Decline in CD4+ cell numbers in cats with naturally acquired feline immunodeficiency virus infection.

T-cell subsets were studied by fluorescence-activated cell sorter analysis in 57 feline immunodeficiency virus (FIV)-seropositive cats with naturally acquired FIV infection to see whether CD4(+)-CD8+ alterations were comparable to those observed in human immunodeficiency virus-infected patients. CD4+ values were decreased and CD8+ values were increased. The CD4+/CD8+ ratio was reduced to 1.6, compared with 3.3 in 33 FIV-seronegative control cats. Variance analysis of data showed a significant influence of FIV seropositivity, sex, and spaying of female cats on CD4+ values. CD8+ values were significantly influenced by FIV seropositivity, age, and breed. These findings indicate a similarity between FIV and human immunodeficiency virus infections, as far as alterations of T-cell subsets are concerned.

Polymorphism of the bovine CSN1S1 promoter: linkage mapping, intragenic haplotypes, and effects on milk production traits.

The bovine CSN1S1 5' flanking region (CSN1S1-5') was screened for polymorphisms in different cattle breeds. Single-strand conformation polymorphisms (SSCP) and sequence analyses revealed four alleles (1-4), two of them being new allelic forms (3 and 4). Sequences were deposited in GenBank with accession numbers AF549499-502. In alleles 1 and 4, potential transcription factor binding sites are altered by the mutations. Using SSCP analysis, all four alleles were identified in German Holsteins. Six intragenic haplo-types comprising CSN1S1-5' (alleles 1, 2, 3, 4) and exon 17 (CSN1S1*B and C) genotypes were found. Linkage mapping using half-sib families from the German QTL project positioned CSN1S1 between the markers FBN14 and CSN3, with 5.6 cM distance between CSN1S1 and CSN3. Variance analysis, using family and CSN1S1 promoter genotypes as fixed effects, of breeding values and deregressed proofs for milk production traits (milk, fat, and protein yield and also fat and protein percentage) revealed significant effects on protein percentage when all families and genotypes were considered. Contrast calculations assigned a highly significant effect to genotype 24, which was associated with highest LS-means for protein percentage breeding values. As CSN1S1 is one of the main caseins in milk, this could be an effect of mutations in regulatory elements in the promoter region. An effect on milk yield breeding values was indicated for genotype 12, but is probably caused by a linked locus.

Potassium currents in hair cells isolated from the cochlea of the chick.

1. Potassium currents were characterized in tall hair cells of the chick's cochlea. Outward potassium currents were found to flow through two distinct classes of channels. 2. Individual hair cells were isolated from 200 microns long segments of the apical half of the chick's cochlea. Whole-cell voltage-clamp and current-clamp recordings were made from these cells. 3. Voltage responses to injected current ranged from high-frequency (100-250 Hz) oscillations in some cells, to slowly repetitive Ca2+ action potentials or slow oscillations (5-20 Hz) in others. 4. Ionic currents recorded in voltage clamp also varied in different hair cells. Cells with high-frequency voltage oscillations had rapidly activating Ca2(+)-dependent outward K+ current, IK(Ca). Cells that generated action potentials had slow delayed rectifier outward K+ current, IK, and inward rectifier current, IIR. All hair cells had inward Ca2+ current. 5. IK(Ca) activated positive to -45 mV. Tail currents reversed at the K+ equilibrium potential. This current was eliminated in Ca2(+)-free solutions, or when exposed to 10 mM-TEA. This outward current was fully activated within 1-3 ms at 0 mV. The whole-cell current was noisy and ensemble variance analysis suggested a single-channel conductance of 63 pS near 0 mV. 6. IK activated positive to -50 mV. Tail currents reversed at the K+ equilibrium potential. This current was not eliminated in Ca2(+)-free solutions, and was relatively resistant to external TEA. IK activated slowly, reaching peak values in 10-20 ms at 0 mV. This current showed little variance and the average single-channel conductance based on macroscopic noise near 0 mV was 8 pS. 7. External tetraethylammonium (TEA) or Ca2(+)-free saline eliminated the high-frequency voltage oscillations seen in many basal cells. In contrast TEA had little effect on slow action potentials (or low-frequency oscillations) seen in cells with IK. 8. IK(Ca) was prominent in hair cells originating 1.0-2.0 mm from the cochlear apex. IK and IIR dominated the membrane conductance of tall hair cells originating within 0.5 mm of the cochlear apex. 9. The frequency of voltage oscillation in apical cells was temperature-dependent, nearly doubling for each 10 degrees C rise in temperature. 10. IIR activated at membrane potentials negative to -75 mV. The average time constant of activation at -100 mV was 2 ms. Tail currents reversed at the K+ equilibrium potential and did not depend on the external Na+ concentration. IIR was blocked by 5 mM-Cs+ or 100 microM-Ba2+ in the external saline.

[Morphometric analysis of the inner skull base and the brain stem on median frozen sections of the fetal head, between 14 and 40 weeks of gestational age].

1. In median sections through frozen heads of 75 human fetuses, between 14 and 40 weeks of age, a total of 12 finite lines was determined at the inner skull-base, at the brain-stem, and in relation of the brain-stem to the inner skull-base. The fetuses were classified into 5 age groups. The data were obtained by means of variance analysis. 2. So far there has not been any information on human fetuses and measured quantities in the available literature. Comparing the mean values of the finite lines of the above mentioned five age groups explains something about the increase of growth. 3. In evaluating the results obtained and comparing the qualitative data reported in the literature, the importance of the different development of medullary functional units on the one hand and the topographic changes in the sense of additive processes on the other hand is emphasized. 4. The comparatively larger increase in the standard deviations between the 29th and the 40th week of fetal age suggests the tendency towards a plateau that may be due to the full development of microstructural systems in the fields examined.

[In vitro semi-automated morphometry of preserved human corneal endothelium].

Cellular morphometry allows us to quantify the structural alterations of the corneal endothelium. Our study was a semi-automatized in vitro analysis of fresh and preserved corneal endothelium (MK medium cryopreservation). The equipment used was the Quantimeter 720 connected to a Digital PDP1123 computer. Nineteen corneas were studied and the following cellular parameters were established: perimeter, surface area, form factor and polymorphism coefficient. The computer program used was the SPSS/PC (Statistical Package for the Social Sciences) and the principal statistical tests were the Bartlett test and variance analysis. The statistical computerized study allowed us to compare the four cellular parameters and to establish the incidence rate of the conservation process on endothelial morphometry. The statistical and morphometry methods have proved to be reproducible and reliable and carry a supplementary quantitative element to analyse the influence of the conservation process.

[Drug supplies in primary care units at the Ministry of Health in Mexico].

OBJECTIVE: To identify the availability of some essential drugs (ED) at primary health care units of the Mexican Ministry of Health (SSA), using data from a research work conducted in 1996 and 1997. MATERIALS AND METHODS: A random sample of the 18 sanitary jurisdictions of states participating in the Extension of Coverage Program (ECP) was drawn. All primary care units from selected jurisdictions were evaluated through an inspection visit, using a checklist that included 36 ED. In phase I of the analysis, the absolute number and proportion of units without a single item of any of the 36 drugs were calculated. In the units with available drugs, the median of the distribution was also calculated. In phase II, the medians according to the type of health unit and state was obtained. The statistic utilized for the comparison of the medians was the Scheffé test with one way variance analysis. In addition, the drugs were classified according to their therapeutic indication and the medians and proportion of available drugs were calculated. The differences in proportion were evaluated with the statistic chi 2. RESULTS: During the visits, on average, 18 of the 36 drugs included in the list of the study were found in the health units. The availability of antibiotics, antituberculosis drugs, and antimalarial drugs was particularly poor. In contrast, oral rehydration salts, family planning methods and vaccines were usually available. In general, the PAC3 states presented the best availability figures. CONCLUSIONS: The Ministry of Health of México will have to develop an enormous effort to overcome the obstacles related to the supply of essential drugs in primary health care units. Otherwise, all other efforts directed to meet the needs of the non-insured population will end up being useless, and enormous amounts of the already scarce resources of the health sector will be wasted since drugs are a vital component of the long chain of health care.

Bioequivalence of two oral cyclosporine preparations.

A comparison of bioequivalence of two cyclosporine (CAS 59865-13-3) preparations was performed. Ten cyclosporine treated patients with transplanted kidneys were included. Criteria were successful transplantation and minimum period from transplantation of at least 6 months. Two months before the experiment, cyclosporine concentrations had to be in therapeutic range without significant oscillation, and kidney function stabile. There had to be no signs of cyclosporine nephrotoxicity. During the objective biochemical analysis it was not allowed to find malfunction in any of the patient's organ important for cyclosporine pharmacokinetics. Cyclosporine concentrations in whole blood were measured with a specific fluoroimmunoassay. Cyclosporine and metabolites concentrations were measured with radioimmunoassay with non-specific antibody. Mean value and standard deviations and shape of distribution were calculated for all numeric data of patients, measured biochemical and other laboratory parameters. Variance analysis for all measured cyclosporine concentrations according to sampling times (C0 to C12, maximal concentrations C(M), time to maximal concentrations t(M), times of absorption delaying t(Lag) and area under the measured concentration curves (AUC) were statistically checked. According to these data it is concluded that the preparations are bioequivalent; a time to reach maximum concentration was slightly shorter for test preparation (2.5 and 3.2 h, respectively), but not statistically significant. There are no significant differences between the areas under the concentration curves (1667 and 1665 ng.h/ml, respectively). After the calculation of pharmacokinetic parameters of concentration data measured by a non-specific method a significant difference for areas under concentration curves was seen (3709 and 4600 ng.h/ml, respectively).(ABSTRACT TRUNCATED AT 250 WORDS)

[Spatial distribution pattern and fractal analysis of Larix chinensis populations in Qinling Mountain].

In this paper, the fractal characters of Larix chinensis populations in Qinling Mountain were studied by contiguous grid quadrate sampling method and by boxing-counting dimension and information dimension. The results showed that the high boxing-counting dimension (1.8087) and information dimension (1.7931) reflected a higher spatial occupational degree of L. chinensis populations. Judged by the dispersal index and Morisita's pattern index, L. chinensis populations clumped at three different age stages (0-25, 25-50 and over 50 years). From Greig-Smiths' mean variance analysis, the figure of pattern scale showed that L. chinensis populations clumped in 128 m2 and 512 m2, and the different age groups clumped in different scales. The pattern intensities decreased with increasing age, and tended to reduce with increasing area when detected by Kershaw's PI index. The spatial pattern characters of L. chinensis populations may be their responses to environmental factors.

Histomorphometrical and comparative analysis of three muscles of buffalo (Bubalus bubalis L.).

Three muscles were analyzed, Longissimus dorsi, Semimembranosus and Caput longum Tricipitis brachii taken from nine cow buffaloes, by examining the histochemical and morphometrical characteristics of different muscle fibres types and their distribution inside the examined muscles. Cross sectional area, perimeter, maximum and minimum diameter of about 200 fibres were measured for each muscle, and fast-twitch glycolytic fibres (FG), fast-twitch oxidative-glycolytic fibres (FOG), slow-twitch oxidative fibres (SO) were histochemically differentiated. The data have been elaborated with the SPSS software. The variance analysis indicates that there are not significant differences about dimensions between FG and FOG fibres, while the average values of transversal section area and perimeter are greater than the oxidative fibres in all examined muscles. The Semimembranosus muscle in comparison to the Longissimus dorsi and to the Caput longum Tricipitis brachii muscles has muscle fibres with the smallest value of transversal section area and perimeter. The balanced distribution and intense myofibrillar adenosine triphosphatose and succinic dehydrogenase activities of the three fibres types in Caput longum Tricipitis brachii muscle can be justified by the function performed by this muscle which, together with the other heads of the Triceps brachii acts essentially as extensor of the forearm in fact, differences in the dimensions of the different fibre types inside the three examined muscles have been underlined; this fact can be justified for every muscle performs different motor functions.

Mitochondrial DNA diversity in the Polish Roma.

Mitochondrial DNA variability in the Polish Roma population has been studied by means of hypervariable segment I and II (HVS I and II) sequencing and restriction fragment-length polymorphism analysis of the mtDNA coding region. The mtDNA haplotypes detected in the Polish Roma fall into the common Eurasian mitochondrial haplogroups (H, U3, K, J1, X, I, W, and M*). The results of complete mtDNA sequencing clearly indicate that the Romani M*-lineage belongs to the Indian-specific haplogroup M5, which is characterized by three transitions in the coding region, at sites 12477, 3921 and 709. Molecular variance analysis inferred from mtDNA data reveals that genetic distances between the Roma groups are considerably larger than those between the surrounding European populations. Also, there are significant differences between the Bulgarian Roma (Balkan and Vlax groups) and West European Roma (Polish, Lithuanian and Spanish groups). Comparative analysis of mtDNA haplotypes in the Roma populations shows that different haplotypes appear to demonstrate impressive founder effects: M5 and H (16261-16304) in all Romani groups; U3, I and J1 in some Romani groups. Interestingly, haplogroup K (with HVS I motif 16224-16234-16311) found in the Polish Roma sample seems to be specific for Ashkenazi Jewish populations.

Continuous glucose monitoring by means of fiber-based, mid-infrared laser spectroscopy.

The application of mid-infrared laser spectroscopy to the reagent-free quantification of the concentration of glucose was investigated using cryogenically cooled lead salt lasers or, alternatively, quantum cascade lasers operating at room temperature. The concentration of glucose in aqueous solutions was quantified by means of fiber-based attenuated total reflection (ATR) spectroscopy (fiber-optical evanescent field analysis, FEFA) as well as fiber-based transmission spectroscopy. Both methods have the potential to be utilized by small fiber sensors, which can be inserted into the subcutaneous tissue in order to continuously measure the local concentration of glucose. In our in vitro experiments, noise-equivalent concentrations as low as 10 mg/dL were achieved. The mid-term stability of the measurement schemes was investigated by means of Allan variance analysis. Based on the research presented in this manuscript, an all-room-temperature measurement scheme using quantum cascade lasers, miniaturized fiber-optic sensors, and pyroelectric detectors appears well suited for the continuous monitoring of glucose concentrations at physiological levels.

[Nuclear volume of cells in the stratum granulosum and theca folliculi interna of the largest dominant and atretic tertiary follicles in sheep during the selection process].

According to available literary data, an increased nuclear volume of endocrine gland cells, various structures of the diencephalon and endocrine ovarial structures could be observed when the respective organs and structures were more active; a decreased nuclear volume was revealed when the activity of the respective organ or its structure was suppressed (Mess, 1962; Maracek and Arendarcik, 1976, 1978). Attention was paid to the karyometric analysis of follicular cells of the stratum granulosum (SG) and secretory cells of the theca folliculi interna (TFI) during selection of the dominant ovulatory follicle (DOF). The aim of this study was to determine differences in hte nuclear volumes of the granulosa follicular cells and the secretory cells of the inner theca of the DO1F in comparison with the largest atretic follicles (AF) as well as to verify the use of karyometric variance analysis for the evaluation of the selection process of the dominant ovulatory tertiary follicle. Histological sections were prepared from large DOF and AF from the ovaries of 18 sheep aged 3-5 years and held under standard conditions of a controlled herd. Control group I (n = 3) consisted of Wallachian sheep on day 15 of the sexual cycle (day 0). Excisions of ovaries from the animals of the experimental group (n = 3) were made on day 16 of the cycle after i.m. treatment with 120 micrograms GnRH 10 and 7 hours prior to sampling. Control group II (n = 3) consisted of Tsigai sheep at the 9th or 10th day of the oestrous cycle (hour 0) and three experimental groups (n = 3 each) consisted of Tsigai sheep 24, 48 and 72 hours after i.m. treatment with 125 micrograms cloprostenol. After excision the ovaries were fixed in neutral formaline, cut into 4 mm thick transverse segments and subjected to standard histological processing. After staining with Harris' HE the 5-7 microns thick sections were karyometrically evaluated at a magnification of 2200 x according to the method of Palkovits (1961) using the PC programme Karyotest 03 (Maracek et al., 1991). 200 cells were evaluated from each sample and both cell types; altogether 7200 cells were examined. GnRH treatment (Dirigestran inj. Spofa) increased the nuclear volume of follicular cells (SG) in the dominant ovulatory follicle in the process of selection (Tab. I, Fig. 1). The former, however, reduced the nuclear volume not only of the SG follicular cells of LAF (Tab. II, Fig. 2) but also of the secretory cells of both the DOF and AF theca interna (Tab. II, Figs. 3, 4). Cloprostenol treatment (Oestrophan inj. Spofa) affected the follicular cells of the granulosa in the dominant ovulatory follicle.(ABSTRACT TRUNCATED AT 400 WORDS)

Normal values of colloidal-osmotic pressure in healthy adults in Mexico city.

The normal value of colloid-osmotic pressure (COP) was determined in male and female adults between 20 and 40 years old. Any differences of this value related to sex and age were investigated. It was found the statistical correlativeness of COP obtained with an oncometer and that calculated with Landis-Pappenheimer equation (F1) and the modified equation (F2) of the same authors. Two groups, each of 50 males and 50 females were formed. Those persons with altered routine laboratory tests that could modify COP were discarded. COP was determined with IL 186 Weil Oncometer and also calculated with both Landis-Pappenheimer equations (F1 and F2). An statistical analysis was made to see if age had any influence on COP. Results in males, measured COP was 27.22 +/- 2.51 torr, calculated COP by F1 was 26.646 +/- 2.38 torr, and calculated COP by F2 was 27.814 +/- 2.43 torr. In females, measured COP was 26.8 +/- 1.71 torr, calculated COP by F1 was 26.9476 +/- 2.005 torr, and calculated COP by F2 was 28.09 +/- 2.0984 torr. Measured COP regardless sex was 27.01 +/- 2.15. Using the variance analysis and the minimal significative differences (MSD) it was seen that measured COP and calculated COP by F1 can be used either one with 95 per cent of credibility in healthy ambulant adult. Using the aforementioned statistical procedure it was noted a lower figure between 30 and 35 years that at any other studied age.

Effect of trimebutine on colonic myoelectrical activity in IBS patients.

The effect of trimebutine 100 mg i.v. and placebo on colonic myoelectrical activity was investigated in 10 patients with the irritable bowel syndrome (IBS) (5 constipated and 5 diarrhoeic), using an intraluminal probe supporting 8 groups of electrodes. At each site examined from transverse to sigmoid colon, the electromyograms exhibited two kinds of spike bursts: short spike bursts (SSB) localized at one electrode site and appearing rhythmically at 10.3/min, and long spike bursts (LSB), isolated or propagated orally or aborally. Computerized analysis of the duration of each kind of spike burst showed that, as compared to the control, trimebutine 100 mg, selectively inhibited by 43 to 73% the mean duration of LSB activity in the transverse, descending and sigmoid colon, from 0 to 30 min after administration. The inhibitory effect was similar in constipated and diarrhoeic patients. Placebo injection did not significantly affect (p greater than 0.05) the duration of LSB and SSB activity. Variance analysis indicated that the inhibitory effect of trimebutine was significantly greater (p less than 0.05) on LSB activity in the transverse than the descending colon, and that it was absent from the sigmoid colon. The results suggest that trimebutine selectively inhibits the propulsive activity of the proximal two thirds of the colon in IBS patients, and that this effect cannot entirely explain its therapeutic efficacy in IBS.

[Eradication of Helicobacter pylori infection in Europe: a meta-analysis based on congress abstracts, 1997-2002].

BACKGROUND: Meta-analyses evaluated several aspects of Helicobacter pylori eradication based on the randomised controlled trials. AIM: to perform a meta-analysis of the papers presented at the European Helicobacter Pylori Study Group and United European Gastroenterology Week meetings from 1997 to 2002. METHODS: Abstracts dealing with the eradication of Helicobacter pylori have been reviewed and the randomised, controlled studies from European countries were included. The studies were classified into groups based on eradication schedules, antibiotics used and country of provenience. The pooled eradication rates were calculated and the differences were assessed by multiple variance analysis. RESULTS: One-hundred and two studies were accepted comprising 25,644 cases and 398 treatment arms. The eradication rate of proton pump inhibitor-based first line triple therapies was 80.4% (confidence interval: 78.9-81.8); no difference was observed between the five proton pump inhibitors (p > 0.05). Ranitidine bismuth citrate based regimens were efficient in 79.9% (75.7-84.0) (p = 0.95 vs PPI). H2 blockers-based therapies achieved 68.6% (59.0-78.1) (p = 0.0007 vs proton pump inhibitor and p = 0.005 vs ranitidine bismuth citrate-based regimens). Proton pump inhibitor-based double combinations were efficient in 47.1 (31.9-62.4) (p = 0.001 vs triple regimens). Clarithromycin+amoxicillin/nitroimidazole combinations achieved rates of 79.6% and 84.1%, respectively, while amoxicillin-nitroimidazole regimens were less efficient (72.5%, 66.6-78.5) (p = 0.006). The pooled eradication rate of second-line triple regimens was 75.5% (69.9-86.4)(p = 0.08 vs primary treatment). Quadruple therapies were successful in 81.1% (76.6-85.6) of cases as first-line and 73.8% (61.2-86.4) as second-line regimens (p = 0.77 and p = 0.02 vs triple regimens). The pooled eradication rates varied from 58% to 92% in the European countries. CONCLUSIONS: The pooled eradication rate of the primary proton pump inhibitor/ranitidine bismuth citrate-based triple regimens are comparable with the results of meta-analyses. H2 blocker-based triple and proton pump inhibitor-based double regimens are of lower efficacy. Quadruple regimens were not better than triple therapies. The eradication rates per country varied, approaching 80% in most places. The results confirm in part post-hoc the validity of the Maastricht consensus recommendations.

[Research on the normal value of total blood cholesterol in the adult population of Palermo].

We chose, as typical values of serum cholesterol, the mean levels of the unimodal distribution of frequency with asymmetrical form, for different age groups, from some quarters of Palermo. We assumed as normal range estimate the interval between the 2.5 and 97.5 percentiles, with associated nonparametric 90% confidence intervals for normal limits for the true percentile. For this biometric analysis of ethnic physiology, we controlled the laboratory measurements in several inspection cycles. As the observed frequencies are not a real statistical sample, neither the 95% of the actual reference population: we smoothed the observed frequencies interpolating, with an algebraic polynomial, the mean empiric levels (and the chosen percentiles) of total serum cholesterol of the examined population - since the measurements taken on Mondays, the days after holidays, in January and April were chosen to see, experimentally, how the collected data depend on environmental alimentary factors; the degree of polynomial which gave the best approximation was chosen, on the grounds of the variance analysis; we tested the goodness of fit of the function to the referred frequencies. Moreover, the choice of the function was based on: a) the test of the hypothesis that the dependent variable reading the various specific ages, is normal, so we estimated the kurtosis and the asymmetry of the single frequency distributions; b) the simulation of the hypothesis that the measurements were made at random from gaussian distribution, so that the variances homogeneity of the examined character at various specific ages was examined by Bartlett's test. To identify the stages within which the cholesterolemic levels vary, due to growth and to the sexual maturation, we formed three age groups for males and four for females, whose delimitation is not arbitrary, since the best interpolation of the mean levels observed within such delimited age groups was again obtained by an algebraic polynomial of 2nd degree.

Spatial training in a complex environment and isolation alter the spine distribution differently in rat CA1 pyramidal cells.

The hippocampus is critically involved in spatial learning. Spatial training in adult rats, which improved their spatial learning ability, increased the number of excitatory hippocampal CA1 spine synapses on basal dendrites as compared with either isolated or standardly housed animals (Moser et al. [1994] Proc. Natl. Acad. Sci. USA 91:12673-12675). In this article, we report that spine synapses on oblique apical dendritic branches do not increase in density or number after the same type of training. When examining the variability of the spine density on basal CA1 dendrites by using variance component analysis, the variance associated with the cells was twice as large in all three groups as that coupled to the rats. Analysis of the spine density plots shows that the enhanced spine density after spatial training is found in most cells recorded from the trained group but that a small subset of CA1 neurones are particularly well supplied with spines. The trained group had a significant right-skewed tail of the spine distribution, i.e., training caused high spine density to occur in a small subset of dendritic segments. Conversely, the isolated group had a significant left-skewed spine distribution, indicating that some of the dendritic segments were undersupplied with spines, whereas the paired group displayed no asymmetry.

Size-distribution analysis of macromolecules by sedimentation velocity ultracentrifugation and lamm equation modeling.

A new method for the size-distribution analysis of polymers by sedimentation velocity analytical ultracentrifugation is described. It exploits the ability of Lamm equation modeling to discriminate between the spreading of the sedimentation boundary arising from sample heterogeneity and from diffusion. Finite element solutions of the Lamm equation for a large number of discrete noninteracting species are combined with maximum entropy regularization to represent a continuous size-distribution. As in the program CONTIN, the parameter governing the regularization constraint is adjusted by variance analysis to a predefined confidence level. Estimates of the partial specific volume and the frictional ratio of the macromolecules are used to calculate the diffusion coefficients, resulting in relatively high-resolution sedimentation coefficient distributions c(s) or molar mass distributions c(M). It can be applied to interference optical data that exhibit systematic noise components, and it does not require solution or solvent plateaus to be established. More details on the size-distribution can be obtained than from van Holde-Weischet analysis. The sensitivity to the values of the regularization parameter and to the shape parameters is explored with the help of simulated sedimentation data of discrete and continuous model size distributions, and by applications to experimental data of continuous and discrete protein mixtures.
