Mould spores and yeasts in outdoor air.

Most inviable spore counts of outdoor air in Europe are based on Burkard spore trap results. For viable counts different kinds of slit samplers and Andersen sampler are used. Results of viable and inviable sampling methods are not completely comparable, but the results complete each other. Spore frequencies in outdoor air are almost equal in different Nordic countries. Cladosporium is the most important spore type everywhere. In South Finland it accounts for ca. 85% of viable and ca. 50% inviable spore counts. The frequencies are lower in the country than in town. Cladosporium frequencies decrease northwards so that in Lapland the percentage is only ca. 5 and the maxima are 1/4-1/5 of those in South Finland. The frequencies of Alternaria spores are 4-5 times lower in Nordic countries than in Middle Europe. E.g. in Turku weekly maxima have been ca. 250 spores X m-3, in the country even lower. Penicillium accounts for ca. 4% of all colonies in viable counts in South Finland, Botrytis 2%, and yeasts also ca. 2%. The most important yeasts are Cryptococcus albidus, Cr. laurentii, Sporobolomyces roseus and Rhodotorula species. Actinomycetes very seldom occur on culture plates, only 0.02% of all.

[New analytic criteria for the characterization of wines].

Isotopic analysis, determination of radioactivity are factors allowing a better characterization of wines. The analysis of the composition in stable isotopes of the constituants of wine can provide new analytic criteria for characterizing wines. Thus, the concentration of 13C in ethanol can allow the disclosure of an addition of cane sugar or maize sugar to the musts. A study of the composition in stable isotopes of the water of wines of a certain region made it possible to show a correlation between the amount of D and 18O in the water of wine, and the alcohlic concentration. The setting up of the curve showing the decrease of the radioactivity of 14C in the Girondewines allows a determination of the date, accurate to within one year, of these wines. The enzymatic determination of the gluconic acid present in the wines produced from crops attacked by Botrytis Cinerea allows one to estimate the type of rottenness of the grape; the gluconic acid, which is only present in very small quantities in wines produced from sound grapes, represents about 50 p. 100 of the fixed acides of the grapes affected by "noble rottenness" and up to 90 p. 100 in the case of adultarated grapes. Thanks to the recults obtained in 1976, standards are proposed to allow the characterization of the wines produced from grapes affected by "noble rottenness", so that they can be distinguished from, on the one hand, the wines produced from sound grapes, and on the other hand, the wines produced from adultarated grapes.

Incidence of fungal spores at the homes of allergic patients in an agricultural community. III. Associations with local crops.

A predominantly agricultural community in California was surveyed for prevalent fungal spores during a 12-month period. Alternaria, Macrosporium and Stemphylium were recovered during asparagus and strawberry harvesting times year-round. Fusarium and Botrytis were less frequently associated with the strawberry harvest and were recovered only during the first quarter of the year. Epicoccum was recovered in the north end of the Salinas valley in low numbers throughout the year and was strongly associated with the strawberry and artichoke harvest. Aureobasidium (Pullularia) recovery occurred in different locations according to season, correlating somewhat with the cabbage harvest as well as with the harvest of strawberries. Recovery of the pigmented yeats showed strong correlation with the local growing season for lettuce. Cladosporium was prevalent year-round but did not appear to be signficantly affected by changing argicultural conditions. These data have permitted the predictability of mold aeroallergens with medical applications.

[Antifungal and antimicrobial activity of beta-ionone and vitamin A derivatives].

The antifungal and antimicrobic activity of some derivatives of beta-ionon and vitamin A was studied. These compounds (citral, pseudo-ionon. beta-ionon aldehyde C14, ketone C18 and its derivatives--4,18-diketone, alcohol C18, semicarbazide ketone C18), as well as vitamin A and its derivatives--retinal, acetate, retinoic acid--differ in composition, structure and substituents of C-atoms in beta-ionon ring and in polyenoic chain. Fusarium solani, Botrytis cenerea and Verticillum dahliae II, race 447 were used as test organisms when studying the antifungal activity. When studying the antimicrobic activity, ketone C18 and alcohol C18 were tested using the museum strains Staphylococcus aureus 209 P, hemolytic Streptococcus pyogenes FF-38, Streptococcus sp. TOM-1606, Micrococcus luteus 2665, as well as the strains Staphylococcus aureus isolated from pyoderma patients. The antifungal activity was determined by inhibition of spore germination, and the antimicrobic activity--by the value of areas of growth inhibition of the test organisms on the agar medium. All the compounds mentioned above possessed the antifungal activity against all the phytopathogenes used. The degree of this activity depends on the composition and structure of both the cyclic part and polyenoic chain. on the number of conjugated unsaturations, substituents and end groups of C-atom. Ketone C18 is the most active one. It inhibits spore germination by 100-94% at concentrations ranging from 0.05 to 0.005% and 24 h exposure, and by 100% at the concentration of 0.1% and 72 h exposure. Alcohol C18 possesses almost the same antifungul activity.(ABSTRACT TRUNCATED AT 250 WORDS)

A volumetric-aerobiologic study of seasonal fungus prevalence inside and outside dwellings of asthmatic patients living in northeast Netherlands.

Air sampling with an Andersen sampler was carried out over a period of 21 mo, in 28 houses of asthmatic patients with an allergy for moulds. Inside, the livingroom, bedroom, and kitchen were studied, and an additional outside sample was always taken. Indoors, Penicillium showed the highest concentration, especially in the autumn and winter months, while Aspergillus and yeasts were also found frequently. The outside air was dominated by Cladosporium and Alternaria during summer months and Botrytis, Aspergillus, Mycelia sterilia and Basidiomycetes all year round. The patients showed high sensitisation towards Aspergillus, irrespective of the concentrations of the airborne fungi.

Persistence of carbaryl residues in and on unprocessed and processed cauliflower curds.

Carbaryl was applied three times at 15 days interval to cauliflower (Brassica oleracea var. botrytis L.), and its persistence was determined spectrophotometrically at 0, 1, 3, 6, 10, and 15 days after the first and last treatments. In unprocessed cauliflower the prescribed tolerance level for carbaryl residues of 5 ppm was only reached on the sixth day, in processed cauliflower on the third day after application. Pretreatment with washing + boiling/cooking on the other hand led to a considerable decrease of the carbaryl residues already on the day of application. The lower carbaryl values encountered in all variants after the last application are attributed by the authors to a thinning effect due to an increased cauliflower mass.

[Antifungal activity of 3,3'-diindolyl-2,2'-tetrasulfide].

The compound 3,3'-diindolyl-2,2'-tetrasulfide was studied for in vitro and in vivo fungistatic activity against various fungi causing human and plant diseases. The compound showed marked activity in vitro especially against dermatophytes and in the agrarian field against Botrytis cinerea. When the low phytotoxicity of the compound is taken into account, it is concluded that the compound has considerable potential as an anticryptogam.

Characterization of two DNA polymerases from cauliflower inflorescence.

Two distinct DNA polymerases, A and B, isolated from a rapidly growing apical tissue of cauliflower (Brassica oleracea var. botrytis) inflorescence have been characterized, and compared with DNA polymerases, alpha and beta, from mouse myeloma. Polymerase-A bears a strong resemblance to polymerase-alpha from mammalian cells in all properties examined. The character of polymerase-B is also quite similar to polymerase-beta of mammalian cells in chromatographic elution properties, template-primer utilization, sensitivity to inhibitors, response to KCl or KPi concentration, and other requirements for maximal activity, although it has a higher molecular weight (approx. 78,000) even in the presence of 0.25 M KCl that polymerase-beta (mol. wt. less than or equal to 50,000) of mammalian cells. This type of DNA polymerase has not been reported to exist in the plant system.

Chemoprevention of azoxymethane-induced colon carcinogenesis by dietary feeding of S-methyl methane thiosulfonate in male F344 rats.

Modifying effects of dietary exposure of S-methyl methane thiosulfonate (MMTS) isolated from cauliflower Brassica oleracea L. var. botrytis on rat colon carcinogenesis induced by azoxymethane (AOM) and on the expression of cell proliferation biomarkers were investigated in two experiments. In experiment 1, male F344 rats were given three s.c. injections of AOM (15 mg/kg body weight) and fed 100 ppm MMTS for 5 weeks, starting 1 week before the first dose of AOM. The frequency of colonic aberrant crypt foci was determined at 5 weeks after the start. Feeding of 100 ppm MMTS for 5 weeks significantly decreased the number of aberrant crypt foci/colon. Colonic mucosal ornithine decarboxylase activity and the number of silver-stained nucleolar organizer regions per nucleus in colonic epithelium were significantly decreased by MMTS treatment compared with those of AOM alone. In experiment 2, effects of dietary feeding of MMTS at two doses (20 and 100 ppm) during the postinitiation phase on intestinal tumorigenesis initiated with AOM were investigated by using a long-term experiments in male F344 rats. Incidence of intestinal neoplasms of rats fed MMTS-containing diets after AOM exposure were reduced in a dose-dependent manner. Feeding of MMTS during the postinitiation phase decreased the number of aberrant crypt foci/colon, colonic ornithine decarboxylase activity, 5-bromodeoxyuridine-labeling index in colonic epithelium, and polyamine level in blood compared with those of AOM alone. These results suggest that MMTS might be a possible chemopreventive agent for intestinal neoplasia.

Antifungal activity of Myxococcus species 1 production, physicochemical and biological properties of antibiotics from Myxococcus fulvus S110 (Myxobacterales).

Chloroform extracts of the culture supernatant of a strain of Myxococcus fulvus isolated from soil were fungistatic and prevented germination of spores of Botrytis cinerea. The antibiotics were produced during the exponential phase of growth and the effects of altering medium composition are described. The activity was fractionated into neutral, acidic and basic fractions. The neutral fraction had a molecular weight of 244 and was tentatively identified as 11-phenyl-undecadiene-2-ol. The acidic fraction was primarily n-hexadecanoic acid. The basic fraction contained components [N-(2-phenylethyl)-acetamide, and compounds tentatively identified as 1-isoquinoline carbonitrile and substituted quinolines] which have not previously been described in the myxobacteria.

Isolation and characterization of a benomyl-resistant form of beta-tubulin-encoding gene from the phytopathogenic fungus Botryotinia fuckeliana.

As an initial step to develop a DNA-mediated transformation system using benomyl resistance as a dominant selectable marker in the phytopathogenic fungus Botryotinia fuckeliana (anamorph, Botrytis cinerea), we have constructed a phage lambda genomic DNA library of a benomyl-resistant strain 91T-1, and a beta-tubulin-encoding gene benA was isolated, cloned and sequenced. Southern blot analysis suggested that a single copy of benA is present in the genome of B. fuckeliana. The benA gene is composed of seven exons which are separated by six introns of 52 to 135 bp. The intron consensus sequences are similar to those of other fungal genes. The deduced amino acid sequence (447 amino acid residues) is highly homologous to those of other fungal beta-tubulin-encoding genes. Comparison of the sequences around codons 198 and 200 in benomyl-resistant and sensitive strains revealed that the benAHR allele from the benomyl-resistant strain 91T-1 contained a mutation at codon 198 from GAG (Glu) to GCG (Ala), which has been correlated with high resistance to benzimidazole fungicides including benomyl in various filamentous fungi.

Aeroallergen sensitivity of Thai patients with allergic rhinitis.

The aim of this study was to determine the aeroallergen sensitivity of allergic rhinitis patients. A total of 100 cases (female: 59, male: 41, aged between 10-59 years, mean age 27.9 years) who were diagnosed with allergic rhinitis by history and clinical presentation, underwent a prick skin test with 30 aeroallergens, and the important sensitizing allergens were assessed. Skin test reactivity showing > or = 3 mm wheal with erythema as the positive skin test, was recorded. The results of patients with positive skin tests follow. TREES: acacia 19%, mango 16%, coconut 12%. GRASSES: bermuda 17%, johnson 21%, timothy 16%, bahia 16% orchard 18%. WEEDS: pigweed 16%, kochia 14%. MOLDS: alternaria 11%, cladosporium 11%, aspergillus 12%, penicillium 16%, helminthosporium 16%, botrytis 15%, rhodotorula 20%, fusarium 26%, curvularia 26%, smut mix 11%, rust 9%. EPIDERMALS: cat 29%, dog 28%, feathers 37%. INDOOR ALLERGENS: house dust 72%, D. pteronyssinus 76%, D. farinae 79%, American cockroach 60%, German cockroach 41%, kapok 30%. Eighty-five percent of patients sensitive to house dust mites were positive to both D. pteronyssinus and D. farinae, indicating substantial cross-reactivity. The study shows that the house dust mite and the cockroach are important aeroallergen sensitizers among the Thai population, since more than half the patients were skin-test positive to the house dust mite and the cockroach.

Distribution and persistence of emamectin benzoate at efficacious concentrations in pine tissues after injection of a liquid formulation.

In an earlier paper the authors reported the creation of a novel emamectin benzoate 40 g litre(-1) liquid formulation (Shot Wan Liquid Formulation). The injection of this formulation exerted a preventative effect against the pine wilt disease caused by the pine wood nematode, Bursaphelenchus xylophilus (Steiner & Buhrer) Nickle, and this effect lasted for at least 3 years. The present study was carried out to show experimentally that the marked effect of this formulation was due to the presence and persistence in pine tissues of sufficient amounts of emamectin benzoate to inhibit nematode propagation. A cleanup procedure prior to quantitative analysis of emamectin benzoate by fluorescence HPLC was devised. The presence of the compound in concentrations sufficient to inhibit nematode propagation in the shoots of current growth and its persistence for 3 years explained the marked preventative effect. Non-distribution of emamectin benzoate in some parts of the lower trunk suggested that the formulation should be injected at several points for large trees in order to distribute the compound uniformly to lower branches.

Genetic variation in Beauveria bassiana populations associated with the darkling beetle, Alphitobius diaperinus.

A study was conducted to assess genetic variation within and among populations of Beauveria bassiana (Deuteromycotina: Hyphomycetes) associated with the darkling beetle, Alphitobius diaperinus (Coleoptera: Tenebrionidae), using RAPD markers. A hierarchical collection of samples (strains from the same insect specimen, from insects from the same location, and from insects from different locations) was obtained from infected beetles from North Carolina (NC) and West Virginia (WV), USA. Ten primers resolved 81 strains into 80 distinct multiband phenotypes reflecting the substantial amount of variation that was present. Variation present within populations was evident not only in the separation of each strain as a distinct multiband phenotype but also in the separation of strains within a population into separate clusters. Among populations, a group sharing more than 89% similarity was observed among all the strains from Martin Co. and Greene Co., NC and 61% of the strains collected from WV. Some genetic differentiation was present among the other populations but the separation was not distinct with a few strains from some populations showing greater affinity to strains from other collection sites.

NaAGP4 is an arabinogalactan protein whose expression is suppressed by wounding and fungal infection in Nicotiana alata.

Arabinogalactan proteins (AGPs) are proteoglycans secreted by plant cells that have been implicated in plant growth and development. Most AGPs cloned to date possess highly labile glycosylphosphatidylinositol (GPI) lipid anchors. These anchors transiently attach AGPs to the plasma membrane before they are released into the cell wall following GPI anchor hydrolysis. We have isolated and partially sequenced the protein core of an AGP purified from styles of Nicotiana alata. The protein sequence data were utilised to clone the AGP's gene, NaAGP4. This AGP shares about 78% sequence identity with the tomato AGP LeAGP-1. RNA gel blot analyses of different plant organs indicate that NaAGP4 is expressed in the same tissues and at similar levels as LeAGP-1. Furthermore, NaAGP4 like LeAGP-1 is rapidly suppressed by tissue wounding and by pathogen infection. We believe NaAGP4 and LeAGP-1 are the first described examples of orthologous AGPs from different plant species. In contrast, another AGP from N. alata, NaAGP1, is comparatively unaffected by wounding and pathogen infection, although this AGP is expressed in similar tissues and at similar levels as NaAGP4.

Novel oxindole derivatives and their biological activity.

The antifungal activity of fourteen novel derivatives of oxindole with side chain was studied using representatives of toxinogenic, phytopathogenic and dermatophytic filamentous fungi. Derivatives with exocyclic C=C bond in position C-3 exhibited a higher antifungal activity compared with derivatives with an exocyclic C-C bond in the same position. The strongest antifungal effects were shown by 3-(-2-thienoylmethylidene)-indol-2(3H)-ones.

Expression regulation of the endochitinase chit36 from Trichoderma asperellum (T. harzianum T-203).

The presence of the endochitinase CHIT36 from Trichoderma harzianum TM was assessed in several antagonistic Trichoderma strains belonging to different molecular taxonomic groups. CHIT37 from T. harzianum CECT 2413 was sequenced and found to display 89% homology with CHIT36 at the amino acid level. Northern analysis showed that chit36Y from T. asperellum is regulated both by glucose and nitrogen levels. Stress conditions, colloidal chitin and N-acetyl-glucosamine are effective inducers of this gene. The promoter of chit36Y was cloned and was used to direct expression of a gfp reporter gene in Trichoderma transformants. Confrontation experiments with the plant pathogen Rhizoctonia solani revealed that direct contact between the fungi is not necessary for gfp expression. The R. solani-inducing factor appears to be a soluble molecule capable of diffusing through a dialysis membrane (<12 kDa). CHIT36 recombinant protein from the yeast Pichia pastoris was active against different phytopathogens, confirming the importance of this endochitinase in the mycoparasitic activity of Trichoderma antagonistic strains.
