Retinoblastoma protein is frequently absent or phosphorylated in anaplastic large-cell lymphoma.

The possible role of retinoblastoma protein (Rb) in the pathogenesis of anaplastic large-cell lymphoma (ALCL) is unknown. We investigated Rb protein expression, both total (phosphorylated and underphosphorylated) and active (underphosphorylated), in four anaplastic lymphoma kinase (ALK)-positive ALCL cell lines (Karpas 299, JB-6, SU-DHL1, and SR-786) by Western blot analysis, and in 67 ALCL tumors (30 ALK-positive, 37 ALK-negative) using immunohistochemical methods. We also used fluorescence in situ hybridization and polymerase chain reaction methods to assess for loss of heterozygosity of the rb gene. The findings were correlated with apoptotic rate assessed by the terminal dUTP nick-end labeling assay. Immunoblots showed high total Rb levels in Karpas 299, SU-DHL1 and SR-786 and relatively lower levels in and JB-6. Underphosphorylated Rb was negative or expressed at low levels in all cell lines. In ALCL tumors, total Rb was detected in 44 (66%) and absent in 23 (34%). The mean apoptotic rate was 3.2% in Rb-negative tumors compared with 2.7%, 2.2%, and 1.2% in tumors with <10%, 10 to 50%, and >50% Rb-positive cells, respectively (P = 0.2, Kruskall-Wallis test). In a subset of 25 total Rb-positive tumors we assessed for underphosphorylated Rb, which was detected in 12 tumors. The detection of only total Rb in the remaining 13 tumors suggests that Rb was phosphorylated. Fluorescence in situ hybridization showed allelic loss of the rb gene in 10 (40%) of 25 tumors analyzed and was significantly associated with absence of Rb expression (P = 0.003). Similar results were obtained for loss of heterozygosity of the 13q14 locus. Five-year progression-free survival for patients with Rb-negative ALCL was 89.4% compared with 47.7% for patients with total Rb-positive ALCL (P = 0.006, log-rank test). Similar trends for progression-free survival held true for patients with ALK-positive and ALK-negative tumors analyzed separately. In conclusion, Rb is absent or phosphorylated in most ALCL cell lines and tumors and absence of Rb expression is associated with better clinical outcome in patients with ALCL.

Matrix metalloproteinase-9-dependent exposure of a cryptic migratory control site in collagen is required before retinal angiogenesis.

Retinal neovascularization is a leading cause of human blindness. However, little is known concerning the molecular mechanisms controlling retinal neovascularization in vivo. Here we provide evidence that exposure of a collagen type IV cryptic epitope detected by monoclonal antibody (mAb) HUIV26, delineates sites of vascular bud formation and represents one of the earliest structural remodeling events required before vessel out-growth. Exposure of these cryptic sites was inhibited in matrix metalloproteinase (MMP)-9-deficient but not MMP-2-deficient mice implicating MMP-9 in their exposure. Retinal endothelial cell interactions with the HUIV26 epitopes induced endothelial cell migration, which was blocked by mAb HUIV26. Importantly, subcutaneous administration of mAb HUIV26 potently inhibited retinal angiogenesis in vivo. Taken together, these findings suggest a novel mechanism in which MMP-9 facilitates exposure of HUIV26 cryptic sites, thereby promoting retinal endothelial cell migration and neovascularization in vivo.

Biological and dietary antioxidants protect against DNA nitration induced by reaction of hypochlorous acid with nitrite.

Nitryl chloride, formed by reaction of hypochlorous acid with nitrite, might contribute to nitrative damage of biomolecules in addition to peroxynitrite. Damage of DNA by these reactive nitrogen oxide species is implicated in carcinogenesis associated with chronic infections and inflammation. Nitrated DNA adducts, such as 8-nitroguanine and 8-nitroxanthine, are not stable in DNA since they undergo spontaneous depurination, leading to apurinic site formation. In this report, we investigate the protective effect of biological and dietary antioxidants in inhibiting DNA nitration induced by nitryl chloride. The effect of inhibition was evaluated by decrease of 8-nitroxanthine and 8-nitroguanine formation. Among the 21 compounds examined, dihydrolipoic acid is the most effective in preventing DNA nitration, followed by N-acetyl-L-cysteine and folic acid. For sulfur-containing compounds, the more highly reduced compounds are stronger inhibitors of DNA nitration. The major product of N-acetyl-L-cysteine reaction with nitryl chloride is characterized as the (R)-2-acetylamino-3-sulfopropionic acid, a physiologically irreversible product, suggesting that nitryl chloride is a strong oxidizing agent.

Correlations between functional and occlusal tooth-surface areas and food texture during natural chewing sequences in humans.

The dental-arch surfaces preferentially used in mastication were studied by measuring functional and occlusal surface areas and comparing these to the number of chews required to swallow foods of different texture properties. The functional surface of the teeth was defined as the total area of visible wear facets on post-incisal teeth, adding to it the contacting areas of restored teeth where no facets were visible. Occlusal surface area was taken as the total area of the occluding parts of post-incisal teeth. Both surfaces were measured with computer image processing on dental-stone casts of the teeth of 31 young adults. Functional surface areas (mean 168 mm(2), four quadrants) were positively correlated with occlusal surface areas (mean 739 mm(2), four quadrants). The left:right area ratios were more variable for functional than for occlusal surfaces. Functional surface-area ratios markedly different from 1.0 might reflect functional side-preponderance of masticatory activity. Correlations between tooth surface area and the number of cycles were examined with five different food samples of known texture during side-imposed mastication. Depending on the elastic moduli of the foods, significant negative correlations were found between the left:right ratios of functional or occlusal surface areas and the left:right ratios of cycle numbers. The rheological properties of the food particles chewed were assumed to be the key factor in the correlations with either the functional or anatomical occlusal surfaces.

Structural and functional characterization of the GalNAc/Gal-specific lectin from the phytopathogenic ascomycete Sclerotinia sclerotiorum (Lib.) de Bary.

The lectin found in mycelium and sclerotes of the phytopathogenic fungus Sclerotinia sclerotiorum is a homodimer consisting of two identical non-covalently bound subunits of 16,000 Da. CD spectra analysis revealed that the S. sclerotiorum agglutinin (SSA) contains predominantly beta-sheet structures. SSA exhibits specificity towards GalNAc whereby the hydroxyls at positions 4 and 6 of the pyranose ring play a key role in the interaction with simple sugars. The carbohydrate-binding site of SSA can also accommodate disaccharides. The N-terminal sequence of SSA shares no significant similarity with any other protein except a lectin from the Sclerotiniaceae species Ciborinia camelliae. A comparison of SSA and the lectins from C. camelliae and some previously characterized lectins indicates that the Sclerotiniaceae lectins form a homogeneous family of fungal lectins. This newly identified lectin family, which is structurally unrelated to any other family of fungal lectins, is most probably confined to the Ascomycota.

Neuronal excitability and conduction velocity changes in hippocampal slices from streptozotocin-treated diabetic rats.

The effects of streptozotocin-induced diabetes on the electrophysiological properties of central neurones were investigated in rat hippocampal slices. No differences in membrane potential, input resistance or action potential parameters could be detected in pyramidal or granule cells. However, a slowing of conduction velocity in granule cells and an increase in excitability of fibre volleys in both the perforant pathway and granule cell mossy fibre projections was observed. The lack of effect on evoked fEPSPs in these pathways is also consistent with diabetes-dependent changes in the voltage-dependence of synaptic strength.

An aldosterone synthase gene variant is associated with improvement in left ventricular ejection fraction in dilated cardiomyopathy.

OBJECTIVE: To assess whether renin-angiotensin-aldosterone (RAA) system gene polymorphisms shown to be associated with alterations in the activity of the system, may predict cardiac function changes subsequent to initiating medical therapy in heart failure. METHODS: The impact of RAA system genotypes on left ventricular ejection fraction (LVEF) following therapy to patients with idiopathic dilated cardiomyopathy (IDC) and class II-III heart failure was assessed. In 107 patients LVEF and LV dimensions were determined using radionuclide ventriculography and echocardiography prior to and subsequent to receiving furosemide, digoxin and angiotensin-converting enzyme (ACE) inhibitor therapy. Patients and controls were genotyped for variants of the ACE (insertion-deletion polymorphism), angiotensinogen (AGT; M235T polymorphism) and the aldosterone synthase (CYP11B2, C-344T polymorphism) genes. RESULTS: RAA system genotypes were not significantly associated with LVEF prior to initiating medical therapy. However, the CYP11B2 gene variant (P=0.0064 on covariate analysis [adjusted for multiple genotyping] with a 1-2% chance of false positive data), but neither the ACE, nor the AGT variants, predicted improvement in LV ejection fraction in patients on medical therapy. CONCLUSION: A CYP11B2 gene variant predicts the variable improvement in LV ejection fraction that occurs subsequent to initiating medical therapy in IDC. These data suggest a role for the aldosterone synthase locus in regulating the progression of heart failure.

Lipoyl dehydrogenase catalyzes reduction of nitrated DNA and protein adducts using dihydrolipoic acid or ubiquinol as the cofactor.

Inflamed tissues generate reactive nitrogen oxide species (RNO(x)), such as peroxynitrite (ONOO-)and nitryl chloride (NO2Cl), which lead to formation of nitrated DNA and protein adducts, including 8-nitroguanine (8NG), 8-nitroxanthine (8NX), and 3-nitrotyrosine (3NT). Once formed, the two nitrated DNA adducts are not stable in DNA and undergo spontaneous depurination. Nitration of protein tyrosine leads to inactivation of protein functions and 3NT has been detected in various disease states. We herein report that reduction of these nitro adducts to their corresponding amino analogues can be catalyzed by lipoyl dehydrogenases (EC 1.8.1.4) from Clostridium kluyveri (ck) and from porcine heart (ph) using NAD(P)H as the cofactor. We also found that dihydrolipoic acid (DHLA) and ubiquinol can be used as effective cofactors for reduction of 8NG, 8NX, and 3NT by these lipoyl dehydrogenases. The reduction efficiency of the mammalian enzyme is higher than the bacterial isozyme. The preference of cofactors by both lipoyl dehydrogenases is DHLA>NAD(P)H>ubiquinol. In all the systems examined, the nitrated purines are reduced to a greater extent than 3NT under the same conditions. We also demonstrate that this lipoyl dehydrogenase/antioxidant system is effective in reducing nitrated purine on NO2Cl-treated double stranded calf thymus DNA, and thus decreases apurinic site formation. The nitroreductase activity for lipoyl dehydrogenase might represent a possible metabolic pathway to reverse the process of biological nitration.

A Mars bar is not an adequate fatty meal--a comparison with Calogen.

A prospective study was performed to compare the contractility of the gall bladder ultrasonically after ingestion of a 65 g Mars bar or 60 ml of Calogen. Using a repeated measure design, 27 healthy subjects were studied fasting and at 10 min intervals after ingestion of the fatty meal. Results show that there is a significant difference between Mars bar and Calogen (ANOVA, P < or = 0.01), with Calogen producing greater gall bladder contraction overall, and that the ejection fraction is greatest at 40 min (t = 2.23, P < or = 0.01). It is suggested that 60 ml of Calogen is used as the standard fatty meal to assess gall bladder contractility and that the patient is rescanned 40 min post fatty meal.

Separation and identification of water-soluble food dyes by ion-exchange and soap thin-layer chromatography.

Eighteen water-soluble food dyes have been studied by chromatography on thin layers of anion-exchange (AG 1-X4, DEAE-cellulose, PAB-cellulose and chitosan) and cation-exchange (Dowex 50-X4, Rexyn 102 and humic acid) materials; layers of silanised silica gel impregnated with cationic or anionic detergent were also used. Fourteen of the dyes were separated, but the two orange and the two black dyes were not. Some applications of the techniques to commercial products are reported.

Validated high-performance liquid chromatographic method for quantitation of neohesperidine dihydrochalcone in foodstuffs.

An analytical method to detect and quantitate neohesperidine dihydrochalcone in foodstuffs has been developed and validated in soft-drink applications. The method was shown to be sufficiently precise, accurate, selective and rugged in quantitating neohesperidine DC both at flavouring (1-5 mg/kg) and sweetening (5-50 mg/kg) levels. Applications of the method to determine neohesperidine DC in foodstuffs other than soft-drinks is also described.

Qualitative and quantitative evaluation of mono- and disaccharides in D-fructose, D-glucose and sucrose caramels by gas-liquid chromatography-mass spectrometry. Di-D-fructose dianhydrides as tracers of caramel authenticity.

The monosaccharide (D-fructose, D-glucose, anhydrosugars), disaccharide (glucobioses) and pseudodisaccharide (di-D-fructose dianhydrides) content of D-fructose, D-glucose and sucrose caramels has been determined by gas-liquid chromatography-mass spectrometry (GLC-MS) of their trimethylsilyl (TMS) or TMS-oxime derivatives. The chromatographic profiles revealed significant differences in the disaccharide/pseudodisaccharide distribution depending on the caramel source: a D-fructose caramel contains prominent proportions of di-D-fructose dianhydrides, a D-glucose caramel mainly D-glucobioses, and a sucrose caramel similar proportions of both disaccharide/pseudodisaccharide series. It is noteworthy that di-D-fructose dianhydrides are found in all three types of caramels and might then be used as specific tracers of the authenticity of caramel, i.e., a product resulting from the controlled heat treatment of food-grade carbohydrates for use as food additives.

Breast milk distribution of theobromine from chocolate.

Six nursing mothers ingested 113 gm of Hershey's milk chocolate containing 240 mg of theobromine. Samples of plasma, saliva, and breast milk were assayed for theobromine by high pressure liquid chromatography. Peak theobromine concentrations of 3.7 to 8.2 mg/l were found in all fluids at 2 to 3 hour after ingestion of chocolate. The disposition half-life of theobromine averaged 7.1 +/- 2.1 hours, body clearance was 65 +/- 20 ml/hour/kilogram, and the apparent volume of distribution was 0.62 +/- 0.13 l/lk. Theobromine is only slightly bound to plasma and milk proteins and concentrations in milk and saliva matched plasma data closely. The mean concentration ratios were 0.82 +/- 0.17 for milk/plasma and 0.92 +/- 0.17 for saliva/plasma. If a mother ate a 4-ounce chocolate bar every 6 hours and the infant nursed when the theobromine concentration in milk was at its peak, the infant could ingest about 10 mg of theobromine per day.

A randomised controlled trial of a psycho-educational intervention to aid recovery in infectious mononucleosis.

OBJECTIVES: Glandular fever is associated with an approximate fivefold increase in fatigue at 6 months. Reduced levels of fitness and illness beliefs may be important predictors of fatigue following glandular fever. We therefore developed a brief psycho-educational intervention aimed at improving recovery from infectious mononucleosis, and piloted a randomised controlled trial to evaluate the intervention. METHODS: We performed a randomised-controlled trial in primary health care in Southeast London and Kent. Sixty-nine patients aged between 16 and 45 years who were diagnosed, serologically and clinically, with acute infectious mononucleosis between December 1999 and December 2000 were randomised. The control group received a standardised fact-sheet about infectious mononucleosis, which gave no advice on rehabilitation. Patients who were randomised to the intervention received an individual treatment session, two follow-up telephone calls, and an information booklet. Fatigue score 6 months after the onset of infectious mononucleosis was the main outcome measure. RESULTS: Sixty-nine out of 139 patients referred were recruited and randomised. Eighty-seven percent of those recruited completed the Fatigue Questionnaire at 6 months. The intervention was acceptable to all who received it. There were fewer fatigue cases in the intervention group than the control group at 6 months follow-up (odds ratio 0.31, 95% confidence interval 0.09-0.91). CONCLUSIONS: A brief intervention at the diagnosis of infectious mononucleosis is acceptable, and may help prevent the development of chronic fatigue. Definitive randomised controlled trials are required to test the intervention.

CNQX increases GABA-mediated synaptic transmission in the cerebellum by an AMPA/kainate receptor-independent mechanism.

GABA(A) receptor-mediated inhibitory synaptic transmission within the CNS is often studied in the presence of glutamate receptor antagonists. However, for nearly a decade it has been known that, in the hippocampus, one of the most commonly used alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA)/kainate receptor antagonists, 6-cyano-7-nitroquinoxaline-2,3-dione (CNQX), can increase the frequency of spontaneous GABA(A) receptor-mediated postsynaptic currents (sIPSCs). In the present study we examined the effect of CNQX and related compounds on GABA-mediated synaptic transmission in the cerebellum. At various stages of development, low concentrations of CNQX increased the frequency of sIPSCs recorded from granule cells. This effect was independent of the blocking action of CNQX on ionotropic glutamate receptors, as it was not observed with the broad-spectrum glutamate receptor antagonist kynurenate. No increase in sIPSC frequency was observed with the NMDA receptor antagonists D-AP5 or 7-ClK, the selective AMPA receptor antagonists GYKI 52466 or GYKI 53655, or the kainate receptor antagonist NS-102. In contrast, two other quinoxaline derivatives, NBQX and DNQX, were capable of increasing sIPSC frequency. These results demonstrate that the novel excitatory action of CNQX, unrelated to blockade of ionotropic glutamate receptors, is not restricted to the hippocampus and can be observed with structurally related compounds.
