Five pesticides were evaluated against laboratory colonies of Leptotrombidium fletcheri (Womersly and Heaslip) by the Pasteur pipet technique. The pesticides were dieldrin (LC50 = 3.6 ppm, LC99 = 18.2 ppm), bromopropylate (LC50 = 9.2 ppm, LC99 = 239.6 ppm), dicofol (LC50 = 27.8 ppm, LC99 = 118.1 ppm), fenthion (LC50 = 15.4 ppm, LC99 = 29.7 ppm), and malathion (LC50 = 84.7 ppm, LC99 = 313.9 ppm). Dieldrin was the most toxic. Dicofol was recommended for further evaluation in field trials.

OBJECTIVE: To construct recombinant plasmids containing the truncated gene of the major surface antigen sta56 of Orientia tsutsugamushi (Ot.) Karp strain for expression antigen in E. coli so as to compare the expression efficiency in different systems. METHODS: From the recombinant plasmid TOPO-sta56 containing sta56 of Orientia tsutsugamushi Karp strain, several truncated genes of sta56 with different length were amplified and subcloned into the expression vectors pPROEX HTb and pET30a. These genes were expressed in E. coli DH5alpha and BL21(DE3) respectively when induced by IPTG. The expressed recombinant proteins were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Western blot. RESULTS: Six recombinant plasmids containing truncated sta56 genes of different length were constructed as follow: pHTbOt957, pHTbOt498, pHTbOt342 and pETOt957, pETOt498, pETOt342. The recombinant sta56 proteins were highly expressed as 6 x His fusion proteins in E. coli DH5alpha and BL21(DE3) respectively. The fusion proteins showed as different bands of different molecular weight respectively when analyzed with SDS-PAGE. Western blot demonstrated that the recombinant proteins were recognized by the positive serum of Ot. patients. CONCLUSION: The sta56 gene of Orientia tsutsugamushi Karp strain could be highly expressed in E. coli and its expression showed better efficiency in pET30a than in pPROEX HTb. The recombinant sta56 antigen with immunoreactivity could be used as diagnostic reagent for Ot. infection.

In this retrospective study, 140 serum samples collected from 85 scrub typhus-negative patients in Kinmen Island in 2000 were tested for antibodies to hantavirus using enzyme-linked immunosorbent assay. Seven patients (8.23%) were confirmed as having hantavirus infection as demonstrated by increased hantavirus-specific immunoglobulin M and/or immunoglobulin G antibodies in their convalescent serum samples. Analysis of indirect immunofluorescence assay showed that Seoul type was the etiologic agent. Serosurvey of rodents caught in the resident township of these hantavirus-infected human cases showed that the seroprevalence of antibodies to hantavirus among Rattus norvegicus, Mus musculus, and R. flavipectus was 50% (4/8), 20% (1/5), and 2% (7/348), respectively. Molecular analysis showed that these reservoir hosts carried a Seoul type hantavirus. To our knowledge, this is the first report demonstrating indigenous hantavirus cases in Kinmen.
A number of febrile diseases imported from the tropics, above all malaria, may, if left untreated, endanger the life of the patient. Microscopy of malaria slides without delay is by far the most important diagnostic tool in all patients who may have been exposed to infection. An accurate, detailed travel history, a knowledge of infections prevalent in areas where the patient has been and a thorough physical examination are essential. However, the majority of persons with fever after travel have a cosmopolitan etiology to their disease. If investigations suggest a possibly exotic yet not clear-cut cause of the febrile condition, it may be desirable to consult a physician with expertise in tropical medicine.

An accurate diagnosis of Tsutsugamushi disease (TD) can be made within a few hours after receiving serum specimens by detecting the specific IgG and IgM antibodies in the patient's serum. Using the indirect immunoperoxidase test we found a total of 730 cases of TD in 32 out of 47 prefectures when serum samples from 2,224 cases of suspected disease or fever of unknown origin or from patients with febrile exanthema of unknown cause were examined during a 10 years period (from May 1980 till December 1989). In addition, 27 cases of spotted fever group rickettsial infection were confirmed and one case of African spotted fever group infection was also detected in 1988. Furthermore, possibly the first case of TD was found in People's Republic of Congo (Africa) in 1989.

OBJECTIVE: To investigate the possibility of Hantavirus (HV) and Orientia tsutsugamushi (Ot) coinfection in their hosts. METHODS: HV and Ot were used to infect Vero E6 cells cultured in vitro singly, simultaneously or successively. Genes of HV and Ot were identified in different generation cells with RT-PCR. RESULTS: Five experiment groups of infected Vero E6 cells were tested, the results were as follows: HV and Ot were both positive in infected Vero E6 cells passaged 2 times and the positive rate increased following the passaged times in HV and Ot infection groups, simultaneously or successively. However, in the groups which were infected with HV and Ot separately, the gene of HV or Ot could be detected in infected Vero E6 cells passaged only once and the positive rate increased following the times of the passaged. The positive rate was higher in the singly infected groups than in those infected simultaneously or successively. CONCLUSION: Coinfection of HV and Ot did exist in the hosts while HV and Ot could inhibit each other in the initial infection stage.

The influence of tsutsugamushi fever infection on the metabolism of blood lymphocytes of mice differing in susceptibility to Rickettsia tsutsugamushi and of rifampicine and tetracycline treatment was studied. In susceptible mice, the activity of oxidation-reduction processes in lymphocytes increased at 7--10 days of the disease and became normal upon recovery of the animals. In mice with low susceptibility this increase of the activity began and terminated much earlier, at 3--5 days. Treatment of the infection with antibiotics resulted in a rapid arrest of the disease and normalization of the lymphocyte activity. The relationship between susceptibility of the animals to R. tsutsugamushi and the activity of oxidation-reduction enzymes is discussed on the example of succinate dehydrogenase.
