[Effect of huangqi zengmian powder on interstitial response in patients with esophageal cancer at peri-operational period].

OBJECTIVE: To observe the effect of Huangqi Zengmian Powder (HQZMP) on interstitial response of esophageal cancer at peri-operational period. METHODS: Thirty-seven patients of esophageal cancer were treated with HQZMP for 4 weeks peri-operationally. The pathological interstitial changes (including mastocyte, microvessels and lymphocytic response) in resected esophageal cancer sample were observed using toluidine blue stain. RESULTS: As compared with the non-HQZMP treated control group, quantity of interstitial mastocyte, focal lymphocytic aggregation and interstitial microvessel damage in the HQZMP treated group were more obvious (P < 0.01). CONCLUSION: HQZMP could improve the immune function of esophageal patients in peri-operational period.

Tannic acid is an inhibitor of CXCL12 (SDF-1alpha)/CXCR4 with antiangiogenic activity.

PURPOSE: Increasing evidence suggests that interaction between the chemoattractant CXCL12/stromal cell-derived factor-1alpha and its receptor CXCR4 plays a pivotal role in the metastasis of various tumors. Our previous studies showed that multi-component Chinese herbal medicines inhibited the effects of CXCL12/CXCR4. As a result of sequential chromatographic fractionation of one herbal medicine ingredient, Lianqiao (fruit of Forsythia suspensa), we observed that tannins were, at least in part, responsible for this activity. The aim of this study was to assess the anti-CXCL12/CXCR4 activity of a commercial tannic acid and evaluate its potential to inhibit tumor cell migration and angiogenesis in vitro. EXPERIMENTAL DESIGN: The inhibitory effect of tannic acid on CXCL12/CXCR4 was measured by chemotaxis assay, ligand binding assay, and fluorescence-activated cell sorter analysis. The antiangiogenic effect of tannic acid was assessed by in vitro endothelial cell tube formation. RESULTS: Tannic acid, at nontoxic concentrations, specifically inhibited CXCL12-induced human monocyte migration (IC(50), 7.5 micro g/ml) but did not inhibit CCL2-, CCL3-, CCL5-, formylmethionylleucylphenylalanine (fMLP)-, or C5a-induced migration. The compound markedly blocked CXCL12 binding to THP-1 cells (IC(50), 0.36 micro g/ml). Tannic acid also inhibited CXCL12-induced, but not epidermal growth factor-induced, migration of MDA 231 breast tumor cells. Additionally, 0.5 micro g/ml of tannic acid selectively inhibited CXCL12-mediated, but not basic fibroblast growth factor- or endothelial cell growth supplement-mediated, bovine aorta endothelial cell capillary tube formation. CONCLUSION: These studies indicate that tannic acid is a novel selective CXCL12/CXCR4 antagonist and consequently may provide a mechanistic basis for the reported antitumor and anti-inflammatory properties of tannic acid.

[Influence of huangqi on TGF-beta 1 inducing extracellular matrix secretion of human peritoneal mesothelial cell].

OBJECTIVE: To study the effect of huangqi on peritoneal sclerosis and its possible mechanism. METHODS: Isolated human peritoneal mesothelial cells (HPMC) was cultured. Cultured HPMC were treated with F12 without serum (control group), F12 with TGF-beta 1 (TGF-beta 1 group), F12 with TGF-beta 1 and huangqi 100 micrograms/ml (huangqi 1 group), F12 with TGF-beta 1 and huangqi 1 mg/ml (huangqi 2 group). Fibronectin(FN) and plasminogen activator inhibitor-1 (PAI-1) were detected by ELISA method. The expression of FN mRNA, PAI-1 mRNA and bFGF mRNA was detected by RT-PCR method. RESULTS: 1. HPMC expressed FN, PAI-1, and bFGF. 2. Fn and PAI-1 significantly increased compared with control with 5 ng/ml TGF-beta 1 stimulated at 24 hours and 48 hours(P < 0.05); 3. The different concentration of huangqi decreased FN and PAI-1 expression compared with TGF-beta 1 group, especially 1 mg/ml huangqi at 24 hours(P < 0.05). 4. FN, PAI-1 and bFGF mRNA expression were higher in 12 hours after stimulation with TGF-beta 1 compared with control. FN, PAI-1 and bFGF mRNA expression were lower in 12 hours after stimulation with different concentration of huangqi (100 micrograms/ml and 1 mg/ml) than those in the TGF-beta 1 group. CONCLUSION: TGF-beta 1 promotes extracellular matrix synthesis and secretion of HPMC. Huangqi partly counteracts the synthesis of human peritoneal mesothelial cell's extracellular matrix by the stimulation of TGF-beta 1.

[Determination of trace sulfur and mercury in the Chinese traditional medicine].

This paper reports that the sulfur compounds of traditional Chinese medicine were reduced to S2- by Auger reagent, and determined by cold atomic fluorescence. At the same time the trace mercury was determined. The conditions the for quantitative determination of Hg2+ and S2- were obtained. The linear range of the determination is 11.20-55.60 mg.mL-1, the correlation coefficient is 0.999 78. In this method, little reagent is needed, the treatment of the sample is simple, the process of determination is convenient, and the sensitivity is high.

[Effect of Milkvetch Injection on cardiac function and hemodynamics in children with tetralogy of Fallot after radical operation].

OBJECTIVE: To evaluate the effect of Milkvetch Injection (MI) on cardiac function and hemodynamics in children with tetralogy of Fallot after radical operation. METHODS: Thirty-two children with tetralogy of Fallot were divided into two groups, the 20 patients in the control group treated with conventional treatment and the 12 patients in the treated group treated with 15 ml of MI every 12 hrs for 7 days. The cardiac function and hemodynamic parameters were determined by echocardiography and thermodilution catheter inserted in pulmonary artery. RESULTS: Patients' cardiac function and hemodynamic parameters were improved significantly 1 hr after administering 15 ml of MI Injection, but restored to the level before medication in 2-4 hrs. Administering MI injection for successive 7 days could gradually improve patients' cardiac function and hemodynamic parameters after operation, and the improvement became more evident with the prolonging time of medication. The intubation time, intensive care unit duration and the time of intravenously administration of inotropic drugs in the treated group were significantly shorter than those in the control group (P < 0.01). CONCLUSION: MI could significantly improve the hemodynamics and cardiac function, and shorten the recovery time in children with tetralogy of Fallot after radical operation.

Antidiabetic drugs of plant origin used in China: compositions, pharmacology, and hypoglycemic mechanisms.

The paper reviewed compositions and pharmacological effects of eight antidiabetic herbal drugs that have been approved by health regulatory agency for commercial use in China. Investigators attributed the hypoglycemic effect of these products to their ability to restore the functions of pancreatic tissues and cause an increase in insulin output, to inhibit the intestinal absorption of glucose, or to the facilitation of metabolites in insulin-dependent processes. Treatment with herbal drugs has an effect on protecting beta cells and smoothing out fluctuations in glucose levels. The use of these naturally derived agents in conjunction with conventional drug treatments such as an chemical agent or insulin permits the use of lower doses of the drug and/or decreased frequency of administration which decreases the side effects most commonly observed.

[Therapeutic effect of "anti-hepatic-fibrosis 268" on hepatic fibrosis in rats].

OBJECTIVE: To investigate the effects of the proprietary Chinese Medicine "anti-hepatic-fibrosis 268" on hepatic fibrosis and the related mechanisms. METHODS: The model of CCl4-induced hepatic damage was established in SD rats. 54 male rats were divided into four groups, namely high dose and low dose "anti-hepatic-fibrosis 268" groups, colchicine control group, and model control group. Using Masson stain and light microscope, the authors examined the rats' hepatic tissues and counted the hepatic fiber components, then examined and counted TGF-beta1, alpha-SMA, FN, Type I, III collagen by means of immunohistochemical technique. The groups were compared and the internal relationships of the data were analyzed. RESULTS: The levels of FN, LN, Type I and III collagen, TGF-beta1, and alpha-SMA of the CCl4 damaged rats increased (P<0.01). After 3 weeks of high dose "anti-hepatic-fibrosis 268" treatment, the levels of TGF-beta1, alpha-SMA, FN, LN, Type I and III collagen decreased (P<0.01) and the degree hepatic fibrosis took a favorable turn significantly (P<0.05) as compared with the model control. In the rats of the low dose group, the levels of TGF-beta1, alpha-SMA, FN, Type III collagen significantly decreased (P<0.05), the levels of LN, Type I collagen were not different from the model control; The hepatic fibrosis improved to a certain extent (P<0.05). CONCLUSION: The mechanism of reversing hepatic fibrosis by "anti-hepatic-fibrosis 268" in this experiment is that the medicine regulates TGF-beta1 and further affects alpha-SMA, thus resulting in the decline of FN, Type I, III collagen levels in liver extracellular matrix.

[Effect of milkvetch injection on immune function of children with tetralogy of Fallot after radical operation].

OBJECTIVE: To evaluate the effect of milkvetch injection (MI) on immune function of children with tetralogy of Fallot (TOF) after radical operation. METHODS: Forty-children with TOF were divided into two groups, the 20 in the control group treated with conventional treatment alone and the 20 in the treated group treated with conventional treatment plus 15 ml of MI every 12 hrs for 14 days. Changes of immunoglobulin, complements, lymphocyte phenotypes and cytokines were observed. RESULTS: In the treated group, the abnormally increased levels of IgG, IgM, C3, C4, CD8+ and CD19+ began to lower at lst-2nd week after treatment, and basically restored to the levels of normal at 3rd-4th week; while the decreased levels of IgA, CD3+, CD4+, CD4+/CD8+ ratio, CD3+/HLA-DR+ and CD3+/CD16+ -CD56+ raised gradually from the 1st week and restored to normal range at 2nd-3rd week. The IL-6 and tumor necrosis factor-alpha (TNF-alpha) levels in the plasma and supernatant, produced in vitro by peripheral blood mononuclear cells (PBMC) decreased gradually at 1st week and restored to the normal level at 3rd-4th weeks. The different value before and after treatment of the above-mentioned indexes in the treated group were superior to those in the control group (P<0.05 or P<0.01). CONCLUSION: MI could significantly improve the immune function of children with TOF after radical operation.
