The effects of intravenous infusion of solutions with different colloid osmotic pressures on rat mesenteric lymph pressure.

The effects of intravenous infusion of solutions of varied colloid osmotic pressure on mesenteric lymph pressure were measured by a glass micropipette. The lymph pressure was 4.0 +/- 1.6 cm H2O between the second and third valve of rat mesenteric lymphatic vessels with diameter of 101 +/- 25 micron (mean +/- SD). Intravenous infusion of 1 ml/100g body weight (B.W.) of isotonic saline, 5% glucose solution, and 20% glucose solution increased the lymph pressure by 1.6 +/- 0.5 cm H2O, 1.5 +/- 0.3 cm H2O and 2.0 +/- 1.0 cm H2O, respectively. Infusion of 2 ml/100g B.W. of the same solutions increased the lymph pressure by 3.3 +/- 1.3 cm H2O, 3.3 +/- 1.6 cm H2O and 3.7 +/- 0.8 cm H2O, respectively. Infusion of hydroxyethyl starch solution, with colloid osmotic pressure of 25 mmHg, minimally altered the lymph pressure. One ml/100g B.W. infusion of low molecular weight dextran solution, with colloid osmotic pressure of 162 mmHg, decreased the lymph pressure by 1.3 +/- 0.9 cm H2O, and 2 ml/100g B.W. infusion of this solution decreased the lymph pressure by 2.0 +/- 1.0 cm H2O. The different effects on lymph pressure derive from the differences in colloid osmotic pressure of these solutions in conformity with the Starling hypothesis.

[Increase in the number of colloidal droplets in the hypothalamus of Natrix maura caused by dehydration].

The influence of osmotic stress on the number of colloid droplets in the magnocellular neurosecretory nuclei of the water snake Natrix maura, has been studied. Five experimental groups of five specimens each, were submitted to dehydration by immersion in sea water for several periods of time (3 to 60 h). The number of colloid droplets, identified by histochemical procedures, was counted in serial sections of the retrochiasmatic nuclei. The study of the mean of each group revealed that the amount of colloids increased with the time of permanence in the hyperosmotic environment (48 h elicited the greatest response). As a conclusion, dehydration seems to activate the hypothalamic magnocellular neurosecretory nuclei of N. maura and, consequently, increases the production of colloid droplets.

Fine needle aspiration of the thyroid. Distinction between colloid nodules and follicular neoplasms using cell blocks and 21-gauge needles.

Although fine needle aspiration (FNA) of the thyroid has been found to be useful in selecting patients for surgery in most cases, the cytologic differentiation of cellular colloid nodules from follicular neoplasms has not been possible because of the scanty amount of tissue obtained by this technique and because of the infrequent preparation of cell blocks, thus precluding appreciation of the tissue architecture. Review of the histology of nodular goiters and follicular neoplasms and comparison of their morphology in smears and cell blocks from FNA specimens with the histology of the surgically excised specimens in 74 cases revealed the presence of hyperplastic papillae and fragments of dilated follicles in the aspirates of most cases of colloid nodules. These two features appear to be specific for colloid nodules and are useful for distinguishing colloid nodules from follicular neoplasms. The merits of using a 21-gauge needle and of preparing cell blocks in thyroid studies are also discussed.

[A case of colloid cyst of the third ventricle].

Colloid cysts of the third ventricle are rare, accounting for less than 1% of all intracranial neoplasms. However, its incidence will increase with increased use of CT scan and MRI. The symptomatology is principally acute or chronic hydrocephalus obstructing the foramen of Monro, and the origin of this cyst is controversial, neuroepithelial or endodermal. We report a case of colloid cyst of the third ventricle, associated with cavum veli interpositi. The patient, a 46-years-old female, had suffered from headache, ataxic gait and tremor lasting 6 months. CT scan and homogenous high signal intensity by SR (2450/450) MR image, detected a small round mass in the anterior part of the third ventricle that was of homogenous high density, but without the effect of enhancement. The lateral ventricle was markedly dilated, and a large cavum veli interpositi was found between the third ventricle and the corpus callosum. The cyst was totally removed by transventricular approach, and the result was satisfactory. We reviewed literatures and discussed MRI findings of colloid cysts of the third ventricle. Because of the presence of microvilli covered with coating materials seen in electron microscopic study, we suggested that the colloid cyst was endodermal in origin.

Preservation of canine hearts after warm ischemia (zero to thirty minutes) and one to two days of hypothermic storage. A comparative analysis of crystalloid and colloid solutions with different osmolarity and ion composition.

The effect of extracellular crystalloid (Ringer's) and colloid (silica gel fraction [SGF]) solutions, and intracellular crystalloid (Sacks) and colloid (modified silica gel fraction [MSGF]) solutions for canine heart preservation in a 24 to 48 hour model of hypothermic storage and zero to 30 minutes of warm ischemia was compared. Canine hearts flushed with an intracellular colloid solution (MSGF) had better survival rates after transplantation than did the hearts flushed with intracellular crystalloid solutions (Sacks). Better survival results also were observed in the group of hearts flushed with extracellular colloid (SGF) solutions than extracellular crystalloid (Ringer's) solutions. The most important theoretical factor in heart preservation appears to be hyperosmolarity and elevated concentration of potassium, proteins, and glucose.

[Hepatic scintigraphy using 99mTc-colloid in liver cirrhosis in children].

A total of 26 children aged 2 to 14 with the initial (6), formed (14) and terminal (6) stages of liver cirrhosis were examined by a method of radionuclide scintigraphy with 99mTc-colloid. 34 children aged 7 to 14 examined in the catamnesis of virus hepatitis, were controls. A set of indices characterizing function of the reticuloendothelial system (RES), the effective hepatic blood flow, metric parameters of the liver and spleen were obtained from an analysis of the curves of the heart, liver and spleen area, and digital imaging of the liver with the marked costal arch. It was shown that at the initial stage of disease indices of the time course of the radioactive colloid were of compensated nature. Spleen function was elevated, liver and spleen sizes were increased. The formed stage was characterized by the signs of subcompensation of liver function: changes of indices of RP retention in the blood, a decrease in the indices of the total and hepatic radioactive colloid. The terminal stage was characterized by marked disorder of liver RES function which was not compensated for by a high splenic uptake, image deformation and focal RP distribution. Irrespective of a stage of disease the syndrome of portal hypertension was shown to manifest itself in splenomegaly and an increase in the radioactive colloid uptake by the liver over 15%. The accuracy of the set of signs was 90%.

Immunoglobulin IgG detected by ELISA in bovine pituitary intraglandular colloid, a non-vascular holocrine secretion of intermediate lobe cells.

The purpose of this study was to determine the presence of immunoglobulin G in bovine pituitary intraglandular colloid (IGC). Intraglandular colloid, housed in the intraglandular lumen (residual lumen), is the holocrine secretion of the marginal layers of intermediate lobe (IL) cells. Immunoglobulin G was detected and quantitated in bovine IGC using an Enzyme Linked Immunosorbent Assay (ELISA). This study establishes the presence of immunoglobulin G in the intraglandular colloid at levels that are of the order of 10 micrograms per mg of colloid. The existence of this molecule in material scrupulously prepared to exclude the possibility of serum contamination in this normally avascular compartment is a unique phenomenon.

Colloid shift following blunt trauma.

Thirty-three patients who underwent spleen imaging with [99mTc]sulfur colloid following blunt trauma to the left chest and/or left upper abdominal quadrant were evaluated. Nineteen of these patients were found to have reversed liver-spleen activity ratios (colloid shift). The incidence of this finding was surprising in view of the absence of any predisposing factors in these patients. Colloid shift was seen as early as 2 hr after trauma and was found to persist as long as 188 days after the traumatic event. The explanation for colloid shift in this group of patients is unknown, but the results suggest that it is secondary to reticuloendothelial system stimulation.

Use of 99mTc-sulfur colloid to assess lymphatic dysfunction in filarial infection.

Investigations of lymphatic dysfunction in animals infected with filarial parasites has been hampered by a paucity of techniques to measure efficiency of lymphatic drainage. In this study a 99mTc-sulfur colloid technique was used to assess the efficiency of lymphatic drainage in Patas monkeys infected with filarial nematodes. In all 15 uninfected hind limbs there was rapid and consistent appearance of labeled colloid in the primary lymph node (popliteal) and subsequently in the secondary nodes (abdomino-pelvic) in 11 of 15 limbs. In contrast, in all eight limbs tested 1-9 months after infection there was reduced rate of migration of the colloid and initial appearance in the abdomino-pelvic region: subsequent accumulation was seen in the popliteal region in only four of the limbs. This data indicated that lymphatic vessels were blocked and that collateral vessels channeled the colloid to the secondary lymph nodes. The lymph flow patterns demonstrated by the isotope technique were supported at autopsy.

Bovine pituitary intraglandular colloid: a transport medium.

Bovine pituitary intraglandular colloid is formed by the cyclic degeneration of marginal cells lining the intermediate lobe and is housed in the intraglandular lumen (residual lumen). The lumen communicates with the subarachnoid cerebrospinal fluid space by well defined channels. Electrophoresis in acrylamide gel shows bovine pituitary intraglandular colloid as having double protein bands identical to the protein in bovine and human cerebrospinal fluid. These studies demonstrate two distinct bands in the gamma region for colloid, not apparent in the normal bovine or human cerebrospinal fluid due to the low concentration of gamma globulins. We conclude that pituitary colloid, laden with immunoreactive fragments of various pituitary hormones, is discharged from the hypophyseal intraglandular space, directly into the subarachnoid cerebrospinal fluid space.

Adjuvant therapy of Dukes' C colon cancer by intra-arterial P-32 colloid for internal radiation therapy of the liver.

To prevent probable occult metastatic liver cancer from progressing to clinical disease, the author used internal radiation therapy as an effective adjuvant to surgical excision of primary Dukes' C colonic cancer. A calculated radiation dose of 5000 rads was delivered to the liver by injecting radioactive 32-P chromic phosphate colloid through the superior mesenteric and celiac arteries. When this was done, the colloid passed through the intestines and was mixed thoroughly with the blood and delivered to the liver by the portal vein. The Kupffer cells in the liver trapped the colloid, and a minimum amount passed through the liver and got into the general circulation. This kept the amount of colloid deposited in the bone marrow to a minimum. In a phase-I pilot study in which nine patients were treated, no serious side effects were noted. In eight patients, the liver has remained free of cancer for more than 1 year.

Localization of osteomyelitis with 99mtechnetium sulphur colloid.

Thirty-two patients with fractures of the limbs were studied 15-30 min following 99mtechnetium sulphur colloid injection. In 17 of the 19 patients where osteomyelitis was superimposed on the various fractures, significant uptake of sulphur colloid was noted at the fracture sites. The sensitivity for the test was 89.5%. In 12 of the 13 cases where osteomyelitis was absent, the sulphur colloid scan was negative giving a specificity of 92.3%. Hence a technetium sulphur colloid scan has shown to be a successful method for detecting osteomyelitis complicating a limb fracture.

[Vascular disorders in irritable hip disclosed by dynamic scintigraphy with radioactive colloids].

The aim of this survey was to determine the vascular disturbances occurring during transient synovitis (observation hip, "hanche irritable"). Within these last two years thirty children were admitted with painful hip, limp and restriction of the passive motion in the hip joint. Legg-Calve-Perthes disease was excluded by roentgenographic analysis and bone scintigraphy. Osteoblastosis was studied by using 99mTc methyl-diphosphonate. Another tracer was necessary to investigate the blood supply into the affected femoral head : 99mTc marked colloids were injected. Immediately after the intravenous administration of the isotope comparative activity was recorded every ten seconds. Computerized information allowed the description of four different stages, i.e. arterial supply, filling peak, venous elimination and later recirculation. Nineteen (63%) affected children presented a decrease in isotopic activity during the first two stages. The proposed interpretation consists in a selective decrease in blood supply in the femoral head. Venous circulation was rarely disturbed. Treatment consisted in bedrest with skin traction. In this series, no aspiration of the joint was performed. Eight days after admission dynamic scintigraphy with marked colloids revealed a highly significant decreased asymmetry between the two femoral heads. Only seven patients (23%) presented arterial hypoperfusion. Fifteen days later only three cases remained abnormal. Two of them presented some weeks after the regression of the symptoms an asymmetrical bone scintigraphy and secondarily the roentgenographic features of Legg-Calve-Perthes disease. Radioactive colloids seem to be useful to diagnose transient synovitis and to determine the prognosis. Although its etiology is still unknown, transient synovitis seems to be characterized by vascular disturbances consisting in a decrease in blood supply in the affected femoral head. Persistence or recurrence of abnormal colloid scintigraphy is indicative of a poor prognosis.

The effect of repeated injections of 99mTc-antimony-sulphide colloid on the RES of the rat--histopathologic study.

Experiments on inbred albino rats were performed to test the effect of repeated administrations of 99mTc-antimony sulphide colloid on the integrity of the RES. Experiments were carried out in two batches, one with the preformed colloid given in increasing doses, and the other with the labelled colloid in increasing doses. In all, 9 groups of animals including a control group were tested. Studies included: organ weights of liver, spleen, lungs and kidneys, histopathology, contact macroautoradiography, microautoradiography and 99mTc activity distribution in organs. Results showed no macroscopic or microscopic evidence of tissue damage after repeated injections up to a total dose of 18 mCi 99mTc, 142.2 mg PVP, and 693 microgram Sb, given in 9 equivalent doses at one week intervals. It is concluded that repeated administrations of 99mTc-antimony sulphide colloid for liver scintigraphy are safe.

Lymphatic clearance of radioactive sulfur colloid.

Lymphatic clearance of radioactive sulfur colloid is measured in the hind limb of five mongrel dogs. A solid state Si(Li) detector is placed onto the skin to continuously record activity over the site of subcutaneous injection. Decrease in activity follows an exponential decay which is modeled as a dual decay occurring from both the radioactive decay of the tracer and the lymph clearance of the tagged sulfur colloid. The calculated decay constants for lymph clearance flow per tissue volume result in a mean value of 0.233 +/- 0.077 ml/hr/ml which is consistent with results of other investigators. Adjacent lymph nodes are monitored with a scintillation detector to show that the colloid is absorbed by the lymph vessels. The carotid artery and liver are similarly monitored to show that there is little or no capillary absorption of the tagged colloid.

[Measurement of the colloid osmotic pressure from plasma density by means of the mechanical oscillator technique (author's transl)].

The colloid osmotic pressure of plasma is a clinically and physiologically important variable. Like the plasma density, it is a function of the total concentration of plasma proteins. Thus, plasma colloid osmotic pressure can be determined from measurement of the plasma density. Minor variations are caused by variations of the albumin-globulin ratio and, much less frequently, by increased concentration of low molecular substances in the plasma. The direct measurement of the colloid osmotic pressure is a time consuming and complex procedure. The determination of the plasma density using the mechanical oscillator technique is a simple and fast procedure. From our results we were able to derive and prove the applicability of an equation which permits the calculation of the colloid osmotic pressure from the measured values of the plasma density. The plasma volume needed for the measurement is 0.2 ml, the measuring time amounts to a few seconds.

Thyrotropin stimulated colloid droplet formation in the thyroid of hypophysectomised mice after calcitonin administration.

The influence of calcitonin on colloid droplet formation in the thyroid gland of hypophysectomised mice after thyrotropin (TSH) stimulation was investigated. Calcitonin was injected 30 min before TSH administration. 30 min after TSH the colloid droplet index in the thyroid cells was twice as low in the calcitonin group in comparison with the control one (0.111 +/- 0.020, 0.228 +/- 0.025 respectively). 60 min after thyrotropin treatment the number of colloid droplets was higher in the animals treated with calcitonin (0.135 +/- 0.093) in comparison with the control group (0.083 +/- 0.025). The possible mechanism of calcitonin action on colloid droplet formation is discussed.

[Limited value of colloidal tests in CSF in the diagnosis of multiple sclerosis].

In 136 clinically definite MS cases the comparison was performed of intrathecal IgG production and detection of oligoclonal bands, evaluated by immunological methods with results of colloidal tests. The obtained results show, that the left side precipitation in colloidal tests indicates the presence of intrathecal IgG production, what was a definite value in the diagnosis of MS. However, the value of colloidal tests is very much limited by several times lower sensitivity of colloidal tests in detection of intrathecal IgG production in comparisons with the evaluation of intrathecal production and detection of oligoclonal bands by the immunological methods.
