Some metabolic aspects of tolerance to bacterial endotoxin.

Berry, L. Joe (Bryn Mawr College, Bryn Mawr, Pa.), and Dorothy S. Smythe. Some metabolic aspects of tolerance to bacterial endotoxin. J. Bacteriol. 90:970-977. 1965.-The tolerance to bacterial endotoxins which is produced in mice given a series of daily injections of heat-killed Salmonella typhimurium failed to occur when actinomycin D was administered with the heat-killed cells. Neither ethionine nor 2-thiouracil, when given with endotoxin, altered the development of tolerance. An injection of endotoxin, actinomycin D, or ethionine lowered the activity of the liver enzyme tryptophan pyrrolase more significantly at either 4 or 17 hr postinjection in normal mice than in tolerant mice. Similarly, an injection of either saccharated iron oxide or Thorotrast lowered liver tryptophan pyrrolase activity more extensively in normal than in tolerant animals. Activation of the reticuloendothelial system (RES) of tolerant mice, as determined by an accelerated rate of carbon clearance from the blood, was observed, but this was prevented by the appropriate dose of actinomycin D. Similar results were obtained when saccharated iron oxide, rather than endotoxin, was used to activate the RES, but these animals were not resistant to endotoxin and their tryptophan pyrrolase was normally diminished after an injection of endotoxin. Thus, RES activation may occur without tolerance developing. A more nearly normal level of enzyme activity appears to be characteristic of the tolerant state.

Acriflavine-binding capacity of Escherichia coli in relation to acriflavine sensitivity and metabolic activity.

Nakamura, Hakobu (Konan University, Kobe, Japan). Acriflavine-binding capacity of Escherichia coli in relation to acriflavine sensitivity and metabolic activity. J. Bacteriol. 92:1447-1452. 1966.-Inheritance of the acriflavine resistance gene by an acriflavine-sensitive strain of Escherichia coli K-12 resulted in a reduction in the cellular accumulation of acriflavine and other basic dyes, but had no effect on the accumulation of acid dyes. In both acriflavine-resistant and -sensitive strains, the cooling of cells from 37 to 0 C increased acriflavine accumulation. This increased accumulation was released from the cells after restoration to 37 C. Acriflavine accumulation was increased by carbon shortage and by metabolic disturbance caused by potassium cyanide, arsenite, puromycin, chloramphenicol, 2-thiouracil, and 8-azaguanine. The functional relation of acrifiavine accumulation to the acriflavine concentration of the medium suggests that adsorption is involved in the accumulation.

Gastric mucinous secretion under various conditions of stimulation in hypothyroid dogs.

Principal glycoproteins and amino acids of the secretions from Heidenhain pouches, were studied in normothyroid and hypothyroid Labrador dogs in the resting state, following injection of pentagastrin, histamine and after feeding. Hypothyroidism was produced by total thyroidectomy and chronic treatment with thiouracil. All samples showed the presence of galactose, glucosamine, galactosamine, fucose, mannose, uronic acid and sulfate. Galactose and glucosamine were found in equimolar amounts and the galactose to galactosamine ratio was 2:1. Concentrations of most carbohydrate components studied were higher in normothyroid than in hypothyroid dogs. Concentrations of most amino acids analyzed were also higher in normothyroid than in hypothyroid dogs, following stimulation with secretagogues.

Recovery of tobacco mosaic virus RNA Replication after incubation at 40 degrees.

Incubation of tobacco mosaic virus (TMV)-infected tissue at 40 degrees for 12 h destroyed the capacity for TMV RNA replication. Upon shift of the tissue from 40 to 25 degrees, synthesis of TMV RNA did not resume. Incubation at 40 degrees for 1 h did not destory the TMV RNA replicase, because viral RNA synthesis resumed upon return to 25 degrees. However, upon further incubation at 25 degrees, the synthesis rate of TMV RNA gradually declined. Upon still further incubation at 25 degrees (16-20 h), the synthesis of TMV RNA in tissue incubated at 40 degrees for either 1 or 12 h recovered. This recovery was inhibited by cycloheximide but not by 2-thiouracil.

Mode of action of the antiphytoviral compound 2,4-dioxohexahydro-1,3,5-triazine (5-azadihydrouracil).

The time course of inhibition of potato virus X ( PVX ) synthesis by the newly developed antiphytoviral compound 2,4- dioxohexahydro -1,3,5-triazine (DHT) was determined in mechanically inoculated leaves of Nicotiana tabacum L. cv. ' Samsun '. At the permissive temperature (22 +/- 3 degrees) DHT inhibited the synthesis of PVX almost 100% within 8 h postinoculation, after which the inhibition declined. In leaves maintained at a temperature of 5 +/- 2 degrees for 5 days, DHT inhibited PVX synthesis about 65% immediately after the shift to permissive temperature. This time course of inhibition was almost identical to that of 2-thiouracil. Uracil completely overcame the inhibition of PVX replication caused by DHT when added within 2 h after incubation of leaf disks with DHT. These results indicate that DHT acts as an analogue of the pyrimidine base uracil.

Synthesis and antiviral activity of 1,3-disubstituted uracils against HIV-1 and HCMV.

The development of new non-nucleoside reverse transcriptase inhibitors (NNRTIs) is an efficient strategy for finding new therapeutic agents against human immunodeficiency virus (HIV). A large number of 6-substituted uracil derivatives have been prepared in order to explore new NNRTIs. However, there are few approaches to anti-HIV agents from 1,3-disubstituted uracil derivatives. Therefore, we tried to prepare several 1,3-disubstituted uracils, which were easily obtainable from uracil by preparation under alkali and Mitsunobu conditions, and examined their antiviral activity against HIV-1 and human cytomegalovirus (HCMV). We found that 1-benzyl-3-(3,5-dimethylbenzyl)uracil and 1-cyanomethyl-3-(3,5-dimethylbenzyl)-4-thiouracil showed powerful inhibition against HCMV and HIV-1, respectively.

Blood capillary enlargement during the development of thyroid hyperplasia in the rat.

To study blood capillary growth in the hyperplastic thyroid gland, rats were fed 0.25% thiouracil in a low iodine diet for time intervals up to 100 days. Thyroids were fixed by vascular perfusion and embeded in Epon. Whole lobes were sectioned from pole to pole and slides were prepared of sections every 0.3 mm. Capillaries were clearly enlarged by 3 days and they enlarged progressively thereafter. By 3 days, the cells of many neighboring capillaries came into close apposition and from this time on, there was evidence of fusion of capillary walls in the form of partial septa in the capillary lumens. Fusion continued until 20 days, when follicles were almost completely surrounded by a continuous endothelial sheet and unfused capillary walls were separated by connective tissue. The vascular pattern around peripheral follicles changed in a way similar to interior follicles, except that in places, capillaries were gradually excluded from the space between epithelium and thyroid capsule. Vascular enlargement was restricted to the thyroid blood vessels. There was no obvious enlargement of the blood capillaries of the parathyroid despite its close proximity to the thyroid.

Lobulo-alveolar development of mouse mammary glands is regulated by thyroid hormones.

The mammary glands of euthyroid female C3H/HeN mice undergo a series of morphological changes during development. In glands from immature animals, the epithelial component consists of a sparse ductal system with few branches which fills about one fourth of the fat pad. In the adult virgin gland, the epithelial component fills the fat pad with a highly branched ductal system and a few alveoli. In contrast, glands from adult animals maintained in a hypothyroid state by ingestion of thiouracil since weaning retain the primitive ductal appearance while filling the fat pad. The glands from animals made hyperthroid by adding 2 micrograms T4/ml drinking water have extensive lobulo-alveolar development. Glands from animals made hypothyroid during 7 weeks of involution after lactation have the same degree of deveopment as the euthyroid controls. When explants of tissue from adult hypothyroid virgin animals are cultured in serum-free medium containing insulin, hydrocortisone, and PRL, the specific milk protein, alpha-lactalbumin, is induced. The level of alpha-lactalbumin, measured as lactose synthetase activity, found per ng epithelial DNA is the same as that found in explants from glands of euthyroid virgins. These results suggest that thyroid hormones, in concert with PRL, play an important role in the regulation of development of the mouse mammary gland. Decreased levels of thyroid hormones in the serum result in retarded growth of the ductal system and little or no alveolar development. However, the resulting epithelial component of glands from hypothyroid mice is fully capable of differentiating in vitro when exposed to the proper hormonal environment.

Lipid-lowering effects of N-[4-methylbenzylthiocarbonyl]-L-phenylalanine (KF1492), a new phenylalanine derivative.

Lipid-lowering effects of KF1492, N-[4-methylbenzylthiocarbonyl]-L-phenylalanine, were evaluated in comparison with clofibrate. This compound lowered serum cholesterol (s-CL) and triglyceride(s-TG) in cholesterol-fed, Triton-injected and glycerol-fed rats as well as in normal rats. The dose of KF1492 required to show these effects was almost equal to that of clofibrate. In addition, KF1492 produced significant reductions of s-CL and s-TG in thiouracil-fed rats and decreasing phase of Triton-induced hyperlipemia of rats. In these models, clofibrate produced no significant reductions. Clofibrate produced a marked increase of liver size and shortened the pentobarbital-induced sleeping time in rats. On the contrary, the increase of liver size by KF1492 was less marked than clofibrate, and KF1492 caused no change in the sleeping time. Thus, it is apparent that KF1492 is a new lipid-lowering compound with less hepatic effect than clofibrate and that the lipid-lowering profile of KF1492 differs from that of clofibrate in some points.

Unrestricted accessibility of short oligonucleotides to RNA.

The propensity of RNA to fold into higher-order structures poses a major barrier to the use of short probes (<15 nucleotides) by preventing their accessibility. Introduction of the pseudo-complementary bases 2-aminoadenine (nA) and 2-thiouracil (sU) and the destabilizing base 7-deazaguanine (cG) into RNA provides a partial solution to this problem. While complementary in hydrogen bonding groups, nA and sU cannot form a stable base pair due to steric hindrance, and are thus pseudo-complementary. Each, however, recognizes the regular T/U and A complements, allowing pairing with oligonucleotides. Short pseudo-complementary RNAs can be prepared by in vitro transcription. Relative to standard transcripts, the modified transcripts possess reduced secondary structure and increased accessibility to short (8-mer) probes in the locked nucleic acid (LNA) configuration. They also hybridize to complementary probes with increased specificity and thermostability. Practical application of this strategy to oligonucleotide-based hybridization assays will require engineering of RNA polymerase for more efficient utilization of pseudo-complementary nucleoside triphosphates.

Effects of thyroid hormone on the renal dopaminergic system.

This study determined the effects of thyroid hormone on the renal dopaminergic system. Surgical thyroidectomy (Tx) and treatment with 2-thiouracil (Thio) decreased renal cortex Na+/K+ ATPase activity and urinary volume. Tx also decreased urinary Na+ and urinary L-DOPA without changing urinary excretion of Dopamine (DA). Thio treatment decreased slightly urinary L-DOPA and Na+, but increased urinary excretion of DA. In both models of thyroid hormone deficiency, the ratio urinary DA/DOPA increased. Changes after Thio treatment were reversed after one month of drug withdrawal. Treatment with T3 via osmotic minipump increased Na+/K+ ATPase activity and urinary L-DOPA, did not change urinary DA, and increased the ratio DA/DOPA. To further analyze the effects of thyroid hormone deficiency, we administered selective DA1 (SCH-23390), DA2 (Sulpiride), and a non selective (Haloperidol) DA receptor antagonists to Thio treated and control animals. The DA1 antagonist decreased diuresis, natriuresis and urinary L-DOPA in control, but had no effect in Thio treated rats. Sulpiride had no effect in either group. The combination of SCH-23390 plus Sulpiride decreased urinary L-DOPA and urinary volume only in Thio treated animals. Haloperidol decreased urinary volume in Thio treated animals, but had no effect in controls. Our findings suggest that renal DA synthesis is to some extent dependent on thyroid hormone levels, and that the response of DA receptors is altered by thyroid hormone deficiency, indicating a role of this hormone in the regulation of the renal dopaminergic system.

Effect of thyroid status on the thyrotrophin-releasing hormone-degrading activity of rat serum.

The TRH-degrading activity of rat serum in vitro is five times more potent than that of human serum. In rats, it is significantly reduced in hypothyroidism (thiouracil-induced) and significantly increased in hyperthyroidism (T3 or T4-induced). This suggests a possible role in the regulation of adenohypophysial-thyroid function which is probably, in turn, dependent on thyroid hormone, rather than TSH, levels.

Expression of the endothelin-1 gene in the rat thyroid gland and changes in its peptide and mRNA levels in goiter formation and iodide-induced involution.

Endothelin-1 (ET-1) is a major vasoconstrictor peptide, first found in endothelial cells, and later in many other tissues, including the thyroid gland. We analysed the expression of the ET-1 gene in the rat thyroid gland and changes in ET-1 mRNA and peptide levels in goiter development and involution, two circumstances characterised by vascular changes. Thyroid hyperplasia was induced in adult Wistar rats by feeding a low iodine diet (LID) supplemented with 0.25% thiouracil for 10 days, and LID alone for 2 further days (H.12 group). Involution was induced by injecting 100 micrograms iodide and refeeding a normal diet during 6 h, 12 h, and 24 h (I.6h, I.12h, I.24h groups). Rats fed a normal iodine diet were used as controls. A specific 488 bp cDNA corresponding to the known sequence of pre-pro ET-1 was found by RT-PCR from RNA extracts in all thyroid experimental groups, as well as in lung and kidney which were used as positive controls. RP-HPLC analysis showed that ET-1 immunoreactivity eluted similarly as mature ET-1. During hyperplasia, ET-1 mRNA and peptide levels were increased 3.5- and 5-fold respectively. The relative volume of the vascular bed was more than doubled. During iodide-induced involution, the glandular ET-1 mRNA level remained elevated. The concentration of ET-1 peptide increased and was significantly greater at 12 h involution than in the H.12 group. At this time, the capillary reticulum reverted to individual capillaries and the vascular bed was significantly reduced. These data demonstrate that the ET-1 gene is expressed in the rat thyroid gland and that the ET-1 mRNA and peptide levels are increased during thyroid hyperplasia and remain elevated during a phase of rapid iodide-induced involution. These data suggest that changes in ET-1 production may play a role in control of thyroid gland trophic regulation and vascularity.

The proteins of normal and abnormal tectorial membranes.

The proteins of the normal mouse tectorial membrane were analysed using SDS polyacrylamide gel electrophoresis. Three major protein bands were found with molecular weights of 145 000, 155 000 and 165 000, together with a number of minor components. Abnormal tectorial membranes from thiouracil-treated mice and from Deafness, Jerker, and Varitint-Waddler mutant mice were also examined in this way, but no evidence could be found to support the hypothesis that the distortion of the membranes might be due to an abnormal protein constitution. No PAS-positive material could be demonstrated in gels of normal membranes.

Effects of dietary supplemented amino acids on endogenous hypercholesterolemia in rats.

Effects of additions of amino acids to a 20% casein diet on serum cholesterol (Ch) were studied in hypothyroid and hepatoma-bearing rats with endogenous hypercholesterolemia as well as in normal rats. In normal Wistar rats, methionine (Met) was hypercholesterolemic at the "nutritional" level (0.2-0.4%), but hypocholesterolemic at the "excess" level (1.2-2.4%). In Wistar rats with hypothyroidism induced by thiouracil, the addition of excess (1.2%) Met to the 20% casein diet reduced an endogenous hypercholesterolemia due to hypothyroidism by suppressing an elevation in (VLDL + LDL)-Ch with no significant influence on HDL-Ch. In Donryu rats received a subcutaneous implantation of AH109A cells (an ascites hepatoma line), either 1.2% Met, 1.2% cystine (Cys), or 1.2% Met and 2.5% glycine (Gly) in combination improved a hepatoma-induced hypercholesterolemia and abnormal serum lipoprotein profiles by suppressing a hepatoma-induced increase in (VLDL + LDL)-Ch. From Ch turnover studies in hepatoma-bearing rats, an impaired catabolism of Ch in the liver was suggested to be one cause for the hepatoma-induced elevation in (VLDL + LDL)-Ch. One of the dietary manipulations. met and Gly in combination (Met + Gly), was found to improve the impaired Ch catabolism, this leading to a reduction of the (VLDL + LDL)-Ch level by Met + Gly in hepatoma-bearing rats.

Involvement of the thyroid gland in the response of young chickens to heat stress.

The involvement of the thyroid gland in the response of young chickens to heat stress was studied. Mean survival times when exposed to 50 C were significantly shorter for 3-week-old broiler male chickens injected with 40 micrograms L-thyroxine (T4)/100 g body weight compared to chickens injected with 30, 20, or 0 microgram of T4/100 g body weight. A significant reduction in heat stress survival time was obtained when T4 was injected 12, 18, or 24 hr before heat stress but not 6 hr before. Triiodothyronine (T3) reduced heat stress survival time when administered 12 to 24 hr before the stress but not when given 6 hr before. Chickens made hypothyroid by radiothyroidectomy had significantly longer heat stress survival times than controls. Likewise, chickens fed a diet containing .2% thiouracil 10 days before heat stress at 28 days had significantly longer survival times than control birds.

Capillary zone electrophoresis determination of thyreostatic drugs in urine.

In this work, a new capillary zone electrophoresis (CZE) method has been developed for the analysis of thyreostatic drugs (TD) such as methylthiouracil (MTU), propylthiouracil (PTU), and thiouracil (TU) in urine specimens. An untreated fused-silica capillary tube (75 microns i.d., 57 cm total length, 49.5 cm length to the detector) was used in all of the experiments. Optimal conditions were found using 50 mM borate buffer, pH 7.6, applied voltage 15 kV, 25 degrees C, and 30 sec of hydrodynamic injection. UV detection at 276 nm was employed. Ethylacetate extraction of the compounds was made before the analysis of each urine sample. An electrostacking procedure to increase the sensitivity was applied. The method developed was used for the analysis of several thousands of samples as part of a veterinary control procedure (Institute for State Control of Veterinary Biological and Medicaments, Brno, Czech Republic). The following detection limits were reached: 0.3 ppm (PTU and MTU) and 0.5 ppm (TU). The standard deviation for PTU, MTU, and TU determination was 1.12, 0.98, and 3.8% rel., respectively.

Pulsepolarographic determination of 6-benzyl-2-thio-uracil.

Polarographic (DPP) activity of 6-benzyl-2-thiouracil (6-benzyl-2-mercapto-4-pyrimidinol, BTU) has been examined in a wide range of pH values and it has been discovered that the number and the height of observed peaks depends on composition of supporting electrolyte and concentration of the thiol. Two different types of signals can be obtained. One of them is controlled by diffusion while the other by adsorption. The procedure for the determination of BTU in pure samples in 0.1 mol.dm-3 sodium hydroxide solution has been worked out. The measurements can be performed in a range 3.10(-4)-3.10(-3) mol.dm-3 of BTU. The detection limit is however lower and reaches 5.10(-6) mol.dm-3. The method has been used in the determination of BTU in an antithyroid drug Basdène.
