An epidemic of urinary retention caused by dimethylaminopropionitrile.

An epidemic of urinary retention among workers in a polyurethane manufacturing plant was discovered in the spring of 1978. The most severely affected workers had neurogenic bladders confirmed by cystometrograms and mild sensory peripheral neuropathy. A survey of the plant disclosed increased incidence of urinary retention, muscle weakness, paresthesia, insomnia, and sexual dysfunction in exposed workers. A catalyst containing dimethylaminopropionitrile was identified as the probable causative agent, and after its removal no new cases occurred.

[Lathyrism and collagens I and III (author's transl)].

The administration of lathyrogenic agents (BAPN) at weak and prolonged doses (9 weeks, 1 g/kg/day) induced in the rat simultaneous alterations of aortic and cutaneous connective tissues comparable to the modifications observed during ageing in man. The lathyrogens produced this effect of inhibiting lysine-oxydase, the enzyme responsible for cross-linking collagen and elastin which leads to an increase in extractable collagen in the tissues. But BAPN did not appear to affect synthesis of collagen either in amount or in types I and III collagen ratio.

Tissue interactions in the organization and maintenance of the muscle pattern in the chick limb.

Recent investigations on a hereditary muscular dysgenesis (cn/cn) in the chicken (Kieny, Mauger, Hedayat & Goetinck, 1983) have suggested that limb muscle pattern development and subsequent maintenance are two independent steps in the formation of the musculature. The respective activities or muscle cells and connective tissue cells in the ontogeny of the musculature have been investigated in avian embryos 1) by in ovo administration of drugs interfering with collagen biosynthesis, and 2) by heterogenetic somite-exchange experiments between normal and mutant embryos. None of the drugs administered to the chick embryo caused any disturbance of muscle pattern formation or maintenance whether treatment occurred before (5 days) or after (7.5 days) the muscle splitting period. Heterogenetic implantations were performed at 2 days of incubation either at the leg or at the wing level. Somitic mesoderm from non-mutant quail embryo was grafted to replace a piece of somitic mesoderm in putative mutant (cn/cn) chick embryos. The introduction of normal myogenic cells into a mutant leg or wing led to a normally patterned musculature, which demonstrates that the muscular dysgenesis cn/cn results from a defect of the somitic myogenic cell line.

Therapeutic implications of collagen metabolism in myelofibrosis.

Myelofibrosis is an idiopathic condition of man associated with increased deposition of bone marrow collagen. This is directly seen on bone marrow reticulin or collagen staining, and reflected by increased levels of serum procollagen III aminoterminal peptide (28). It is but one of a number of diseases in which collagen, and an aberration of the connective tissue matrix, influence normal anatomy and physiology. Animal experiments have shown that the therapeutic manipulation of the intra- and extracellular processing of collagen with various agents can influence its deposition and the degree of end-organ damage. Several of these drugs have been used therapeutically for various clinical conditions and deserve clinical trials to evaluate their effectiveness in myelofibrosis. Bone marrow fibrosis, and peripheral serum markers which reflect collagen metabolism could be followed as experimental parameters. Development of an effective and safe antifibroblastic therapy is awaited eagerly. An attempt to prevent the fibrosis found in myelofibrosis, as opposed to managing its complications, would revolutinize our approach to managing these patients.

Influence of beta-amino-propionitrile on elastogenesis in the chick embryo (light and electron microscopic study).

Beta-amino-propionitrile was administered in a single dose into the chorioallantoic cavity of 14 days old chick embryos. The effects on the elastic fibre production were examined on the 3rd or 7th day following the injection. By light microscopy dissecting aneurysm, aortic ruptures were found. The elastic fibres were poorly developed and exhibited irregularities and structural anomalies. Electron microscopically we observed reduction of the number or complete absence of elastic aggregates and fibres. The elastic structures exhibited abnormal phosphotungstic acid binding. This was considered as a sign for disturbances in the development of cross linkages and in the incorporation of elastin into the elastic fibres.

Effect of parathyroid extract and of glutaurine (gamma-L-glutamyl-taurine) on the manifestations of osteolathyrism.

BAPN-induced osteolathyrism in young and older male rats was examined for its responses to parathormone-free, gel-filtered bovine parathyroid extract (extract P) and to glutaurine (gamma-L-glutamyl-taurine), the latter being a hitherto unknown putative hormone isolated from bovine parathyroid and reproduced by synthesis. In case of the daily administration of 20 mg BAPN + 0.2 mg extract P/100 g body weight and of 40 mg BAPN + 0.2 micrograms glutaurine/100 g body weight, both substances were found to attenuate the manifestations of osteolathyrism, to a significant extent, without, however preventing their development completely. The significantly decreased Ca, P, chondroitine sulphate and chrondroprotein values due to the administration of BAPN showed a significant, or highly significant, increase for the cartilage in response to extract P, and for both cartilage and bone in response to glutaurine, without however, attaining the control values. Additional evidence has been furnished by the present study that synthetic glutaurine is equal to the parathormone-free, gel-filtered parathyroid extract (extract P) in its attenuating effect on experimental osteolathyrism.

The effect of beta-aminoproprionitrile on the lingual artery and aorta of the rat: a light microscopic study.

The effects of chronic toxicity of beta-aminoproprionitrile (BAPN) on the lingual artery and aorta of the rat were compared. Thirty-two three week old weanling male white Wistar rats were given 1 g/kg/day (for five days weekly) of a 40% saturated solution of BAPN fumarate by intragastric tube. The animals were divided into four groups of eight animals each and sacrificed after one, three, six and nine weeks respectively. For each group, four controls were used. Cross-sections of the lingual artery and aorta stained with hematoxylin-erythrosin-safranin, alcian blue, Masson's trichrome and Verhoeff's iron hematoxylin were studied. BAPN had no effect on the lingual artery. Early changes were seen in the aorta at six weeks. Characteristic lesions were found at nine weeks involving the elastic tissue, myocytes and ground substance. The elastic lamellae were thinner, lost their normal orientation, and became swollen and finally fragmented. The myocytes of the media changed orientation and migrated into the intima. The proteoglycans increased and fibrosis supervened at the end stage. Failure to induce lesions in the lingual artery may be explained by early structural maturation and a low metabolic turnover rate in muscular arteries, decreased exposure to or greater protection from mechanical stress.

The histology and collagen content of cotton pellet and polyvinyl sponge-induced granulomas in normal and drug-treated rats.

Comparison of cotton wool and sponge-induced granulomas in the rat showed that collagen contents were very similar despite the different pattern of granuloma formation. There was significantly less variation of individual results in the sponge. Although collagen accounted for less than 10% of the dry weight, there was a positive correlation between the 2 parameters in both models of 9 days but only in the pellet at 15 days. Hydrocortisone treatment significantly decreased dry weight and collagen content. There was rapid cell proliferation and collagen synthesis in the sponge granuloma from Day 5, reaching a peak between the ninth and twelfth day, when DNA levels dropped and collagen increased more slowly to plateau by Day 15. Day 9 was chosen as the most suitable stage to study drug effects. Dexamethasone significantly decreased collagen synthesis whereas indomethacin and phenylbutazone did not. Soluble collagen levels were increased with beta-amino-propionitrile and d-penicillamine; a dose-related effect was obtained with d-penicillamine.

[Preventive treatment in caustic stenosis of the oesophagus (author's transl)].

The authors review the varioux forms of treatment used experimentally and clinically in an attempt to prevent oesophageal stenosis following ingestion of caustic substances. Corticosteroid therapy is still used by certain authors though its effectiveness is far from obvious. However, promising results have been obtained experimentally using substances such as amino-nitriles, which alter the properties of newly formed collagen.
