If the carcinogen was dissolved in dimethylformamide, the removal of B[a]P was slower, so that only 18% of the injected B[a]P was removed from the organism and 0.36% accumulated in the body outside the head 24 hr after injection. The high level of B[a]P in gills and hemolymph 4 hr after injection and the kinetics of the decrease of its concentration with time indicate that these two organs could be involved in the excretion of B[a]P from the body. The B[a]P/DNA adducts characteristic for vertebrates could not be demonstrated in gills, skin, brain, hepatopancreas, and lymphocytes of the cuttlefish 24 hr after injection of B[a]P. The dose of the carcinogen injected into the cuttlefish was 2-4 times higher than the dose resulting in the formation of a high level of B[a]P/DNA adducts in the vertebrates. A different metabolism of B[a]P in the tissue of cephalopods, compared to vertebrates, could be less favorable to the process leading to malignant transformation and could explain the absence from the literature of reports of tumors in cephalopods.

Preclinical chemotherapy on human head and neck cancer xenografts grown in athymic nude mice.

This study was undertaken to investigate the potential role of xenografts established from human head and neck squamous cell carcinoma (HNSCC) in the selection of new anticancer agents for phase II clinical trials. Eight HNSCC tumor lines were established in NMRI nude mice. The tumor-bearing animals were then treated with drugs at the maximum tolerated dose level. Treatment with drugs known for their activity in 15%-30% of HNSCC patients [cisplatin (CDDP), bleomycin (BLEO), 5-fluorouracil (5-Fu), cyclophosphamide (CY), and doxorubicin (DOX)] caused strong responses in up to 38% and moderate responses in 50%-67% of the HNSCC tumor lines. Methotrexate (MTX), known to cause remissions in about 40% of HNSCC patients, was only minimally active in this model system. A clinically ineffective drug, amsacrine (m-AMSA), was included as a negative control and showed no or minimal activity in all four HNSCC lines tested. A number of experimental drugs that have promising preclinical activity were also tested. Brequinar sodium (Dup 785) and 10-ethyl, 10-deaza-aminopterin (10-EdAM) showed activity in three of five, and two of the four tested tumor lines respectively. N,N-dimethylformamide (DMF) and 5-aza-2'-deoxycytidine (5-aza-dCyd), agents with the capacity to induce differentiation in in vitro systems, showed moderate activity in 43% and 40%, and strong activity in 14% and 40% of the lines, respectively. Our results indicate that the nude mouse xenograft model may play a role in the screening of new drugs, and in particular, it could be of help in the selection of drugs to be tested in phase II HNSCC clinical trials.

Enhancement of lung-colonizing ability of cloned low-metastatic Lewis lung carcinoma cells by treatment with highly polar compounds.

Treatment of low-metastatic Lewis lung carcinoma cells (P-29) with dimethylsulfoxide (DMSO) in vitro enhanced their lung-colonizing ability. The effects of other highly polar compounds on the lung-colonizing ability of P-29 cells were examined. The following compounds were found to enhance the lung-colonizing ability of the cells: acetamide, N-methylacetamide, N-methylformamide, N,N-dimethylformamide, piperidone and hexamethylphosphoric triamide. Treatment of P-29 cells with DMSO or other polar compounds resulted in increases in activities of degradative enzymes, such as cathepsin B and plasminogen activator. The increases in cathepsin B and plasminogen activator activities were apparent after a 24 h treatment with DMSO and were suppressed by simultaneous treatment with cycloheximide, which suggested that they were due to syntheses of new proteins. DMSO-treated P-29 cells degraded [3H]leucine-labelled subendothelial matrix much more than did untreated cells. P-29 cells treated with DMSO or other polar compounds became attached to culture dishes more rapidly and were more resistant to detachment by trypsin treatment than untreated cells. A significant correlation was found between the degrees of adhesiveness of P-29 cells treated with various polar compounds and their lung-colonizing abilities.
