Effects of subsoiling on lateral roots, sucrose metabolizing enzymes, and soil ergosterol in two Jeffrey pine stands.

We determined the effects of subsoiling on woody lateral roots and enzyme activities involved in stem carbon metabolism of 90- to 100-year-old Jeffrey pine (Pinus jeffreyi Grev. and Balf.) growing on the eastern side of the California Sierra Nevada Range. Twelve 1.0-ha plots were established on each of two sites. Four site treatments-thinning and subsoiling entire plots, thinning and no subsoiling, thinning and subsoiling skid trails only, and no thinning or subsoiling (undisturbed control)-were replicated three times on each site. Root excavations and assays of stem cambium sucrose metabolizing enzymes were carried out during the summer and fall of 1994 and 1995. Subsoiled plots had more roots < 1 cm in diameter exhibiting dieback than undisturbed plots. No relationship was found for stem cambial sucrose synthase (SS) activity among treatments or sites; however, a strong seasonal pattern was observed. Activity of pyrophosphate-dependent phosphofructokinase (PPi-PFK) was seasonal, whereas ATP-dependent phosphofructokinase (ATP-PFK) activity was constant throughout the seasons. Neither of the PFK activities responded to treatment or site. Among plots, ergosterol, a surrogate for estimating live fungal biomass, was present in highest concentrations in soil and rhizosphere samples from subsoiled plots.

Diameter growth of Scots pine (Pinus sylvestris) trees grown at elevated temperature and carbon dioxide concentration under boreal conditions.

We investigated the impacts of elevated temperature and carbon dioxide concentration ([CO2]) on diameter growth of Scots pine (Pinus sylvestris L.), aged about 20 years, grown with a low nitrogen supply in closed chambers at (i) ambient temperature and [CO2] (AT+AC), (ii) ambient temperature and elevated [CO2] (AT+EC), (iii) elevated temperature and ambient [CO2] (ET+AC), and (iv). elevated temperature and [CO2] (ET+EC). Each treatment was replicated four times. Diameter growth was monitored with a band dendrograph at 15-min intervals throughout the growing seasons of 1997, 1998 and 1999. Over the monitoring period, diameter growth began 2-3 weeks earlier in trees in the ET+EC and ET+AC chambers than in trees in the AT+AC and AT+EC chambers. However, the cessation of growth occurred about a week later in trees in the ET+EC, ET+AC and AT+EC chambers compared with the AT+AC chambers. The duration of the growing season was 115 and 108 days in the ET+EC and ET+AC chambers, respectively, and 95 and 84 days in the AT+EC and AT+AC chambers, respectively. The ET+AC and ET+EC treatments enhanced diameter growth most early in the growing season, whereas in trees in the AT+AC and AT+EC treatments diameter growth rate was highest in the middle of the growing season. Diameter growth rate leveled off more slowly in trees in the ET+EC and AT+EC treatments than in the other treatments. The growth response to elevated T, elevated [CO2] or both decreased with time and it was less than the maximum observed in other studies for small seedlings and under optimal growth conditions. Nevertheless, cumulative diameter growth for the 3-year period was 67% greater in trees in the ET+EC treatment, and 57 and 26% greater in trees in the AT+EC and ET+AC treatments, respectively, compared with trees in the AT+AC treatment. Over the 3 years, [CO2] had a statistically significant (P < 0.10) effect on both absolute and relative diameter growth, but the interaction between [CO2] and temperature was not significant.

Flooding, root temperature, physiology and growth of two Annona species.

The effects of root zone temperature (RZT) and flooding on physiology and growth of Annona glabra L. (pond apple) and A. muricata L. (soursop) were investigated. Trees planted in containers were exposed to RZTs of 5, 10, 20, 25 or 35 degrees C in controlled root temperature chambers. Trees at each RZT were either non-flooded (control) or continuously flooded. There were four replications over time for each treatment combination. Pond apple was more flood-tolerant than soursop. A combination of flooding and RZTs of 5 and 10 degrees C resulted in tree mortality of both species by Week 4. Only trees that appeared to develop morphological adaptations survived continuous flooding. In both species, net CO2 assimilation (A) decreased to nearly zero within 1 week following exposure to RZTs of 5 or 10 degrees C and became consistently negative over the remaining experimental period. Flooding reduced leaf chlorophyll index (measured with a SPAD meter), A and plant growth, and increased root electrolyte leakage from soursop. Optimum growth occurred at RZTs of 25 to 35 degrees C for non-flooded pond apple trees and at 20 to 25 degrees C for flooded trees. Soursop exhibited maximum growth at RZTs of 35 degrees C under non-flooded conditions and at 25 degrees C under flooded conditions.

Central actions of neuropeptide-Y may provide a neuromodulatory link between nutrition and reproduction.

Central injection of neuropeptide-Y (NPY) has been shown to attenuate secretion of LH in ovariectomized rats, rabbits, and monkeys. Several investigators have reported elevated concentrations of NPY in the central nervous system of undernourished animals. The relationship between nutrition and reproduction positions NPY as a potential neuromodulator involved in nutritionally induced changes in secretion of LH. Three experiments were conducted with the following objectives: 1) to examine the effects of NPY on secretion of LH in ovariectomized (OVX) ewes and the influence of estradiol-17 beta (E) on these effects; 2) to determine whether NPY may act through direct effects on the pituitary to influence secretion of LH; and 3) to determine changes in concentrations of NPY in laterocerebro-spinal fluid (CSF) of food-restricted ewes compared to well-fed ewes. In Experiment 1, OVX ewes with s.c. implants of E (OVX + E, n = 4) or no steroid treatment (OVX, n = 4) were fitted with intracerebroventricular (i.c.v.) and jugular cannulae. One of 4 doses of porcine NPY (pNPY; 0, 0.5, 5, or 50 micrograms) was injected i.c.v. and blood samples were collected every 10 min for 4 h prior to and following i.c.v. injection. Blood serum was assayed for LH. The experiment was replicated four times such that each ewe received each dose of pNPY. Mean concentrations of LH as well as frequency and amplitude of pulses of LH were attenuated in response to i.c.v. injection of pNPY in a dose-related manner in both OVX and OVX + E ewes.(ABSTRACT TRUNCATED AT 250 WORDS)

De novo protein synthesis by bovine uterine tube (oviduct) epithelial cells changes during co-culture with bull spermatozoa.

Polypeptides secreted by uterine tube epithelial cells (UTEC) may facilitate sperm cell capacitation in vivo. This experiment evaluated the effect of sperm-UTEC co-culture on de novo protein synthesis by epithelial cells of the tubal isthmus. Comparisons of the patterns of proteins secreted into medium were made between four culture groups incubated for 24 h in the presence of 35S-methionine: group 1, sperm cells alone; group 2, control UTEC monolayers; group 3, UTEC co-cultured with sperm cells; and group 4, UTEC partitioned by a diffusible membrane from sperm cells during culture. Two-dimensional PAGE followed by fluorography was used to analyze conditioned medium containing secreted proteins from each group. The experiment was replicated four times. Sperm cells alone secreted no detectable proteins, whereas control UTEC monolayers produced a wide array of polypeptides. Sperm cells attached to UTEC in co-culture within minutes, and the resultant protein profile for these UTEC differed markedly from that of the control UTEC. Several new proteins were seen only from co-cultured cells, whereas other protein groups that were present with UTEC alone were absent in the co-culture medium of group 3. The protein pattern expressed by UTEC partitioned from sperm cells (group 4) was intermediate between that of the group 2 controls and that of co-cultured UTEC (group 3). In summary, the attachment of sperm cells to the UTEC during co-culture changed the types and quantities of proteins secreted into the conditioned medium as compared to those of control UTEC monolayers.(ABSTRACT TRUNCATED AT 250 WORDS)

Mechanical aspects of epidural catheter insertion--prevention of catheter buckling by means of a needle hub insert.

Six commercially available epidural catheters were tested in a special apparatus designed to simulate epidural catheter insertion and quantitatively measure the buckling strength of these catheters. The experimental apparatus utilized a modified Tuohy needle and a specially calibrated force transducer. Catheters were inserted through the Tuohy needle in a manner similar to that employed clinically, and the maximum forces developed against the surface of the force transducer were recorded electronically. In addition, a custom-designed "needle hub insert" was tested for its ability to prevent catheter buckling during insertion of an epidural catheter through a Tuohy needle. Catheter buckling forces were measured with new catheters and also with catheters that had been damaged by previous insertion attempts. Both the new and damaged catheters were tested with and without the presence of a needle hub insert. Each combination of experimental conditions was replicated four times. The results showed a marked difference in the effective buckling strength of the catheters tested. Buckling strengths ranged from a minimum of 201 +/- 21 g (SD) to 418 +/- 15 g of force. The largest forces were obtained with a Deseret type catheter that had an internal plastic stylet. When comparing only new catheters without stylets, the buckling forces ranged from 201 to 285 g. All catheters, whether new or damaged showed an increase in the maximum buckling force with the use of the needle hub insert. Increased precision using countermatching in nested case-control studies.

Nested case-control studies in occupational cohorts are often used to estimate exposure effects when development of detailed exposure estimates for all cohort members is too costly. Duration of exposure, which can act as a surrogate for cumulative exposure, is often readily available for all cohort members. Langholz and others have recently proposed a method of control selection called countermatching, which uses data on the surrogate to determine which controls are selected from the risk set for a given case. This method may increase precision relative to the usual random sampling of the risk set. We compare countermatching with random sampling in a nested case-control study of silicosis among miners. Data on cumulative exposure were in fact available for all cohort members, enabling estimation of the parameter of interest in the full cohort. We conducted nested case-control analyses using 100, 20, 10, and 3 controls per case using random sampling and additional analyses using 3 controls per case with two different methods of countermatching. All analyses were replicated 50 times to explore the statistical properties of the estimated exposure parameter. We found that one of the countermatching methods markedly increased efficiency compared with random sampling. Countermatching using 3 controls per case yielded an approximate 25% increase in relative efficiency compared with random sampling; it was approximately equivalent to random sampling using 10 controls.

Season of birth and schizophrenia in Northeast Brazil: relationship to rainfall.

Although the association of schizophrenia and winter birth has been replicated many times in the Northern hemisphere, studies in the Southern hemisphere have been less consistent in their findings. A study from NE Brazil indicated the period between May and July, 3 months after the peak in rainfall, as a risk period for schizophrenia birth. We report findings from a random selection of charts (N = 1789) from the only public inpatient psychiatric facility in Ceará, Brazil (2 degrees to 5 degrees S). We compared the seasonality of birth in the schizophrenia group (N = 406) to multiple control groups: (1) psychosis not otherwise specified (N = 868), (2) other psychiatric diagnoses (N = 515), and (3) the average monthly birth rate in the general population. Patients with schizophrenia had a significantly greater risk of being born in the risk period compared with any of the control groups. There is a significant association between rainfall in a month and schizophrenia births 3 (p = 0.03) and 4 months (p = 0.01) later. This study corroborates findings of a significant seasonality in schizophrenia births in northeast Brazil.

An epigenetic state associated with areas of gene duplication.

Asynchronous DNA replication is an epigenetically determined feature found in all cases of monoallelic expression, including genomic imprinting, X-inactivation, and random monoallelic expression of autosomal genes such as immunoglobulins and olfactory receptor genes. Most genes of the latter class were identified in experiments focused on genes functioning in the chemosensory and immune systems. We performed an unbiased survey of asynchronous replication in the mouse genome, excluding known asynchronously replicated genes. Fully 10% (eight of 80) of the genes tested exhibited asynchronous replication. A common feature of the newly identified asynchronously replicated areas is their proximity to areas of tandem gene duplication. Testing of other clustered areas supported the idea that such regions are enriched with asynchronously replicated genes.

Incorporation of [S]Sulfate and [C]Bicarbonate into Karyotype-specific Polysaccharides of Chondrus crispus.

Gametophytic and sporophytic tissues of Chondrus crispus (Stackhouse) cultured in vitro were labeled with (35)S and (14)C. The major sulfated polysccharides isolated from the two karyotypes were characterized by KCI fractionation, immunoprecipitation, and infrared spectroscopy. Reproducibility of data has been demonstrated by an experiment using gametophytic T(4) strain with five replicates per time point. The rate of sulfate uptake was similar in haploid and diploid plants from a given area cultured for a similar time. Cultures from different sources cultured for different times showed different uptake and incorporation levels. Although sulfate uptake did not appear to be karyotype-related, the pattern of incorporation of (35)S and (14)C into polysaccharides was ploidy-specific.

Genetically based polymorphisms in morphology and life history associated with putative host races of the water lily leaf beetle, Galerucella nymphaeae.

A host race is a population that is partially reproductively isolated from other conspecific populations as a direct consequence of adaptation to a specific host. The initial step in host race formation is the establishment of genetically based polymorphisms in, for example, morphology, preference, or performance. In this study we investigated whether polymorphisms observed in Galerucella nymphaeae have a genetic component. Galerucella nymphaeae, the water lily leaf beetle, is a herbivore which feeds and oviposits on the plant hosts Nuphar lutea and Nymphaea alba (both Nymphaeaceae) and Rumex hydrolapathum and Polygonum amphibium (both Polygonaceae). A full reciprocal crossing scheme (16 crosses, each replicated 10 times) and subsequent transplantation of 1,001 egg clutches revealed a genetic basis for differences in body length and mandibular width. The heritability value of these traits, based on midparent-offspring regression, ranged between 0.53 and 0.83 for the different diets. Offspring from Nymphaeaceae parents were on average 12% larger and had on average 18% larger mandibles than offspring from Polygonaceae parents. Furthermore, highly significant correlations were found between feeding preference of the offspring and the feeding preference of their parents. Finally, two fitness components were measured: development time and survival. Development time was influenced by diet, survival both by cross type and diet, the latter of which suggest adaptation of the beetles. This suggestion is strengthened by a highly significant cross x diet interaction effect for development time as well as for survival, which is generally believed to indicate local adaptation. Although no absolute genetic incompatibility among putative host races was observed, survival of the between-host family offspring, on each diet separately, was lower than the survival of the within-host family offspring on that particular host. Survival of offspring of two Nymphaeaceae parents was about two times higher on Nymphaeaceae than on Polygonaceae, whereas survival of offspring of two Polygonaceae parents was 11 times higher on Polygonaceae than on Nymphaeaceae (based on untransformed data). Based on these results, we conclude that genetically determined polymorphisms in morphology and feeding preference exist in G. nymphaeae, resulting in differential performance. Furthermore, in each diet separately, offspring of between-host family crosses were less fit than offspring of within-host family crosses. These results support the hypothesis that within this species two host races can be distinguished.

The complete analysis of oxytetracycline pharmacokinetics in farmed Pacific white shrimp (Litopenaeus vannamei).

Lack of dosing information of the major antibiotics known as oxytetracycline (OTC) for the Pacific white shrimp (Litopenaeus vannamei) could have harmful impact on aquaculture in Thailand. The aim of this study was to detail complete pharmacokinetic information of OTC in the Pacific white shrimp. Sixty-four male L. vannamei weighing 14-22 g with carapace length of 2.30-3.00 cm in the standardized moulting stage of C-D(0) were used for the investigations. Single dose, 10 microg/g body weight OTC solution was administered intra-sinusally (i.s.), and the shrimps were then sampled in three replicates at time intervals of 0.25, 0.5, 2, 4, 6, 9, 12, 24, 48, 72, 170, 336 and 504 h postdose. OTC levels with time intervals in biological matrices including the hemolymph, abdominal muscle, and digestive gland of each sample were determined by validated high-performance liquid chromatography, and were analyzed with noncompartment and compartment models. A simplified two-compartment model was employed rather than a more complicated model, with additional digestive compartment if necessary. A significant portion of the OTC was found in the digestive glands, even though the OTC was administered i.s. The model indicated that the OTC was thus not only distributed into the tissue compartment, but also to the digestive gland, from where it was eliminated from the shrimp's body. The dispositional half-lives of all compartments was found to be 14-21 h. Approximately 60% of the drug elimination took place in digestive gland, which is proposed to be the major route of elimination.

Induction of dominant lethal mutations for control of the facefly, Musca autumnalis DeGeer.

1. Application of the sterile insect technique (SIT) to the facefly, Musca autumnalis DeGeer, is considered. 2. Six-day-old pupae were exposed to ionizing irradiation at doses in the range 100-1600 rads, each treatment being replicated six times. 3. Eclosion was unaffected but fecundity and fertility was inversely proportional to the radiation dose. 4. 1600 rads gave 97% dominant lethality of sperm in treated males and suppressed ovarian development in females. Irradiated flies did not recover fertility. 5. Irradiation of males and females did not affect insemination rates. 6. Sterile males showed a decline in longevity and reduced mating competitiveness. 7. Local eradication of M. autumnalis is considered to be feasible if sufficient diapausing sterile flies are stockpiled and released in early spring, followed by additional releases of non-diapausing flies in late spring and early summer, with further releases of diapausing sterile flies in late summer and autumn.

Divided genomes: negotiating the cell cycle in prokaryotes with multiple chromosomes.

Historically, the prokaryotic genome was assumed to consist of a single circular replicon. However, as more microbial genome sequencing projects are completed, it is becoming clear that multipartite genomes comprised of more than one chromosome are not unusual among prokaryotes. Chromosomes are distinguished from plasmids by the presence of essential genes as well as characteristic cell cycle-linked replication kinetics; unlike plasmids, chromosomes initiate replication once per cell cycle. The existence of multipartite prokaryotic genomes raises several questions regarding how multiple chromosomes are replicated and segregated during the cell cycle. These divided genomes also introduce questions regarding chromosome evolution and genome stability. In this review, we discuss these and other issues, with particular emphasis on the cholera pathogen Vibrio cholerae.

The pp67 mRNA assay in treatment and monitoring of cytomegalovirus disease in renal transplant patients in India.

The present report describes use of nucleic acid sequence-based amplification (NASBA) technology to detect pp67 mRNA of cytomegalovirus (CMV) in transplant patients in India. In our experience, pp67 mRNA assay was an accurate, rapid, and effective diagnostic tool to detect active CMV disease in 40.7% (50/123) of symptomatic transplant cases. This assay also allowed us to monitor CMV therapy. As part of the immunosuppressive regimen mycophenolate mofetil was found to increase the risk of developing CMV disease. All positive cases with this assay were subjected to antiviral therapy, with complete remission of the disease. At our center CMV NASBA assay has become the gold standard for the diagnosis of CMV disease in transplant patients.

An experimental analysis of a neurobehavioral motor intervention.

This study reports the effects of a neurobehavioral intervention approach on the motor skills of four children with cerebral palsy between 21 and 34 months of age. The intervention is based on the merging of neuromotor and behavioral approaches. The behavioral approach, using principles of 'how to teach', addressed motivational issues and allowed for precise definitions of expected outcomes which assisted in measuring acquisition and generalization of behaviors. The neuromotor approach provided 'what to teach' by focusing the intervention on underlying movement components (e.g., trunk rotation, weight bearing) rather than discrete milestone skills. The impact of the approach was assessed using a multiple-baseline design replicated three times. The children demonstrated the movement component by using it to perform both a treated exemplar skill (i.e. a motor skill requiring the movement component for execution) and an untreated exemplar skill.

Influence of cholinergic blockade on the development of epinephrine-induced ventricular arrhythmias in halothane- and isoflurane-anesthetized dogs.

The arrhythmogenic effects of anesthetic drugs are assessed using the arrhythmogenic dose of epinephrine (ADE) model. The purpose of this study was to determine the influence of cholinergic blockade (CB) produced by glycopyrrolate (G) on ADE in 1.5 minimum alveolar concentration (MAC) halothane (H)- and isoflurane (I)-anesthetized dogs. Eight dogs (weighing between 12.5 and 21.5 kg) were randomly assigned to four treatment groups (H, HG, I, and IG) and each treatment was replicated three times. Anesthesia was induced and maintained with H (1.31%, end-tidal [ET]) or I (1.95%, ET) in oxygen. Ventilation was controlled (carbon dioxide [PCO2] 35 to 40 mmHg, ET). G was administered 10 minutes before ADE determination at a dose of 22 microgram/kg (11 microgram/kg, intravenous [IV] and 11 micrograms/kg, intramuscular [IM]). The ADE was determined by IV infusion of epinephrine at sequentially increasing rates of 1.0, 2.5, and 5.0 micrograms/kg/min; and defined as the total dose of epinephrine producing at least four ectopic ventricular contractions (EVCs) within 15 seconds during a 3-minute infusion and up to 1 minute after the end of the infusion. Total dose was calculated as the product of infusion rate and time to arrhythmia. Data were analyzed using a randomized complete block analysis of variance. When significant (P < .05) F values were found a least significant difference test was used to compare group means. Values are reported as means +/- standard error. The ADE (micrograms/kg) for H, HG, I, and IG were 1.53 +/- 0.08, 3.37 +/- 0.46, 1.61 +/- 0.21, and > 15.00, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)

Use of small group with chronically ill children.

Using small groups with elementary school children is a valuable tool for dealing with the physical, social and emotional aspects of chronic disease. The self-esteem of the participants was improved and a means of providing peer support developed. Also, this particular format helped the children talk about their diseases, their feelings and encouraged them to talk with their parents. We would encourage others to try this approach. The model needs to be replicated many times before it could be fully validated because this was undertaken with a small sample of students. One recommended addition is to include at least one session with both parents and their children. This would provide a structured setting for parents and children to better understand each other. It also would be beneficial to have more group sessions with the parents to further explore some of the many issues that were raised.
